β-Catenin-regulated ALDH1A1 is a target in ovarian cancer spheroids.

Cancer cells form three-dimensional (3D) multicellular aggregates (or spheroids) under non-adherent culture conditions. In ovarian cancer (OC), spheroids serve as a vehicle for cancer cell dissemination in the peritoneal cavity, protecting cells from environmental stress-induced anoikis. To identify new targetable molecules in OC spheroids, we investigated gene expression profiles and networks upregulated in 3D vs traditional monolayer culture conditions. We identified ALDH1A1, a cancer stem cell marker as being overexpressed in OC spheroids and directly connected to key elements of the β-cate

Wnt/β-catenin signalling: from plasma membrane to nucleus.

Wnt/β-catenin signalling plays essential roles in embryonic development as well as tissue homoeostasis in adults. Thus abnormal regulation of Wnt/β-catenin signalling is linked to a variety of human diseases, including cancer, osteoporosis and Alzheimer's disease. Owing to the importance of Wnt signalling in a wide range of biological fields, a better understanding of its precise mechanisms could provide fundamental insights for therapeutic applications. Although many studies have investigated the regulation of Wnt/β-catenin signalling, our knowledge remains insufficient due to the complexity

Identification of Wnt/β-catenin modulated genes in the developing retina.

PURPOSE: During mammalian eye development, the restriction of Wnt/β-catenin signaling at the junction of the neural retina and the retinal pigment epithelium in the peripheral eyecup is required for the development of the ciliary margin, a non-neural region of the eyecup that is the precursor of the ciliary body and iris of the adult eye. METHODS: To identify genes that are modulated by β-catenin activity in the embryonic retina, we performed gene expression profiling in Li(+)-treated retinal explants, a pharmacological model of β-catenin activation. The Li(+)-modulated gene data set was sear

HGF/c-Met related activation of β-catenin in hepatoblastoma.

BACKGROUND: Activation of beta-catenin is a hallmark of hepatoblastoma (HB) and appears to play a crucial role in its pathogenesis. While aberrant accumulation of the beta-catenin is a common event in HB, mutations or deletions in CTNNB1 (beta-catenin gene) do not always account for the high frequency of protein expression. In this study we have investigated alternative activation of beta-catenin by HGF/c-Met signaling in a large cohort of 98 HB patients enrolled in the SIOPEL-3 clinical trial. METHODS: We performed immunohistochemistry, using antibodies to total beta-catenin and tyrosine654-

beta-Catenin promotes respiratory progenitor identity in mouse foregut.

The mammalian respiratory system, consisting of both trachea and lung, initiates from the foregut endoderm. The molecular program that instructs endodermal cells to adopt the respiratory fate is not fully understood. Here we show that conditional inactivation of beta-Catenin (also termed Ctnnb1) in foregut endoderm leads to absence of both the trachea and lung due to a failure in maintaining the respiratory fate. In converse, conditional expression of an activated form of beta-Catenin leads to expansion of Nkx2.1, an early marker for the trachea and lung, into adjacent endoderm including the s

Wnt/beta-catenin signaling promotes renal interstitial fibrosis.

Wnts compose a family of signaling proteins that play an essential role in kidney development, but their expression in adult kidney is thought to be silenced. Here, we analyzed the expression and regulation of Wnts and their receptors and antagonists in normal and fibrotic kidneys after obstructive injury. In the normal mouse kidney, the vast majority of 19 different Wnts and 10 frizzled receptor genes was expressed at various levels. After unilateral ureteral obstruction, all members of the Wnt family except Wnt5b, Wnt8b, and Wnt9b were upregulated in the fibrotic kidney with distinct dynamic

[The specific features of the expression of E-cadherin and beta-catenin in adenomyosis].

The expression of E-cadherin and beta-catenin in the eutopic and ectopic endometrium was studied in 40 patients with adenomyosis and 12 without this condition, by using an immunohistochemical test and enzyme immunoassay. There was increased expression of E-cadherin and beta-catenin in the eutopic and ectopic endometrium in adenomyosis. The cytoplasmic expression of beta-catenin was also revealed in the smooth muscle cells surrounding the foci of adenomyosis. The E-cadherin concentration measured by enzyme immunoassay was significantly higher in the endometrium and myometrium of patients with a

Canonical WNT/beta-catenin signaling is required for ureteric branching.

WNT/beta-catenin signaling has an established role in nephron formation during kidney development. Yet, the role of beta-catenin during ureteric morphogenesis in vivo is undefined. We generated a murine genetic model of beta-catenin deficiency targeted to the ureteric bud cell lineage. Newborn mutant mice demonstrated bilateral renal aplasia or renal dysplasia. Analysis of the embryologic events leading to this phenotype revealed that abnormal ureteric branching at E12.5 precedes histologic abnormalities at E13.5. Microarray analysis of E12.5 kidney tissue identified decreased Emx2 and Lim1 ex

Beta-catenin activation and epithelial-mesenchymal transition in the pathogenesis of pterygium.

PURPOSE: To investigate whether beta-catenin activation and epithelial-mesenchymal transition (EMT) is involved in the pathogenesis of pterygium. METHODS: beta-Catenin and E-cadherin expression were examined in surgically excised tissue and eye bank corneas with intact pterygium. Snail and Slug, the transcriptional repressors of E-cadherin, and matrix metalloproteinase (MMP)-7, a down-stream gene regulated by beta-catenin were also investigated. Epithelial cells undergoing EMT-like changes were identified by double immunostaining for alpha-smooth muscle actin (SMA)/vimentin and cytokeratin 14

Role of connexin32 and beta-catenin in tumor promotion in mouse liver.

Tumor promoters are nonmutagenic chemicals that increase the probability of cancer by accelerating the clonal expansion of cells transformed during tumor initiation. The molecular mechanisms underlying this process are only partly understood but interference with signaling pathways regulating cell division and/or cell death is likely to be important. Ras- and beta-Catenin-dependent signaling is important for both of these processes and ras and beta-catenin genes are known mutational targets in mouse hepatocarcinogenesis. About 80% of liver tumors generated in mice by a promotional regimen incl

Evaluation of E-cadherin/catenin complex in primary and secondary glomerulonephritis.

Catenins (alpha-, beta-, gamma-catenin, p120(ctn)) are cytoplasmic proteins initially identified in a complex with E-cadherin (ECD). The latter belongs to a superfamily of transmembrane glycoproteins important for cell adhesion in normal and disease states. Catenins and p120(ctn), in particular, are substrates for growth factor receptor tyrosine kinases. Cell adhesive mechanisms have an impact on cell migration and proliferation and thus are potentially involved in the pathogenesis of glomerulonephritides (GNs). Using appropriate monoclonal antibodies, we investigated the immunohistochemical e

Nuclear localization of beta-catenin is an important prognostic factor in hepatoblastoma.

In this study, mutational and immunohistochemical analyses of beta-catenin were performed in 30 hepatoblastomas, to assess the prevalence of alterations of the Wnt pathway with respect to clinicopathological parameters and survival. Four missense mutations of beta-catenin (13.3%) were detected and there was strong immunoreactivity for beta-catenin in the cytoplasm and/or the nucleus in 97% of hepatoblastomas. Nuclear and cytoplasmic staining was demonstrated in 19 of 30 tumours (63%), while ten revealed only cytoplasmic staining. Statistically, this nuclear beta-catenin staining was significan

Presenilin affects arm/beta-catenin localization and function in Drosophila.

Presenilin is an essential gene for development that when disrupted leads to a neurogenic phenotype that closely resembles Notch loss of function in Drosophila. In humans, many naturally occurring mutations in Presenilin 1 or 2 cause early onset Alzheimer's disease. Both loss of expression and overexpression of Presenilin suggested a role for this protein in the localization of Armadillo/beta-catenin. In blastoderm stage Presenilin mutants, Arm is aberrantly distributed, often in Ubiquitin-immunoreactive cytoplasmic inclusions predominantly located basally in the cell. These inclusions were no

Frequent association of beta-catenin and WT1 mutations in Wilms tumors.

The etiology of Wilms tumor, an embryonic kidney tumor, is genetically heterogeneous. One Wilms tumor gene, WT1, which encodes a zinc finger transcription factor, is mutated in 10-20% of Wilms tumors, but it is still not clear what critical cellular pathway(s) is affected by these mutations. Recently beta-catenin mutations have been reported in 6 of 40 (15%) of Wilms tumors. Beta-catenin is the central effector in the Wnt signal transduction pathway, and deregulation of beta-catenin signaling is critical in the development of a number of malignancies. The observation of beta-catenin mutations

The role of beta-catenin stability in mutant PS1-associated apoptosis.

Most early onset cases of familial Alzheimer's disease (FAD) are caused by mutations in presenilin-1 (PS1) and presenilin-2 (PS2). These mutations lead to increased beta-amyloid formation and induce apoptosis when expressed in vitro. Recently, PS1 has been reported to associate with beta-catenin, an armadillo repeat protein. PS1 may regulate the function of beta-catenin, and mutant PS1 may disrupt this regulation. In the present study, we confirm that PS1-WT, as well as mutant PS1, associates with beta-catenin, and that mutant PS1 expression decreases the stability and/or enhances the degradat

WNT/β-catenin signaling: implications for homeostasis and tumorigenesis in the esophagus.

The WNT/β-catenin signaling pathway plays a central role in the development, homeostasis, and tumorigenesis of the gastrointestinal tract. During embryonic development, dynamic regulation of β-catenin activity is essential for foregut formation and for the transition of the esophageal epithelium from columnar to stratified squamous morphology, with inhibitory signals ensuring proper lineage specification. In the adult esophagus, however, the precise role of WNT/β-catenin signaling in basal progenitor cell proliferation and differentiation remains incompletely understood, although dysregulation

Novel 3D models for β-catenin-driven liver cancers.

BACKGROUND & AIMS: Constitutive activation of β-catenin is a key feature in the pathogenesis of HCC and hepatoblastoma (HB). Activating alterations in the CTNNB1 gene and, to a lesser extent, inactivating alterations in the APC gene are observed in 30-40% of HCC cases and 80-90% of HB cases. For both tumours, therapeutic management remains suboptimal. Therefore, relevant experimental models are needed to improve our understanding and to test new therapeutic approaches. METHODS: Liver organoids (n=17 mice) and tumouroids (n=14 mice) were established from APCΔhep and βcatΔex3 mouse models, whic

β-catenin expression as a strong predictor of recurrence in juvenile angiofibroma.

BACKGROUND: Juvenile angiofibroma (JA) is a rare fibrovascular tumor affecting young males. Although histologically benign, JA is clinically aggressive, with high morbidity, mortality, and recurrence rates. While β-catenin expression is known to contribute to JA growth, its role in recurrence remains uncertain. OBJECTIVE: This study investigates the relationship between β-catenin expression and recurrence risk in JA patients. METHOD: An analytical observational case-control study was conducted at Dr. Cipto Mangunkusumo Hospital, analyzing 33 JA patients who underwent tumor excision (2013-202
