Balantioides coli is a ciliated protist that can cause dysentery in humans, pigs and nonhuman primates and may have the potential for zoonotic transmission. Its diagnosis is routinely performed through conventional parasitological techniques, and few studies have used culturing techniques to isolate it, applying molecular tools for the characterization of this protozoan. Thus, the objective of this study was to confirm B. coli diagnosis using molecular tools and to characterize the genetic variants of this parasite isolated from pigs kept on family farms in Brazil using three different culture

Scuticociliatosis, caused by ciliated protozoa in the subclass Scuticociliatia of the phylum Ciliophora, can cause fatal disease in teleost fish species. However, information on scuticociliatosis in elasmobranchs is still scarce. In this report, we describe a case of locally extensive meningoencephalitis caused by Miamiensis avidus (syn. Philasterides dicentrarchi) in a 2 yr old captive zebra shark Stegostoma fasciatum. Granulocytic meningoencephalitis was observed through histological assessment. Inflammation was confined to the ventral aspect of the brain with a large number of ciliated prot

Initiation of the innate immune response requires recognition of pathogen-associated molecular patterns by pathogen recognition receptors such as Toll-like receptors (TLRs). MyD88 adaptor-like (Mal) is an adaptor that responds to TLR activation and acts as a bridging adaptor for MyD88. In the present study, the open reading frame of Mal was identified in orange-spotted grouper (Epinephelus coioides), and named EcMal. It contained 831 bp encoding 276 aa, and was encoded by a 1299 bp DNA sequence with three exons and two introns. EcMal and the Mal sequence of other species shared different degre

As a pivotal checkpoint molecule in immune system, programmed death-ligand 1 (PD-L1) orchestrates T cell immunity by suppressing T cell proliferation/activation, promoting apoptosis/anergy/exhaustion, and inducing regulatory T cell differentiation. Although teleosts possess a PD-L1 homologue capable of executing analogous functions, its immunological mechanisms remain largely uncharacterized. Here, we characterized a PD-L1 homologue (LcPD-L1) in a commercially significant species, Larimichthys crocea. Structural analysis reveals that it possesses conserved extracellular IgV-like and IgC-like d

This study describes a co-infection of Kudoa islandica (Myxozoa) and Nucleospora cyclopteri (Microsporida) in farmed lumpfish, Cyclopterus lumpus L., in Norway. Several other parasites (Cryptocotyle sp., protozoan ciliates and Gyrodactylus sp.) were also found in gills. In June 2013, the mortality in a farmed lumpfish population increased to 65%. Lumpfish showed erratic swimming behaviour and loss of weight. At necropsy, nodules in the kidney were the only visible lesions. Histologically, all fish showed severe changes with gill inflammation and necrosis in the spleen, kidney and liver. Haemor

Sandy beaches are dynamic ecosystems where terrestrial and marine organisms coexist, resulting in diverse communities where microeukaryotes (protists, microalgae, and meiofauna) are key drivers. Understanding their diversity patterns and responses to stressors is essential for elucidating ecological processes, yet information on this subject remains limited. Herein, microeukaryotic diversity in a sandy beach in Acapulco, Guerrero, was analyzed via ITS1 metabarcoding assessing community structure changes following hurricane Otis. Overall, the number of ASVs decreased substantially from 260 befo

Plastic debris in inland waters is less studied than in marine systems, particularly regarding its interactions with biota. Despite being problematic pollutants, plastics may serve as habitats for freshwater organisms. A research gap exists regarding how plastic physical properties and season influence colonization. We addressed this by investigating macroplastics colonization by aquatic biota in a German reservoir over a full year. Plastics were categorized by type (hard, foam, soft, and film) and surface texture (porous vs. smooth), and polymer composition was determined by near-infrared spe

Cells exhibit a wide range of behaviors due to their diverse molecular systems, which have been established through the molecular evolution of individual species. Protein families generated through gene duplications may be involved in this process. The actin cytoskeleton is no exception. The majority of eukaryotes express not only actin, but also several types of actin-related proteins (Arps). The ciliate Tetrahymena thermophila possesses a species-specific Arp, named tArp, which localizes to cilia. However, the evolutionary origin of the gene and its detailed cellular functions remain unclear

Ichthyophthirius multifiliis remains a devastating ciliate parasite in global aquaculture, yet sustainable therapies are critically scarce. While many plant-derived compounds display potent in vitro efficacy, their practical application is frequently hindered by the failure of immersion treatment to reach tissue-embedded parasites. To overcome this challenge, following a screening of 20 natural compounds, we identified shikonin (SK) as a highly promising orally bioavailable candidate with a novel mechanism. In vitro, SK demonstrated potent efficacy with EC50 values of 0.24, 0.37, and 4.05 mg/L

Parasitic diseases are considered to be a major problem in olive flounder (Paralichthys olivaceus) farms in South Korea, with scuticociliate infections being especially problematic due to their year-round occurrence and difficulty in control. In this study, we collected scuticociliates from flounder showing typical symptoms of scuticociliasis, such as ulcers, hemorrhages, and necrosis, at farms in Busan and Jeju, South Korea. Four species of scuticociliates were identified: Mesanophrys carcini, Miamiensis avidus, Paralembus digitiformis, and Pseudocohnilembus persalinus, the M. carcini and P.

The concentration of immunoglobulin (Ig) E is the lowest among serum Igs, but it can induces type I hypersensitivity and plays an important role in anti-parasitic infection. The present study aimed to explore the residence characteristics of IgE+ cells in the sheep small intestine and the impact of Moniezia benedeni infection on them. The recombinant plasmids pET-28a-IgE were constructed and induced and expressed in Escherichia coli. BL21 (DE3). The rabbit anti-sheep IgE polyclonal antibody was prepared using the obtained recombinant protein as antigen. Finally, the levels of IgE+ cells in the

Nodular gill disease (NGD) is a serious proliferative gill condition that affects farmed salmonids, particularly in Europe. While the cause of NGD remains unknown (and maybe multifactorial), various amoebae are often isolated from the gills of affected fish and can in some cases be seen associated with lesions by histopathology. The present study aimed to quantify the abundance of different amoeba species directly from the gills of rainbow trout affected by NGD and healthy controls. An 18S rRNA amplicon metagenomic approach was employed to profile the diversity and abundance of micro-eukaryote

Eunice woodwardiBaird, 1869, originally described from the Ría de A Coruña (NW Iberian Peninsula), has been overlooked and never reported from the Atlantic coast of Spain after original description and the subsequent redescription of the holotype. In the present study, we revised comparatively the holotype, newly collected specimens of this species and specimens identified as Eunice vittata (Delle Chiaje, 1829) from western Mediterranean Sea. The validity of E. woodwardi is supported and previous descriptions are complemented after a throughout study of the external morphology by means of ligh

Balantioides coli (formerly Balantidium coli) is a ciliate protozoan causing balantidiasis in humans and non-human primates (NHPs), yet it remains underrecognized in zoological settings. Morphologically similar ciliates described as Buxtonella-like organisms have been reported in NHPs and are generally considered asymptomatic. In this study, we report symptomatic infection in mandrills (Mandrillus sphinx) and investigate the prevalence and genetic diversity of Balantioides spp. and Buxtonella-like ciliates across multiple NHP species housed in Polish zoological gardens. Faecal samples were col

Sessilids (Oligohymenophorea, Peritrichia, Sessilida) reportedly colonize the gut of certain "higher termites" (family Termitidae), but only a single species, Termitophrya africana from Jugositermes tuberculatus (subfamily Apicotermitinae), has been described based on a drawing. Similar ciliates were observed in other Apicotermitinae but remained unspecified. Our SSU rRNA gene-based survey of peritrich ciliates in a broad range of Termitidae recovered distinct phylotypes from several Apicotermitinae (Astalotermes, Jugositermes, and Phoxotermes), albeit only from samples collected in Cameroon a

Loxodes is one of the best ecologically characterized ciliate genera with numerous intriguing physiological abilities, including gravity-sensing organelles and nitrate respiration. However, these cells have been considered challenging to cultivate in bulk, and are poorly preserved by conventional fixatives used for fluorescence microscopy. Here we describe methods to grow and harvest Loxodes cells in bulk with liquid soil extract medium, as well as a new fixative called ZFAE (zinc sulfate, formaldehyde, acetic acid, ethanol) that can fix Loxodes cells more effectively than buffered formaldehyd

Morphological studies of Sicuophora (Syn. Wichtermania) multigranularis Xiao et al., 2002, from the rectum of the frog, Quasipaa spinosa, performed using silver impregnation and scanning electron microscopy, confirmed the following newly recognized features: (1) only one apical suture on the right surface; (2) two naked regions at the posterior end of both the left and the right side of the body. Phylogenetic analysis based on the SSU-rRNA gene showed that S. multigranularis is a sister to a clade comprising all other Clevelandellida, strongly supporting the validity of the genus Sicuophora. Ciliates can form an important link between the microbial loop and higher trophic levels primarily through consumption by copepods. This high predation pressure has resulted in a number of ciliate species developing rapid escape swimming behaviour. Several species of these escaping ciliates also possess a long contractile tail for which the functionality remains unresolved. We use high-speed video, specialized optics and novel fluid visualization tools to evaluate the role of this contractile appendage in two free-swimming ciliates, Pseudotontonia sp. and Tontonia sp., and compare the performa

The evolution of sophisticated differentiations of the gastro-intestinal tract enabled herbivorous mammals to digest dietary cellulose and hemicellulose with the aid of a complex anaerobic microbiota. Distinctive symbiotic ciliates, which are unique to this habitat, are the largest representatives of this microbial community. Analyses of a total of 484 different 18S rRNA genes show that extremely complex, but related ciliate communities can occur in the rumen of cattle, sheep, goats and red deer (301 sequences). The communities in the hindgut of equids (Equus caballus, Equus quagga), and eleph

We report that exo- and endogenous guanosine 3',5'-cyclic monophosphate (cGMP) specifically influenced the photophobic response. In behavioral experiments the slowly hydrolyzable and membrane-permeable analogs of cGMP (8-bromo-cGMP [Br-cGMP] and N6,2'-o-dibutyryl-cGMP) dramatically prolonged the time for ciliary stop response and decreased the duration of ciliary reversal in a dose-dependent manner. When analogs of adenosine 3',5'-cyclic monophosphate (cAMP) (8-bromo-cAMP or N6,2'-o-dibutyryl-cAMP) were used, no essential effects were detected on the kinetics of the photophobic response. Both
