Statins are administered daily, but because of their long half-lives, atorvastatin and rosuvastatin can be given every other day (q.o.d.). We examined the effect of q.o.d. statin therapy in 25 patients treated with either atorvastatin (mean dose, 18.8 mg q.o.d.) or rosuvastatin (mean dose, 9.7 mg q.o.d.). Low-density lipoprotein cholesterol decreased 43%, triglycerides (mean +/- SEM) decreased 22%+/-6%, and high-density lipoprotein cholesterol increased 9%+/-6% with q.o.d. atorvastatin (n=9; p<0.05 for all). Low-density lipoprotein cholesterol decreased 28%, triglycerides decreased 15%+/-6%, a

BACKGROUND AND AIMS: Sex differences in the longitudinal evolution of low-density lipoprotein cholesterol (LDL-C) management among patients with chronic coronary syndromes (CCS) are not well known.
METHODS: In the international CLARIFY registry, which included patients with CCS, LDL-C levels were monitored annually over the 5-year follow-up period. The analysis included patients with available LDL-C measurement at baseline. Target LDL-C was set at 100 mg/dL, in line with prevailing recommendations at enrolment. Sex-specific differences in LDL-C were adjusted for age and geographical region. I

BACKGROUND AND AIM: The Portfolio Diet has demonstrated clinically meaningful reductions in LDL-C concentrations. Variations in ABCA1, ABCG8, APOA5, ANGPTL3, and APOC1 have also been associated with LDL-C and other blood lipid concentrations. The interaction between these genetic variations and the Portfolio Diet on LDL-C is unclear. Therefore, we examined whether variations in genes involved in lipid metabolism modify the association between the Portfolio Diet Score (PDS) and its components and LDL-C concentrations.
METHODS AND RESULTS: This cross-sectional analysis included 1490 young adult

BACKGROUND: Accurate estimation of low-density lipoprotein cholesterol (LDL-C) is important for guiding cholesterol-lowering therapy. Different methods currently exist to estimate LDL-C.
OBJECTIVES: This study sought to assess discordance of estimated LDL-C using the Friedewald, Sampson, and Martin/Hopkins equations.
METHODS: Electronic health record data from patients with atherosclerotic cardiovascular disease and triglyceride (TG) levels of <400 mg/dL between October 1, 2015, and June 30, 2019, were retrospectively analyzed. LDL-C was estimated using the Friedewald, Sampson, and Martin/Ho

AIMS: To investigate the relative contribution of on-treatment low-density lipoprotein (LDL) cholesterol and C-reactive protein (CRP) to the risk of recurrent stroke and transient ischemic attack (TIA) in patients with history of ischemic stroke.
METHODS: A total of 1095 patients with non-cardioembolic ischemic stroke were randomized into two groups: control and patients receiving 10 mg of pravastatin per day. After excluding 18 patients who did not have baseline CRP data, the effects of LDL cholesterol and CRP on recurrent stroke and TIA were prospectively assessed in 1077 patients.

AIM: Our previous "J-BENEFIT (Japan BEzafibrate cliNical EFfectIveness and Tolerability)" study demonstrated that bezafibrate improves blood lipid profiles and glucose control in dyslipidemic patients with diabetes. However, bezafibrate did not significantly improve low-density lipoprotein cholesterol (LDL-C), although some patients showed decreases while others showed increases in the LDL-C levels. Therefore, a subgroup analysis of the J-BENEFIT study was conducted to identify factors influencing the bezafibrate-induced changes in the LDL-C levels.
METHODS: Of the 3,316 patients in the J-BEN

Research into the management of dyslipidaemia has increasingly focussed on the importance of high-density lipoprotein cholesterol (HDL-C) levels. It is well known that low levels of HDL are associated with an increased risk of cardiovascular events. Recent data have highlighted the anti-atherogenic actions of HDL such as anti-inflammatory, anti-oxidative activities, and of course, reverse cholesterol transport. This latter observation may explain certain effects on atherosclerotic plaques, in particular the observation that plaque regression can occur when treatments augment HDL-C levels. This

BACKGROUND AND AIMS: Chronic inflammatory diseases (CID) are associated with a profound increase in cardiovascular (CV) risk resulting in reduced life expectancy. However, LDL-cholesterol is reported to be low in CID patients which is referred to as the "LDL paradoxon". The aim of the present study was to investigate whether LDL-particles in CID exhibit an increased content of the highly atherogenic small-dense LDL subfraction (sdLDL).
METHODS AND RESULTS: In this prospective, single center, observational study we enrolled 141 patients with CID (RA n = 59, inflammatory bowel disease (IBD) n =

BACKGROUND AND AIMS: Low-density lipoprotein cholesterol (LDL-C) is the primary therapeutic target for lipid lowering, yet it may incompletely reflect atherosclerotic cardiovascular disease (ASCVD) risk in patients with chronic kidney disease (CKD). Non-high-density lipoprotein cholesterol (non-HDL-C) and remnant cholesterol (RC) capture cholesterol carried by a broader spectrum of atherogenic lipoproteins and may provide complementary information for risk stratification in this population. This study aimed to compare the associations of LDL-C, non-HDL-C, and RC with ASCVD risk and to evaluate

Background: The advent of newer pharmacological agents, particularly proprotein convertase subtilisin/kexin type 9 (PCSK-9) inhibitors, in combination with conventional lipid-lowering treatments, has allowed for the significant lowering of low-density lipoprotein cholesterol (LDL-C). However, it is unclear if very low LDL-C levels achieved with the use of PCSK-9 inhibitors are associated with increased adverse events that may outweigh potential benefits. Methods: A systematic search of PubMed, Medline, and Cochrane databases was conducted from their inception to 21 February 2025, for randomize

BACKGROUND: Although guidelines recommend low-density lipoprotein cholesterol (LDL-C) to be < 70 mg/dL in patients with atherosclerotic cardiovascular disease (ASCVD), the rate of achieving this goal remains suboptimal. We sought to understand real world contemporary practice patterns of LDL-C management in patients with ASCVD, and whether LDL-C testing influenced management across US health systems.
METHODS: A retrospective cohort study utilizing electronic medical record data from five health systems participating in the CardioHealth Alliance was performed on patients with an LDL-C measurem

Background: Dyslipidaemia represents a group of disorders of lipid metabolism, characterized by either an increase or decrease in lipid particles, usually associated with triglycerides, LDL cholesterol (LDL-C) and/or HDL cholesterol (HDL-C). Most hyperlipidaemias and HDL deficiencies confer an increased cardiovascular risk, while hypolipidaemia, such as abeta or hypobetalipoproteinemia, may present different manifestations ranging from poor weight progression to neurological manifestations. The aim of this study is to present 7 cases with rare dyslipidaemias associated with low LDL or low HDL

PURPOSE: The photoreceptor outer segment is a highly specialized ciliary structure essential for phototransduction, rendering photoreceptors especially vulnerable to ciliary dysfunction. WDR34, a key component of the retrograde intraflagellar transport machinery, has been implicated in rod-cone dystrophy. However, the pathogenic mechanisms linking WDR34 deficiency to photoreceptor degeneration remain elusive. In this study we aim to investigate the in vivo function of Wdr34 in the photoreceptor cells using conditional knockout allele of Wdr34.
METHODS: We generated retina-specific Wdr34 knock

PURPOSE OF REVIEW: Acquired hypocholesterolaemia occurs more commonly than inherited hypocholesterolaemia but has received little attention in the literature. In this review, we discuss the causes and underlying mechanisms of acquired hypocholesterolaemia and its relevance to safety of therapeutically induced decreased LDL cholesterol levels.
RECENT FINDINGS: Hypocholesterolaemia is increasingly identified as cholesterol testing becomes more widespread in the assessment of cardiovascular risk. Lower therapeutic targets for LDL cholesterol are also being achieved more regularly with the introd

BACKGROUND: Coronary artery disease (CAD) is a major problem worldwide. Atherosclerosis and thrombosis underlying CAD involve multiple cell types. New and useful diagnostic markers are required. MicroRNAs (miRNAs) are a class of noncoding RNAs that posttranscriptionally regulate the gene expressions involved in various cellular processes. Endothelial dysfunction is implicated in early processes of athero-thrombosis. Thus, it was hypothesized that the level of vascular endothelium-enriched miRNAs would be altered in plasma samples of CAD patients.
METHODS: Vascular endothelium-enriched miRNA (

Proprotein convertase subtilisin/kexin type 9 (PCSK9) promotes the degradation of the LDL receptor (LDLR), thereby increasing LDL cholesterol (LDL-C) levels and elevating the risk of cardiovascular disease. Current antibodies and siRNA target and inhibit secreted PCSK9 but are clinically costly, noncatalytic in action, and induce feedback upregulation. High-affinity cyclic peptides (CP1/CP2) developed by Merck and Novartis block the PCSK9-LDLR interaction but induce compensatory PCSK9 elevation. Herein, we report cyclic peptide-based lysosome-targeting chimeras (LYTACs), LY1 and LY2, construct

AIM: Intensive low-density lipoprotein cholesterol (LDL-C) lowering is a cornerstone of secondary prevention after percutaneous coronary intervention (PCI). Although most evidence has focused on achieved LDL-C levels, the prognostic significance of relative LDL-C reduction has been less well characterized. The joint influence of achieved LDL-C levels and relative reduction in real-world PCI populations remains unclear.
METHODS: We retrospectively analyzed consecutive patients who underwent PCI between 2006 and 2016 at a single tertiary center. Relative LDL-C reduction was calculated from base
