Ceruloplasmin and regulation of transferrin iron during Neisseria meningitidis infection in mice.

The role of ceruloplasmin (ferroxidase I; EC 1.16.3.1) in iron metabolism during experimental Neisseria meningitidis infection was investigated. Plasma ceruloplasmin activity was found to increase greatly in mice during the convalescence phase of iron-controlled infection and after a plasma hypoferremia had occurred. Ceruloplasmin activity-deficient animals became hypoferremic as a result of an impaired release of iron from the reticuloendothelial system as shown by impaired return of reticuloendothelial system-processed heme iron in these mice. Hypoferremia in ceruloplasmin activity-deficient

Copper and ceruloplasmin (Cp) concentrations during the acute inflammatory process in the rat.

We have measured the concentration of copper and ceruloplasmin in serum of rats with carrageenan foot-oedema and in serum and exudate of rats with carrageenan pleurisy. Both serum copper and ceruloplasmin were found to increase in both models of inflammation. In these experimental conditions we have shown a strong positive correlation between copper and ceruloplasmin, in the serum of both normal and inflamed rats.

Circulating ceruloplasmin is an important source of copper for normal and malignant animal cells.

The relative uptake of copper from ceruloplasmin and non-ceruloplasmin plasma pools, by normal and malignant cells, was investigated in vivo and in vitro, using 64Cu and 67Cu. 1. Most of the copper administered intravenously to normal and tumor-bearing rats was removed within 1 h, a substantial portion entering the liver. There were differences in the apparent avidity of individual tissue for ceruloplasmin vs. ionic copper, but when calculated on the basis of actual microgram absorbed, all showed a preference for ceruloplasmin. 2. Appreciable amount of copper from either source were also absor

Hepatic SEL1L-HRD1 ER-associated degradation regulates systemic iron homeostasis via ceruloplasmin.

Iron homeostasis is critical for cellular and organismal function and is tightly regulated to prevent toxicity or anemia due to iron excess or deficiency, respectively. However, subcellular regulatory mechanisms of iron remain largely unexplored. Here, we report that SEL1L-HRD1 protein complex of endoplasmic reticulum (ER)-associated degradation (ERAD) in hepatocytes controls systemic iron homeostasis in a ceruloplasmin (CP)-dependent, and ER stress-independent, manner. Mice with hepatocyte-specific Sel1L deficiency exhibit altered basal iron homeostasis and are sensitized to iron deficiency w

Serum copper, ceruloplasmin and 24-h urine copper evaluations in celiac patients.

The aim of the study is to evaluate the serum copper, ceruloplasmin and 24-h urine copper levels in celiac patients. Serum copper, ceruloplasmin and 24-h urine measurements were evaluated in patients with celiac (n = 32), Crohn's (n = 25), Wilson's (n = 11) and in a healthy group (n = 35). Serum and 24-h urine zinc levels, AST, ALT, BUN, creatinine, iron, hemoglobin, hematocrit, lymphocyte, sedimentation and CRP levels were also measured. Results were evaluated statistically and significance was accepted as meaningful if P < 0.05. In celiacs, levels of urine copper were high (52 +/- 29 microg/

Hepatic copper content, urinary copper excretion, and serum ceruloplasmin in liver disease.

Increased liver copper concentration and raised serum ceruloplasmin were demonstrated in primary biliary cirrhosis and disorders of the biliary tract, and occasionally in chronic active hepatitis and cirrhosis of the liver. Eight of 13 patients with primary biliary cirrhosis had liver copper content as high as seen in patients with hepatolenticular degeneration (is greater than 250 mjg/g dry weight). Normal liver content was found in patients with acute hepatitis, steatosis of the liver, hepatic amuloidosis, haemochromatosis, and Gilbert's syndrome. The urinary copper excretion was increased (

Ceruloplasmin and neurodegeneration: A link between copper dysregulation, iron imbalance, and neuronal damage.

Ceruloplasmin (Cp) is a multifunctional multicopper oxidase principally produced in the liver and, to a lesser extent, in the central nervous system. It regulates iron homeostasis and oxidative balance, Cp oxidizes ferrous iron (Fe²⁺) to ferric iron (Fe³⁺), thereby enabling its safe integration into transferrin and averting reactive oxygen species formation. Beyond its ferroxidase activity, Cp also serves as a major copper carrier in plasma and contributes to antioxidant defense mechanisms. Although, the imbalance of ceruloplasmin has been gradually recognized as a key marker in the pathogenes

Retinoids rescue ceruloplasmin secretion and alleviate oxidative stress in Wilson's disease-specific hepatocytes.

Wilson's disease (WD) is a copper metabolic disorder caused by a defective ATP7B function. Conventional therapies cause severe side effects and significant variation in efficacy, according to cohort studies. Thus, exploring new therapeutic approaches to prevent progression to liver failure is urgent. To study the physiology and pathology of WD, immortalized cell lines and rodent WD models have been used conventionally; however, a large gap remains among different species as well as in genetic backgrounds among individuals. We generated induced pluripotent stem cells (iPSCs) from four WD patien

Interleukin-1--stimulated induction of ceruloplasmin synthesis in normal and copper-deficient rats.

Regulation of ceruloplasmin synthesis by interleukin-1 (IL-1) as influenced by dietary copper status was examined in rats. In copper-sufficient rats, ceruloplasmin oxidase activity did not peak in the serum until at least 24 h after IL-1 was given. The rates of ceruloplasmin synthesis, as measured by pulse labeling with [3H]leucine and immunoprecipitation, peaked 12 h after IL-1 and returned to basal rates by 24 h. Copper had to be given to copper-deficient rats for the IL-1 to induce oxidase activity. This activity was stimulated by copper alone in the deficient rats but not to the same level

alpha 1-Acid glycoprotein, alpha 1-antitrypsin, and ceruloplasmin in human intestinal helminthiases.

alpha 1-acid glycoprotein, alpha 1-antitrypsin, and ceruloplasmin levels were followed in 150 children from the rural tropics after arrival in the temperate zone. It is postulated that multiple intestinal helminthiases, especially ancylostomiasis, cause an acute phase type of reaction which is enhanced by immunization with T.A.B.-cholera vaccine.

Complete amino acid sequence of a histidine-rich proteolytic fragment of human ceruloplasmin.

The complete amino acid sequence has been determined for a fragment of human ceruloplasmin [ferroxidase; iron(II):oxygen oxidoreductase, EC 1.16.3.1]. The fragment (designated Cp F5) contains 159 amino acid residues and has a molecular weight of 18,650; it lacks carbohydrate, is rich in histidine, and contains one free cysteine that may be part of a copper-binding site. This fragment is present in most commercial preparations of ceruloplasmin, probably owing to proteolytic degradation, but can also be obtained by limited cleavage of single-chain ceruloplasmin with plasmin. Cp F5 probably is an

A cross-sectional assessment of the diagnostic value of serum ceruloplasmin for Wilson's disease in children.

Wilson's disease (WD) is a rare autosomal recessive disorder of copper metabolism requiring early diagnosis to prevent severe hepatic and neurological damage, particularly in children, where diagnostic challenges are pronounced. Serum ceruloplasmin (CPN) is a critical biochemical marker, yet its diagnostic accuracy and optimal cutoff value in pediatric populations need further evaluation in specific regional contexts. This study evaluates the diagnostic performance of serum CPN in diagnosing WD among children at Damascus University Children's Hospital and determines an optimal diagnostic cutof

The divergent effects of astrocyte ceruloplasmin on learning and memory function in young and old mice.

Ceruloplasmin (CP) plays an important role in maintaining iron homeostasis. Cp gene knockout (Cp-/-) mice develop a neurodegenerative disease with aging and show iron accumulation in the brain. However, iron deficiency has also been observed in 3 M Cp-/- mice. The use of systemic Cp gene knockout is insufficient to reveal specific functions for CP in the central nervous system. Considering recent discoveries that astrocytes synthetize the majority of brain CP, we generated astrocyte conditional Cp knockout (CpGfapcKO) mice, and found that iron contents decreased in the cerebral cortex and hipp
