Expression of basement membrane antigens in spindle cell melanoma.

Spindle cell melanoma (SCM) is an uncommon form of melanoma that may be confused histologically with other tumors, including malignant peripheral nerve sheath tumors (MPNST). Tumors with neural differentiation and melanocytic nevi may both show basement membrane immunohistochemically and at the ultrastructural level. However, most ultrastructural studies of melanoma have failed to demonstrate well formed basement membrane around tumor cells. The presence of basement membrane has been used by some authors as evidence favoring MPNST, as opposed to SCM. To evaluate this distinction immunohistoche

Modulation of cell membrane potential in cultured vascular endothelium.

The present study explores the role of different ionic conductances in the regulation of membrane potential under resting conditions and after bradykinin (BK) or thapsigargin (TG) stimulation of cultured bovine aortic endothelial cells. Under resting conditions, the cell membrane potential observed was -62+/- 5 mV. The main conductance under these conditions is an inwardly rectifying potassium (IRK) channel. Application of 50 nM BK induced a transient hyperpolarization to -87 +/- 4 mV followed by sustained depolarization to -35 +/- 5 mV. The transient hyperpolarization was eliminated by 1 micr

[Hereditary high red cell membrane phosphatidylcholine hemolytic anemia (HPCHA)].

HPCA is a rare disease among hemolytic anemias. A dominantly transmitted chronic anemia characterized by an increase of erythrocyte membrane phosphatidylcholine (PC) and cholesterol with decrease of phosphatidylethanolamine (PE). On peripheral blood smears stomatocytes and target cells are evident. The MCV may be increased, but the life span of red cells is decreased. The osmotic fragility is normal or increased, but splenectomy is not effective. These two points are different from other hereditary hemolytic anemias. As in other hemolytic anemias, gallstone and biliary tract disease more frequ

Decreased cell membrane magnesium in some essential hypertension patients.

The concentrations of total ([T-Mg]), ultrafilterable ([UF-Mg]), and protein-bound or nonfilterable ([NF-Mg]) magnesium were measured in the plasma and in the intracellular compartment of blood from 8 essential hypertensive patients and 9 normotensive subjects. In the former, [T-Mg] was unchanged in the plasma but decreased in whole blood due to decreases of both [UF-Mg] and [NF-MG]; [UF-Mg] was increased in plasma but decreased intracellularly while [NF-Mg] was decreased in plasma and unchanged intracellularly. These concentrations correlated significantly with the average blood pressures. De

[Cytoskeleton anomalies in disorders of red cell membrane proteins].

Cytoskeletal abnormalities in red cells were studied in 250 patients at our laboratory, especially on hereditary elliptocytosis (HE), hereditary spherocytosis (HS), and band 4.2 anomalies. First of all, on HE, we have found two patients of a dominantly-inherited trait of a new beta-spectrin variant with 216 kDa peptide. The contents of abnormal beta'-spectrin to the total spectrins were 7.6% in propositus, and 10.5% in her mother. As a functional abnormality, abnormal alpha beta'-spectrin dimer could not be converted to tetramer in both patients. Thus, our patients may differ from HE Nice. Sec

Cross bonding and stiffening of the red cell membrane.

Cross bonding and stiffening of the human red cell membrane was studied using treatments with SH, amino, and carboxyl reagents, oxidizing and denaturing treatments and acidification. Membrane cross bonding was initiated when, after red cell treatment, opposite areas of the cytoplasmic face of the red cell membrane were brought into contact by cell shrinking. Membrane cross bonding was detected by light microscopy when this contact persisted upon swelling the cells in a hypotonic medium. Membrane stiffening was recorded as a decrease in elongation of red cells in the shear field of a viscous de

Calcium ions, drug action and the red cell membrane.

Intracellular calcium regulates a number of membrane functions in the erythrocyte, including control of shape, membrane lipid composition and cation permeability. Measurement of total red cell calcium has yielded values between 5 and 15 nmol/ml cells, and these low values in part reflect the absence of Ca2+ -containing organelles. Most intracellular Ca2+ is bound and the low cell ionized Ca2+ concentration (approximately 0.2 microM) is maintained by a combination of low membrane permeability and a powerful Ca2+ -pump. This pump has been identified with a (Ca2+ + Mg2+)-stimulated ATPase, and bo

Role of membrane-bound haemoglobin products in oxidative damage in sickle cell membranes.

Even though the development of the membrane abnormalities in sickle cell erythrocytes is due to the presence of a mutant gene product, sickle cell anaemia is in a sense a membrane disease. In particular, the presence of iron-containing breakdown products of haemoglobin in the sickle cell membranes provides a source of continued oxidative damage. An understanding of the primary causes of these membrane abnormalities may be useful in the development of effective therapies.

Retinal glial cell removing during internal limiting membrane peeling is associated with higher-stage idiopathic epiretinal membrane.

BACKGROUND: Histological examination of surgically excised internal limiting membrane (ILM) specimens may provide insights into retinal tissue changes, but their relationship to retinal imaging findings remains unclear. The aim of this study was to correlate retinal imaging findings with histological evidence of retinal tissue components in surgically excised ILM specimens from eyes with idiopathic epiretinal membrane (iERM) or full-thickness macular hole (FTMH). METHODS: This retrospective clinicopathological study included 42 eyes that underwent pars plana vitrectomy with membrane peeling a

Advances in Biomimetic Cell Membrane Nanoplatforms for Renal-Targeted Theranostics: From Pathophysiological Basis to Membrane-Stratified Design.

Kidney diseases represent a major global health burden, with current clinical treatments impeded by short circulatory half-lives, inadequate renal targeting, and the risk of systemic off-target toxicity. Conventional synthetic nanocarriers face critical bottlenecks, including rapid immune clearance and insufficient targeting specificity within the complex renal pathophysiological microenvironment. While biomimetic cell membrane nanoplatforms (BCM-NPs) offer a promising solution, to the best of our knowledge, no previous review has systematically integrated the pathophysiological basis of renal

Absence of Specific Chlamydia trachomatis Inclusion Membrane Proteins Triggers Premature Inclusion Membrane Lysis and Host Cell Death.

Chlamydia trachomatis is a human pathogen associated with significant morbidity worldwide. As obligate intracellular parasites, chlamydiae must survive within eukaryotic cells for sufficient time to complete their developmental cycle. To promote host cell survival, chlamydiae express poorly understood anti-apoptotic factors. Using recently developed genetic tools, we show that three inclusion membrane proteins (Incs) out of eleven examined are required for inclusion membrane stability and avoidance of host cell death pathways. In the absence of specific Incs, premature inclusion lysis results

Voltage-dependent changes in specific membrane capacitance caused by prestin, the outer hair cell lateral membrane motor.

In the outer hair cell (OHC), membrane capacitance principally derives from two components - that associated with lateral membrane sensor/motor charge movement, and that proportional to the membrane surface area (C(sa)). We used measures of membrane capacitance to test a model hypothesis that OHC lateral membrane molecular motors, recently identified as the protein prestin, fluctuate between two area states. By measuring membrane capacitance in native OHCs or prestin-transfected HEK cells at extreme voltages (+/-200 mV) where motor-derived charge movement is small or absent, we observed that C

Feedback regulation between plasma membrane tension and membrane-bending proteins organizes cell polarity during leading edge formation.

Tension applied to the plasma membrane (PM) is a global mechanical parameter involved in cell migration. However, how membrane tension regulates actin assembly is unknown. Here, we demonstrate that FBP17, a membrane-bending protein and an activator of WASP/N-WASP-dependent actin nucleation, is a PM tension sensor involved in leading edge formation. In migrating cells, FBP17 localizes to short membrane invaginations at the leading edge, while diminishing from the cell rear in response to PM tension increase. Conversely, following reduced PM tension, FBP17 dots randomly distribute throughout the

Ektacytometry Analysis of Post-splenectomy Red Blood Cell Properties Identifies Cell Membrane Stability Test as a Novel Biomarker of Membrane Health in Hereditary Spherocytosis.

Hereditary spherocytosis (HS) is the most common form of hereditary chronic hemolytic anemia. It is caused by mutations in red blood cell (RBC) membrane and cytoskeletal proteins, which compromise membrane integrity, leading to vesiculation. Eventually, this leads to entrapment of poorly deformable spherocytes in the spleen. Splenectomy is a procedure often performed in HS. The clinical benefit results from removing the primary site of destruction, thereby improving RBC survival. But whether changes in RBC properties contribute to the clinical benefit of splenectomy is unknown. In this study w

Listeria Membrane Protrusion Collapse: Requirement of Cyclophilin A for Listeria Cell-to-Cell Spreading.

BACKGROUND: Listeria generate actin-rich tubular protrusions at the plasma membrane that propel the bacteria into neighboring cells. The precise molecular mechanisms governing the formation of these protrusions remain poorly defined. METHODS: In this study, we demonstrate that the prolyl cis-trans isomerase (PPIase) cyclophilin A (CypA) is hijacked by Listeria at membrane protrusions used for cell-to-cell spreading. RESULTS: Cyclophilin A localizes within the F-actin of these structures and is crucial for their proper formation, as cells depleted of CypA have extended actin-rich structures t

Effects of Poloxamer 188 on red blood cell membrane properties in sickle cell anaemia.

Vaso-occlusive crisis (VOC) is the main acute complication in sickle cell anaemia (SS) and several clinical trials are investigating different drugs to improve the clinical severity of SS patients. A phase III study is currently exploring the profit of Velopoloxamer in SS during VOCs. We analysed, in-vitro, the effect of poloxamer (P188) on red blood cell (RBC) properties by investigating haemorheology, mechanical and adhesion functions using ektacytometry, microfluidics and dynamic adhesion approaches, respectively. We show that poloxamer significantly reduces blood viscosity, RBC aggregation

Type II integral membrane protein, TM of J paramyxovirus promotes cell-to-cell fusion.

Paramyxoviruses include many important animal and human pathogens. Most paramyxoviruses have two integral membrane proteins: fusion protein (F) and attachment proteins hemagglutinin, hemagglutinin-neuraminidase, or glycoprotein (G), which are critical for viral entry into cells. J paramyxovirus (JPV) encodes four integral membrane proteins: F, G, SH, and transmembrane (TM). The function of TM is not known. In this work, we have generated a viable JPV lacking TM (JPV∆TM). JPV∆TM formed opaque plaques compared with JPV. Quantitative syncytia assays showed that JPV∆TM was defective in promoting c

Detection of anti-liver cell membrane antibody using a human hepatocellular carcinoma cell line.

A radioimmunometric technique for the detection of autoantibodies to liver membrane antigens has been developed using Alexander cells, a human hepatocellular carcinoma cell line. After incubation of Alexander cells with serum, antimembrane antibodies were detected by addition of 125I-labeled Protein A. Binding ratios in 15 children with uncontrolled autoimmune chronic active hepatitis and in seven children with primary sclerosing cholangitis were significantly higher than in 18 age-matched normal controls. Nine patients with inactive autoimmune chronic active hepatitis, 13 with alpha 1-antitry
