Behavioural pharmacology of the α5-GABAA receptor antagonist S44819: Enhancement and remediation of cognitive performance in preclinical models.

Previous work has shown that S44819 is a novel GABAA receptor (GABAAR) antagonist, which is selective for extrasynaptic GABAARs incorporating the α5 subunit (α5-GABAARs). The present study reports on the preclinical neuropsychopharmacological profile of S44819. Significantly, no sedative or pro-convulsive side effects of S44819 were found at doses up to 30 mg/kg i.p. Object recognition (OR) memory in intact mice was enhanced by S44819 (0.3 mg/kg p.o.) given before the acquisition trial. Mice treated with phencyclidine for two weeks and tested six days after the cessation of treatment failed to

Enhanced cognitive performance in older adults through combined cognitive training and transcranial direct current stimulation.

Cognitive decline in older adults has significant implications for autonomy and quality of life. This study evaluated the combined effects of cognitive training (CT) and transcranial direct current stimulation (tDCS) on cognitive performance in older adults. Two CT approaches-core training and strategy training-were paired with either active or sham anodal tDCS over the left DLPFC. Eighty-five participants were assigned to three groups: (1) CT with active tDCS, (2) CT with sham tDCS, and (3) a control group attending memory-related lectures. Participants underwent 16 intervention sessions and

Enhancing Social Cognition in Mild Cognitive Impairment with Non-Invasive Brain Stimulation: A Randomized Clinical Trial.

BackgroundThe effectiveness of non-invasive neuromodulation to improve social cognition (SC) in neurological disorders remains unclear. However, repetitive transcranial magnetic stimulation (rTMS) shows promise for treating cognitive abnormalities by promoting neuroplasticity.ObjectiveIn this randomized, double-blind, sham-controlled study, we investigated the effects of high-frequency rTMS on the medial prefrontal cortex (mPFC) and right temporal parietal junction (rTPJ) in patients with mild cognitive impairment (MCI) to enhance SC abilities and other cognitive and functional abilities relat

Auditory stimulation at individual gamma frequency enhances cognitive performance.

Personalized auditory gamma stimulation may sharpen human cognition; however, behavioral evidence remains sparse. Here, we recruited 29 healthy adults (15 men; age range, 20-49 years) with no self-reported history of psychiatric disorders, and identified each participant's dominant gamma frequency (IGF), and then investigated whether auditory stimulation at that frequency-delivered through individualized IGF music-modulated cognitive performance. We first estimated each participant's IGF based on electroencephalogram during a 5‑min chirp‑music sweep. The identified frequency was embedded into

Preclinical Characterization of Vindeburnol: Pharmacokinetics, Safety, and Cognitive-Enhancing Properties.

Cognitive impairment is a common symptom of many psychiatric disorders and can significantly reduce the patients' quality of life. Vindeburnol, a synthetic derivative of eburnamine-vincamine alkaloids, has shown potential in models of depression and neurodegeneration; however, its comprehensive properties and molecular mechanisms of action have remained unexplored. We conducted a comprehensive preclinical assessment of vindeburnol, including a subchronic toxicity study (20 and 80 mg/kg/day, orally, 14 days), pharmacokinetic analysis (40 mg/kg), behavioral tests for memory and learning, and tra

Simulated visual hallucinations in virtual reality enhance cognitive flexibility.

Historically, psychedelic drugs are known to modulate cognitive flexibility, a central aspect of cognition permitting adaptation to changing environmental demands. Despite proof suggesting phenomenological similarities between artificially-induced and actual psychedelic altered perception, experimental evidence is still lacking about whether the former is also able to modulate cognitive flexibility. To address this, we measure participants' cognitive flexibility through behavioral tasks after the exposure to virtual reality panoramic videos and their hallucinatory-like counterparts generated b

Endogenous Acetylcholine and Its Modulation of Cortical Microcircuits to Enhance Cognition.

Acetylcholine regulates the cerebral cortex to sharpen sensory perception and enhance attentional focus. The cellular and circuit mechanisms of this cholinergic modulation are under active investigation in sensory and prefrontal cortex, but the universality of these mechanisms across the cerebral cortex is not clear. Anatomical maps suggest that the sensory and prefrontal cortices receive distinct cholinergic projections and have subtle differences in the expression of cholinergic receptors and the metabolic enzyme acetylcholinesterase. First, we briefly review this anatomical literature and t

Enhancing creative cognition with a rapid right-parietal neurofeedback procedure.

The present article describes an innovative neurofeedback training (NFT) procedure aimed at increasing creative cognition through the enhancement of specific brain activities previously associated with divergent thinking. We designed and tested two NFT protocols based on training alpha and beta EEG oscillations selectively measured over the right parietal region. A total of 80 participants were involved, 40 in the alpha NFT protocol and 40 in the beta NFT protocol. The NFT loop was closed on a video stream that would advance only when oscillation power exceeded a normalized threshold. The tota

Cognitive and Performance Enhancing Medication Use to Improve Performance in Poker.

Use of neuroenhancers has been studied in groups ranging from students to surgeons; however, use of cognitive and performance enhancing medications (CPEMs) to improve performance in poker has remained largely overlooked. To assess the use of CPEMs to improve poker performance, a survey of poker players was conducted. Participants were recruited via Internet poker forums; 198 completed the online survey. Approximately 28 % of respondents used prescription CPEMs, with the most commonly used including: amphetamine/dextroamphetamine (62 %), benzodiazepines (20 %), and methylphenidate (20 %). CPEMs

Cognitive remediation to enhance mobility in older adults: the CREM study.

Mobility disabilities represent the most prevalent disability among seniors. Emerging evidence indicates that executive functions play an important role in maintaining mobility. However, the use of cognitive remediation programs to enhance mobility has not been investigated in a full-scale randomized control trial. The CREM study is a single-blind randomized control trial to examine the effect of computerized cognitive remediation versus computer-based health education training on mobility in 420 seniors. The primary outcome is change in gait speed during normal walking and walking-while-talki

Neuro-cognitive performance is enhanced during short periods of microgravity.

There is increasing interest in the effects of microgravity on cognitive processing, particularly as it relates to the potential for human space travel. While findings to date are quite inconsistent, studies reporting a decrement in cognitive performance have generally not been able to distinguish between the direct influence of microgravity, and any associated influence of stress. Furthermore, the currently available findings are primarily based on behavioral observations, and there is a need to better understand the underlying neurophysiological responses. The current study aimed to determin

Traumatic brain injury decreases AMP-activated protein kinase activity and pharmacological enhancement of its activity improves cognitive outcome.

Prolonged metabolic suppression in the brain is a well-characterized secondary pathology of both experimental and clinical traumatic brain injury (TBI). AMP-activated kinase (AMPK) acts as a cellular energy sensor that, when activated, regulates various metabolic and catabolic pathways to decrease ATP consumption and increase ATP synthesis. As energy availability after TBI is suppressed, we questioned if increasing AMPK activity after TBI would improve cognitive outcome. TBI was delivered using the electromagnetic controlled cortical impact model on male Sprague-Dawley rats (275-300 g) and C57

The enhancement of video feedback by cognitive preparation in the treatment of social anxiety. A single-session experiment.

The present study replicates and extends Harvey, Clark, Ehlers and Rapee's (Behav. Res. Therapy 38 (2000) 1183-1192) single-session experiment of video feedback with cognitive preparation in individuals with high social anxiety. Forty participants high on social anxiety were asked to present a speech before a video camera and then received video feedback of their speech. Half of the participants received cognitive preparation prior to the video feedback, whereas the other half only received video feedback. The cognitive preparation procedure asked the participants to (1) predict in detail what

Resident Registered Nurse Anesthetists' Use of Cognitive Enhancers in the U.S.

Graduate students, including medical students, use cognitive enhancer (CE) medications with or without legal prescription to aid academic performance despite little evidence that they improve performance in individuals without a medical diagnosis and subsequent prescription. However, there is limited information on CE medication use, with or without legal prescription, by resident registered nurse anesthetists (RRNAs). This cross-sectional survey assessed prevalence of CE medication use with or without legal prescription and its potential predictors (depression, anxiety, Attention Deficit Hype

Enhancing Mood, Cognition, and Quality of Life in Pediatric Multiple Sclerosis.

Pediatric-onset multiple sclerosis (POMS), representing approximately 5% of all MS cases, affects the central nervous system during its ongoing development. POMS is most commonly diagnosed during adolescence but can occur in younger children as well. For pediatric patients with MS, it is critical to manage the full impact of the disease and monitor for any effects on school and social functioning. Disease management includes not only disease-modifying therapies but also strategies to optimize wellbeing. We review the interventions with the highest evidence of ability to improve the disease cou
