[Fundamental and clinical studies on ceftazidime in the field of obstetrics and gynecology].

Fundamental and clinical studies were performed on ceftazidime ( CAZ ), a new cephem antibiotic. Following a single intravenous administration of 1 g dose of CAZ , the transfer of CAZ to the internal genital organs was good. The transfer of CAZ to retroperitoneal fluid was excellent. In a clinical trial, CAZ was given to 6 patients with obstetrical and gynecological infections. The efficacy was evaluated as excellent in 3 cases and good in the other 3 cases. No adverse effects were observed in any of the patients treated with CAZ .

Ceftazidime in the treatment of chronic suppurative otitis media in children.

Seventeen children aged 5 months to 3 years with recurrent or chronic otitis media complicated by Pseudomonas aeruginosa were treated with ceftazidime. All patients had been previously treated with several antibiotics. All strains of Pseudomonas were initially sensitive to ceftazidime. In four of the six cases which failed bacteriologically, the Pseudomonas was found to be of intermediate sensitivity after treatment. In 11 patients the Pseudomonas was eradicated during treatment but reappeared in 4 during the follow-up period. All 11 patients were clinically cured or improved. Three of the six

Studies on absorption, distribution, metabolism and excretion of ceftazidime in Japan.

Ceftazidime achieved mean serum levels of 54.6 and 119.7 mg/l at 5 min and 0.9 and 1.9 mg/l at 8 h after an intravenous bolus injection of 0.5 and 1 g in healthy male adult volunteers, showing a dose-related increase in serum levels. Serum half-lives were in the range from 1.6 to 1.7 h. About 90% of the given dose was excreted in the urine during the first 24 h. Following a 1-h intravenous drip infusion of 1 and 2 g, ceftazidime attained peak serum levels of 69.5 and 150.8 mg/l at the end of the infusion. Eighty-three and 90%, respectively, of the given doses were excreted in the urine during

Ceftazidime/Avibactam Monotherapy Versus Other Antibiotics: Where Do We Stand?

The global rise of multi-drug resistant (MDR) pathogens, including the widespread resistance to beta-lactams through the production of β-lactamases, like extended spectrum β-lactamases (ESBLs), has led to the increasing use of last-line antibiotics such as carbapenems. Subsequently, the worldwide emergence of carbapenemase-producing pathogens poses a formidable challenge. The combination ceftazidime/avibactam (CAZ/AVI) has emerged as a pivotal agent in the management of multidrug-resistant Gram-negative infections. Avibactam, a novel β-lactamase inhibitor, demonstrates a wider spectrum of acti

Use of Ceftazidime-Avibactam and Ceftolozane-Tazobactam After Lung Transplantation.

INTRODUCTION: After lung transplantation (LTx), infections caused by multidrug-resistant (MDR) bacteria are frequent and difficult to treat. Some new antibiotics seem to be effective in treating these infections. MATERIAL AND METHODS: We describe our experience in treatment of Klebsiella pneumoniae MDR and Pseudomonas aeruginosa MDR infections with ceftazidime-avibactam (CEF-AVI) and ceftazidime-tazobactam (CEFT-TAZ) in patients who underwent LTx. RESULTS: In 3 patients who underwent double LTx and in 4 patients who underwent single LTx, strains of K. pneumoniae and P. aeruginosa were isolat

Interactions between Avibactam and Ceftazidime-Hydrolyzing Class D β-Lactamases.

Class D β-lactamases exhibit very heterogeneous hydrolysis activity spectra against the various types of clinically useful β-lactams. Similarly, and according to the available data, their sensitivities to inactivation by avibactam can vary by a factor of more than 100. In this paper, we performed a detailed kinetic study of the interactions between two ceftazidime-hydrolyzing OXA enzymes and showed that they were significantly more susceptible to avibactam than several other class D enzymes that do not hydrolyze ceftazidime. From a clinical point of view, this result is rather interesting if o

[Evaluation of ceftazidime monotherapy in Pseudomonas aeruginosa bacteremias. Prospective study].

Twenty-six episodes of Pseudomonas aeruginosa bacteremia treated with intravenous ceftazidime, 4-6 g/day were evaluated. Treatment was begun within the first 24 hours after the isolation of the microorganism and was maintained for 10-12 days. In two patients with neutropenia amikacin was added during the initial 48-72 hours until the susceptibility to ceftazidime was known. All isolates were sensitive to ceftazidime. The most common underlying diseases were neoplasia (12), diabetes with stroke (4), neurosurgical and vascular procedures (4), rheumatoid arthritis (2), burns (2), cor pulmonale (1

Antimicrobial activity, pharmacokinetics, therapeutic indications and adverse reactions of ceftazidime.

Ceftazidime is an aminothiazolyl cephalosporin with potent activity against gram-negative bacteria including multiresistant strains of Pseudomonas aeruginosa. It has limited activity against gram-negative anaerobes, is less active against some gram-positive cocci than other newer beta-lactam compounds and is inactive against Streptococcus faecalis and methicillin-resistant Staphylococcus aureus. Ceftazidime is stable against common plasmid and chromosomally mediated beta-lactamase produced by Enterobacteriaceae and Pseudomonas sp. Its pharmacokinetic properties are similar to those of moxalact

Time-Kill Evaluation of Antibiotic Combinations Containing Ceftazidime-Avibactam against Extensively Drug-Resistant Pseudomonas aeruginosa and Their Potential Role against Ceftazidime-Avibactam-Resistant Isolates.

Ceftazidime-avibactam (CZA) has emerged as a promising solution to the lack of new antibiotics against Pseudomonas aeruginosa infections. Data from in vitro assays of CZA combinations, however, are scarce. The objective of our study was to perform a time-kill analysis of the effectiveness of CZA alone and in combination with other antibiotics against a collection of extensively drug-resistant (XDR) P. aeruginosa isolates. Twenty-one previously characterized representative XDR P. aeruginosa isolates were selected. Antibiotic susceptibility was tested by broth microdilution, and results were int

Optimal ceftazidime/avibactam dosing exposure against KPC-producing Klebsiella pneumoniae.

OBJECTIVES: Infections due to carbapenem-resistant Enterobacterales are considered urgent public health threats and often treated with a β-lactam/β-lactamase inhibitor combination. However, clinical treatment failure and resistance emergence have been attributed to inadequate dosing. We used a novel framework to provide insights of optimal dosing exposure of ceftazidime/avibactam. METHODS: Seven clinical isolates of Klebsiella pneumoniae producing different KPC variants were examined. Ceftazidime susceptibility (MIC) was determined by broth dilution using escalating concentrations of avibacta

Ceftazidime alone for treating Pseudomonas aeruginosa septicaemia in neutropenic patients.

Twenty-nine patients with primary or secondary hypoplastic bone-marrow were treated successfully with ceftazidime alone for established septicaemia caused by Pseudomonas aeruginosa of which 38% strains were gentamicin-resistant . All the patients were neutropenic at the start of therapy; most were cured with microbiological confirmation before the bone marrow had regenerated. One patient died of cerebral haemorrhage due to profound thrombocytopenia without evidence of infection at autopsy. Significant toxicity was not observed. Ceftazidime alone is, therefore, a safe and effective treatment fo

Sequential ventricular fluid concentrations of ceftazidime--report of three cases.

Three adult subjects, each with a ventriculostomy, received ceftazidime 2g iv q8h for three doses. Serial samples of serum and CSF ventricular fluid were obtained following the third dose; ceftazidime concentrations were measured by high pressure liquid chromatography. In one patient without inflammatory cells in the CSF, ceftazidime CSF concentrations were only approximately 0.3 micrograms/ml. In two other patients who had inflammatory cells and blood in the CSF, concentrations of ceftazidime in ventricular fluid demonstrated a slow rise and decline over an eight-hour period. Although contami

In vitro susceptibility of Burkholderia cepacia to ceftazidime-avibactam: A systematic review.

BACKGROUND: Burkholderia cepacia complex (BCC) is a group of multi-drug resistant (MDR) pathogens that are challenging to treat, especially in cystic fibrosis (CF) patients. Ceftazidime-avibactam is a promising antibiotic combination for treating BCC infections, but its efficacy requires further in vitro evaluation. METHODS: This systematic review was conducted following the PRISMA guidelines to assess the in vitro susceptibility of BCC strains to ceftazidime-avibactam. We systematically searched PubMed, Web of Science, Scopus, and Embase databases. Inclusion criteria required original articl

Ceftazidime-resistance in pediatric melioidosis: A case report and literature review.

We report a first case of ceftazidime-resistant pediatric melioidosis involving a previously healthy seven-year-old boy who presented with right lobar pneumonia complicated with a 5-cm lung abscess. Ceftazidime was initiated on Day-6 of admission when Burkholderia pseudomallei (ceftazidime-susceptible, minimum inhibitory concentration [MIC] 1.0 mcg/mL) was isolated from blood. Despite ceftazidime therapy at the recommended dosage, he developed fulminant septic shock and respiratory failure on Day-18 of hospitalization, requiring invasive ventilation, hemodynamic support, and continuous renal r

Ceftazidime-Avibactam as a Salvage Treatment for Severely Infected Immunosuppressed Children.

BACKGROUND: Multidrug-resistant Gram-negative bacteria (MDR-GNB) are becoming increasingly common around the world, with carbapenems frequently serving as a last resort but being threatened by the growing incidence of carbapenemase-producing bacteria. Ceftazidime-avibactam (CAZ/AVI) is a potential agent against MDR-GNB but with limited clinical experience, particularly in critically ill immunosuppressed children. METHODS: This study analyzed the use of CAZ/AVI as salvage treatment in severely infected immunosuppressed children from September 2019 to July 2022. Patients with confirmed GNB infe
