Stress-induced formation of cell wall-deficient cells in filamentous actinomycetes.

The cell wall is a shape-defining structure that envelopes almost all bacteria and protects them from environmental stresses. Bacteria can be forced to grow without a cell wall under certain conditions that interfere with cell wall synthesis, but the relevance of these wall-less cells (known as L-forms) is unclear. Here, we show that several species of filamentous actinomycetes have a natural ability to generate wall-deficient cells in response to hyperosmotic stress, which we call S-cells. This wall-deficient state is transient, as S-cells are able to switch to the normal mycelial mode of gro

Cell wall glycoproteins participate in the adhesion of Sporothrix schenckii to epithelial cells.

Sporothrix schenckii is the etiologic agent of sporotrichosis. This fungal infection is an emerging disease potentially fatal in immunocompromised patients. The adhesion to host cells is a crucial early event related with the dissemination of pathogens. In order to clarify the mechanisms of adhesion of S. schenckii yeast cell to epithelial cells, we studied the biochemical basis of this process. The electrophoretic analysis of cell wall protein from S. schenckii coupled at ConA and stained with HRP, revealed nine different proteins with MW ≥ 180, 115, 90, 80, 58, 40, 36, 22 and 18 kDa. Cas5 Regulates the Exposure of β-Glucan, the Cell Surface Hydrophobicity, and the Expression of Cell Wall Proteins to Remodel the Candida albicans Cell Wall and Participates in the Recruitment of Neutrophils.

Candida albicans (C. albicans) is a major opportunistic fungal pathogen that causes life-threatening infections, particularly in immunocompromised individuals, underscoring the critical need to understand its pathogenic mechanisms. This study investigates the role of Cas5, a key transcription factor, in regulating C. albicans cell wall remodeling, virulence, and host interactions. Genetic manipulation and biochemical assays were used to examine the effects of Cas5 depletion on C. albicans cell wall structure, adhesion to host cells, morphology transition, innate immune cells recruitment, and p

Ligation of arabinogalactan to peptidoglycan in the cell wall of Mycobacterium smegmatis requires concomitant synthesis of the two wall polymers.

To study the late events of cell wall assembly in Mycobacterium smegmatis, specific in vivo radiolabelling of exponentially growing liquid cultures over periods of less than one cell generation were carried out. N-Acetyl-[(14)C]glucosamine was used to label peptidoglycan and [(14)C]glucose to label arabinogalactan and arabinomannan. Over periods of several generations, radioactive cell wall material was turned over as soluble autolysis products into the culture fluid. However, turnover of newly synthesized and labelled cell wall was delayed for about one cell generation, implying inside-to-out

A versatile bacterial cell wall-based nanomedicine for combination treatment of oral squamous cell carcinoma.

Oral squamous cell carcinoma (OSCC), an aggressive malignancy characterized by high recurrence and metastasis rates, continues to pose a significant clinical challenge to human health. Bacterial immunotherapy has emerged as a promising strategy for cancer treatment by activating multimodal immune responses. Porphyromonas gingivalis (Pg), an oral pathogenic bacterium strongly associated with periodontitis, can produce abundant μ-oxo bisheme on its cell wall (CW) to avoid the oxidative damage induced by H2O2 during inflammatory processes. Owing to the presence of μ-oxo bisheme, the CW extracted

E69K mutation in β-tubulin 2 blocks cell wall integrity signaling during plant cell elongation.

The intact cell wall is the prerequisite for plant cell morphogenesis. Loss of function in FRA1/KINESIN-4A, which encodes a microtubule-based kinesin motor, causes dwarfed growth phenotypes with reduced cell wall mechanics. However, the underlying mechanisms remain elusive. Here, using genetic screening, we identify a suppressor of fra1 (sofa1) mutation that specifically suppresses the dwarf phenotype of the fra1 mutant. The sofa1 carries an E69K mutation in β-Tubulin 2 (TUB2), and the dominant suppressive effect of E69K mutation is conserved among β-tubulins. We further reveal that incorporat

Immunolabeling Sieve Element Cell Walls with the LM26 Antibody.

Sieve elements in the phloem transport carbon and small molecules, such as RNA and phytohormones, throughout the plant body. Understanding the physical dimensions of sieve elements and phloem tissue is thus crucial for predicting how much carbon can be moved at any given time. Quantification of sieve element diameters and areas has previously been performed using transmission electron microscopy, scanning electron microscopy, and light microscopy, but sieve element identification is difficult because the phloem is a heterogeneous tissue. The recently identified LM26 antibody labels a pectin in

Understanding the microbiology of the Aspergillus cell wall and the efficacy of caspofungin.

The efficacy of echinocandins against Aspergillus species has been established through in vitro assays, in animal models of infection, and in clinical practice. Caspofungin is an inhibitor of 1,3-β-D glucan synthesis (GS) that produces dramatic morphological changes, but incomplete clearing, in cultures of growing hyphae. Despite the apparent fungistatic in vitro activity against Aspergillus species, compounds in this class have strong efficacy in vivo. For example, caspofungin prolonged survival in chronically immunosuppressed mice with induced disseminated aspergillosis, even when neutropeni

Characterization of the outer hair cell's lateral wall membranes.

We examined the properties of outer hair cell (OHC) lateral wall membranes by application of 2 fluorescent membrane probes. The markers, C6-NBD-Ceramide and DiOC6, have been used in other cell types to label Golgi apparatus and endoplasmic reticulum, respectively. In living isolated OHCs NBD-Ceramide demonstrated uninterrupted fluorescence along the OHC lateral wall, while DiOC6 labeling proved punctate and notably less uniform in this region. In aldehyde-fixed isolated OHCs both probes exhibited distinct, continuous lateral wall fluorescence. Fixed preparations of the organ of Corti labeled w

Uptake and retention of metals by cell walls of Bacillus subtilis.

Isolated walls of Bacillus subtilis Marburg, prepared in a manner which avoided metal contamination other than by the growth medium, were incubated in dilute metal solutions, separated by membrane filtration (0.22 mum), and monitored by atomic absorption to give uptake data for 18 metals. Substantial amounts of Mg2+, Fe3+, Cu2+, Na+, and K+ (amounts which were often visible as Au3+, and Ni2+ (the higher atomic-numbered elements also visible as electron scattering), and small amounts of Hg2+, Sr2+, Pb2+, and Ag+ were taken into the wall. Some (Li+, Ba2+, Co2+, and Al3+) were not absorbed. Most

Mitochatting - If only we could be a fly on the cell wall.

Mitochondria, cellular metabolic hubs, perform many essential processes and are required for the production of metabolites such as ATP, iron-sulfur clusters, heme, amino acids and nucleotides. To fulfill their multiple roles, mitochondria must communicate with all other organelles to exchange small molecules, ions and lipids. Since mitochondria are largely excluded from vesicular trafficking routes, they heavily rely on membrane contact sites. Contact sites are areas of close proximity between organelles that allow efficient transfer of molecules, saving the need for slow and untargeted diffus

Cell wall glycopolymers of Firmicutes and their role as nonprotein adhesins.

Cell wall glycopolymers (CWGs) of gram-positive bacteria have gained increasing interest with respect to their role in colonization and infection. In most gram-positive pathogens they constitute a large fraction of the cell wall biomass and represent major cell envelope determinants. Depending on their chemical structure they modulate interaction with complement factors and play roles in immune evasion or serve as nonprotein adhesins that mediate, especially under dynamic conditions, attachment to different host cell types. In particular, covalently peptidoglycan-attached CWGs that extend well

Recent advances in the understanding of the Aspergillus fumigatus cell wall.

Over the past several decades, research on the synthesis and organization of the cell wall polysaccharides of Aspergillus fumigatus has expanded our knowledge of this important fungal structure. Besides protecting the fungus from environmental stresses and maintaining structural integrity of the organism, the cell wall is also the primary site for interaction with host tissues during infection. Cell wall polysaccharides are important ligands for the recognition of fungi by the innate immune system and they can mediate potent immunomodulatory effects. The synthesis of cell wall polysaccharides

Interaction of the CdSe quantum dots with plant cell walls.

There is an increasing application of quantum dots (QDs) in plant science, as markers for the cells or their cell walls (CWs). In a plant cell the CW is a first target place for external agents. We studied interaction of CdSe QDs with CWs isolated from a conifer -Picea omorika (Panč) Purkynĕ branch. Binding of CdSe QDs was followed by using fluorescence microscopy, fluorescence and FT-IR spectroscopy. The aim of the study was to see whether the QDs induce structural changes in the CW, as well as to find out which kind of interactions between QDs and CWs occur and to which particular constituen

Structure of ristocetin A in complex with a bacterial cell-wall mimetic.

Antimicrobial drug resistance is a serious public health problem and the development of new antibiotics has become an important priority. Ristocetin A is a class III glycopeptide antibiotic that is used in the diagnosis of von Willebrand disease and which has served as a lead compound for the development of new antimicrobial therapeutics. The 1.0 A resolution crystal structure of the complex between ristocetin A and a bacterial cell-wall peptide has been determined. As is observed for most other glycopeptide antibiotics, it is shown that ristocetin A forms a back-to-back dimer containing conca

The immune response to the cell wall of Mycobacterium bovis BCG.

Mice were immunized with the cell wall of BCG suspended in an oil-in-saline emulsion, and examined against time for the emergence of T cell-mediated acquired immunity. Evidence is presented that shows that levels of acquired resistance expressed in these animals over the first month following inoculation, and which enabled them to substantially resist an intravenous challenge infection with Mycobacterium tuberculosis, were completely nonspecific in nature, in that they were equally well expressed in normal and T cell-deficient mice, and were present at a time when no protective T cell activity

Immunologically active cell wall polysaccharides of green microalga Dictyosphaerium chlorelloides (Chlorellacea).

Dictyosphaerium chlorelloides is a green microalga from the Chlorella clade that produces highly viscous exocellular polysaccharides. The cell wall polysaccharides of this alga have not been studied in detail. In this article, water-soluble polysaccharides from D. chlorelloides biomass were extracted with hot water and purified by preparative chromatography. The composition, structural features and molecular masses of subsequently eluted fractions F1, F2, F3, F4 and F5 (minor) were determined. Three high-yield products F1, F3 and F4 consisted mainly of galactopyranosyl, 2-O-methyl-galactopyran

Immune Activation and Glycolytic Responses to Cutibacterium acnes Cell Wall Polysaccharides.

Carbohydrates are key components of many microbial cell walls and play a versatile role in immune recognition. In this study, we analyzed the carbohydrate cell wall composition of Cutibacterium acnes strains associated with healthy skin (denoted as CH) and acne-prone skin (denoted as CA) to understand their influence on host immune responses in acne. We identified glucose, mannose, and galactose as the primary monosaccharides, with minor amounts of fucose, N-acetylgalactosamine, and N-acetylglucosamine. Linkage analysis revealed structural variations between CH and CA strains: CH strains showe
