Chlamydia trachomatis control requires a vaccine.

As the most common reported communicable disease in North America and Europe, Chlamydia trachomatis is the focus of concerted public health control efforts based on screening and treatment. Unexpectedly control efforts are accompanied by rising reinfection rates attributed in part to arresting the development of herd immunity. Shortening the duration of infection through the testing and treatment program is the root cause behind the arrested immunity hypothesis and because of this a vaccine will be essential to control efforts. Advances in Chlamydia vaccinomics have revealed the C. trachomatis

Forward genetic approaches in Chlamydia trachomatis.

Chlamydia trachomatis, the etiological agent of sexually transmitted diseases and ocular infections, remains poorly characterized due to its intractability to experimental transformation with recombinant DNA. We developed an approach to perform genetic analysis in C. trachomatis despite the lack of molecular genetic tools. Our method involves: i.) chemical mutagenesis to rapidly generate comprehensive libraries of genetically-defined mutants with distinct phenotypes; ii.) whole-genome sequencing (WGS) to map the underlying genetic lesions and to find associations between mutated gene(s) and a

Chlamydia trachomatis Infection and Photosensitive Dermatoses.

BACKGROUND: Photosensitivity to Chlamydia trachomatis has been described in almost 50% of chronic cases of lymphogranuloma venereum (LGV) caused by L1, L2 or L3 serovars. Photosensitivity in non LGV strains of C trachomatis has not been studied. We studied the association of various photosensitive dermatoses with C trachomatis infection in non LGV cases. METHODS: Sera of all the cases of photosensitivity, melasma, chronic actinic dermatitis (CAD), polymorphic light eruption (PLE), actinic prurigo (AP) and rosacea were tested for the presence of IgM, IgG and IgA antibodies to C trachomatis by

Shotgun proteomic analysis of Chlamydia trachomatis.

Chlamydiae are widespread bacterial pathogens responsible for a broad range of diseases, including sexually transmitted infections, pneumonia and trachoma. To validate the existence of hitherto hypothetical proteins predicted from recent chlamydial genome sequencing projects and to examine the patterns of expression of key components at the protein level, we have surveyed the expressed proteome of Chlamydia trachomatis strain L2. A combination of two-dimensional gel analysis, multi-dimensional protein identification (MudPIT) and nanocapillary liquid chromatography-tandem mass spectrometry allo

Modern diagnostics of Chlamydia trachomatis infections.

Chlamydia trachomatis (C. trachomatis) is the most common agent of sexually transmitted infections. The clinical spectrum of the disease ranges from urethritis to infertility in women and to trachoma. Intracellular localisation of the pathogen creates a challenge for routine diagnostics. In this review possible diagnostic tests have been presented, varying from classic cell culture analysis and serodiagnostics (Enzyme-linked Immunoassays, Indirect Immunofluorescence) to the most sophisticated nucleic acid analyses (hybridisation, Polymerase Chain Reaction, Transcription Mediated Amplification,

The aim of the research was to determine the prevalence of C. trachomatis (C.t.) conjunctivitis and urogenital infections coexistence in adults. The study was carried out on 222 patients (109 women and 113 men) with clinical signs of conjunctivitis. Conjunctival swabs were taken from all patients for evaluation and urogenital swabs as well as blood from the patients with confirmed infection. Direct diagnostics of chlamydial infections involved IF and LCR methods while serologic diagnostics (specific IgG antibodies) used EIA and indirect IF methods. The control group for conjunctival swabs incl

Chlamydia trachomatis: impact on human reproduction.

Chlamydia trachomatis infections are the most prevalent bacterial sexually transmitted infections (STI) recognized throughout the world. Worldwide, the magnitude of morbidity associated with sexually transmitted chlamydial infections is enormous. C.trachomatis is a common cause of urethritis and cervicitis, and sequelae include pelvic inflammatory disease (PID), ectopic pregnancy, tubal factor infertility, epididymitis, proctitis and reactive arthritis. The sharp worldwide increase in the incidence of PID during the past two decades has led to the secondary epidemics of tubal factor infertilit

Chlamydia trachomatis--role in tubal infertility.

The prevalence of humoural IgG and IgM antibodies to Chlamydia trachomatis was determined in 110 infertile women and compared to 87 healthy pregnant women without any known fertility problem. Overall antibodies to chlamydia were detected in 45% of infertile women. Antibodies were found in significantly more patients with tubal factor infertility (65%) than in women whose infertility was due to other causes (22%) (p less than 0.005). These findings are consistent with the hypothesis that C. trachomatis is a major cause of tubal factor infertility. In addition the prevalence of antibody in patie

Quantitative culture of endocervical Chlamydia trachomatis.

We examined the number of Chlamydia trachomatis inclusions produced in the initial passage of cell cultures of endocervical specimens from 1,231 women with positive chlamydial cultures who attended a sexually transmitted diseases clinic. Youth, white race, oral contraceptive use, and concurrent infection by Neisseria gonorrhoeae were associated with high chlamydial inclusion counts. Youth, white race, and oral contraceptive use were independent determinants of a high chlamydial inclusion count in women without concurrent gonorrhea but not in women with gonorrhea. Results of our study suggest t

Chlamydia trachomatis genital infections in Tahiti.

The rate of Chlamydia trachomatis infection was determined in three populations in Tahiti by means of a direct immunofluorescence test performed in specimens, tissue culture and detection of chlamydial antibody in serum specimens using a single-serotype indirect immunofluorescence test. Chlamydia trachomatis was recovered in 53% of 53 bar girls, 24% of 75 women attending a public maternity clinic for routine care, and 37% of 71 men attending a sexually transmitted disease clinic with acute or subacute urethritis. The presence of chlamydial antibody in a high proportion of the groups studied co

Diagnosis of Chlamydia trachomatis genitourinary infections.

The lack of a widely available diagnostic test for genital infections with Chlamydia trachomatis, coupled with their often nonspecific clinical nature, have been important factors contributing to the increasing incidence of these infections. Recent studies have more clearly defined the clinical manifestations of C. trachomatis infections, especially mucopurulent cervicitis and pelvic inflammatory disease. In addition, methods for the direct detection of chlamydial antigen in genital secretions have been developed. Although less sensitive than traditional cultural methods, these noncultural met

Serologic diagnosis of Chlamydia trachomatis infections.

The serology of various infections often caused by Chlamydia trachomatis including complications such as pelvic inflammatory disease (PID) and infertility was investigated comprehensively among diverse patient groups in a developing country using initially an indirect immunofluorescent antibody test (IFA). Any positives detected were further examined by a micro-immunofluorescence (MIF) method for the presence of type specific anti-chlamydial IgG/IgM antibodies. Conventional cell culture was carried out concurrently to compare culture results with serologic results. Among 416 patients (107 male

Chlamydia trachomatis and cervical intraepithelial neoplasia.

57 patients with viral condylomata of the uterine cervix and/or cervical intraepithelial neoplasia (CIN) were examined evaluating the presence of Chlamydia Trachomatis (Ct) cervical infection and serum IgG anti-Ct. The prevalence of Ct cervical infection was globally 7.22%, in CIN 3 group the rate was higher (16.67%). In addition, in this group the rate of IgG anti-Ct was significantly higher (66.67% vs 11.76%; p = 0.01) in comparison to the whole group. The role of the association between Human Papillomaviruses and Ct infection was finally discussed.

Chlamydia trachomatis serology in ankylosing spondylitis.

Demonstration of chlamydial antibodies in patients with ankylosing spondylitis (AS) could show an etiological role of Chlamydia trachomatis in this condition. We studied serum specimens from 50 HLA-B27 positive patients with AS (Group I), 34 HLA-B27 positive patients with other rheumatic diseases (Group II), 67 HLA-B27 positive healthy blood donors (Group III) and 37 healthy untyped blood donors. Chlamydia trachomatis infection and pregnancy outcome.

Chlamydia trachomatis is now recognized as the most common sexually transmitted disease organism in the United States. Although the potential for vertical transmission of C. trachomatis from pregnant women to their infants is well established, the extent to which infection adversely affects pregnancy and causes perinatal complications remains controversial. We report herein the results of a prospective study of 270 pregnant women with endocervical C. trachomatis compared with 270 matched control subjects (age +/- 1 year, race, and socioeconomic status). Among the entire group (n = 540), the ra
