In alpha-chloralose anaesthetized spinal cats, Met-enkephalin amide (M-ENKA) and substance P were administered electrophoretically in the substantia gelatinosa while studying the excitation of deeper spinal neurones by noxious and non-noxious cutaneous stimuli. At sites where Met-enkephalin selectively reduced excitation by noxious skin stimuli, substance P was without effect. This result does not support the hypothesis that enkephalins are released at axo-axonic synapses on the terminals of substance P releasing primary afferent fibres.

In dogs anaesthetized with chloralose, cardiac receptors were stimulated by injections of veratridine into the aortic root and the effects studied on resistance and capacitance in the vascularly isolated and perfused abdominal circulation. Unless carotid sinus pressure was high, veratridine injections consistently resulted in dilatation of resistance and capacitance vessels. However, at high carotid pressures, the resistance response to veratridine was inhibited and the capacitance response was usually reversed. These results indicate that stimulation of cardiac receptors can affect resistance

Reactions of neurons without background activity ("silent" neurons) on long-lasting stimulation of mesenteric nerves were studied in cats anesthetized with chloralose 65-70 mg/kg) and paralyzed with flaxedile. It is shown that the "silent" neurons of the parietal and first projections areas are characterized by habituation during long-lasting stimulation of visceral nerves. Habituation developed faster in the parietal cortex and in the focus of maximal activity of the first somatosensory areas as compared to the second somatosensory area. The particularities of neuronal habituation in the cort

Two types of vagal evoked potentials were recorded in chloralose-anesthetized rats: a positive-negative one and a negative-positive one in granular and agranular insular cortical subareas, respectively, the former being more susceptible to barbiturate. Both potentials consisted of two subtypes which were distinct as to threshold and latency. Peripheral conduction velocities as well as thresholds of these two subtypes corresponded to myelinated and non-myelinated primary afferents, respectively. Thus, these two primary vagal afferents are probably both represented in two insular cortical subare

1. In cats anaesthetized with chloralose, perfusion of tubocurarine through the anterior horn of a lateral cerebral ventricle produces a rhythmic discharge of high voltage negative spikes recorded from an electrode inserted into this horn. The discharge spreads to the cerebral cortex where it gives rise to synchronous surface negative deflexions of low voltage. They have previously been described as ;slow waves'.2. The discharge results from an excitatory action of tubocurarine on the anterior limbic area, which is a cortical structure in the medial wall of the anterior horn lying rostral to t

Previous data have shown that apomorphine-induced respiratory depression can be reversed by the opiate antagonist, naloxone. The present study investigates the influence of naloxone on cardiovascular changes and vomiting elicited by apomorphine in dogs. In chloralose-anaesthetized animals, naloxone (0.02 mg/kg i.v.) failed to modify either the decrease in blood pressure and the biphasic changes (bradycardia followed by a long-lasting tachycardia in heart rate or the characteristics (occurrence, latency, duration) of the emesis elicited by apomorphine (200 micrograms/kg i.v.). In contrast, in c

Pentobarbitone, thiopentone and phenobarbitone in concentrations ranging from 10(-4) M to 10(-3) M reduced the contractile responses of isolated canine and feline trachealis muscle to electrical field stimulation without affecting contractile responses to acetylcholine. The order of potency was thiopentone, pentobarbitone and phenobarbitone. Experiments using carbachol and neostigmine or physostigmine suggested that the barbiturates act by preventing the release of acetylcholine from nerve endings. In vivo, pentobarbitone (2.5-20 mg/kg i.v.) produced dose-related reductions in the increase in

In conscious animals, the intravenous administration of enkephalins increases heart rate (HR) and mean systemic arterial blood pressure (MAP); however, when given during barbiturate anesthesia, enkephalins reduce HR and MAP. We have investigated the potential role of the gamma-aminobutyric acid (GABA) complex (consisting of chloride-ion channel and binding sites for GABA, benzodiazepine, and barbiturate/picrotoxin) as the site of modulation of enkephalin responses by certain anesthetic agents in our chronically instrumented dog model. In our model, methionine-enkephalin (Met5-ENK) (35 microgra

BACKGROUND: This study examined the effects of halothane on arterial pressure after central nervous system (CNS) pressor site stimulation in anesthetized cats, cats rendered unconscious by midcollicular transection, and conscious cats.
METHODS: Two anesthetized groups and two nonanesthetized groups were used. Cats were anesthetized with either alpha-chloralose and urethane or pentobarbital. Nonanesthetized groups were cats with midcollicular transections or conscious cats with chronically implanted electrodes. Stimulating electrodes were placed into vasomotor areas of the hypothalamus (HYP),

Trazodone, a non-tricyclic drug with antidepressant properties, diminished the central hypotensive response to clonidine in a dose-dependent manner. Both drugs were infused into the left vertebral artery of chloralose-anaesthetized cats. Intravenously injected trazodone did not reduce the central hypotensive effect of clonidine. The antagonism probably occurs at the level of central alpha-adrenoreceptors in the brain stem, where clonidine is the alpha-agonist and trazodone the alpha-antagonist. Such an antagonism, already known for clonidine and the tricyclic drugs also holds for non-tricyclic

In experiments on cats anesthetized with a mixture of chloralose and pentobarbital, reversible cooling of the 1st sensomotor area (SM-1) unilaterally to between 25 and 23 degree C was accompanied by a simultaneous and generalized change in the activity of other parts of the cortex. These changes were expressed either as the appearance of slow high - amplitude activity or, on the other hand, by a marked decrease in amplitude of the EEG. Warming the cooled region of the cortex was followed by restoration of the normal EEG of the other parts of the cortex. These changes in electroencephalographic

Acupuncture therapy has a positive role in the prevention and treatment of some related diseases by regulating autonomic nervous function. But it has been found in experimental researches that the modulatory effects of acupuncture on autonomic nervous system are not always consistent with each other. Because in the animal studies on the mechanism of acupuncture intervention, the anesthetic agent has to be used and definitely affects the activities of the autonomic nervous system while playing its pharmacological effects. Hence, it is very significant to explore the rational application of anes

The aim of the study was to examine sympathetic nerve activity, heart rate and blood pressure during naloxone-precipitated withdrawal reactions in morphine-dependent rats. In two groups of rats, one group conscious and the other anaesthetized with chloralose, renal sympathetic nerve activity (rSNA), heart rate (HR) and mean arterial blood pressure (MAP) were recorded before and during naloxone-precipitated abstinence. The conscious rats showed a biphasic pattern in the withdrawal responses. Initially, after small doses of naloxone, rSNA and HR increased and increased somatomotor activity inclu

Although static contraction of the hindlimb muscles of anesthetized cats is known to reflexly increase arterial pressure and heart rate, the cardiovascular effects of rhythmic contractions of these muscles is unclear. To help clarify this issue, we determined, in chloralose-anesthetized cats, the effects on arterial pressure and heart rate of rhythmically contracting the hindlimb muscles at a frequency of 5 Hz. In addition, we determined the effect of rhythmic contractions on the impulse activity of group III and IV muscle afferents whose activation is known to increase cardiovascular function

Biphalin (0.3 mg/kg) administered intravenously (i.v.) to urethane-chloralose anaesthetized rats consistently evoked apnoea, followed by breathing at subnormal respiratory rate with increased tidal volume. Mean arterial pressure and heart rate were lowered. Naloxone completely antagonized the respiratory and cardiovascular responses to biphalin. Midcervical vagotomy prevented all respiratory effects of biphalin, and nearly abolished the fall in blood pressure and attenuated bradycardia. These results indicate that mu opioid receptors distributed in areas supplied by vagal afferents (e.g. the l

Pesticide intoxications are mainly accidental with a benign course, such as ingestions of diluted fertilizers or low concentration antivitamin K rodenticides, ant-killing products or granules of molluscicides containing 5% metaldehyde. Voluntary intoxications by chloralose, strychnine, organophosphorus or organochlorine insecticides, concentrated anti-vitamin K products, herbicides such as paraquat, chlorophenoxy compounds, glyphosate or chlorates may be severe. Toxicity is due to active substances but also to solvents or surfactants included in the composition. Analysis done in a toxicology l

An extracorporeal veno-venous bypass utilizing an oxygenator to alter mixed venous blood gases was used in experiments on chloralose anesthetized cats. The gas mixture in the oxygenator was alternate 95% O2 or 95% N2 with additional 5% CO2. The produced difference of pO2 in the right ventricle (pO2v-) was 1.3 kPa. The arterial acid base status and the blood gases were not affected by this procedure. Comparing the alterations of pO2v- the higher level of pO2v- was connected with an increase of the mean arterial blood pressure, of the clearances of inulin and PAH, and of the sodium and the potas

In chloralosed cats, usually with ligated adrenals and paralysed with gallamine, the thoracic dorsal roots were electrically stimulated and gastric motor responses were recorded. Thoracic dorsal root stimulation regularly elicited gastric contractions after pretreatment with guanethidine and/or hexamethonium which were readily blocked by atropine. It was established that these gastric motor responses were due to peripheral stimulation of the dorsal root and not to spread to the ventral roots. Together with other results (Delbro & Lisander 1980) the findings suggest an antidromic activation of
