CaPDX1, a Novel Protein, Positively Regulates Cold Stress Tolerance via Interaction with CaSnRK2.4 in Pepper (Capsicum annuum L.).

Capsicum annuum is a Solanaceae crop that is sensitive to cold, which affects its growth and development upon prolonged exposure and ultimately reduces yield. In response, a complex regulatory network of cold-responsive genes is activated. Earlier studies have shown that SnRKs play a positive role in enhancing cold tolerance in different crops, including peppers; however, the underlying molecular mechanisms and downstream targets have yet to be fully elucidated. In this study, yeast hybrid screening using CaSnRK2.4 identified a potential interacting partner CaPDX1. The interaction between CaPD

Lipid Transfer Proteins (LTPs) Partially Purified from Capsicum chinense Jacq. Seeds: Antifungal Properties and α-amylase Inhibitory Activity.

In this study, we identified and partially purified antimicrobial peptides belonging to the family of lipid transfer proteins (LTPs) from Capsicum chinense seeds (UENF 1751 accession). Fractions rich in LTPs were obtained via ion exchange chromatography and subsequently purified via reverse-phase chromatography in an HPLC system. Therefore, two fractions were revealed: C1 (the nonretained fraction) and C2 (the retained fraction in ion-exchange chromatography). Fraction C1 was subjected to reverse-phase chromatography via a C18 column on an HPLC system, and ten fractions were obtained (P1-P10),

Unveiling the Biomedical Applications of Novel Coumarins Isolated From Capsicum Annuum L. Seeds by a Multivariate Extraction Technique.

For the first time, kinetic thermomagnetic extraction is a novel approach presented in this work. It required the application of four distinct variables: rotation speed (50, 75, and 100 rpm), magnetic field (0.8, 1.2, and 1.6 T), time interval (30, 60, and 90 min), and temperature (45, 55, and 65 °C). Numerous phytochemical categories were detected in the 81 crude chloroform extracts of green sweet bell pepper seeds that were collected, according to phytochemical analysis. Nine extracts were discovered to be linked to the coumarin chemical class and to have the same two extraction parameters:

Thyme essential oil fostering the efficacy of aqueous extract of licorice against fungal phytopathogens of Capsicum annuum L.

Capsicum annuum L. production is impeded by various biotic factors, including fungal diseases caused by Colletotrichum capsici, Pythium aphanidermatum, and Fusarium oxysporum. Various plant extracts and essential oils are increasingly used to control different plant diseases. In this study, licorice (Glycyrrhiza glabra) cold water extract (LAE) and thyme (Thymus vulgaris) essential oil (TO) were found to be highly effective against the C. annuum pathogens. LAE at 200 mg ml-1 demonstrated the maximum antifungal activity of 89.9% against P. aphanidermatum, whereas TO at 0.25 mg ml-1 showed 100%

CaWRKY58, encoding a group I WRKY transcription factor of Capsicum annuum, negatively regulates resistance to Ralstonia solanacearum infection.

WRKY transcription factors are encoded by large gene families across the plant kingdom. So far, their biological and molecular functions in nonmodel plants, including pepper (Capsicum annuum) and other Solanaceae, remain poorly understood. Here, we report on the functional characterization of a new group I WRKY protein from pepper, termed CaWRKY58. Our data indicate that CaWRKY58 can be localized to the nucleus and can activate the transcription of the reporter β-glucuronidase (GUS) gene driven by the 35S core promoter with two copies of the W-box in its proximal upstream region. Molecular interactions between capsicum protein and capsanthin: enhancement of complex formation and physicochemical stability by ultrasound and microwave treatments.

To investigate the mechanisms by which ultrasound (US), microwave (MW), and ultrasound-microwave combined (UM) treatments enhance the stability of capsanthin (Cap) in chili products, this study examined the effects of US, MW, and UM on Cap stability within the capsicum protein (CP)-Cap complex system. The characteristics and molecular structure of the complex (CCAP) were also analyzed. Results indicated that all treatments improved CP-Cap binding rate and CCAP stability, enhancing the resistance of Cap to light, heat, and acid-base stress, with MW showing the strongest effect. Identification of the pepper SAR8.2 gene as a molecular marker for pathogen infection, abiotic elicitors and environmental stresses in Capsicum annuum.

Pepper ( Capsicum annuum L.) SAR8.2 genes, designated CASAR82A, B and C, which are induced by all the biotic and abiotic stresses, were isolated from a pepper cDNA library constructed with the mRNAs from pepper plants infected with Xanthomonas campestris pv. vesicatoria. The pepper CASAR82A, B and C gene products, which are very similar to each other in amino acid sequences, have 43-50% homology with those of tobacco SAR8.2 genes. The CASAR8.2 genes were not constitutively expressed in any of the organs of healthy pepper plants. In contrast, the CASAR82A gene was locally or systemically induce

[Effect of EGTA and La2+ on induction of sesquiterpene cyclase gene expression in leaves of Capsicum annuum by several abiotic elicitors].

To destroy Ca2+ signal system, the detached leaves of Capsicum annuum were pretreated with EGTA, a commonly used Ca2+ chelator, or La3+, an ion which competitively inhibits Ca2+ movement through plasma membrane channels. The induction of sesquiterpene cyclase gene expression in the pretreated pepper leaves under the treatments of CuCl2, HgCl2 and UV was conducted in this paper. The result showed that no hindrance of sesquiterpene cyclase enzyme activity was detected in the EGTA or LaCl3 pretreated leaves, compared to the controls treated with CuCl2, HgCl2, and UV without the pretreatment, whil

Comparative effects of Capsicum annuum selenium nanoparticles and sodium selenite on growth, antioxidant activity, bioaccumulation, and DNA damage in quails.

Selenium nanoparticles (Se-NPs), a nanoscale form of selenium, and sodium selenite (SS), an inorganic selenium source, were included in the diet of quails to evaluate their effects on growth performance, selenium accumulation, antioxidant status, and DNA damage. A total of 420 quails (14-day-old) were distributed into 5 groups. Each group consisted of 84 birds, arranged into 3 replicates (28 quails per replicate). The first group served as the control and received a basal diet, while the second and third groups were supplemented with Se-NPs at 0.2 and 0.4 mg/kg, respectively. The fourth and fi

Cellulase applied to the leaves of sweet pepper (Capsicum annuum L. var. grossum) upregulates the production of salicylic and azelaic acids.

Treating the leaves of sweet pepper (Capsicum annuum L. var. grossum) with an aqueous solution of cellulase resulted in a four-fold increase in the salicylic acid level compared to a control plant. The level of endogenous azelaic acid was also elevated by the cellulase treatment. Azelaic acid has recently been reported to act as a mobile "priming" agent to arm plants against pathogenic attack. Our results are consistent with this and that the cellulase treatment enhanced the ability of sweet pepper to withstand viral attack.

CaSnRK2.4-interacting CaUDP-D (UDP-D-apiose/UDP-D-xylose synthase) positively regulates cold tolerance in Pepper (Capsicum annuum L.).

Pepper (Capsicum annuum L.) is highly sensitive to low-temperature stress, which inhibits plant growth, impairs photosynthesis, disrupts membrane integrity, promotes excessive reactive oxygen species (ROS) accumulation, and ultimately reduces productivity and yield. UDP-D-xylose synthase plays an important role in plant development and abiotic stress responses. However, its underlying mechanism and functional role in response to cold stress in pepper are not well explored. In this study, CaUDP-D was identified as a candidate interacting protein screened from CaSnRK2.4 and subsequently confirme

Overexpression of CaWRKY27, a subgroup IIe WRKY transcription factor of Capsicum annuum, positively regulates tobacco resistance to Ralstonia solanacearum infection.

WRKY proteins are encoded by a large gene family and are linked to many biological processes across a range of plant species. The functions and underlying mechanisms of WRKY proteins have been investigated primarily in model plants such as Arabidopsis and rice. The roles of these transcription factors in non-model plants, including pepper and other Solanaceae, are poorly understood. Here, we characterize the expression and function of a subgroup IIe WRKY protein from pepper (Capsicum annuum), denoted as CaWRKY27. The protein localized to nuclei and activated the transcription of a reporter GUS

Polyphenolic and Anthocyanin Responses to Postharvest Fungal Pathogen Infection in Purple and Green near Isogenic Pepper (Capsicum annuum) Lines.

Postharvest fungal pathogens, such as Botrytis cinerea, Alternaria alternata and Fusarium culmorum, pose major challenges for pepper (Capsicum annuum) storage and shelf-life. To explore the basis of induced resistance, near isogenic lines (NILs) differing in pigmentation (green vs. purple fruits and their red ripe counterparts) were artificially inoculated and evaluated for disease severity by phenotyping and by qPCR, and metabolite composition by spectroscopy and by HPLC. Infection severity was strongly dependent on whether purple or green NILs were infected and on ripening stage: economicall

Zinc priming enhances Capsicum annuum immunity against infection by Botrytis cinerea- From the whole plant to the molecular level.

Micronutrient manipulation can enhance crop resilience against pathogens, but the mechanisms are mostly unknown. We tested whether priming Capsicum annuum plants with zinc (5 μM Zn) or manganese (3 μM Mn) for six weeks increases their immunity against the generalist necrotroph Botrytis cinerea compared to deficient (0.1 μM Zn, 0.02 μM Mn) and control conditions (1 μM Zn, 0.6 μM Mn). Zinc priming reduced the pathogen biomass and lesion area and preserved CO2 assimilation and stomatal conductance. Zinc mobilization at the infection site, visualized by micro-X-ray fluorescence, was accompanied by

Structural and Biochemical Characterization of Three Antimicrobial Peptides from Capsicum annuum L. var. annuum Leaves for Anti-Candida Use.

The emergence of resistant microorganisms has reduced the effectiveness of currently available antimicrobials, necessitating the development of new strategies. Plant antimicrobial peptides (AMPs) are promising candidates for novel drug development. In this study, we aimed to isolate, characterize, and evaluate the antimicrobial activities of AMPs isolated from Capsicum annuum. The antifungal potential was tested against Candida species. Three AMPs from C. annuum leaves were isolated and characterized: a protease inhibitor, a defensin-like protein, and a lipid transporter protein, respectively
