The Microbiota Dynamics of Alfalfa Silage During Ensiling and After Air Exposure, and the Metabolomics After Air Exposure Are Affected by Lactobacillus casei and Cellulase Addition.

Both inoculants treatment and enzyme treatment promote the reproduction of lactic acid bacteria (LAB) to produce enough lactic acid to lower pH in silage. The present study investigated the microbial community and metabolome in cellulase, Lactobacillus casei, and air treated alfalfa silage. Chopped and wilted alfalfa (first cutting, 29% dry matter) was ensiled without (CON) or with L. casei (1 × 106 cfu g-1 fresh matter) (LC) or cellulase (20,000IU, 0.5% of fresh matter) (CE) for 56 days, then exposed to air for 3 days (PO). Greater ensiling quality was observed in LC and CE, which had lower p

Synergistic enhancement of physicochemical and immunomodulatory properties of Cyclocarya paliurus polysaccharides through co-culture solid-state fermentation with endophytic cellulase-producing fungi.

Cyclocarya paliurus (C. paliurus) polysaccharides (CP) exhibit hypoglycemic, antihypertensive, and antioxidant activities, yet their extraction efficiency and bioactivity are limited by the intact plant cell wall. Microbial fermentation, especially using endophytic fungi, offers a green strategy to degrade cell wall polysaccharides and modify their structure. In this study, two high-cellulase-producing endophytes (PZ8 and PM2) were screened from C. paliurus and used for co-culture solid-state fermentation. The fermentation significantly increased CP yield to 29.16 mg/g, reduced the molecular w

Exploring the aerobic stability, antioxidant and microbial community of Broussonetia papyrifera ensiled with ferulic acid esterase-producing Lactiplantibacillus plantarum in combination with cellulase and/or xylanase.

BACKGROUND: Ferulic acid possesses certain antioxidant and antibacterial properties. Additionally, ferulic acid esterase (FAE) and cellulolytic enzymes have been associated with synergistic degradation of ferulic acid ester bonds, thereby facilitating greater release of ferulic acid from lignocellulose, which could have important effects on silage quality and aerobic stability. METHODS: This study examined the effects of ensiling Broussonetia papyrifera with FAE-producing Lactiplantibacillus plantarum (LP), cellulase (CE) and xylanase (XY) under aerobic exposure conditions. The following trea

Antimicrobial and anti-dust mite efficacy of Cinnamomum camphora chvar. Borneol essential oil using pilot-plant neutral cellulase-assisted steam distillation.

Cinnamomum camphora chvar. Borneol essential oil (BEO) was efficiently extracted by using pilot-plant neutral cellulase-assisted steam distillation (NCSD). Borneol, β-cadinene and α-caryophyllene were identified as major components. Bacillus subtilis was the most sensitive bacteria to BEO with the lowest minimal inhibition concentration (MIC) and minimal bactericial concentration (MBC) at 1·75 and 3·50 mg ml-1 , respectively. Antimicrobial activity of the BEO was also reasonably high against Salmonella typhimurium, Escherichia coli and Staphylococcus aureus, but not sensitive against two fungi

Purification and biochemical properties of a thermostable, haloalkaline cellulase from Bacillus licheniformis AMF-07 and its application for hydrolysis of different cellulosic substrates to bioethanol production.

A thermophilic strain AMF-07, hydrolyzing carboxymethylcellulose (CMC) was isolated from Kerman hot spring and was identified as Bacillus licheniformis based on 16S rRNA sequence homology. The carboxymethylcellulase (CMCase) enzyme produced by the B. licheniformis was purified by (NH4)2SO4 precipitation, ion exchange and gel filtration chromatography. The purified enzyme gave a single band on SDS- PAGE with a molecular weight of 37 kDa. The CMCase enzyme was highly active and stable over broad ranges of temperature (40-80ºC), pH (6.0-10.0) and NaCl concentration (10-25%) with an optimum at 70º

Dietary fiber modification by cellulase-AP3 supplementation tailors oat digestion under elderly gastrointestinal conditions with attenuated starch hydrolysis and enhanced polyphenol bioaccessibility.

The resilient fiber matrix of oats limits nutrient release and moderates glycemic response. This study employed a dynamic in vitro stomach-intestine model to investigate how Cellulase-AP3 enzyme supplementation alters oat digestion under simulated elderly conditions. Cellulase-AP3 supplementation caused significant oat matrix disintegration by enzymatically converting insoluble fiber (54.84% to 32.18%) into soluble fiber (50.56% to 59.63%), which in turn elevated digestive fluid viscosity and delayed gastric emptying. This modified physiological environment hindered amylase action, thereby sub

An integrated ultrasonic and cellulase pretreatment strategy: simultaneously enhancing medium-chain fatty acid production and reducing antibiotic resistance genes in anaerobic digestion of animal manure.

Rapid expansion of animal husbandry poses significant environmental challenges, including waste management and pollution. While medium-chain fatty acid (MCFA) production from livestock manure via chain elongation (CE) process in anaerobic digestion (AD) offers a promising and economically valuable waste management strategy, the associated risks of antibiotic resistance genes (ARGs) propagation through AD products are often overlooked. Additionally, the impact of substrate pretreatments on MCFA production and ARGs abundances in livestock manure during CE process remains unclear. This study asse

Biochar composite derived from cellulase hydrolysis apple branch for quinolone antibiotics enhanced removal: Precursor pyrolysis performance, functional group introduction and adsorption mechanisms.

In this study, magnetic biochar (MAB) and humic acid (HA)-coated magnetic biochar produced from apple branches without and after cellulase hydrolysis (HMAB and CHMAB, respectively) were prepared and tested as adsorbents of enrofloxacin (ENR) and moxifloxacin (MFX) in aqueous solution. Compared with MAB and HMAB, novel adsorbent CHMAB possessed a superior mesoporous structure, greater graphitization degree and abundant functional groups. When antibiotic solutions ranged from 2 to 20 mg L-1, the theoretical maximum adsorption capacities of CHMAB for ENR and MFX were 48.3 and 61.5 mg g-1 at 35 °C

Effects of Ultra-High Pressure-Cellulase Pretreatment on the Structural Characteristics and Functional Properties of Soluble Dietary Fiber from Perilla Seed Hulls and Its Application in Wheat-Corn Composite Dough.

Perilla seed hulls are a promising but underutilized source of dietary fiber, yet little is known about how pretreatment-induced structural changes in soluble dietary fiber (SDF) derived from these hulls affect its functionality in cereal dough. This study compared SDF extracted from untreated, ultra-high-pressure (UHP)-pretreated, cellulase-pretreated, and sequentially UHP-cellulase-pretreated perilla seed hulls and evaluated their structural, physicochemical, and dough-modifying properties. The combined treatment increased the SDF content of the hull material from 3.71% to 8.63% and decrease

Synergistic effects of ferulic acid esterase-producing lactic acid bacteria, cellulase and xylanase on the fermentation characteristics, fibre and nitrogen components and microbial community structure of Broussonetia papyrifera during ensiling.

BACKGROUND: The high fibre content of whole plants of Broussonetia papyrifera limits its efficient utilization. Ferulic acid esterase (FAE), in combination with xylanase, can effectively cleave the lignin-carbohydrate complex, promoting the function of cellulase. However, little is known about the impact of these additives on silage. To effectively utilize natural woody plant resources, FAE-producing Lactiplantibacillus plantarum RO395, xylanase (XY) and cellulase (CE) were used to investigate the dynamic fermentation characteristics, fibre and nitrogen components and microbial community struc

Application of statistical designs strategy to improve cellulase production using agro-waste residue by a novel isolate Bacillus licheniformis strain-MA1 and assessing the enzyme effect on apple juice quality.

BACKGROUND: Cellulose is the major part of lignocellulosic biomass. It can be hydrolyzed into glucose units via specific enzymes called cellulases that have been applied in many commercial fields. There are several studies illustrate the influence of enzymes on apple juice clarification. However, to the best of our knowledge, the effect of microbial cellulase on volatile compounds of apple juice is not well known. The present study aimed to assess the effect of cellulase from a new bacterial isolate on the physicochemical properties of apple juice as well as volatile compounds. The hydrolysis

Application of cross-linked carboxymethyl cellulose degradation by beta-glucosidase and vaginal microbes to toxic shock syndrome.

Eleven bacterial and two yeast strains, four of which were previously identified as having activity on a lightly cross-linked carboxymethyl cellulose (CLD-2) found in one type of superabsorbent tampon, were grown on a variety of substrates, most containing cellulosics. None produced detectable amounts of cellulases, but all elaborated beta-glucosidase. None of these 13 strains nor 3 commercially obtained beta-glucosidase preparations could hydrolyze CLD-2, although a commercial cellulase and two other bacterial preparations known to produce cellulases could. Based on these results, it appears

Enhanced secretion of extracellular proteins in Trichoderma reesei through targeted deletion of a kinesin encoding gene.

Trichoderma reesei is an important industrial cellulase producer, but its protein secretion is limited by elongated hyphae during bioreactor cultivation. Physical and process-based controls are widely applied to modulate fungal morphology, but genetic strategies remain poorly explored. Here, we show that deletion of kinesin gene kin1 alters hyphal growth and boosts cellulase production in T. reesei. Deletion of kin1 led to pronounced morphological changes: shorter, highly branched hyphae, compact colony architecture, and increased biomass. Under cellulase-inducing conditions, the Δkin1 strain

Creating a large designer cellulosome in yeast to boost ethanol production.

Cellulosic biomass represents a promising feedstock for biofuel and biochemical production. However, its recalcitrant structure strongly hinders enzymatic degradation. Cellulosomes are large multienzyme complexes, highly efficient at degrading cellulose. A cellulase in a cellulosome has a dockerin domain that binds to a cohesin module on the CipA (cellulosome integrating protein A). In a native cellulosome all cohesins are identical, so that the cellulase types and their positions in a CipA cannot be controlled. Here, we constructed the largest designer CipA known to date. Using innovative tec

Multiple domains in endoglucanase B (CenB) from Cellulomonas fimi: functions and relatedness to domains in other polypeptides.

Endoglucanase B (CenB) from the bacterium Cellulomonas fimi is divided into five discrete domains by linker sequences rich in proline and hydroxyamino acids (A. Meinke, C. Braun, N. R. Gilkes, D. G. Kilburn, R. C. Miller, Jr., and R. A. J. Warren, J. Bacteriol. 173:308-314, 1991). The catalytic domain of 608 amino acids is at the N terminus. The sequence of the first 477 amino acids in the catalytic domain is related to the sequences of cellulases in family E, which includes procaryotic and eucaryotic enzymes. The sequence of the last 131 amino acids of the catalytic domain is related to seque

Respiratory allergy to Aspergillus-derived enzymes in bakers' asthma.

Baking and food industry workers are exposed to several powdered Aspergillus-derived enzymes with carbohydrate-cleaving activity that are commonly used to enhance baked products. We describe a retrospective study of sensitization to fungal alpha-amylase and cellulase on bakers. Five bakers in whom respiratory allergy symptoms developed when they were exposed to bread "improvers" that contained fungal alpha-amylase and cellulase were investigated by in vivo and in vitro tests. Type I hypersensitivity to these enzymes was demonstrated in the five patients by means of skin testing, histamine rele
