Chemical Ecology of the North American Newt Genera Taricha and Notophthalmus.

The North American newt genera Taricha and Notophthalmus (order Caudata) are well known for the combination of potent toxicity, aposematic coloration, and striking defense postures that protects these animals from predation. This suite of traits is centered around the neurotoxin tetrodotoxin, which causes paralysis and death in metazoans by disrupting the initiation and propagation of electrical signals in the nerves and muscles. Tetrodotoxin defends newts from predation across multiple life history stages and its role in generating arms-race coevolution between Taricha newts and garter snake

Modification of the radioreceptor assay technique for estimation of serum neuroleptic drug levels leads to improved precision and sensitivity.

This report describes a series of modifications in the radioreceptor assay procedure for monitoring of serum neuroleptic levels which resulted in a substantial increase in precision and in a higher sensitivity than is the case with existing assays. The use of a dopamine receptor preparation from pig striatum yielded a lower limit of sensitivity for haloperidol of 0.08 ng/ml, when 0.5 ml serum was used in the assay; the intra- and inter-assay coefficients of variation were 3 and 9%, respectively. The relative antipsychotic potency of various neuroleptics correlated with their optimal daily dosa

Alpine salamanders at risk? The current status of an emerging fungal pathogen.

Amphibians globally suffer from emerging infectious diseases like chytridiomycosis caused by the continuously spreading chytrid fungi. One is Batrachochytrium salamandrivorans (Bsal) and its disease ‒ the 'salamander plague' ‒ which is lethal to several caudate taxa. Recently introduced into Western Europe, long distance dispersal of Bsal, likely through human mediation, has been reported. Herein we study if Alpine salamanders (Salamandra atra and S. lanzai) are yet affected by the salamander plague in the wild. Members of the genus Salamandra are highly susceptible to Bsal leading to the leth

[Metamorphosis in the Urodelan amphibians: patterns, regulation mechanisms, and evolution].

Patterns of metamorphosis and mechanisms of its regulation in primitive and advanced salamanders are compared. It is found that urodelan evolution was characterised by the following trends: 1) increase in the number of metamorphosing systems; 2) increase in the amplitude of metamorphic transformations of each particular system due to the progressive divergence of the larval and the adult morphology; 3) synchronization of metamorphic transformations and their concentration within a relatively short period of ontogeny; 4) increase in the role of the thyroid hormones (TH) in the regulation of met

Motion sickness in amphibians.

We explored the question of whether amphibians get motion sickness by exposing anurans (frogs) and urodeles (salamanders) to the provocative stimulus of parabolic aircraft flight. Animals were fed before flight, and the presence of vomitus in their containers after flight was used to indicate motion-induced emesis. None of the species that we studied vomited during the 8 to 10 parabolas of each flight. However, at least one specimen from each of the anuran species Rana rugosa, Rana nigromaculata, Hyla japonica, and Rhacophorus schlegelii vomited in a period of 0.5 to 42 h after flight. Some sp

Selective antileishmania activity of 13,28-epoxy-oleanane and related triterpene saponins from the plant families Myrsinaceae, Primulaceae, Aceraceae and Icacinaceae.

Maesa saponins with the 13,28-epoxy-oleanane triterpene core skeleton were described recently to possess strong and selective in vitro and in vivo antileishmania activity. In the absence of direct chemical derivatization possibilities, a structure-based literature search was carried out to explore a structure-activity relationship. Crude alcohol extracts from several plant species of Myrsinaceae, Primulaceae, Aceraceae and Icacinaceae were evaluated for in vitro activity against Leishmania infantum intracellular amastigotes and cytotoxicity on MRC-5(SV2) cells, while the saponin content was ev

Triassic stem caecilian supports dissorophoid origin of living amphibians.

Living amphibians (Lissamphibia) include frogs and salamanders (Batrachia) and the limbless worm-like caecilians (Gymnophiona). The estimated Palaeozoic era gymnophionan-batrachian molecular divergence1 suggests a major gap in the record of crown lissamphibians prior to their earliest fossil occurrences in the Triassic period2-6. Recent studies find a monophyletic Batrachia within dissorophoid temnospondyls7-10, but the absence of pre-Jurassic period caecilian fossils11,12 has made their relationships to batrachians and affinities to Palaeozoic tetrapods controversial1,8,13,14. Here we report

Bone conduction pathways confer directional cues to salamanders.

Sound and vibration are generated by mechanical disturbances within the environment, and the ability to detect and localize these acoustic cues is generally important for survival, as suggested by the early emergence of inherently directional otolithic ears in vertebrate evolutionary history. However, fossil evidence indicates that the water-adapted ear of early terrestrial tetrapods lacked specialized peripheral structures to transduce sound pressure (e.g. tympana). Therefore, early terrestrial hearing should have required nontympanic (or extratympanic) mechanisms for sound detection and loca

Genetic basis for an evolutionary shift from ancestral preaxial to postaxial limb polarity in non-urodele vertebrates.

In most tetrapod vertebrates, limb skeletal progenitors condense with postaxial dominance. Posterior elements (such as ulna and fibula) appear prior to their anterior counterparts (radius and tibia), followed by digit-appearance order with continuing postaxial polarity. The only exceptions are urodele amphibians (salamanders), whose limb elements develop with preaxial polarity and who are also notable for their unique ability to regenerate complete limbs as adults. The mechanistic basis for this preaxial dominance has remained an enigma and has even been proposed to relate to the acquisition o

Aspects on the relief of living surfaces using atomic force microscopy allow "art" to imitate nature.

The visualization of the surface of biological samples using an atomic force microscope reveals features of the external relief and can resolve very fine and detailed features of the surface. We examined specimens from the skin of the amphibians Salamandra salamandra Linnaeus, 1758, Lyciasalamandra luschani basoglui Baran & Atatür, 1980 and Mesotriton alpestris Laurenti, 1768, and from the surface of pollen grains of the plant species Cyclamen graecum Link, 1835 and Cistus salviifolius Linnaeus, 1753, which exhibit certain interesting features, imaged at the nanoscale level. It is likely that

Why does the yellow-eyed Ensatina have yellow eyes? Batesian mimicry of Pacific newts (genus Taricha) by the salamander Ensatina eschscholtzii xanthoptica.

Color patterns commonly vary geographically within species, but it is rare that such variation corresponds with divergent antipredator strategies. The polymorphic salamander Ensatina eschscholtzii, however, may represent such a case. In this species, most subspecies are cryptically colored, whereas E. e. xanthoptica, the Yellow eyed ensatina, is hypothesized to be an aposematic mimic of highly toxic Pacific newts (genus Taricha). To test the mimicry hypothesis, we conducted feeding trials using Western Scrub-Jays, Aphelocoma californica. In every feeding trial, we found that jays, following pr

Tongue evolution in lungless salamanders, family plethodontidae. III. Patterns of peripheral innervation.

Innervation of the tongue and associated musculature in plethodontid salamanders was studied using Palmgren stained sectioned materials, fresh dissection, and whole mounts of experimental specimens treated with horseradish peroxidase (HRP). Species studied were chosen to represent modes of tongue projection recognized by Lombard and Wake ('77). Special attention was given to species of the genera Plethodon, Batrachoseps, Pseudoeurycea, and Hydromantes, but representatives of other genera were investigated. As expected we found that cranial nerves IX and X and spinal nerve 1 supplied the muscle

DNA metabarcoding analysis of stomach flushing contents reveals the exceptionally diverse diet of the golden alpine salamander.

Amphibians, particularly salamanders, are facing dramatic population declines worldwide; therefore, investigating their role as predators through diet analysis could inform targeted conservation programs. DNA metabarcoding significantly outperforms morphology-based identification of prey taxa in other vertebrates, but has rarely been applied to amphibians, and then only for faecal samples. Here, we chose the golden Alpine salamander Salamandra atra aurorae, an endangered endemism of the Italian Alps, to (i) develop a new metabarcoding protocol for amphibian stomach flushing samples; (ii) inves

How will warming affect the growth and body size of the largest extant amphibian? More than the temperature-size rule.

Declining body size is a universal ecological response to global warming in ectotherms. Ectotherms grow faster but mature at a smaller size at higher temperatures. This phenomenon is known as the temperature-size rule (TSR). However, we know little about the details of the relationship between temperature and size. Here, this issue was studied in the Chinese giant salamander (Andrias davidianus), one of the largest extant amphibians and a flagship species of conservation in China. Warm-acclimated A. davidianus larvae (25 °C) had accelerated development but little superiority in body growth whe

The trophic niche of subterranean populations of Speleomantes italicus.

The determination of a species trophic niche can clarify its functional role within a food web and how prey resources are used in relation with the spatial and temporal variability of environmental conditions. This information may result particularly useful for the implementation of conservation plans of endangered species having a cryptic behaviour or living in places difficult to be surveyed. Here we present the first long-term study on the trophic niche of the Italian cave salamander Speleomantes italicus, a strictly protected facultative cave species that seasonally exploits surface enviro
