The effect of manganese on chloroplast structure and photosynthetic ability of Chlamydomonas reinhardi.

Manganese deficiency was induced in mixotrophically-grown cultures of the green alga Chlamydomonas reinhardi and the effects of this deficiency on photosynthetic activity and cell structure were investigated. Manganese-deficient cells are unable to carry out, at normal rates, photosynthetic reactions involving system II of the photosynthetic electron transport chain. Reactions requiring only system I are not inhibited. Normal system II activity returns within 2 hr in the light following addition of manganous ions to deficient cells.Significant structural alterations for deficient cells were ob

SOME PROPERTIES OF PROTOPLASMIC GELS : I. TENSION IN THE CHLOROPLAST OF SPIROGYRA.

The chloroplast of Spirogyra is a long, spirally coiled ribbon which may contract to form a short, nearly straight rod. This happens under natural conditions and it can also be produced by a variety of inorganic salts and by some organic substances. It also occurs when the chloroplast is freed by centrifugal force from the clear peripheral protoplasm which is in contact with the cellulose wall. It would therefore seem that the chloroplast may be passively stretched by the action of the clear protoplasm and hence it contracts as soon as it is set free. This contraction happens in dead as well a

Unique variegation characteristics of camellia cultivar 'Kumagai', in which chloroplasts disappear only in palisade tissue.

Previous studies reported that some types of variegation are caused by reduction of chlorophyll and infection of pathogens, and the variegated leaves showed lower photosynthetic capacity. To elucidate the mechanisms that control the variegation, we observed a variegated individual of camellia cultivar 'Kumagai' in which leaves are variegated only on the adaxial side. We found for the first time in seed plants that the amount of chloroplasts was decreased only in palisade tissue while healthy chloroplasts were maintained in spongy tissue. Variegated parts of leaves of variegated plant of 'Kumag

Chloroplasts alter their morphology and accumulate at the pathogen interface during infection by Phytophthora infestans.

Upon immune activation, chloroplasts switch off photosynthesis, produce antimicrobial compounds and associate with the nucleus through tubular extensions called stromules. Although it is well established that chloroplasts alter their position in response to light, little is known about the dynamics of chloroplast movement in response to pathogen attack. Here, we report that during infection with the Irish potato famine pathogen Phytophthora infestans, chloroplasts accumulate at the pathogen interface, associating with the specialized membrane that engulfs the pathogen haustorium. The chemical

Dengue virus tetra-epitope peptide expressed in lettuce chloroplasts for potential use in dengue diagnosis.

Dengue virus causes about 100 million cases of dengue disease per year in the world. Laboratory diagnosis is done mainly by serological techniques, which in many cases use crude virus extracts that may cause cross-reactions to other flaviviruses. These undesirable cross-reactions can be reduced or eliminated by using recombinant proteins based on restricted epitopes. Aiming to decrease flaviviral cross-reactions and non-specific interactions in dengue serological assays, a plant expression system was chosen for recombinant antigen production as a reliable and inexpensive dengue diagnostic tool

H2O2-triggered retrograde signaling from chloroplasts to nucleus plays specific role in response to stress.

Recent findings have suggested that reactive oxygen species (ROS) are important signaling molecules for regulating plant responses to abiotic and biotic stress and that there exist source- and kind-specific pathways for ROS signaling. In plant cells, a major source of ROS is chloroplasts, in which thylakoid membrane-bound ascorbate peroxidase (tAPX) plays a role in the regulation of H(2)O(2) levels. Here, to clarify the signaling function of H(2)O(2) derived from the chloroplast, we created a conditional system for producing H(2)O(2) in the organelle by chemical-dependent tAPX silencing using

Effects of the herbicide 2,4-DB and fungicide captan on reactions of mitochondria and chloroplasts.

The effects of the herbicide 4(2,4-dichlorophenoxy)butyric acid (2,4-DB) and fungicide N-(trichloromethyltio)-4-cyclohexene-1,2-dicarboximide (captan) on electron transport processes of mitochondria and chloroplasts have been investigated. Chloroplasts, isolated from spinach leaves (Spinacia oleracea L.), were treated with pesticide prior to the addition of electron acceptor and ADP. White potato (Solanum tuberosum L.) mitochondria were either incubated with pesticide before the addition of substrate, or they were treated with pesticide after the addition of substrate and ADP. Captan inhibited

Subcellular Potassium Ion-Mediated Structural Coordination Between Mesophyll Cells and Chloroplasts Determines CO2 Mesophyll Conductance.

Potassium (K) homeostasis critically orchestrates photosynthetic performance through mesophyll conductance (gm) regulation-a key bottleneck limiting CO2 assimilation under K deficiency. While K deprivation reduces gm via anatomical alterations, the mechanistic interplay between subcellular K+ distribution and chloroplast-level structural determinants remains elusive. Field experiments using two Brassica napus L. cultivars with contrasting K utilization efficiencies (KUtE) were employed to resolve how subcellular K+ dynamics regulate gm through hierarchical structural modifications. Effective plague vaccination via oral delivery of plant cells expressing F1-V antigens in chloroplasts.

The chloroplast bioreactor is an alternative to fermentation-based systems for production of vaccine antigens and biopharmaceuticals. We report here expression of the plague F1-V fusion antigen in chloroplasts. Site-specific transgene integration and homoplasmy were confirmed by PCR and Southern blotting. Mature leaves showed the highest level of transgene expression on the third day of continuous illumination, with a maximum level of 14.8% of the total soluble protein. Swiss Webster mice were primed with adjuvant-containing subcutaneous (s.c.) doses of F1-V and then boosted with either adjuva

Inhibition of electron flow and energy transduction in isolated spinach chloroplasts by the herbicide dinoseb.

The mode of action of dinoseb (2-sec-butyl-4-6-dinitrophenol) on chloroplast reactions was studied. Electron flow from water or from an artificial electron donor, diphenylcarbazide (DPC), to dichlorophenol indophenol (DCPIP) was inhibited at low concentrations of the herbicide (5--10 microM) suggesting a site for dinoseb inhibition at the oxidizing side of photosystem II (PS II). Ferricyanide photoreduction was also inhibited by dinoseb. Cyclic and non-cyclic photophosphorylation and Mg2+-ATPase activity were inhibited by dinoseb, which indicates that this herbicide also acts as an energy tran

[High-efficiency expression of a receptor-binding domain of SARS-CoV spike protein in tobacco chloroplasts].

Chloroplast-based expression system is promising for the hyper-expression of plant-derived recombinant therapeutic proteins and vaccines. To verify the feasibility of obtaining high-level expression of the SARS subunit vaccine and to provide a suitable plant-derived vaccine production platform against the severe acute respiratory syndrome coronavirus (SARS-CoV), a 193-amino acid fragment of SARS CoV spike protein receptor-binding domain (RBD), fused with the peptide vector cholera toxin B subunit (CTB), was expressed in tobacco chloroplasts. Codon-optimized CTB-RBD sequence was integrated into

Expression of Bacillus anthracis protective antigen in transgenic chloroplasts of tobacco, a non-food/feed crop.

The Centers for Disease Control (CDC) lists Bacillus anthracis as a category A agent and estimates the cost of an anthrax attack to exceed US$ 26 billion per 100,000 exposed individuals. Concerns regarding anthrax vaccine purity, a requirement for multiple injections, and a limited supply of the protective antigen (PA), underscore the urgent need for an improved vaccine. Therefore, the 83 kDa immunogenic Bacillus anthracis protective antigen was expressed in transgenic tobacco chloroplasts. The PA gene (pag) was cloned into a chloroplast vector along with the psbA regulatory signals to enhance

Effect of sulfur dioxide and sulfite on photochemical energy storage of isolated chloroplasts--a photoacoustic study.

Photoacoustic spectroscopy was used to study the effect of sulfite and SO(2) on isolated corn mesophyll chloroplasts by monitoring the photochemical energy storage. Sulfite incubation of isolated chloroplasts, either in light or in darkness, caused a decrease in photochemical energy storage. The more pronounced decrease in light indicates a light-dependent sulfite inhibitory site(s) in chloroplasts. Also diphenylcarbazide caused a partial recovery of energy storage in sulfite treated chloroplasts indicating a possible site of damage at the water oxidizing system. Although the chloroplast membr

Specific light-harvesting complexes mediate grana stacking and prevent energy spillover between photosystems in plant chloroplasts.

Land plant chloroplasts are characterized by a prominent morphological feature: the stacking of thylakoid membranes into grana, ie defining a continuous membranous structure surrounded by the stroma and folded into appressed grana and non-appressed domains, enclosing the thylakoid lumen. Photosystem II and its antenna system are concentrated, whereas Photosystem I and ATPase are located in the grana end membranes and, in the stroma, membranes connecting grana. Thylakoid stacking promotes the physical separation of Photosystems I and II, thereby ensuring a balanced flow of excitation energy bet

An Arabidopsis Protein-Flavonoid Interactome Identifies Peroxiredoxin A as a Candidate for Flavonoid Action in Chloroplasts.

The ability of phytochemicals to act as small molecule effectors of protein function is a largely overlooked dimension of plant biochemistry. This is particularly true for the ubiquitous flavonoids where, despite abundant examples of functional interactions with human proteins, biological activities in plants are primarily attributed to ROS scavenging. We used affinity capture to explore the protein interactome of the flavonoid glycoside, rutin, in Arabidopsis seedlings. Unexpectedly, the 397 high-confidence candidates included numerous proteins associated with chloroplasts, where flavonoids a

Drought stress-induced physiological mechanisms, signaling pathways and molecular response of chloroplasts in common vegetable crops.

Drought stress is one of the most adverse abiotic stresses that hinder plants' growth and productivity, threatening sustainable crop production. It impairs normal growth, disturbs water relations and reduces water-use efficiency in plants. However, plants have evolved many physiological and biochemical responses at the cellular and organism levels, in order to cope with drought stress. Photosynthesis, which is considered one of the most crucial biological processes for survival of plants, is greatly affected by drought stress. A gradual decrease in CO2 assimilation rates, reduced leaf size, st
