3D Finite Element Model of Human Ear with 3-Chamber Spiral Cochlea for Blast Wave Transmission from the Ear Canal to Cochlea.

Blast-induced auditory trauma is a common injury in military service members and veterans that leads to hearing loss. While the inner ear response to blast exposure is difficult to characterize experimentally, computational models have advanced to predict blast wave transmission from the ear canal to the cochlea; however, published models have either straight or spiral cochlea with fluid-filled two chambers. In this paper, we report the recently developed 3D finite element (FE) model of the human ear mimicking the anatomical structure of the 3-chambered cochlea. The model consists of the ear c

MECOM promotes supporting cell proliferation and differentiation in cochlea.

Permanent damage to hair cells (HCs) is the leading cause of sensory deafness. Supporting cells (SCs) are essential in the restoration of hearing in mammals because they can proliferate and differentiate to HCs. MDS1 and EVI1 complex locus ( MECOM) is vital in early development and cell differentiation and regulates the TGF-β signaling pathway to adapt to pathophysiological events, such as hematopoietic proliferation, differentiation and cells death. In addition, MECOM plays an essential role in neurogenesis and craniofacial development. However, the role of MECOM in the development of cochlea

Curcumin protects against acoustic trauma in the rat cochlea.

OBJECTIVES: In this study we evaluated the therapeutic utility of curcumin in a rodent model of acoustic trauma using histopathology, immunohistochemical, and distortion product otoacoustic emission (DPOAEs) measurements. METHODS: 28 Wistar albino rats were included in the study and randomly assigned to 4 treatment groups. The first group (group 1) served as the control and was exposed to acoustic trauma alone. Group 2 was the curcumin group. Group 3 was the curcumin plus acoustic trauma group. Group 4 was the saline plus acoustic trauma group. Otoacoustic emission measurements were collected

Sox2 induces neuronal formation in the developing mammalian cochlea.

In the cochlea, spiral ganglion neurons play a critical role in hearing as they form the relay between mechanosensory hair cells in the inner ear and cochlear nuclei in the brainstem. The proneural basic helix-loop-helix transcription factors Neurogenin1 (Neurog1) and NeuroD1 have been shown to be essential for the development of otocyst-derived inner ear sensory neurons. Here, we show neural competence of nonsensory epithelial cells in the cochlea, as ectopic expression of either Neurog1 or NeuroD1 results in the formation of neuronal cells. Since the high-mobility-group type transcription fa

Allopurinol attenuates endolymphatic hydrops in the guinea pig cochlea.

Based on the hypothesis that Ca(2+) overload in the scala media may produce endolymphatic hydrops and generate free oxygen radicals (FOR), allopurinol, a xanthine oxidase inhibitor and free radical scavenger, was administered to guinea pigs after the surgical obliteration of the endolymphatic duct. Allopurinol was given intraperitoneally (50 mg/kg/day) for 15 days starting 1 day prior to the surgical blockage procedure. Measurements from histological serial sections of these temporal bones showed that the total volume of the scala media was significantly reduced (p = 0.007) compared with contr

Hair cell loss in the aged guinea pig cochlea.

The various effects of ageing on the auditory system, collectively termed presbycusis, are being studied across a wide range of animal species, including humans. One contributing factor to presbycusis is thought to be losses of the sensory hair cells in the cochlea. In this study, hair cell counts were obtained from cochleas of pigmented guinea pigs (Cavia porcellus) at ages ranging from 11 days to 4 years 7 months, using scanning electron microscopy to visualize the organ of Corti. Representative samples of the basal, middle and apical turn of the cochlea were photographed for analysis. Hair

[The cochlea: modern physiologic insights into sensorineural hearing loss].

The cochlea is organized tonotopically: each frequency between 20 Hz and 20 kHz has its own location on the basilar membrane. In addition the cochlea has a large dynamic range (sound intensity). Active movements of the outer hair cells of the sensory organ of Corti play a crucial part in determining hearing acuity and frequency selectivity. Long-known properties such as combination tones and more recent observations like spontaneous and evoked otoacoustic emissions (the cochlea as sound source) fit in the same concept of a cochlea acting as a non-linear fine-tuned amplifier. Most sensorineural

Primary neural disorders in the deaf white cat cochlea.

Eleven white kittens were investigated from 2 days after birth up to an age of 4 months. After their hearing ability or deafness had been tested electrophysiologically, electron microscopic work was done at the level of the cochlea. Previous histological data indicated that the hereditary process of degeneration begins at the epithelial and sensory elements of the cochlea, and that the neural degeneration is only a secondary and very slow process. Results presented here indicate that, at least in some white cats, this assumption needs to be modified. Early failure in the myelination of the lam

Pax3 deficiency diminishes melanocytes in the developing mouse cochlea.

Cochlear melanocytes are intermediate cells in the stria vascularis that generate endocochlear potentials required for auditory function. Human PAX3 mutations cause Waardenburg syndrome and abnormalities of melanocytes, manifested as congenital hearing loss and hypopigmentation of skin, hair and eyes. However, the underlying mechanism of hearing loss remains unclear. During development, cochlear melanocytes in the stria vascularis are dually derived from Pax3-Cre+ melanoblasts migrating from neuroepithelial cells including neural crest cells and Plp1+ Schwann cell precursors originated from al

Imaging the Aging Cochlea with Light-Sheet Fluorescence Microscopy.

Deafness is the most common sensory impairment, affecting approximately 5% or 430 million people worldwide as per the World Health Organization1. Aging or presbycusis is a primary cause of sensorineural hearing loss and is characterized by damage to hair cells, spiral ganglion neurons (SGNs), and the stria vascularis. These structures reside within the cochlea, which has a complex, spiral-shaped anatomy of membranous tissues suspended in fluid and surrounded by bone. These properties make it technically difficult to investigate and quantify histopathological changes. To address this need, we d

Static magnetic field affects oxidative stress in mouse cochlea.

OBJECTIVE: It has been shown that oxidative stress plays an important role in development of noise induced hearing loss. Since static magnetic fields (SMF) exposure may alter dynamics of oxidative processes in the tissue, the aim of the study was to assess the influence of SMF on noise-induced alteration in the cochlear level of reactive oxygen species (ROS) and hearing thresholds. MATERIALS AND METHODS: Auditory brainstem response (ABR), lipid peroxidation (LPO) levels, super-oxide dismutase (SOD) activity and catalase activity were assessed in the cochlea prior to, and at five time-points o

Cell division in the gerbil cochlea after acoustic trauma.

The recent discovery of hair cell regeneration in the avian inner ear raises the possibility that hair cell regeneration might occur in the mammalian cochlea as well. The authors used 3H-thymidine labeling to detect mitotic activity in the cochleas of normal 3-week old gerbils exposed to acoustic trauma. Following an acoustic insult that caused progressively more severe damage in an apical to basal progression, 3H-thymidine was injected for 5 days. Control animals were not exposed to the acoustic insult. The gerbils' cochleas were sectioned and processed for autoradiography. In the control coc

Reduced active protection to the cochlea during physical exercise.

The present investigation was designed to evaluate whether dynamic physical exercise is able to modify stapedius reflex (SR) activity and, as a consequence, to influence the amount of temporary threshold shift (TTS). Experimental subjects were 10 normal-hearing male volunteers, aged 27-34 years (mean 30.8). SR parameters in the time-domain were assessed before, during and after submaximal exercise test, performed for 10 min on an ergometer cycle at 50% of maximum work capacity. The same parameters were also investigated in the absence of exercise, at time-matched intervals. In addition, the co

Muscarinic acetylcholine receptor binding in the guinea pig cochlea.

Because of the possibility that olivocochlear bundles may use acetylcholine as an inhibitory transmitter, we examined the cochlea of the guinea pig for cholinergic receptor binding sites. Binding of the potent muscarinic antagonist [3H]quinuclidinyl benzilate (3H-QNB) showed that the cochlea has sites with the pharmacological specificity of muscarinic cholinergic receptors. Specific 3H-QNB binding was saturated at 0.8 nM and was half-saturated at 0.03 nM concentrations. Total 3H-QNB binding was reduced 70% in the presence of saturating doses of acetylcholine, oxotremorine and atropine, and hal

Study on damage of the macrostructure of the cochlea under the impact load.

Due to the tiny and delicate structure of the cochlea, the auditory system is the most sensitive to explosion impact damage. After being damaged by the explosion impact wave, it usually causes long-term deafness, tinnitus, and other symptoms. To better understand the influence of impact load on the cochlea and basilar membrane (BM), a three-dimensional (3D) fluid-solid coupling finite element model was developed. This model accurately reflects the actual spatial spiral shape of the human cochlea, as well as the lymph environment and biological materials. Based on verifying the reliability of t

Rack1 regulates cellular patterning and polarity in the mouse cochlea.

Rack1 features seven WD40 repeats that fold into a multifaceted scaffold used to build signaling complexes in a context-dependent manner. Previous in vitro studies have revealed associations between Rack1 and many other proteins. Rack 1 is required for establishing planar cell polarity (PCP) in zebrafish and Xenopus. However, any molecular role of Rack1 in protein complexes or polarity regulation remains unclear. Here, we show that Rack1 is an essential gene in mice. Conditional knockout of Rack1 shortened the cochlear duct and induced cellular patterning defects characteristic of defective co

Transcriptomic characterization of dying hair cells in the avian cochlea.

Sensory hair cells are prone to apoptosis caused by various drugs including aminoglycoside antibiotics. In mammals, this vulnerability results in permanent hearing loss because lost hair cells are not regenerated. Conversely, hair cells regenerate in birds, making the avian inner ear an exquisite model for studying ototoxicity and regeneration. Here, we use single-cell RNA sequencing and trajectory analysis on control and dying hair cells after aminoglycoside treatment. Interestingly, the two major subtypes of avian cochlear hair cells, tall and short hair cells, respond differently. Dying sho
