Sex differences in glucocorticoid production are associated with increased responsiveness of the adrenal gland in females. However, the adrenal-intrinsic mechanisms that establish sexual dimorphic function remain ill defined. Glucocorticoid production is gated at the molecular level by the circadian clock, which may contribute to sexual dimorphic adrenal function. Here we examine sex differences in the adrenal gland using an optical reporter of circadian clock function. Adrenal glands were cultured from male and female Period2::Luciferase (PER2::LUC) mice to assess clock function in vitro in r

The zebrafish constitutes a powerful model organism with unique advantages for investigating the vertebrate circadian timing system and its regulation by light. In particular, the remarkably early and rapid development of the zebrafish circadian system has facilitated exploring the factors that control the onset of circadian clock function during embryogenesis. Here, we review our understanding of the molecular basis underlying functional development of the central clock in the zebrafish pineal gland. Furthermore, we examine how the directly light-entrainable clocks in zebrafish cell lines hav

An association between circadian clock function and mood regulation is well established and has been proposed as a factor in the development of mood disorders. Patients with depression or mania suffer disturbed sleep-wake cycles and altered rhythms in daily activities. Environmentally disrupted circadian rhythms increase the risk of mood disorders in the general population. However, proof that a disturbance of circadian rhythms is causally involved in the development of psychiatric disorders remains elusive. Using clock gene mutants, manipulations of sleep-wake and light-dark cycles, and brain

The circadian clock generates daily cycles of gene expression that regulate physiological processes. The liver plays an important role in xenobiotic metabolism and also has been shown to possess its own cell-based clock. The liver clock is synchronized by the master clock in the brain, and a portion of rhythmic gene expression can be driven by behavior of the organism as a whole even when the hepatic clock is suppressed. So far, however, there is relatively little evidence indicating whether the liver clock is functionally important in modulating xenobiotic metabolism. Thus, mice lacking circa

In mammals, circadian rhythms are generated by delayed negative feedback, in which period (PER1-PER3) and cryptochrome (CRY1, CRY2) proteins gradually accumulate in the nucleus to suppress the transcription of their own genes. Although the importance of nuclear import and export signals for the subcellular localization of clock proteins is well established, little is known about the dynamics of these processes as well as their importance for the generation of circadian rhythms. We show by pharmacological perturbations of oscillating cells that nuclear import and export are of crucial importanc

Although the circadian clock is a self-sustaining oscillator having a periodicity of nearly 1 d, its period length is not necessarily 24 h. Therefore, daily adjustment of the clock (i.e., resetting) is an essential mechanism for the circadian clock to adapt to daily environmental changes. One of the major cues for this resetting mechanism is light. In the unicellular green alga Chlamydomonas reinhardtii, the circadian clock is reset by blue/green and red light. However, the underlying molecular mechanisms remain largely unknown. In this study, using clock protein-luciferase fusion reporters, w

Comparative studies on cellular and molecular clock mechanisms have revealed striking similarities in the organization of the clocks among different animal groups. To gain evolutionary insight into the properties of the clock network within the Drosophila genus, we analyzed sequence identities and similarities of clock protein homologues and immunostained brains of 10 different Drosophila species using antibodies against vrille (VRI), PAR-protein domain1 (PDP1), and cryptochrome (CRY). We found that the clock network of both subgenera Sophophora and Drosophila consists of all lateral and dorsa

Circadian and seasonal rhythms are a fundamental feature of all living organisms and their organelles. Biological rhythms are responsible for daily food intake; the period of hunger and satiety is controlled by the central pacemaker, which resides in the suprachiasmatic nucleus (SCN) of the hypothalamus, and communicates with tissues via bidirectional neuronal and humoral pathways. The molecular basis for circadian timing in the gastrointestinal tract (GIT) involves interlocking transcriptional/translational feedback loops which culminate in the rhythmic expression and activity of a set of clo

This article reviews human and animal studies about the circadian patterns of physiological variables involved with the respiratory function. Some measures reflecting the mechanical properties of the lungs, such as functional residual capacity, forced expiratory volumes and airway resistance, change periodically with the time of the day. Also resting pulmonary ventilation (V(E)), tidal volume, and breathing rate follow circadian patterns. In humans, these patterns occur independently of the daily changes in activity, whereas, to some extent, they are linked to changes in the state of arousal.

Neurospora crassa (bdA) mycelia were kept in liquid culture. Without rhythmic conidiation the levels of adenine nucleotides undergo circadian changes in constant darkness. Maxima occur 12-17 hr and 33-35 hr after initiation of the rhythm, i.e., at CT 0-6 hr. Pulses of metabolic inhibitors such as vanadate (Na3Vo4), molybdate (Na2MoO4 : 2 H2O), N-ethylmaleimide (NEM), azide (NaN3), cyanide (NaCN) and oligomycin phase shift the circadian conidiation rhythm of Neurospora crassa. Maximal advance phase shifts are observed at about CT 6 with all inhibitors. Pulses of N,N'dicyclohexylcarbodiimide (DC

Misaligned feeding may lead to pancreatic insufficiency, however, whether and how it affects circadian clock in the exocrine pancreas is not known. We exposed rats to a reversed restricted feeding regimen (rRF) for 10 or 20 days and analyzed locomotor activity, daily profiles of hormone levels (insulin, glucagon, and corticosterone) in plasma, and clock gene expression in the liver and endocrine and exocrine pancreas. In addition, we monitored responses of the exocrine pancreatic clock in organotypic explants of mPer2Luc mice in real time to acetylcholine, insulin, and glucocorticoids. rRF pha

The circadian system in mammals is responsible for the temporal coordination of multiple physiological and behavioural processes that are necessary for homeostasis. In the skeleton, it has long been known that metabolic functions of chondrocytes, osteoblasts and osteoclasts exhibit intrinsic circadian rhythms. In addition, results from animal models reveal a close connection between the disruption of circadian rhythms and skeletal disorders such as rheumatoid arthritis, osteoarthritis and osteoporosis. In this review, we summarise the latest insights into the genetic and biochemical mechanisms

In mammals it has been thought that the circadian clock localizes only in the suprachiasmatic nucleus of the hypothalamus. Recent studies have revealed that certain brain regions and peripheral tissues may also have intrinsic circadian clocks. However, the roles played by 'peripheral circadian clocks' have not been fully elucidated. In this study, we investigated their function using mouse pineal glands, and found that expression of the arylalkylamine N-acetyltransferase (Aa-Nat, EC 2.3.1.87, the rate-limiting enzyme of melatonin synthesis) gene after adrenergic receptor stimulation depended o

Long-duration space missions expose astronauts to microgravity, radiation, confinement, fluid redistribution, and circadian disruption, which together induce physiological adaptations, including cardiovascular hemodynamics. Emerging evidence suggests that spaceflight may modulate hemostasis and potentially increase thrombotic risk in the head/neck; however, the association of circadian disruption and hemostasis in space, and its ground-based analogs, has yet to be systematically evaluated. This systematic review aimed to synthesize evidence from real-spaceflight missions and ground-based analo

Disruption of cellular timekeeping systems is a key driver of cancer cell development and progression. The circadian clock is a 24-hour regulatory system that relies on core clock genes - CLOCK, BMAL1, PER, and CRY. Maintaining cellular homeostasis is essential, as it regulates processes such as the cell cycle, metabolism, DNA repair, and apoptosis. Disruption of Circadian rhythms is common in cancer, altering gene expression and causing the mis-timing of cellular events. This disturbance stimulates excessive growth, genetic instability, and resistance to cell death. Circadian rhythms and tumo

Primary cell cultures of avian pinealocytes and the mammalian suprachiasmatic nucleus (SCN), immortalized cell lines derived from the SCN (SCN2.2), and fibroblasts derived from mice and rats have been employed as in vitro models to study the cellular and molecular mechanisms underlying circadian biological clocks. This article compares and contrasts these model systems and describes methods for avian pinealocyte cultures, immortalized SCN2.2 cells, and mouse fibroblast culture. Each of these culture models has advantages and disadvantages. Avian pinealocytes are photoreceptive, contain a circa

The output generated by the endogenous circadian clock to control circadian functions and temporal organization in metazoans is unknown. Redox state perturbations generated by reactive oxygen species (ROS) and antioxidants are known to influence the expression of a number of genes and signal transduction pathways. Evidence has been recently provided that the reduced redox cofactors NAD and NADP both regulate clock gene activity in the suprachiasmatic nucleus (SCN) and are induced by it. Significant periodic variations of lipidperoxidation in human blood with a dip at 04.00 h have been previous

Circadian clocks prepare the organism to cyclic environmental changes in light, temperature, or food availability. Here, we characterized the master clock in the brain of a strongly photoperiodic insect, the aphid Acyrthosiphon pisum, immunohistochemically with antibodies against A. pisum Period (PER), Drosophila melanogaster Cryptochrome (CRY1), and crab Pigment-Dispersing Hormone (PDH). The latter antibody detects all so far known PDHs and PDFs (Pigment-Dispersing Factors), which play a dominant role in the circadian system of many arthropods. We found that, under long days, PER and CRY are

According to an epidemiological survey, myocardial infarction, ischemia-reperfusion injury, and aortic rupture usually occur in the morning and summer and are characterized by more severe symptoms and poorer prognosis. The occurrence, development, treatment difficulties, and poor prognosis of ischemic heart disease are closely related to circadian rhythm disorders and the abnormal transcription of circadian clock genes. Normal circadian rhythms regulate neurohumoral regulation and energy metabolism, thereby playing a protective or modulatory role in ischemic heart disease (IHD). HIF1α links th

Thrombotic events such as myocardial infarction and stroke exhibit circadian variation with early-morning peaks, implicating intrinsic biological regulation, yet the molecular mechanisms coordinating hemostatic function remain unclear. This systematic review synthesizes evidence linking circadian rhythms and clock genes to hemostatic regulation and highlights the underlying molecular pathways. A comprehensive literature search of PubMed, Scopus, and Web of Science was conducted in accordance with Preferred Reporting Items for Systematic Reviews and Meta-Analyses guidelines.
