Cerebellar EEG source localization reveals age-related compensatory activity moderated by genetic risk for Alzheimer's disease.

The apolipoprotein-E (APOE) ε4 allele is the greatest genetic risk factor for late-onset Alzheimer's disease (AD), but alone it is not sufficiently predictive. Because neuropathological changes associated with AD begin decades before cognitive symptoms, neuroimaging of healthy, cognitively intact ε4 carriers (ε4+) may enable early characterization of patterns associated with risk for future decline. Research in the cerebral cortex highlights a period of compensatory recruitment in elders and ε4+, which serves to maintain cognitive functioning. Yet, AD-related changes may occur even earlier in

Exciting the social butterfly: Anodal cerebellar transcranial direct current stimulation modulates neural activation during predictive social mentalizing.

Transcranial Direct Current Stimulation (tDCS) has emerged as a promising tool for enhancing social cognition. The posterior cerebellum, which is part of the mentalizing network, has been implicated in social processes. In our combined tDCS-fMRI study, we investigated the effects of offline anodal cerebellar tDCS on activation in the cerebellum during social action prediction. Forty-one participants were randomly assigned to receive either anodal (2 mA) or sham (0 mA) stimulation over the midline of the posterior cerebellum for 20 min. Twenty minutes post stimulation, participants underwent a

Localization of orofacial representation in the corona radiata, internal capsule and cerebral peduncle in Macaca mulatta.

Subcortical white matter injury is often accompanied by orofacial motor dysfunction, but little is known about the structural substrates accounting for these common neurological deficits. We studied the trajectory of the corticobulbar projection from the orofacial region of the primary (M1), ventrolateral (LPMCv), supplementary (M2), rostral cingulate (M3) and caudal cingulate (M4) motor regions through the corona radiata (CR), internal capsule (IC) and crus cerebri of the cerebral peduncle (ccCP). In the CR each pathway was segregated. Medial motor area fibers (M2/M3/M4) arched over the cauda

Human cerebellar microstructure with 7T MRI: Spherical b-tensor encoding and super resolution reconstruction.

Cerebellar function and structure remain less studied compared with the cerebrum, and while high-resolution 7T gives superior anatomical information, methods to visualise its microstructure in vivo are not widely available. However, improved understanding of cerebellar microstructure is highly clinically relevant: the granular layer of the cerebellum, located in the grey matter, contains the majority of the brain's neurons and has been shown in post mortem studies to be affected by various diseases. The limited spatial resolution of MR does not resolve individual cell layers, but advanced diff

Distinct and shared intrinsic resting-state functional networks in children with idiopathic autism spectrum disorder and fragile X syndrome.

Autism spectrum disorder (ASD) and fragile X syndrome (FXS) are behaviorally overlapped. However, little is known about the functional patterns underlying the cognitive and behavioral characteristics of FXS and ASD. The present study aimed to identify the distinct or/and shared functional networks in young children with FXS and idiopathic ASD. We recruited 150 children consecutively in a group with FXS, a group with idiopathic ASD, and a group with typically developing (TD) children. Resting-state functional magnetic resonance imaging (fMRI) and behavioral data were collected and genetic infor

Technical Modification of Amygdalo-Hippocampectomy in Temporal Lobe Epilepsy Surgery to Further Reduce Severe Neurological Complications: A Clinical-Anatomical Study.

BACKGROUND: Temporal lobe resection (TLR) including amygdalohippocampectomy (AHE) is the most frequent performed procedure in epilepsy surgery. Owing to the close anatomic relationship of the mesial temporal structures and the midbrain and choroidal fissure, the incidence of severe complications, such as postoperative stroke, is as high as 2.5%. METHODS: We developed a modification of the classical technique for AHE that involves entering the choroidal fissure early via the anterior part of the frontomesial temporal horn cleft to identify the crus cerebri, posterior cerebri artery, and oculom

Lobule-Specific Dosage Considerations for Cerebellar Transcranial Direct Current Stimulation During Healthy Aging: A Computational Modeling Study Using Age-Specific Magnetic Resonance Imaging Templates.

OBJECTIVE: Aging is associated with a decline in cognitive and motor performances, which are a part of geriatric syndromes. Since aging is associated with morphological changes in the cerebellum and cerebellar morphology is a good predictor of cognitive and motor performances, so the study of cerebellar role in age-related decline in performance is necessary. Cerebellar transcranial direct current stimulation (ctDCS) has been proposed to study and facilitate the cerebellar function. However, lobule-specific dosing has not been investigated in healthy aging. This is important because the same e

Silent Verb Generation during fMRI Reveals Post-Radiotherapy Alterations in Cerebellar-Cerebral Language Regions in Patients Treated for Medulloblastoma.

BACKGROUND: Medulloblastoma is the most common malignant pediatric brain tumor. Although radiotherapy improves survival, it is often associated with neurocognitive impairments affecting language and executive functions in developing brains. We investigated treatment-related changes in task-evoked activation using silent verb generation fMRI. METHODS: Thirty-one children and adolescents with medulloblastoma enrolled on SJMB12 (18 males; mean age 14.1 ± 4.7 years) completed visual and auditory noun-cued covert verb generation during fMRI before radiotherapy and again within 6 weeks after comple

Abnormal functional lateralization and cooperation in primary angle-closure glaucoma correlate with cell type-specific transcriptional signatures.

BACKGROUND: Primary angle-closure glaucoma (PACG) damages retinal ganglion cells (RGCs) and is associated with neurodegeneration. This study used resting-state functional magnetic resonance imaging (fMRI) to analyze hemispheric lateralization and cooperative functional alterations in PACG. Machine learning assessed the classification efficacy of neuroimaging indicators, while integrated transcriptomics described spatial relationships between neuroimaging changes and genes, neurotransmitters, and cell types. METHODS: Resting-state fMRI data were collected from 101 subjects (44 patients with PA

Consensus Paper: Cerebellum and Ageing.

Given the key roles of the cerebellum in motor, cognitive, and affective operations and given the decline of brain functions with aging, cerebellar circuitry is attracting the attention of the scientific community. The cerebellum plays a key role in timing aspects of both motor and cognitive operations, including for complex tasks such as spatial navigation. Anatomically, the cerebellum is connected with the basal ganglia via disynaptic loops, and it receives inputs from nearly every region in the cerebral cortex. The current leading hypothesis is that the cerebellum builds internal models and

The neurobiological underpinning of the social cognition impairments in patients with spinocerebellar ataxia type 2.

Clinical studies described emotional and social behaviour alterations in patients with cerebellar diseases, proposing a role of specific cerebello-cerebral circuits in social cognition. However, for a long time these difficulties were underestimated, and no studies have addressed the correlation between social cognition deficits and topography of the cerebellar damage. The present study aims to investigate the social cognition impairment and the neuroanatomical alterations in patients with spinocerebellar ataxia type 2 (SCA2) and to analyze their relationship. To this purpose a social cognitio

Localization of arm representation in the cerebral peduncle of the non-human primate.

Motor deficit severity and the potential for recovery in patients with brain injury depend on the integrity of descending corticofugal projections. Clinical assessment of these conditions following subtotal brain trauma requires a comprehensive understanding of the anatomical structures involved in the lesion as well as those structures that are spared. To assist in this endeavor, we investigated motor fiber organization in the crus cerebri of the cerebral peduncle (ccCP) in the rhesus monkey. Fibers originating from the arm representations of the primary (M1), supplementary (M2), rostral cing

Movement preparation and execution: differential functional activation patterns after traumatic brain injury.

Years following the insult, patients with traumatic brain injury often experience persistent motor control problems, including bimanual coordination deficits. Previous studies revealed that such deficits are related to brain structural white and grey matter abnormalities. Here, we assessed, for the first time, cerebral functional activation patterns during bimanual movement preparation and performance in patients with traumatic brain injury, using functional magnetic resonance imaging. Eighteen patients with moderate-to-severe traumatic brain injury (10 females; aged 26.3 years, standard devia

Cerebellar and basal ganglia structural connections in humans: Effect of aging and relation with memory and learning.

INTRODUCTION: The cerebellum and basal ganglia were initially considered anatomically distinct regions, each connected via thalamic relays which project to the same cerebral cortical targets, such as the motor cortex. In the last two decades, transneuronal viral transport studies in non-human primates showed bidirectional connections between the cerebellum and basal ganglia at the subcortical level, without involving the cerebral cortical motor areas. These findings have significant implications for our understanding of neurodevelopmental and neurodegenerative diseases. While these subcortical

The Cerebellar Predictions for Social Interactions: Theory of Mind Abilities in Patients With Degenerative Cerebellar Atrophy.

Recent studies have focused on the role of the cerebellum in the social domain, including in Theory of Mind (ToM). ToM, or the "mentalizing" process, is the ability to attribute mental states, such as emotion, intentions and beliefs, to others to explain and predict their behavior. It is a fundamental aspect of social cognition and crucial for social interactions, together with more automatic mechanisms, such as emotion contagion. Social cognition requires complex interactions between limbic, associative areas and subcortical structures, including the cerebellum. It has been hypothesized that

Cerebellar lesions and prism adaptation in macaque monkeys.

If a laterally displacing prism is placed in front of one eye of a person or monkey with the other eye occluded, they initially will point to one side of a target that is located directly in front of them. Normally, people and monkeys adapt easily to the displaced vision and correct their aim after a few trials. If the prism then is removed, there is a postadaptation shift in which the subject misses the target and points in the opposite direction for a few trials. We tested five Macaque monkeys for their ability to adapt to a laterally displacing prism and to show the expected postadaptation

Association Between Motor Symptoms and Brain Metabolism in Early Huntington Disease.

IMPORTANCE: Brain hypometabolism is associated with the clinical consequences of the degenerative process, but little is known about regional hypermetabolism, sometimes observed in the brain of patients with clinically manifest Huntington disease (HD). Studying the role of regional hypermetabolism is needed to better understand its interaction with the motor symptoms of the disease. OBJECTIVE: To investigate the association between brain hypometabolism and hypermetabolism with motor scores of patients with early HD. DESIGN, SETTING, AND PARTICIPANTS: This study started in 2001, and analysis

[Operative neurosurgery: personal view and historical backgrounds (4). Selective amygdalohippocampectomy SAHE].

Selective amygdalohippocampectomy SAHE has been pioneered by Yasargil et al in the mid 1970 within the scope of surgical treatment for intractable mesial temporal lobe epilepsy MTLE. In this article, the author has emphasized microsurgical points to be kept in mind in carrying out the procedure from the experience of just more than 200 surgeries performed by himself during the last 14 years. Historical backgrounds of development of this technique, necessary topographic anatomy, perioperative management and our results were also presented.
