The parathyroid hormone-related protein associated with malignancy is secreted by neuroendocrine tumors.

We have demonstrated the production of the PTH-related protein (PTHrP) associated with hypercalcemia of malignancy by human neuroendocrine cell lines that also produce calcitonin gene products and chromogranin A. PTHrP was demonstrable in the cells by immunocytochemistry and immunoassay and Northern analysis of the cells revealed the presence of multiple mRNAs for PTHrP. The cell lines also secreted PTHrP in a regulated fashion, with the most potent secretagogue being phorbol. Thus, PTHrP is secreted by neuroendocrine cells and it may have neuroectodermal lineage. The coexpression of calcitoni

Interleukin-1 suppresses follicle-stimulating hormone-induced estradiol secretion from cultured ovarian granulosa cells.

The effect of recombinant human interleukin-1 beta (IL-1 beta) on the follicle stimulating hormone-(FSH) induced secretion of estradiol was investigated using cultured granulosa cells obtained from immature rats with diethylstilbestrol implants. Estradiol secretion was significantly reduced by IL-1 beta in cultures containing FSH and either 10(-7) or 10(-8) M androstenedione as a substrate for estradiol synthesis. However, the inhibition of FSH-induced estradiol secretion by IL-1 beta was more apparent in the presence of 10(-8) M as compared to 10(-7) M androstenedione. IL-1 beta suppressed es

Increased cholera toxin-, and forskolin-induced cyclic AMP accumulations in psoriatic involved versus uninvolved or normal human epidermis.

Psoriatic involved epidermis reveals variously altered receptor-adenylate cyclase responses; among them the most prominent is defective beta-adrenergic adenylate cyclase response, which is normally the major receptor-adenylate cyclase system of human epidermis. It is known that activation of hormone-stimulated adenylate cyclase, a membrane-bound enzyme complex, requires functional coupling of at least 3 distinct subunits: 1) receptor subunit (R), 2) guanine nucleotide binding protein (G), and 3) catalytic subunit (C). The precise nature of the beta-adrenergic defect in the psoriatic epidermis,

Muscarinic acetylcholine receptor function in chick heart cells cultured in serum-free medium.

Primary culture of chick heart cells has proven useful in biochemical and physiological investigations of receptor-mediated myocardial responses. While cell culture provides a system in which the cellular environment can be manipulated in carefully controlled ways, the use of serum as a culture medium supplement introduces variability due to the inherent differences in the composition of different lots of sera. In this report we compare the functional properties of the muscarinic acetylcholine receptor of cultured embryonic chick heart cells grown in serum-free, defined medium to that of cells

Stimulatory effect of forskolin on gastric acid secretion in the rat, in vitro and in vivo.

The effect of forskolin, a potent adenylate cyclase activator, was studied on rat gastric acid secretion in vitro and in vivo. In the isolated gastric mucosa, forskolin was 25 and 100 times more potent than histamine and dibutyryl cyclic AMP respectively, with an EC50 of 2 microM and a maximum effect at 8-10 microM. In the lumen-perfused anaesthetised rat, acid secretion in response to a series of bolus i.v. injections of forskolin failed to show dose dependency, despite dose-related hypotensive effects. Secretory responses to i.v. infusions of forskolin were dose-related and were associated w

Effect of heparin on platelet aggregation inhibited by PGI2, trifluoperazine and verapamil.

The enhancement of platelet aggregation by heparin in the presence of certain inhibitors of aggregation was investigated in an attempt to discern the mechanism through which heparin alters platelet function in plasma. These studies were performed by adding prostaglandin I2 (PGI2), verapamil, or trifluoperazine to platelet-rich plasma (PRP) in the presence or absence of heparin. Adenosine diphosphate (ADP), collagen, or arachidonic acid were used for induction of platelet aggregation. The inhibitory agents reduced platelet aggregation to 5 to 20% of control in the absence of heparin. When prese

Effect of forskolin on action potential, slow inward current and tension of frog atrial fibers.

The new nonhormonal activator of adenylate cyclase forskolin was studied on frog atrial trabeculae by current clamp and voltage clamp methods using a double sucrose gap technique. Forskolin (5 X 10(-6) M to 2 X 10(-5) M) dose-dependently increased action potential duration, the height of the plateau and twitch tension. The time constant for inactivation of the slow inward current and the steady state kinetic variables of calcium channels d infinity and f infinity remained uneffected. Forskolin increased the amplitude of slow inward calcium current isi and of the phasic tension related to it. T

Forskolin stimulates adenylate cyclase and iodine metabolism in thyroid.

Forskolin is a potent activator of the cyclic AMP-generating system in many tissues. In dog thyroid slices, the enhancement of cyclic AMP level was rapid, sustained in the presence of forskolin, but easily reversible after its withdrawal. Contrary to TSH, forskolin induced little apparent desensitization. Forskolin potentiated the effects of TSH, PGE1 and cholera toxin. However, the forskolin-induced cyclic AMP accumulation was still sensitive to inhibitors of dog thyroid adenylate cyclase such as iodide, norepinephrine and adenosine. As fluoride, but contrary to TSH and PGE1, forskolin stimul

Zbtb18 Deficiency Impairs the Hippocampal Development and Neurite Growth.

Previous studies of the transcription factor Zbtb18 have primarily focused on its role in cortical development, whereas its function during hippocampal development remains poorly understood. In this study, we investigated the role of Zbtb18 in mouse hippocampal development using histological, transcriptomic, and functional approaches. Immunostaining demonstrated that Zbtb18 was partially overlapped with the neural progenitor marker Pax6, intermediate progenitor marker Tbr2, postmitotic neuronal marker Tbr1, and neuronal marker NeuN in the hippocampal primordium from embryonic day 13.5. Functional rescue of CFTR in rectal organoids from patients carrying R334W variant by CFTR modulators and PDE4 inhibitor Roflumilast.

BACKGROUND: Many disease-causing variants in the Cystic Fibrosis Transmembrane conductance Regulator (CFTR) gene remain uncharacterized and untreated. Restoring the function of the impaired CFTR protein is the goal of personalized medicine, particularly in patients carrying rare CFTR variants. In this study, functional defects related to the rare R334W variant were evaluated after treatment with CFTR modulators or Roflumilast, a phosphodiesterase-4 inhibitor (PDE4i). METHODS: Rectal organoids from subjects with R334W/2184insA and R334W/2183AA > G genotypes were used to perform the Forskolin-i

Inhibition of CFTR-mediated intestinal chloride secretion by nornidulin: Cellular mechanisms and anti-secretory efficacy in human intestinal epithelial cells and human colonoids.

Secretory diarrhea, a major global health concern, particularly among young children, is often characterized by excessive chloride secretion through the cystic fibrosis transmembrane conductance regulator (CFTR) channel. Nornidulin, a fungus-derived natural product from Aspergillus unguis, has previously been shown to inhibit cAMP-induced Cl- secretion in T84 cells (human intestinal cell lines). However, the cellular mechanism of nornidulin in inhibiting cAMP-induced Cl- secretion and its anti-secretory efficacy is still unknown especially in a human colonoid model, a preclinical model recapit

Natural Terpenes Inhibit the Cytopathogenicity of Naegleria fowleri Causing Primary Amoebic Meningoencephalitis in the Human Cell Line Model.

Naegleria fowleri is one of the free-living amoebae and is a causative agent of a lethal and rare central nervous system infection called primary amoebic meningoencephalitis. Despite the advancement in antimicrobial chemotherapy, the fatality rate in the reported cases is more than 95%. Most of the treatment drugs used against N. fowleri infection are repurposed drugs. Therefore, a large number of compounds have been tested against N. fowleri in vitro, but most of the compounds showed high toxicity. To overcome this, we evaluated the effectiveness of naturally occurring terpene compounds again

RETRACTED: Protective Efficacy of a Dietary Supplement Based on Forskolin, Homotaurine, Spearmint Extract, and Group B Vitamins in a Mouse Model of Optic Nerve Injury.

Источник пометил публикацию как отозванную

Glaucoma is a multifactorial blinding disease with a major inflammatory component ultimately leading to apoptotic retinal ganglion cell (RGC) death. Pharmacological treatments lowering intraocular pressure can help slow or prevent vision loss although the damage caused by glaucoma cannot be reversed. Recently, nutritional approaches have been evaluated for their efficacy in preventing degenerative events in the retina although mechanisms underlying their effectiveness remain to be elucidated. Here, we evaluated the efficacy of a diet supplement consisting of forskolin, homotaurine, spearmint e

Primary culture of embryonic rat olfactory receptor neurons.

Embryonic cells are very robust in surviving dissection and culturing protocols and easily adapt to their in vitro environment. Despite these advantages, research in the olfactory field on cultured embryonic olfactory neurons is sparse. In this study, two primary rat olfactory explant cultures of different embryonic d (E17 and E20) were established, comprising epithelium and bulb. The functionality of these neurons was tested by measuring intracellular calcium responses to cAMP-inducing agents forskolin (FSK) and 3-isobutyl-1-methylxanthine (IBMX) with fluorescence microscopy. For E17, the res

Regulation of cyclic AMP level by progesterone in ovariectomized rat neocortex.

Exposure of neocortical slices to progesterone, without prior treatment with estrogen, augmented forskolin-induced cyclic AMP within 15 min. 30 nM progesterone produced approximately 1/2 the maximal effect but as little as 10 nM progesterone produced a detectable increase in cyclic AMP. When forskolin was replaced by dideoxyforskolin, an analog that does not directly stimulate adenylyl cyclase but shares many of its other actions, progesterone did not augment cyclic AMP. Progesterone also failed to affect increased cyclic AMP that followed exposure to norepinephrine or isoproterenol. The effec

Cholecystokinin (CCK) release from rat brain slices in vitro is enhanced by agents which elevate intracellular cAMP.

Cholecystokinin (CCK) release from rat brain slices in vitro is enhanced by agents which elevate intracellular cAMP. Treatment of several CCK expressing tumor cells in culture with agents which increase intracellular cAMP increased secretion of CCK. The possibility that elevation of cAMP also alters potassium-evoked release of CCK from rat brain slices incubated in vitro was also investigated. Treatment of slices of rat prefrontal cortex and caudate-putamen with 5 microM forskolin plus 0.5 mM IBMX (3-isobutyl-1-methylxanthine, a phosphodiesterase inhibitor) caused a significant elevation of po

Crypt base cells show forskolin-induced Cl- secretion but no cation inward conductance.

Whole-cell patch-clamp studies in base cells of isolated colonic crypts of rats pretreated with dexamethasone were performed to examine the effects of stimulation by forskolin (10 micromol/l). The experiments were designed in order to distinguish between two postulated effector mechanisms: the activation of a non-selective cation channel and the activation of Cl- channels. As shown in an accompanying report, forskolin depolarizes the membrane voltage (Vm) by some 40-50 mV and enhances the whole-cell membrane conductance (Gm) substantially in these cells. In this report all experiments were per

Effects of forskolin on crypt cells of rat distal colon. Activation of nonselective cation channels in the crypt base and of a chloride conductance pathway in other parts of the crypt.

We recently showed that prostaglandin E2 (PGE2) causes depolarization in cells at the base of isolated crypts from rat distal colon by activating nonselective cation channels. In order to investigate whether PGE2 acts via intracellular cyclic adenosine monophosphate (cAMP), the effect of forskolin on cell potential and on whole-cell current was investigated using the slow whole-cell patch-clamp method with nystatin. In addition, effects of forskolin in cells at other sites along the crypt were investigated. At the crypt base, the unstimulated cells had a resting potential of -70.6 +/- 1.3 mV (

Effect of depolarizing and hyperpolarizing agents on the membrane potential difference of primary cultures of rabbit aorta vascular smooth muscle cells.

Vascular smooth muscle cells of rabbit aorta were enzymatically dispersed, kept in primary culture, and studied between days 1 and 7 in a bath rinsed with Ringer-like solution at 37 degrees C. The electrical membrane potential difference (PD) was measured with microelectrodes. The mean value of PD was -50 +/- 0.4 mV (n = 53). Cromakalim (BRL 34915), 1 mumol/l and 10 mumol/l, hyperpolarized the membrane potential by 9 +/- 1 mV (n = 11) and 15 +/- 1 mV (n = 53) respectively. Glibenclamide (10 mumol/l) abolished the hyperpolarizing effect of chromakalim (n = 6). Simultaneous addition of cromakali
