Cold shock, chilling and cryopreservation impact sperm proteasome function.

SUMMARY: A 38-year-old man was admitted because of transient somnolence. Five weeks previously, he had suffered a subarachnoid hemorrhage from a ruptured aneurysm of the anterior communicating artery (ACOM), which was treated by craniotomy and clipping. He had recovered well, although loss of short-term memory and a forehead paresis on the side of craniotomy persisted. Clinical examination on admission showed no new neurological deficits. Cerebral computed tomography with angiography revealed no bleeding or infarction and correctly positioned clips. Laboratory examination showed severe hyperna

StICE1 activates StCBFs expression to regulate cold stress response in potato (Solanum tuberosum L.) seedlings.

Cold stress is a major environmental factor limiting potato yield. Understanding the molecular regulatory networks that govern plant responses to low temperature is therefore crucial for enhancing crop stress tolerance through genetic approaches. In this study, we investigated the functional role and molecular mechanisms of the StICE1 gene in potato under chilling stress, aiming to provide a theoretical basis for the molecular breeding of cold-tolerant varieties. We generated both overexpression (OE) and RNA interference (RNAi) transgenic lines. Cold tolerance among these genotypes was compreh

The role of transcription factor MdGeBP3 in Modulating cold stress tolerance and anthocyanin biosynthesis in apple.

Cold stress is a major environmental constraint limiting crop productivity and quality. Here, we identify a transcription factor MdGeBP3 in apple, which responds to cold stress at both transcriptional and post-translational levels. Functional analysis revealed that overexpression of MdGeBP3 significantly enhances cold tolerance in both apple calli and the model plant Arabidopsis, and the transcript levels of cold-responsive genes were induced in MdGeBP3 overexpression plants. Furthermore, MdGeBP3 was found to participate in anthocyanin biosynthesis. Overexpression of MdGeBP3 in Arabidopsis and

GmSNAT1 enhances cold tolerance in soybean by promoting melatonin biosynthesis and interacting with GmPSYR1.

The GmSNAT1 gene, which encodes a key enzyme involved in soybean melatonin biosynthesis, is crucial for abiotic stress tolerance. In the present study, the molecular mechanism by which GmSNAT1 enhances cold tolerance is elucidated. The cold tolerance of plants was significantly increased by GmSNAT1 overexpression and reduced by CRISPR/Cas9-mediated knockout, a phenotype that was effectively rescued by exogenous melatonin. Integrated transcriptomic, physiological, and biochemical analyses revealed that the GmSNAT1-mediated melatonin pathway activates calcium signaling; coordinates the crosstalk

Physiological and Molecular Mechanisms of Nano-Hydroxyapatite (nHAP) in Regulating Chilling Tolerance of Cucumber Seedlings.

Xishuangbanna (XIS) cucumber (Cucumis sativus) originated from low-altitude southwest China and shows extreme cold sensitivity. Nano-hydroxyapatite (nHAP), known for its high bioavailability and surface reactivity relative to bulk HAP, was applied to enhance the cold tolerance of XIS seedlings. We optimized fertilization timing and fertilizer concentration. By determining physiological parameters alongside gene expression profiling and transcriptomic analysis, we preliminarily dissected the physiological and molecular mechanisms underlying nHAP-enhanced chilling tolerance. nHAP application pre

Molecular mechanisms of cold stress-induced lens opacity: An investigation through multi-omics integrative analysis.

Cold exposure accelerates lens protein aggregation through epigenetic modulation and metabolic dysregulation. However, existing research has primarily focused on the mechanisms of protein phase separation; the molecular mechanisms underlying cold-induced reversible lens opacity remain inadequately characterized. This study aimed to elucidate the dynamic regulatory network of lens responses to cold exposure. Using a male Wistar rat model subjected to cold stress, we investigated molecular alterations through integrated transcriptomic and metabolomic analyses. Our findings demonstrate that cold

Physiological and molecular analysis of pitaya (Hylocereus polyrhizus) reveal up-regulation of secondary metabolites, nitric oxide, antioxidant defense system, and expression of responsive genes under low-temperature stress by the pre-treatment of hydrogen peroxide.

Low-temperature events are one of the leading environmental cues that considerably reduce plant growth and shift species biodiversity. Hydrogen peroxide (H2O2) is a signaling molecule that has a distinguished role during unfavorable conditions and shows outstanding perspectives in low-temperature stress. Herein, we elucidated the protective role and regulatory mechanism of H2O2 in alleviating the deleterious effects of low-temperature stress in pitaya plants. Micropropagated pitaya plants were cultured in Murashige and Skoog media supplemented with different levels of H2O2 (0, 5, 10, and 20 mM

Molecular regulation by H2S of antioxidant and glucose metabolism in cold-sensitive Capsicum.

BACKGROUND: Cold is an important environmental limiting factor affecting plant yield and quality. Capsicum (chili pepper), a tropical and subtropical vegetable crop, is extremely sensitive to cold. Although H2S is an important signaling regulator in the responses of plant growth and development to abiotic stress, few studies have examined its effects on cold-sensitive capsicum varieties. Through biotechnology methods to enhance the cold resistance of peppers, to provide some reference for pepper breeding, investigated molecular regulation by H2S of responses to cold stress in cold-sensitive ca

Hemodynamic response patterns to acute behavioral stressors resemble those to cocaine.

Hemodynamic responses to cocaine vary greatly between animals, and the variability is related to the incidence of cocaine-induced cardiomyopathies and hypertension. The variability in cardiac output and systemic vascular resistance responses to cocaine in individuals is correlated with the responses to acute startle (air jet). This experiment was designed to determine whether responses to cocaine and to air jet are related to those evoked by a conditioned stimulus (tone preceding foot shock) and to an unconditioned stimulus (cold water). We verified the relationship in hemodynamic response pat

Molecular Equilibrist: The Small Heat Shock Protein IbpA from Mycoplasma.

Small heat shock proteins (sHSPs) serve as "first aid" stress-response proteins in both eukaryotes and prokaryotes. Their holdase activity enables binding to partially denatured proteins, maintaining them in a folding-competent state under stress. The sHSP IbpA from the mycoplasma Acholeplasma laidlawii is a unique member of its family, combining the functions of two Escherichia coli sHSPs that typically act in tandem. In this study, we demonstrate for the first time that IbpA forms distinct supramolecular structures under contrasting temperature stresses in crowded environments without any ar

Cold exposure and immune function.

The influence of cold exposure on immune function is reviewed. Data obtained mainly on small mammals suggest that the acute effect of severe chilling is a suppression of several cellular and humoral components of the immune response, including a decrease of lymphocyte proliferation, a down-regulation of the immune cascade, a reduction of natural killer (NK) cell count, cytolytic activity, activation of complement, and the induction of heat shock proteins. However, adaptation to a given cold stimulus appears to develop over the course of 2-3 weeks. Further work is needed to examine interactions

PolyQ-Expanded Mutant Huntingtin Forms Inclusion Body Following Transient Cold Shock in a Two-Step Aggregation Mechanism.

Age-dependent formation of insoluble protein aggregates is a hallmark of many neurodegenerative diseases. We are interested in the cell chemistry that drives the aggregation of polyQ-expanded mutant Huntingtin (mHtt) protein into insoluble inclusion bodies (IBs). Using an inducible cell model of Huntington's disease, we show that a transient cold shock (CS) at 4 °C followed by recovery incubation at temperatures of 25-37 °C strongly and rapidly induces the compaction of diffuse polyQ-expanded HuntingtinExon1-enhanced green fluorescent protein chimera protein (mHtt) into round, micron size, cyt

Effect of different stunning methods on rigor mortis, shape, energetic status and physical characteristics of Salmo carpio fillets.

BACKGROUND: Consumer interest in safeguarding animal welfare and increased demand for fresh aquatic products support the need to understand the effects of stunning methods used in aquaculture on the biochemical process affecting fish fillet quality. The present paper aimed at comparing electrical stunning (ES) and cold shock (ICE) in Salmo carpio, an Italian endemic under-investigated species. Rigor mortis evolution, fillet adenosine 5'-triphosphate (ATP), shape, colour, pH and water holding capacity were assessed by integrating chemical and image analyses. The S100B Protein and Partners in Adipocyte Response to Cold Stress and Adaptive Thermogenesis: Facts, Hypotheses, and Perspectives.

In mammals, adipose tissue is an active secretory tissue that responds to mild hypothermia and as such is a genuine model to study molecular and cellular adaptive responses to cold-stress. A recent study identified a mammal-specific protein of the endoplasmic reticulum that is strongly induced in the inguinal subcutaneous white adipocyte upon exposure to cold, calsyntenin 3β (CLSTN3β). CLSTN3β regulates sympathetic innervation of thermogenic adipocytes and contributes to adaptive non-shivering thermogenesis. The calcium- and zinc-binding S100B is a downstream effector in the CLSTN3β pathways.

Perioperative Management of a Patient with Cold Urticaria.

Cold urticaria consists of an allergic immune response to cold temperatures with symptoms ranging from pruritic wheals to life-threatening angioedema, bronchospasm, or anaphylactic shock. Adequate planning to maintain normothermia perioperatively is vital due to impaired hypothalamic thermoregulation and overall depression of sympathetic outflow during deep sedation and general anesthesia. This case report describes the successful perioperative management of a 45-year-old female with a history of cold urticaria undergoing a laparoscopic Nissen fundoplication for refractory gastroesophageal ref

Phytohormone-Mediated Regulation of Plant Cold Stress Tolerance: Signaling, Hormonal Crosstalk, and Translational Perspectives.

Cold stress (CS) represents a major environmental factor that adversely affects plant growth, development, and productivity. To cope with low-temperature conditions, plants have evolved sophisticated mechanisms for CS perception and response, mediated through complex cellular signaling networks and physiological processes. Central to these adaptive responses are phytohormones, which function either independently or through synergistic and antagonistic interactions to fine-tune CS tolerance. This review synthesizes current knowledge on the roles of major classical phytohormones and signaling me

Metabolomics analysis reveals potential response mechanisms to cold stress in Altay sheep.

Understanding the mechanisms underlying cold tolerance in livestock is crucial for improving welfare and productivity in harsh environments. However, the complex regulatory responses to cold stress (CS) in sheep remain incompletely elucidated. This study investigated the cold adaptation mechanisms in Altay sheep (cold-adapted) compared to Turpan black sheep (heat-adapted) using serum antioxidant assays, histomorphometry of skin and rumen, and multi-tissue metabolomics under CS. Altay sheep exhibited significantly enhanced systemic antioxidant capacity under CS, evidenced by increased serum sup

Peripheral nociceptive mechanisms in an experimental rat model of fibromyalgia induced by repeated cold stress.

Fibromyalgia (FM) is a debilitating disease characterized by generalized and persistent musculoskeletal pain. Although central mechanisms are strongly implicated in the pathogenesis of FM, the involvement of peripheral mechanisms is poorly understood. To understand the peripheral nociceptive mechanisms, we examined muscular nociceptors in an FM model, which was made by exposing rats to repeated cold stress (RCS). A single muscle C-fiber nociceptors were identified through the teased fiber technique using ex vivo muscle-nerve preparations. Response properties of C-fibers to noxious stimuli were

Natural Variation of COLD and CATECHINS REGULATOR 1 Coordinately Fine-Tunes Cold Tolerance and Tea Quality in Tea Plants.

Cold tolerance and tea quality are essential for profitable tea cultivation, yet the genetic interplay between them remains elusive. Here, we systematically evaluated cold tolerance and quantified 12 key tea quality-related metabolites over two consecutive years of 108 geographically diverse tea accessions in China. Multi-trait genome-wide association mapping identified a central hub regulator, COLD AND CATECHINS REGULATOR 1 (CCR1), significantly associated with cold tolerance and catechins biosynthesis in tea plants. Functional analysis demonstrated that CsCCR1 positively regulates cold toler

Identification and functional characteristics of key regulatory genes in Populus alba in response to cold stress.

Forest crops are particularly susceptible to cold stress after dormancy release. Newly emerged shoots are especially vulnerable, and shoot mortality can cause serious loss of biomass production. Revealing the molecular mechanisms of cold stress responses in forest trees is therefore crucial. Populus alba is a frost-resistant tree species distributed worldwide. In this study, we characterized the transcriptome of P. alba in response to cold stress and observed that a large number of genes were up-regulated in both root and leaf tissues. Through Mfuzz clustering and phylogenetic analysis, four e
