Microglial cathepsin B as a key driver of inflammatory brain diseases and brain aging.

Interleukin-1β is a potent proinflammatory cytokine that plays a key role in the pathogenesis of the brain aging and diverse range of neurological diseases including Alzheimer's disease, Parkinson's disease, stroke and persistent pain. Activated microglia are the main cellular source of interleukin-1β in the brain. Cathepsin B is associated with the production and secretion of interleukin-1β through pyrin domain-containing protein 3 inflammasome-independent processing of procaspase-3 in the phagolysosomes. The leakage of cathepsin B from the endosomal-lysosomal system during aging is associate

Cathepsin B as a potential cystatin M/E target in the mouse hair follicle.

Deficiency of the cysteine protease inhibitor cystatin M/E (Cst6) in mice leads to disturbed epidermal cornification, impaired barrier function, and neonatal lethality. We report the rescue of the lethal skin phenotype of ichq (Cst6-deficient; Cst6-/-) mice by transgenic, epidermis-specific, reexpression of Cst6 under control of the human involucrin (INV) promoter. Rescued Tg(INV-Cst6)Cst6ichq/ichq mice survive the neonatal phase, but display severe eye pathology and alopecia after 4 mo. We observed keratitis and squamous metaplasia of the corneal epithelium, comparable to Cst6-/-Ctsl+/- mice,

Cathepsin B Expression and the Correlation with Clinical Aspects of Oral Squamous Cell Carcinoma.

BACKGROUND: Cathepsin B (CTSB), a member of the cathepsin family, is a cysteine protease that is widely distributed in the lysosomes of cells in various tissues. It is overexpressed in several human cancers and may be related to tumorigenesis. The main purpose of this study was to analyze CTSB expression in oral squamous cell carcinoma (OSCC) and its correlation with patient prognosis. METHODOLOGY/PRINCIPAL FINDINGS: Tissue microarrays were used to detect CTSB expression in 280 patients and to examine the association between CTSB expression and clinicopathological parameters. In addition, the

Efficient inhibition of cathepsin B by a secreted type 1 cystatin of Fasciola gigantica.

Cysteine proteases are important antigens in the liver fluke genus Fasciola, essential for infection, protection and nutrition. While their biochemistry, biological roles and application as vaccines have been thoroughly studied there is a lack of data concerning their regulation. In the present study we have continued our investigation of cysteine protease inhibitors in Fasciola gigantica and demonstrate, in comparison with FgStefin-1 and human cystatin C, that a second type 1 cystatin of the parasite, FgStefin-2, has been evolutionary adapted to block cathepsin B. The protein, which unusually

Expression of proteolytic cathepsins B, D, and L in periodontal gingival fibroblasts and tissues.

BACKGROUND: A major feature of gingivitis and periodontitis is the destruction of the collagenous matrix of the surrounding connective tissue. The hypothesis that proteolytic enzymes release from cells adjacent to the site of destruction was recently supported by the presence of increased levels of cathepsin B, D, and L in the gingival crevicular fluid of patients suffering from periodontal disease (PD). EXPERIMENTAL DESIGN: We studied the expression of mRNA of cathepsins B,D, and L in early passaged, adherent gingival fibroblasts of patients with chronic adult PD. In addition, we examined th

Immunocytochemical localization of cathepsins B, H, and L in the rat gastro-duodenal mucosa.

Cathepsins B, H, and L are representative cysteine proteinases in lysosomes of a large variety of cells. Previous immunochemical studies indicated the presence of these enzymes also in the gastrointestinal wall. Using specific antisera, the cellular and subcellular distribution of cathepsins B, H, and L in rat gastric (oxyntic and pyloric part) and duodenal mucosa was investigated by light and electron microscopical immunocytochemistry. The subtypes of cathepsins were distributed differently in the cellular constituents of the epithelia: Cathepsin B was localized to lysosomes of all cells exce

Cathepsin B and cystatin B in HIV-seropositive women are associated with infection and HIV-1-associated neurocognitive disorders.

OBJECTIVE: HIV-1-associated neurocognitive disorders (HAND) is triggered by immune activation of brain cells and remain prevalent during progressive viral infection despite antiretroviral therapy. Cathepsins and cystatins are lysosomal proteins secreted by macrophages and microglia, and may play important roles in neuroregulatory responses. Our laboratory has shown increased secretion and neurotoxicity of cathepsin B from in-vitro HIV-infected monocyte-derived macrophages, and increased expression in postmortem brain tissue with HIV encephalitis and HAND. We hypothesized that cystatin B and ca

Cathepsin B expression during in vitro maturation of buffalo COCs: apoptosis and developmental competence.

PURPOSE: The current study aimed to investigate whether cathepsin B (cath B) inhibition in cumulus cells of morphologically graded buffalo cumulus-oocyte complexes (COCs) during in vitro maturation (IVM) could modulate apoptotic markers and enhance developmental potential. METHODS: Cumulus cells from grade A and B buffalo COCs were collected at 0, 12, and 24 h of IVM, with or without supplementation of the optimized cysteine protease inhibitor E-64. Cathepsin B and selected apoptotic regulators were quantified by qRT-PCR. Cathepsin B protein levels and apoptotic signals were assessed by immun

Targeting Cathepsin B-mediated pyroptosis in fibroblasts alleviate ultraviolet-induced skin inflammation and injury.

Exposure to ultraviolet (UV) radiation is known to cause acute skin damage and inflammation, yet the underlying molecular mechanisms and cellular damage involved in this process remain poorly understood. In this study, we provide compelling evidence that UV-induced fibroblast pyroptosis not only triggers a cascade of macrophage inflammatory responses but also could represents a important mechanism in the pathogenesis of acute sunburn. We further investigate the intricate interplay between pyroptotic fibroblasts (Pyr-Fbs) and macrophages, elucidating the resultant alterations in macrophage phen

Unveiling Cathepsin B inhibition with repurposed drugs for anticancer and anti-Alzheimer's drug discovery.

Alzheimer's disease (AD) is characterized by the aggregation of amyloid β (Aβ) peptides and the formation of plaques in the brain, primarily derived from the proteolytic degradation of amyloid precursor protein (APP). Cathepsin B (CatB) is a cysteine protease that plays a pivotal role in this process, making it a potential target for the development of anti-Alzheimer's therapies. Apart from AD, CatB is implicated in various physiological and pathological processes, including cancer. Given the critical role of CatB in these diseases, identifying effective inhibitors is of significant therapeuti

Cathepsin B Relocalization in Late Membrane Disrupted Neurons Following Diffuse Brain Injury in Rats.

Traumatic brain injury (TBI) has consequences that last for years following injury. While TBI can precipitate a variety of diffuse pathologies, the mechanisms involved in injury-induced neuronal membrane disruption remain elusive. The lysosomal cysteine protease, Cathepsin B (Cath B), and specifically its redistribution into the cytosol has been implicated in cell death. Little is known about Cath B or neuronal membrane disruption chronically following diffuse TBI. Therefore, the current study evaluated Cath B and diffuse neuronal membrane disruption over a more chronic post-injury window (6 h

Toxoplasma Cathepsin Protease B and Aspartyl Protease 1 Are Dispensable for Endolysosomal Protein Digestion.

The lysosome-like vacuolar compartment (VAC) is a major site of proteolysis in the intracellular parasite Toxoplasma gondii Previous studies have shown that genetic ablation of a VAC-residing cysteine protease, cathepsin protease L (CPL), resulted in the accumulation of undigested protein in the VAC and loss of parasite viability during the chronic stage of infection. However, since the maturation of another VAC localizing protease, cathepsin protease B (CPB), is dependent on CPL, it remained unknown whether these defects result directly from ablation of CPL or indirectly from a lack of CPB ma

Immunocapture of circulating Schistosoma mansoni cathepsin B antigen (Sm31) by anti-Sm31 polyclonal antibodies.

Adult Schistosoma mansoni parasites have the capacity to degrade ingested host hemoglobin and other host plasma proteins by using a series of gut proteolytic enzymes, including cathepsin B; this enzyme is released to the host intravascular environment during regurgitations of adult worms. Cathepsin B becomes thus a circulating parasite component that has been shown to be specifically recognized as the Sm31 antigen by antibodies present in most S. mansoni infected patients. Taking advantage of this immunological property, we attempted here to immunocapture Sm31 from sera of infected patients us

Compartment-specific inhibition of cytosolic Cathepsin B mitigates microglial senescence in the aging brain.

Microglial senescence is a hallmark of brain aging, but how lysosomal dysfunction fuels their pro-aging activity remains unresolved. Here we identify cytosolic Cathepsin B (CatB) as a pivotal driver of microglial senescence. In aged mice and senescence-induced models, lysosomal membrane permeabilization releases CatB into the cytosol, where it remains enzymatically active at neutral pH. Cytosolic CatB promotes senescence, and its inhibition mitigates this process, whereas cytosolic delivery of recombinant CatB accelerates it. Mechanistically, cytosolic CatB binds and degrades small nuclear rib

Cathepsin B‑Responsive Phosphatidylinositol 3‑Kinase (PI3K) Prodrug for the Treatment of Pulmonary Fibrosis.

PI3K inhibitors are effective therapeutic agents for pulmonary fibrosis, but they are plagued by systemic toxicity and a narrow therapeutic window. To overcome this limitation, we developed a novel enzyme-responsive prodrug delivery system designed for targeted lung delivery. This system is based on the PI3K inhibitor P001 conjugated via a cathepsin B-cleavable linker (Val-Ala-PAB-MAC) to create the albumin-binding prodrug, PI3K-001. Leveraging albumin's natural lung-targeting properties, PI3K-001 achieves site-specific drug delivery and controlled release. In vivo studies demonstrated that th

Cathepsin B-Oriented Screening, Isolation, and Antitumor Validation of Bioactive Metabolites from Sargassum polycystum.

Marine medicinal algae represent a valuable reservoir of bioactive metabolites for anticancer drug discovery, yet the efficient identification of target-relevant compounds from chemically complex marine matrices remains challenging. In this study, an integrated cathepsin B-oriented strategy was developed to discover, prioritize, isolate, and validate antitumor metabolites from the brown alga Sargassum polycystum. Affinity ultrafiltration LC-MS was first applied to screen CTSB-binding constituents from the crude extract, followed by molecular docking, molecular dynamics simulation, and gray rel

Phytochemicals from Musa acuminata Colla peel exhibit cathepsin B inhibition and anti-inflammatory activity.

Rheumatoid arthritis (RA) is a chronic inflammatory illness in which inflammatory mediators and cathepsin B and other proteases play a role in disease progression. The existing treatments may have negative side effects. Therefore, an alternative natural therapy for RA has been considered safe and beneficial. Secondary metabolites from plants have been suggested as one such alternative. In our present study phytochemicals from Musa acuminata (banana) peel have been explored for their anti-cathepsin B and anti-inflammatory potential. The crude phytochemical extract inhibited cathepsin B with an

Salmonella Typhimurium expressing chromosomally integrated Schistosoma mansoni Cathepsin B protects against schistosomiasis in mice.

Schistosomiasis threatens hundreds of millions of people worldwide. The larval stage of Schistosoma mansoni migrates through the lung and adult worms reside adjacent to the colonic mucosa. Several candidate vaccines are in preclinical development, but none is designed to elicit both systemic and mucosal responses. We have repurposed an attenuated Salmonella enterica Typhimurium strain (YS1646) to express Cathepsin B (CatB), a digestive enzyme important for the juvenile and adult stages of the S. mansoni life cycle. Previous studies have demonstrated the prophylactic and therapeutic efficacy of
