Electron-acceptor loadings affect chloroform dechlorination in a hydrogen-based membrane biofilm reactor.

Chloroform (CF) can undergo reductive dechlorination to dichloromethane, chloromethane, and methane. However, competition for hydrogen (H2 ), the electron-donor substrate, may cause poor dechlorination when multiple electron acceptors are present. Common acceptors in anaerobic environments are nitrate (NO3- ), sulfate (SO42- ), and bicarbonate (HCO3- ). We evaluated CF dechlorination in the presence of HCO3- at 1.56 e- Eq/m2 -day, then NO3- at 0.04-0.15 e- Eq/m2 -day, and finally NO3- (0.04 e- Eq/m2 -day) along with SO42- at 0.33 e- Eq/m2 -day in an H2 -based membrane biofilm reactor (MBfR). Effects of Sulfate Reduction on Trichloroethene Dechlorination by Dehalococcoides-Containing Microbial Communities.

In order to elucidate interactions between sulfate reduction and dechlorination, we systematically evaluated the effects of different concentrations of sulfate and sulfide on reductive dechlorination by isolates, constructed consortia, and enrichments containing Dehalococcoides sp. Sulfate (up to 5 mM) did not inhibit the growth or metabolism of pure cultures of the dechlorinator Dehalococcoides mccartyi 195, the sulfate reducer Desulfovibrio vulgaris Hildenborough, or the syntroph Syntrophomonas wolfei In contrast, sulfide at 5 mM exhibited inhibitory effects on growth of the sulfate reducer

Successful operation of continuous reactors at short retention times results in high-density, fast-rate Dehalococcoides dechlorinating cultures.

The discovery of Dehalococcoides mccartyi reducing perchloroethene and trichloroethene (TCE) to ethene was a key landmark for bioremediation applications at contaminated sites. D. mccartyi-containing cultures are typically grown in batch-fed reactors. On the other hand, continuous cultivation of these microorganisms has been described only at long hydraulic retention times (HRTs). We report the cultivation of a representative D. mccartyi-containing culture in continuous stirred-tank reactors (CSTRs) at a short, 3-d HRT, using TCE as the electron acceptor. We successfully operated 3-d HRT CSTRs

Mercury geochemistry and microbial diversity in meromictic Glacier Lake, Jamesville, NY.

Meromictic lakes are stratified lakes that typically stimulate phototrophic anoxic microbial metabolism, including the transformation of sulphur. Less studied are the transformations of mercury in these environments, and the microorganisms, which mediate these reactions. In order to further an understanding of redox species, mercury and microbial populations in meromictic lakes, we examined the geochemistry and microbiology of Glacier Lake in Jamesville, NY. We found an anoxic transition at a depth of 6 m, followed by active nitrate and sulphate utilization. A chlorophyll a maximum was located

Ferrous-driven microbial synergy enhances reductive dechlorination under sulfide stress.

Chlorinated ethenes (CEs), such as trichloroethene (TCE), frequently co-occur with elevated sulfate in contaminated aquifers, where sulfide produced by sulfate reduction strongly inhibits Dehalococcoides mccartyi (Dhc), the key organohalide-respiring bacterium. In this study, we demonstrate that sulfide is far more toxic to Dhc-mediated TCE dechlorination than sulfate, with activity nearly abolished at concentrations above 5 mM. To overcome this bottleneck, we employed a synergistic system combining Dhc strain CWV2 and the sulfate-reducing bacterium Nitratidesulfovibrio liaohensis strain KPS w

Growth of Dehalococcoides mccartyi species in an autotrophic consortium producing limited acetate.

The dechlorinating Dehalococcoides mccartyi species requires acetate as carbon source, but little is known on its growth under acetate limiting conditions. In this study, we observed growth and dechlorination of a D. mccartyi-containing mixed consortium in a fixed-carbon-free medium with trichloroethene in the aqueous phase and H2/CO2 in the headspace. Around 4 mM formate was produced by day 40, while acetate was constantly below 0.05 mM. Microbial community analysis of the consortium revealed dominance by D. mccartyi and Desulfovibrio sp. (57 and 22% 16S rRNA gene copies, respectively). From

Multi-method assessment of the intrinsic biodegradation potential of an aquifer contaminated with chlorinated ethenes at an industrial area in Barcelona (Spain).

The bioremediation potential of an aquifer contaminated with tetrachloroethene (PCE) was assessed by combining hydrogeochemical data of the site, microcosm studies, metabolites concentrations, compound specific-stable carbon isotope analysis and the identification of selected reductive dechlorination biomarker genes. The characterization of the site through 10 monitoring wells evidenced that leaked PCE was transformed to TCE and cis-DCE via hydrogenolysis. Carbon isotopic mass balance of chlorinated ethenes pointed to two distinct sources of contamination and discarded relevant alternate degra

Reductive dechlorination of tetrachloroethene in marine sediments: Biodiversity and dehalorespiring capabilities of the indigenous microbes.

Chlorinated compounds pose environmental concerns due to their toxicity and wide distribution in several matrices. Microorganisms specialized in leading anaerobic reductive dechlorination (RD) processes, including Dehalococcoides mccartyi (Dhc), are able to reduce chlorinated compounds to harmless products or to less toxic forms. Here we report the first detailed study dealing with the RD potential of heavy polluted marine sediment by evaluating the biodegradation kinetics together with the composition, dynamics and activity of indigenous microbial population. A microcosm study was conducted u

Dechlorination and organohalide-respiring bacteria dynamics in sediment samples of the Yangtze Three Gorges Reservoir.

Several groups of bacteria such as Dehalococcoides spp., Dehalobacter spp., Desulfomonile spp., Desulfuromonas spp., or Desulfitobacterium spp. are able to dehalogenate chlorinated pollutants such as chloroethenes, chlorobenzenes, or polychlorinated biphenyls under anaerobic conditions. In order to assess the dechlorination potential in Yangtze sediment samples, the presence and activity of the reductively dechlorinating bacteria were studied in anaerobic batch tests. Temperature dependence of anaerobic TCE-dechlorination in a highly enriched Dehalococcoides-containing culture.

Temperature dependence of reductive trichloroethene (TCE) dechlorination was investigated in an enrichment culture (KB-1), using lactate or propionate as electron donors at a temperature interval from 4 to 60 degrees C. Dechlorination was complete to ethene at temperatures between 10 and 30 degrees C (lactate-amended) and between 15 and 30 degrees C (propionate-amended). Dechlorination stalled at cis-1,2-dichloroethene (cDCE) at 4 degrees C (lactate-amended), at and below 10 degrees C (propionate-amended), and at 40 degrees C with both electron donors. No dechlorination of TCE was observed at

Anaerobic transformation of tetrachloroethane, perchloroethylene, and their mixtures by mixed-cultures enriched from contaminated soils and sediments.

The focus of this research was to investigate the anaerobic transformation of tetrachloroethane (TeCA), perchloroethylene (PCE), and their mixtures by mixed cultures enriched from contaminated soils or sediments. Batch transformation studies were conducted using TeCA (60 microM), PCE (60 microM), or TeCA + PCE (each added at 60 microM) as electron acceptor(s) and H2 + acetate (each added at 3 mM) or butyrate (3mM) as electron donor(s). A Dehalococcoides spp.-containing, sediment-enrichment dechlorinated PCE rapidly to ethene (ETH) but slowly and incompletely dechlorinated TeCA. Moreover, when

Mitigating Tetracycline antibiotic contamination in chicken manure using ex situ fermentation system.

Excessive use of tetracycline antibiotics in poultry farming results in significant concentrations of these drugs and tetracycline resistance genes (TRGs) in chicken manure, impacting both environmental and human health. Our research represents the first investigation into the removal dynamics of chlortetracycline (CTC) and TRGs in different layers of an ex situ fermentation system (EFS) for chicken waste treatment. By pinpointing and analyzing dominant TRGs-harboring bacteria and their interactions with environmental variables, we've closed an existing knowledge gap. Findings revealed that CT

Epilithic and endolithic microorganisms and deterioration on stone church facades subject to urban pollution in a sub-tropical climate.

Weathering of two church facades in Rio de Janeiro was caused substantially by salts, mainly halite and gypsum, detected by SEM and chemical analyses, which cause physical stresses by deposition within the rock. Biofilm populations, determined by SEM and as operational taxonomic units (OTUs), degraded stone by penetration, solubilization and redeposition of minerals on their surfaces. Endolithic cyanobacteria were associated with gypsum deposits. Microbiomes were typical for high-stress environments, high salt, intense insolation, low water and low nutrients (eg halophilic Rubrobacter, Salinic

Formate: A promising electron donor to enhance trichloroethene-to-ethene dechlorination in Dehalococcoides-augmented groundwater ecosystems with minimal bacterial growth.

Various substrates have been used to stimulate habitat microbes in chloroethene-contaminated groundwater, however, the specific efficiency and minimum growth of microbes have rarely been studied. This study investigated the effects of seven substrates on trichloroethene (TCE) dechlorination by augmentation of groundwater with Dehalococcoides mccartyi NIT01 and its contribution to the microbial community. Three out of eight test groups completed dechlorination of 1 mM TCE-to-ethene in varying durations; groundwater supplemented with formate (FOR) required 78 days, whereas the microcosms with la

Bench-scale evaluation of aerosol delivery for biostimulation and bioaugmentation in the vadose zone.

Aerosol delivery was evaluated for distributing biostimulation and bioaugmentation amendments in vadose zones. This technique involves transporting amendments as micron-scale aerosol droplets in injected gas. Microcosm experiments were designed to characterize reductive dechlorination of trichloroethene (TCE) under unsaturated conditions when delivering components as aerosols. Delivering amendments and/or microbes as aqueous aerosols resulted in complete dechlorination of TCE, similar to controls operated under saturated conditions. Reductive dechlorination was achieved with manual injection o

Genomic Characterization of Dehalococcoides mccartyi Strain JNA That Reductively Dechlorinates Tetrachloroethene and Polychlorinated Biphenyls.

Dehalococcoides mccartyi strain JNA detoxifies highly chlorinated polychlorinated biphenyl (PCB) mixtures via 85 distinct dechlorination reactions, suggesting that it has great potential for PCB bioremediation. However, its genomic and functional gene information remain unknown due to extremely slow growth of strain JNA with PCBs. In this study, we used tetracholorethene (PCE) as an alternative electron acceptor to grow sufficient biomass of strain JNA for subsequent genome sequencing and functional gene identification. Analysis of the assembled draft genome (1 462 509 bp) revealed the presenc

Review of reactive kinetic models describing reductive dechlorination of chlorinated ethenes in soil and groundwater.

Reductive dechlorination is a major degradation pathway of chlorinated ethenes in anaerobic subsurface environments, and reactive kinetic models describing the degradation process are needed in fate and transport models of these contaminants. However, reductive dechlorination is a complex biological process, where many microbial populations including dechlorinating, fermentative, methanogenic, iron and sulfate reducing, interact. In this article the modeling approaches and the experimental data needed to calibrate them are reviewed, classified, and discussed. Model approaches considered includ

Effect of anthraquinone-2,6-disulfonate on the trichloroethene degradation by Dehalococcoides-containing consortium.

This paper reports the findings of an examination on the influence of anthraquinone-2,6-disulfonate (AQDS) on the degradation of trichloroethene (TCE) by Dehalococcoides-containing consortium (designated UC-1). Compared with the control, the results indicated that (i) in 100 μmol/L AQDS, TCE was rapidly degraded. More ethene was produced, while less vinyl chloride (VC) was accumulated. AQDS might improve the activity of organisms in dechlorinating populations which resulted in more ethene being accumulated in the medium; (ii) in 500 μmol/L AQDS, TCE was incompletely degraded. Presumably, 500 μ

From disturbance to resilience: early microbial community establishment in landslide soils of New Zealand.

Landslides strongly reshape mountain landscapes and initiate soil formation, yet microbial succession in newly formed soils remains poorly understood. We analysed bacterial and archaeal communities along a centennial landslide chronosequence in the Western Southern Alps of New Zealand using 16S rRNA gene amplicon sequencing and geochemical data. A core microbial community was established within ∼3 years and persisted for up to ∼100 years. Alpha diversity declined with soil age and was lowest in the oldest soils (Shannon: 4.8 ± 0.4 to 4.0 ± 0.2). Community composition showed strong and consiste

Gut microbiota disorders aggravate terbuthylazine-induced mitochondrial quality control disturbance and PANoptosis in chicken hepatocyte through gut-liver axis.

Terbuthylazine (TBA) is a widely prevalent pesticide pollutant, which is a global concern due to its environmental residual. However, the toxic mechanism of TBA have not been fully solved. Here, we explored that TBA exposure disrupts the intestinal flora and aggravated disturbance of mitochondrial quality control and PANapoptosis in hepatocytes via gut-liver axis. Our findings demonstrated that TBA exposure induced significant damage to the jejunum barrier, evidenced by a marked decrease in the expression of Occludin and ZO-1. Moreover. TBA led to intestinal microflora disorder, manifested as
