Potent inhibition of human organic cation transporter 2 (hOCT2) by β-carboline alkaloids.

1. Beta-carbolines are indole alkaloids with a wide range of pharmacological and toxicological activities. Beta-carbolines are structurally related to the neurotoxin 1-methyl-4-phenylpyridinium (MPP+), a known substrate of organic cation transporters (OCTs). The goal of this study is to determine the interaction of β-carbolines with human OCT1, 2, and 3 (SLC22A1-3). 2. Dose-dependent inhibition studies were performed for five commercially available β-carbolines using a fluorescent substrate assay in HEK293 cells stably expressing hOCT1-3. The substrate potential was evaluated by uptake assays

A case of beta-carboline alkaloid intoxication following ingestion of Peganum harmala seed extract.

Beta-carboline alkaloids harmine, harmaline, and tetrahydroharmine can stimulate the central nervous system by inhibiting the metabolism of amine neurotransmitters, or by direct interaction with specific receptors; they are found in numerous plants, including Peganum harmala, Passiflora incarnata and Banisteriopsis caapi, and in the entheogen preparation Ayahuasca, which is traditionally brewed using B. caapi to enhance the activity of amine hallucinogenic drugs. The ingestion of plant preparations containing beta-carboline alkaloids may result in toxic effects, namely visual and auditory hall

Harman (1-methyl-beta-carboline) is a natural inhibitor of monoamine oxidase type A in rats.

Harman (1-methyl-beta-carboline) displaces [3H]pargyline in vitro from high affinity binding sites on membranes from cerebral cortex, provided that experimental conditions are chosen under which [3H]pargyline labels selectively monoamine oxidase type A. Norharman (beta-carboline) is a much weaker displacing compound. It is well known that the type A enzyme can be blocked irreversibly in vivo by treatment of rats with clorgyline. Under these conditions no specific binding of [3H]harman and [3H]pargyline to monoamine oxidase type A was detected in brain, whereas the specific binding was reduced

Sex differences in open-field behavior in response to the beta-carboline FG 7142 in rats.

Sex differences in animal models of anxiety and depression that employ external stimuli have been previously reported. This study examined the effect of gender on pharmacologically induced anxiety in rats coupled with novel handling, injection, and activity in an open-field test. The anxiogenic compound FG 7142 significantly decreased male open-field exploratory at 5, 10, and 20 mg/kg, while rearing behavior was decreased only at the 20 mg/kg dose. Female rats were more resistant to the effects of FG 7142 on open-field exploratory behavior, decreasing open-field activity, and rearing behavior

A beta-carboline derivative (ZK 93426) counteracts the cardiorespiratory depressant effects of intravenous midazolam.

The purpose of this study was to test the effects of the new beta-carboline ZK 93426 on midazolam-induced cardiorespiratory depression. Seven pentobarbital-anesthetized (35 mg/kg i.p.) cats were treated with intravenous midazolam (2 mg/kg) while monitoring the respiratory minute volume (VE), tidal volume, respiratory rate, blood pressure, heart rate and expired CO2. Midazolam caused significant decreases in VE (p less than 0.05) and blood pressure (p less than 0.05). ZK 93426 (5 mg/kg i.v.) antagonized these effects and produced significant increases in VE and blood pressure that resulted in t

Discriminative stimulus control by the anxiogenic beta-carboline FG 7142: generalization to a physiological stressor.

Drug discrimination was employed to investigate the similarities between FG 7142-induced anxiogenesis and the stress produced by exposure to either a novel environment or to footshock. Eight rats were trained to discriminate between the stimulus properties of the beta-carboline FG 7142 (5.0 mg/kg) and its vehicle in a two-lever, food motivated operant task. Once trained, decreasing doses of FG 7142 produced fewer FG 7142-appropriate responses and the dose-response relationship yielded an ED50 of 1.45 mg/kg. Rats were subsequently subjected to two physiological/environmental stressors, footshoc

Conjugates of γ-Carbolines and Phenothiazine as new selective inhibitors of butyrylcholinesterase and blockers of NMDA receptors for Alzheimer Disease.

Alzheimer disease is a multifactorial pathology and the development of new multitarget neuroprotective drugs is promising and attractive. We synthesized a group of original compounds, which combine in one molecule γ-carboline fragment of dimebon and phenothiazine core of methylene blue (MB) linked by 1-oxo- and 2-hydroxypropylene spacers. Inhibitory activity of the conjugates toward acetylcholinesterase (AChE), butyrylcholinesterase (BChE) and structurally close to them carboxylesterase (CaE), as well their binding to NMDA-receptors were evaluated in vitro and in silico. These newly synthesize

Effects of diazepam and two beta-carbolines on epileptic activity and on EEG and behavior in rats with absence seizures.

In the present series of experiments, effects of a full benzodiazepine receptor agonist (diazepam) are described and compared with those of a partial benzodiazepine receptor agonist (ZK 91296) and an inverse partial benzodiazepine receptor agonist (FG 7142), both compounds of the beta-carboline family. In a rat model for generalized absence epilepsy, the anticonvulsant, the hypnotic and the myorelaxant properties were investigated, as well as effects on on-going behavior and effects on the electroencephalogram (EEG). While diazepam showed all behavioral and electrophysiological changes charact

β-Carboline-Based Fluorescent Probes Sense and Stabilize G-quadruplex DNA Structures.

DNA forms four-stranded G-quadruplex (G4) structures in guanine-rich regions of the genome, regulating various biological functions, including stalling replication and modulating gene expression at both transcription and translation levels. Thus, investigating the folding, dynamics, and subcellular localization of G4s using small-molecule probes is useful to understand their biological functions. In this study, we report a series of fluorescent probes based on β-carboline that emit upon binding to G4 DNA structures. The lead probe BCC exhibits up to an ∼18-fold fluorescence enhancement (40% qu

Host-controlled switching between TADF and RTP in acridone-carboline donor-acceptor dyads.

Purely organic, metal-free emitters that combine thermally activated delayed fluorescence (TADF) and room-temperature phosphorescence (RTP) provide an attractive route to triplet harvesting without heavy-metal complexes. Here, we report two acridone-carboline donor-acceptor emitters, Acr-Me-Cb and Acr-Anisyl-Cb, that exhibit bluish-green emission centered at 422 nm and high photoluminescence quantum yields of up to 85% in toluene. A moderate singlet-triplet energy gap (ΔEST ≈ 0.3 eV) enables competing reverse intersystem crossing and triplet stabilization pathways, allowing delayed fluorescenc

β-Carboline and chloroquine hybrids as potent antiplasmodial agents: Design, synthesis and biological evaluation.

A series of thirteen novel β-carboline and chloroquine (CQ) hybrids was designed and synthesized. The compounds were evaluated for in vitro antiplasmodial activity against Plasmodium falciparum strains 3D7 (CQ-sensitive) and Dd2 (multidrug-resistant). All compounds exhibited potent activity, with IC 50 values in the nanomolar to low micromolar range, while retaining potency against the resistant strain (1.0 to 365.5 nmol L-1 for 3D7 and from 2.2 to 2415.3 nmol L-1 for Dd2 strain). Several compounds were more active against Dd2, then against 3D7 strain with resistance index (RI) lower than 1. M

Investigation of β-Carboline Alkaloid Harmaline Against Cyvirus cyprinidallo3 Infection In Vitro and In Vivo.

Cyvirus cyprinidallo3, also known as Cyprinid herpesvirus 3 (CyHV-3), is a common pathogen of koi and common carp (Cyprinus carpio). Infection of CyHV-3 can lead to high mortality in fry under 4 months of age. CyHV-3 can become latent in recovered fish, and latent CyHV-3 can reactivate under stress conditions and spread the virus. Reactivation of CyHV-3 can also lead to mortality and diseases in latently infected fish. No effective drugs are available to prevent CyHV-3 infection or reactivation from latency. There is a need for the discovery of anti-CyHV-3 drugs. Harmine (HAR) and harmaline (H

Development of dual-function disulfaneyl oxadiazole bactericides by structurally incorporating β-carboline moieties.

Structural derivatization of known leads using the scaffold hopping strategy is a classic approach in pesticide discovery. However, the modificatory universality of lead skeletons profoundly influences the feasibility of this protocol. We herein report development of disulfaneyl oxadiazole bactericides involving β-carboline moieties. Derivative B18 is filtered out to exhibit excellent in vitro antibacterial properties with half-maximal effective concentrations of 0.47 mg/L, 1.72 mg/L, and 5.97 mg/L against Xanthomonas oryzae pv. oryzae, Xanthomonas oryzae pv. oryzicola, and Xanthomonas citri p

Conjugation of Aminoadamantane and γ-Carboline Pharmacophores Gives Rise to Unexpected Properties of Multifunctional Ligands.

A new series of conjugates of aminoadamantane and γ-carboline, which are basic scaffolds of the known neuroactive agents, memantine and dimebon (Latrepirdine) was synthesized and characterized. Conjugates act simultaneously on several biological structures and processes involved in the pathogenesis of Alzheimer's disease and some other neurodegenerative disorders. In particular, these compounds inhibit enzymes of the cholinesterase family, exhibiting higher inhibitory activity against butyrylcholinesterase (BChE), but having almost no effect on the activity of carboxylesterase (anti-target). Design, Synthesis, and Antifungal Activity of Novel Aryl-1,2,3-Triazole-β-Carboline Hybrids.

The copper catalytic azide and terminal alkyne cycloaddition reaction, namely "click chemistry", gives a new and convenient way to create l,4-disubstitutd-l,2,3-triazoles. In this work, 2-pyrrolecarbaldiminato⁻Cu(II) complexes were established as efficient catalysts for the three-component 1,3-dipolar cycloaddition reaction of arylboronic acid and sodium azide (NaN₃) with terminal alkynes in ethanol at room temperature to 50 °C, 1,4-disubstituted 1,2,3-triazoles were synthesized. Following the optimized protocol, two series of new aryl-1,2,3-triazole-β-carboline hybrids have been designed and

9-Methyl-beta-carboline has restorative effects in an animal model of Parkinson's disease.

In a previous study, a primary culture of midbrain cells was exposed to 9-methyl-beta-carboline for 48 h, which caused an increase in the number of tyrosine hydroxylase-positive cells. Quantitative RT-PCR revealed increased transcription of genes participating in the maturation of dopaminergic neurons. These in vitro findings prompted us to investigate the restorative actions of 9-methyl-beta-carboline in vivo. The compound was delivered for 14 days into the left cerebral ventricle of rats pretreated with the neurotoxin 1-methyl-4-phenyl-pyridinium ion (MPP+) for 28 days applying a dose which

Interaction of beta-carboline inverse agonists for the benzodiazepine site with another site on GABAA receptors.

1. We examined the effects of methyl 6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate (DMCM), a beta-carboline inverse agonist for the benzodiazepine site, on gamma-aminobutyric acid (GABA)-induced Cl-currents in several cloned rat GABAA receptor subtypes expressed in human embryonic kidney cells. The Cl- currents were measured in the whole cell configuration of patch clamp techniques. 2. DMCM at low concentrations (< 0.5 microM) occupying only the benzodiazepine site decreased GABA-induced Cl currents in the alpha 1 beta 2 gamma 2 and alpha 3 beta 2 gamma 2 subtypes as expected from an inve

Enhanced sensitivity to beta-carboline inverse agonists in rats chronically treated with FG 7142.

The biochemical and behavioural effects of the chronic administration of the beta-carboline inverse agonist FG 7142 were studied in the rat. Repeated administration of FG 7142 (15 mg/kg IP, twice daily for 10 consecutive days) induced sensitization to the effects of this drug, which from proconvulsant became a full convulsant. Thus, myoclonic seizures were observed in 30% and 80% of the animals by the third and the eighth day of treatment, respectively. The sensitization to the convulsant effect of FG 7142 persisted for up to 50 days after withdrawal and was completely prevented by the concurr

[Comparative study of the effect of 3-carboxy-beta-carboline methylamide and diazepam on rat behavior].

The effects of benzodiazepine receptor agonist, diazepam, and inverse agonist, FG 7142, were examined. Strong antagonism between FG 7142 (10 mg/kg) and diazepam (1 mg/kg) activity was revealed in the open field test. On the other hand, both FG 7142 and diazepam inhibited isolation-induced intraspecies aggressive behaviour of rats. FG 7142 also reduced interspecies aggression of mouse-killing rats. The findings suggest that both diazepam and FG 7142 have antiaggressive properties in the isolation-induced aggression model, which are mediated by benzodiazepine receptors of the central nervous sys
