The Entry Blocker Peptide Produced in Chlamydomonas reinhardtii Inhibits Influenza Viral Replication in vitro.

This year, a respiratory virus caused an emergency pandemic alert in health services around the world, showing the need for biotechnological approaches to fight these diseases. The influenza virus is one of the main viral agents that generate pandemic outbreaks. Currently, the majority of co-circulating influenza A virus (IAV) strains are adamantine- and oseltamivir-resistant strains, and the challenge is to find new antivirals for more efficient treatments. The antiviral entry blocker (EB) peptide is a promising candidate for blocking the virus entry into cells. The aim of this research was t

Controlling the development of the dengue vector Aedes aegypti using HR3 RNAi transgenic Chlamydomonas.

The Aedes aegypti mosquito plays an important role in the spread of diseases, including epidemic ones, such as dengue fever, Zika virus disease, yellow fever, and chikungunya disease. To control the population of Ae.aegypti, we transferred an HR3 RNAi fragment into the microalgae Chlamydomonas, which serves as food for Ae.aegypti larvae. Results showed that the HR3 RNAi transgenic algal strains were lethal to Ae.aegypti. The integumentary system of larvae fed with HR3 RNAi transgenic algal strains was severely damaged. Muscles of the larvae were unevenly distributed and disordered, and their m

Towards early-warning gene signature of Chlamydomonas reinhardtii exposed to Hg-containing complex media.

The potential of using gene expression signature as a biomarker of toxicants exposure was explored in the microalga Chlamydomonas reinhardtii exposed 2 h to mercury (Hg) as inorganic mercury (IHg) and methyl mercury (MeHg) in presence of copper (Cu) and Suwannee River Humic Acid (SRHA). Total cellular Hg (THg = IHg + MeHg) decreased in presence of SRHA for 0.7 nM IHg and 0.4 nM MeHg, but increased for 70 nM IHg exposure. In mixtures of IHg + MeHg and (IHg or MeHg) + Cu, SRHA decreased THg uptake, except for 0.7 nM IHg + 0.4 nM MeHg which was unchanged (p-value>0.05). In the absence of SRHA, 0.

Discovery of CRR1-targeted copper deficiency response in Chlamydomonas reinhardtii exposed to silver nanoparticles.

The molecular mechanisms behind the adaptive responses for interactions between organisms and nanoparticles, such as silver nanoparticles (AgNPs), are of great concern. In this study, the transcriptome of freshwater alga Chlamydomonas reinhardtii was characterized via RNA sequencing (RNA-seq) after exposure to a nontoxic concentration of AgNPs (0.5 mg/L). The cytochrome c6 (CYC6) and ferredoxin-5 (FDX5) genes were identified with the greatest increase in expression level, which were indications of the copper deficiency in the algae. Gene set enrichment analysis also showed significant enrichme

Genome-based approaches to understanding phosphorus deprivation responses and PSR1 control in Chlamydomonas reinhardtii.

The Chlamydomonas reinhardtii transcription factor PSR1 is required for the control of activities involved in scavenging phosphate from the environment during periods of phosphorus limitation. Increased scavenging activity reflects the development of high-affinity phosphate transport and the expression of extracellular phosphatases that can cleave phosphate from organic compounds in the environment. A comparison of gene expression patterns using microarray analyses and quantitative PCRs with wild-type and psr1 mutant cells deprived of phosphorus has revealed that PSR1 also controls genes encod

Field strains of the unicellular alga Chlamydomonas reinhardtii exhibit multicellular characteristics that shape their interactions.

Chlamydomonas reinhardtii is a unicellular green alga long studied as a biological model system but rarely considered from the perspective of its own ecology, thus epitomizing the disconnection between reductionist biology in the laboratory and life in nature. Here, we present insights into its ecology, understood from field strains. We examined bacterial communities that coenriched with C. reinhardtii from the field, revealing specific associations. We then compared the biology of C. reinhardtii field strains to laboratory strains, illuminating strain-level heterogeneity and adaptations to li

The Roles of Cullins E3 Ubiquitin Ligases in the Lipid Biosynthesis of the Green Microalgae Chlamydomonas reinhardtii.

Microalgae-based biodiesel production has many advantages over crude oil extraction and refinement, thus attracting more and more concern. Protein ubiquitination is a crucial mechanism in eukaryotes to regulate physiological responses and cell development, which is highly related to algal biodiesel production. Cullins as the molecular base of cullin-RING E3 ubiquitin ligases (CRLs), which are the largest known class of ubiquitin ligases, control the life activities of eukaryotic cells. Here, three cullins (CrCULs) in the green microalgae Chlamydomonas reinhardtii were identified and characteri

Manganese co-localizes with calcium and phosphorus in Chlamydomonas acidocalcisomes and is mobilized in manganese-deficient conditions.

Exposing cells to excess metal concentrations well beyond the cellular quota is a powerful tool for understanding the molecular mechanisms of metal homeostasis. Such improved understanding may enable bioengineering of organisms with improved nutrition and bioremediation capacity. We report here that Chlamydomonas reinhardtii can accumulate manganese (Mn) in proportion to extracellular supply, up to 30-fold greater than its typical quota and with remarkable tolerance. As visualized by X-ray fluorescence microscopy and nanoscale secondary ion MS (nanoSIMS), Mn largely co-localizes with phosphoru

A flavin binding cryptochrome photoreceptor responds to both blue and red light in Chlamydomonas reinhardtii.

Cryptochromes are flavoproteins that act as sensory blue light receptors in insects, plants, fungi, and bacteria. We have investigated a cryptochrome from the green alga Chlamydomonas reinhardtii with sequence homology to animal cryptochromes and (6-4) photolyases. In response to blue and red light exposure, this animal-like cryptochrome (aCRY) alters the light-dependent expression of various genes encoding proteins involved in chlorophyll and carotenoid biosynthesis, light-harvesting complexes, nitrogen metabolism, cell cycle control, and the circadian clock. Additionally, exposure to yellow

Bioaccumulation and effects of CdTe/CdS quantum dots on Chlamydomonas reinhardtii - nanoparticles or the free ions?

In order to properly assess the environmental risk of engineered nanoparticles (ENP), it is necessary to determine their fate (including dissolution, aggregation, and bioaccumulation) under representative environmental conditions. CdTe/CdS quantum dots (QD), such as those used in medical imaging, are known to release Cd(2+) due (mainly) to the dissolution of their outer shell. In this study, Chlamydomonas reinhardtii was exposed to either a soluble Cd salt or QD at similar concentrations of total Cd. Free Cd concentrations were measured using the Absence of Gradients and Nernstian Equilibrium

Coordinate copper- and oxygen-responsive Cyc6 and Cpx1 expression in Chlamydomonas is mediated by the same element.

Chlamydomonas reinhardtii activates the transcription of the Cyc6 and the Cpx1 genes (encoding cytochrome c(6) and coprogen oxidase) in response to copper deficiency. Mutational analysis of promoter regions of the Cyc6 and Cpx1 genes revealed a four nucleotide sequence, GTAC, which was absolutely essential for copper responsiveness. The Cyc6 promoter contains two copper response elements, each with a functionally important GTAC sequence, whereas the Cpx1 promoter contains only one. This may contribute to the stronger and more tightly regulated expression of the Cyc6 gene. Mutation or deletion

Studies on the photobiological activity of two naturally occurring furochromones, visnagin and khellin, in Chlamydomonas reinhardtii.

Irradiation of arg-1 cells of the green alga Chlamydomonas reinhardtii with UV-A in the presence of visnagin (10 microg/ml) produced weak mutagenic effects when a fluence rate of 5.1 W/m2 and fluences of 1.5-36.7 kJ/m2 were applied. A maximum number of revertants was obtained at approximately 9.2 kJ/m2. When a fluence rate of 20.4 W/m2 was used the photomutagenicity of visnagin was markedly enhanced with fluences of > or = 36.7 kJ/m2. In survival experiments with a fluence rate of 5.1 W/m2 the surviving fraction decreased continuously to approximately 4%. In experiments with a fluence rate of

Base-resolution profiling of 5-glyceryl-methylcytosine in Chlamydomonas reinhardtii via deaminase-assisted sequencing.

5gmC (5-glyceryl-methylcytosine), a vitamin C-derived hypermodified base, has been identified in the genome of Chlamydomonas reinhardtii. However, the global distribution of 5gmC and its role as an epigenetic mark remain poorly understood. In this study, we employed a DNA deaminase to distinguish 5gmC from 5mC (5-methylcytosine) and cytosines, enabling precise profiling of 5gmC across the genome. This deaminase-assisted sequencing demonstrates superior performance compared to the previously proposed TET-coupled bisulfite sequencing. Using both methods, we identified numerous confident 5gmC sit

Hydrogen Sulfide Signaling Protects Chlamydomonas reinhardtii Against Allelopathic Damage From Cyanobacterial Toxin Microcystin-LR.

Cyanobacterial blooms have become more frequent and serious in recent years. Not only do massive blooms cause environmental pollution and nutrient eutrophication, but they also produce microcystins (MCs), a group of toxic cycloheptapeptides, which threaten aquatic ecosystem and human health. As such, clarifying the allelopathic interactions between cyanobacteria and other algae is critical to better understand the driving factors of blooms. To date, however, such studies remain largely insufficient. Here, we treated model alga Chlamydomonas reinhardtii with microcystin-LR (MC-LR) to determine

Chlamydomonas reinhardtii PsbS Protein Is Functional and Accumulates Rapidly and Transiently under High Light.

Photosynthetic organisms must respond to excess light in order to avoid photo-oxidative stress. In plants and green algae the fastest response to high light is non-photochemical quenching (NPQ), a process that allows the safe dissipation of the excess energy as heat. This phenomenon is triggered by the low luminal pH generated by photosynthetic electron transport. In vascular plants the main sensor of the low pH is the PsbS protein, while in the green alga Chlamydomonas reinhardtii LhcSR proteins appear to be exclusively responsible for this role. Interestingly, Chlamydomonas also possesses tw

Systems biology approach in Chlamydomonas reveals connections between copper nutrition and multiple metabolic steps.

In this work, we query the Chlamydomonas reinhardtii copper regulon at a whole-genome level. Our RNA-Seq data simulation and analysis pipeline validated a 2-fold cutoff and 10 RPKM (reads per kilobase of mappable length per million mapped reads) (~1 mRNA per cell) to reveal 63 CRR1 targets plus another 86 copper-responsive genes. Proteomic and immunoblot analyses captured 25% of the corresponding proteins, whose abundance was also dependent on copper nutrition, validating transcriptional regulation as a major control mechanism for copper signaling in Chlamydomonas. The impact of copper deficie

Adenosine 3',5'-cyclic monophosphate in Chlamydomonas reinhardtii. Influence on flagellar function and regeneration.

Adenosine 3',5'-cyclic monophosphate (cAMP) influences both flagellar function and flagellar regeneration in Chlamydomonas reinhardtii. The methylxanthine, aminophylline, which can cause a tenfold increase in cAMP level in C. reinhardtii, inhibits flagellar movement and flagellar regeneration by wild-type cells, without inhibiting cell multiplication. Caffeine, a closely related inhibitor, also inhibits flagellar movement and regeneration, but it inhibits cell multiplication too. Regeneration by a mutant lacking the central pair of flagellar microtubules was found to be more sensitive than wil

Polystyrene nanoplastics alter cadmium toxicity outcomes in Chlamydomonas reinhardtii: Physiological responses and multi-omics insights.

Cadmium (Cd) and micro/nanoplastics frequently coexist in aquatic environments, yet how plastic particles modulate heavy metal toxicity in microalgae remains poorly understood. Here, the freshwater microalga Chlamydomonas reinhardtii was employed to investigate the interaction between polystyrene nanoplastics (PS-NPs) and Cd at physiological, biochemical, ultrastructural, and molecular levels. Among the tested particle sizes, 500 nm PS-NPs exhibited the strongest observed growth inhibition under Cd co-exposure and were therefore selected for subsequent mechanistic investigation. Cd-NPs co-expo

Recombinant production of human papillomavirus type 16 E6 and E7 vaccine antigens in Chlamydomonas reinhardtii.

Cervical cancer remains a major health challenge in low- and middle-income countries, where access to prevention and treatment is limited. Persistent infection with high-risk human papillomavirus (HPV), particularly HPV16, drives malignant transformation through the continuous expression of the viral oncoproteins E6 and E7. These tumor-specific antigens are promising targets for therapeutic vaccine development, but scalable and affordable production systems for recombinant HPV antigens are still lacking. This study establishes Chlamydomonas reinhardtii as a microalgal platform for producing HP

5' UTR introns reduce nuclear transgene silencing and enable robust protein expression in Chlamydomonas reinhardtii.

While Chlamydomonas is a widely used model organism, nuclear transgene expression in this system is well documented to face notorious difficulties. Here, we show that 5' untranslated region (UTR) introns can play a positive role in reducing gene silencing. By screening promoter-5' UTR complexes containing introns (PUTRis) from highly expressed genes, we identify many candidates that outperform the two most widely used promoters, HR and PSAD. Truncation analyses reveal that intron removal increases gene silencing to a similar extent as promoter deletion. These 5' UTR introns promote open chroma
