Apoptotic pathways mobilized in microglia and neurones as a consequence of chromogranin A-induced microglial activation.

Senile plaques of Alzheimer's brain are characterized by activated microglia and immunoreactivity for the peptide chromogranin A. We have investigated the mechanisms by which chromogranin A activates microglia, producing modulators of neuronal survival. Primary cultures of rat brain-derived microglia display a reactive phenotype within 24 h of exposure to 10 nM chromogranin A, culminating in microglial death via apoptotic mechanisms mediated by interleukin-1beta converting enzyme. The signalling cascade initiated by chromogranin A triggers nitric oxide production followed by enhanced microglia

Phylogenetic study on distribution and chromogranin/secretogranin content of histamine immunoreactive elements in the gut.

The distribution of histamine-immunoreactive (HA-IR) elements and possible coexistence of chromogranin A (CgA), chromogranin B (CgB) and secretogranin II (SgII) were immunohistochemically studied in gut specimens of various vertebrate species. In fish, HA-IR cells were distributed mainly within the gastric and duodenal mucosa, and nerve fibres in the gastric myenteric plexus. Only the gastric HA-IR cells co-stored SgII. In frog specimens, HA-IR endocrine cells and nerve fibres were found in the distal stomach wall, but SgII coexisted only in the nerve fibres. In lizard, HA-IR endocrine cells w

Presence and coexistence of chromogranin A and multiple neuropeptides in Merkel cells of mammalian oral mucosa.

By the use of light microscopic (LM) immunohistochemistry, Merkel cells of the mammalian oral mucosa have been examined for the presence and coexistence of some neuropeptides and of the neuroendocrine marker chromogranin A (CG-A). Peptide and CG-A immunophenotypes of oral Merkel cells were found to vary between species and to depend on the developmental stage, as exemplarily revealed in the pig. Oral Merkel cells of adult cat, mouse and pig but not those of adult guinea pig stained for calcitonin gene-related peptide (CGRP), substance P (SP), vasoactive intestinal polypeptide (VIP) and peptide

The amino terminal sequences of bovine and human chromogranin A and secretory protein I are identical.

The amino terminal sequences of bovine and human adrenal medullary chromogranin A have been determined. Their sequences are identical and also identical to the published sequence of secretory protein I from the parathyroid gland. This data indicates that the previously published sequence of chromogranin A is incorrect at residues 2 and 19. These data confirm earlier observations of a substantial similarity between secretory protein I and chromogranin A and, in fact, strongly suggest that they are identical.

Case Report: Feline adrenal pheochromocytoma with a synaptophysin-positive, chromogranin A-negative immunophenotype.

Pheochromocytoma is a rare adrenal medullary neoplasm in cats, with limited published cases and diagnostic challenges due to variable clinical presentation. An 8-year-old neutered male domestic shorthair cat presented with severe watery diarrhea, lethargy, and anorexia. Diagnostic evaluation revealed chronic kidney disease, pyelonephritis, and a large cystic right adrenal mass identified on ultrasonography and computed tomography. Endocrine testing did not support hyperadrenocorticism or hyperaldosteronism, and functional assessment of catecholamine excess was not performed. Surgical adrenalec

Chromogranin B Protects Human Umbilical Endothelial Cells against Oxidative Stress.

Chromogranin B (CgB) is involved in the control of the cardiovascular system through the regulation of catecholamine release. Whether CgB can exert direct actions on the endothelium has not yet been clarified. Here, we aimed to investigate the effects of CgB on cell viability, mitochondrial membrane potential, reactive oxygen species (ROS), glutathione (GSH), nitric oxide (NO) release, and the cytosolic calcium concentration ([Ca2+]c) in human vascular endothelial cells (HUVECs) cultured under both physiological and peroxidative conditions. In HUVECs, experiments were conducted to establish th

Evaluating the usefulness of plasma chromogranin A measurement in cyclic ACTH-dependent Cushing's syndrome.

Cushing's syndrome, a clinical condition characterized by hypercortisolemia, exhibits distinct clinical signs and is associated with cyclic cortisol secretion in some patients. The clinical presentation of cyclic Cushing's syndrome can be ambiguous and its diagnosis is often challenging. We experienced a 72-year-old woman with cyclic ACTH-dependent Cushing's syndrome caused by a pulmonary carcinoid tumor. Diagnosis was challenging because of the extended trough periods, and the responsible lesion was initially unidentified. A subsequent follow-up computed tomography revealed a pulmonary lesion

The chromogranin A-derived peptides catestatin and vasostatin in dogs with myxomatous mitral valve disease.

BACKGROUND: The protein chromogranin A (CgA) is stored and co-released with catecholamines from the stimulated adrenal glands. Increased plasma concentrations of CgA have been shown in people with heart disease. The aim of the study was to investigate whether plasma concentrations of the CgA-derived biologically active peptides catestatin and vasostatin were associated with the severity of myxomatous mitral valve disease (MMVD) in dogs and to assess potential associations between these blood variables and dog characteristics, echocardiographic variables, heart rate (HR), blood pressure (BP) an

Increased salivary chromogranin A in women with severe negative mood states in the premenstrual phase.

The present study investigated whether salivary chromogranin A (CgA), a psychological stress marker associated with sympathetic nervous system activity, changes during the menstrual cycle in women with different degrees of premenstrual psychoemotional symptoms. Forty-five women (28.6 ± 1.3 years) with regular menstrual cycles participated in this study. Salivary CgA and cortisol were measured during the follicular and late-luteal phases. The authors used the Profile of Mood State (POMS) to assess current mood states of subjects in each menstrual phase and divided the subjects into three groups

High levels of human chromogranin A in umbilical cord plasma and amniotic fluid at parturition.

OBJECTIVE: The human placenta is a neuroendocrine organ that produces several hypothalamic and pituitary hormones that are secreted during pregnancy and parturition into maternal and fetal circulation and amniotic fluid. Human chromogranin A (CgA) is a glycoprotein mainly localized to the adrenal medulla and released in response to several stressful events. During pregnancy, intrauterine tissues express and synthesize CgA mRNA and peptide and secret it into the biologic fluids of pregnancy, so we investigated whether maternal, umbilical, and amniotic human CgA levels are affected by the stress

Determination of residues in chromogranin A-(16-40) required for inhibition of parathyroid hormone secretion.

Chromogranin A (CGA), which is cosecreted from the parathyroid gland with PTH in response to low extracellular calcium, can be processed to amino-terminal peptides that, in turn, inhibit PTH secretion. The synthetic peptide KCIVEVISDTLSKPSPMPVSKECFE [CGA-(16-40)] is active in inhibiting secretion from freshly isolated or cultured bovine parathyroid cells. Peptide analogs in which alanine is substituted for classes of residues between the two cysteines have been synthesized and tested for biological activity. Substitution of the lysine, serine, or threonine residues by alanines does not greatly

Sensitivity and specificity of a new ELISA method for determination of chromogranin A in the diagnosis of pheochromocytoma and neuroblastoma.

We have evaluated a new commercially available ELISA kit for determination of plasma chromogranin A with respect to its usefulness in the diagnosis of neuroendocrine tumors, mainly pheochromocytoma. Serum and differently anticoagulated plasmas gave different chromogranin A concentrations. Control values (n = 21) were 18.9 +/- 5.8 units/l. Chromogranin A values > 30.4 units/l (mean + 2 S.D.) were considered elevated. In 22 patients suspected of (but found not to have) pheochromocytoma and in 24 patients with renovascular hypertension, 18% were found to have elevated chromogranin A concentration

Expression of chromogranin A and B and secretoneurin immunoreactivity in neoplastic and nonneoplastic pancreatic alpha cells.

In the endocrine pancreas, chromogranins A and B as well as secretoneurin (a biologically active peptide processed endoproteolytically from secretogranin II) are most intensely expressed in alpha (glucagon) cells. We examined whether the functional status of neoplastic and nonneoplastic human alpha cells is reflected in the expression patterns of chromogranins/secretogranins. Neoplastic alpha cells were analysed immunocytochemically in six functioning glucagonomas and 37 nonfunctioning neuroendocrine tumours (29 with alpha cells) for their immunoreactivity to chromogranin A and B, as well as s

Chromogranin A as a Novel Biomarker of Irritable Bowel Syndrome in Adults: A Systematic Review and Meta-Analysis.

BACKGROUND: Irritable bowel syndrome (IBS) is a chronic condition characterized by recurring abdominal discomfort and irregular bowel movements. Currently, IBS diagnosis lacks specific radiological, biochemical, or endoscopic markers. Chromogranin A (CgA), a gastrointestinal protein, shows variation between patients with IBS and healthy controls. This study aimed to evaluate differences in CgA concentrations between these groups. METHODS: This review was conducted in 2023 using the PRISMA 2020 guidelines. All observational research was retrieved from MEDLINE, EBSCO-Host, ScienceDirect, and Pr

Diagnostic value of CSF chromogranin A to discriminate between Alzheimer's disease and dementia with Lewy bodies.

BACKGROUND: Chromogranin A (CgA) seems to be involved in the pathophysiology of different neurodegenerative pathologies such as Alzheimer's disease (AD) and dementia with Lewy Bodies (DLB). CgA is present in the aggregates of amyloid plaques and in Lewy bodies but CgA also has a function in neuroinflammatory processes via microglia. Our objective was to determine if there is a difference in the CgA concentration in the cerebrospinal fluid (CSF) of AD and DLB patients and whether the CgA concentration can discriminate between the two diseases. METHODS: Using the previously described AlphaLewyM

A surface-enhanced Raman scattering-based probe method for detecting chromogranin A in adrenal tumors.

Aim: We aim to demonstrate that a surface-enhanced Raman spectroscopy (SERS) probe can be effectively used for protein detection in adrenal tumors. Materials & methods: The SERS probe method, which uses Au@Ag core-shell nanoparticles conjugated with a CgA antibody and a SERS reporter, was applied to detect CgA in adrenal tumors. Results: Our data reveal that the results of the CgA-SERS probe method were almost identical to those of western blot and superior to those of traditional immunohistochemistry. Conclusion: This study offers a novel strategy to detect CgA in adrenal tumors and provides
