A Pollen-Enhanced Bionic Mechanoreceptor Induced by Asymmetric Ionic Convection in Hydrogel for Sensory-Augmented Prostheses.

The growing prevalence of age-related limb loss underscores the need for prosthetic technologies that restore not only motor function but also authentic sensory feedback. Current prosthetic systems largely depend on sensory substitution or signal remapping, which fall short of replicating natural somatosensory signals. In this work, we develop a plant-enhanced bionic mechanoreceptor that mimics biological touch by converting mechanical stimuli into ionic signals. Incorporating bio-derived pollen microgels into the hydrogel matrix introduces interfacial ion-anchoring sites that strengthen catio

A Phase 1 Study of Convection-Enhanced Delivery of Intraputaminal AAV2-GDNF in Advanced Parkinson's Disease.

BACKGROUND: Parkinson's disease (PD) is a progressive neurodegenerative disorder. Neurotrophic therapeutic approaches have been limited in part by incomplete delivery to the putamen. We developed image-guided convection-enhanced delivery with real-time monitoring to improve intraputaminal distribution of neurotrophic gene therapy. OBJECTIVES: To evaluate the 5-year safety and tolerability of bilateral putaminal delivery of adeno-associated virus serotype 2 encoding glial cell line-derived neurotrophic factor (AAV2-GDNF), and to describe exploratory long-term clinical outcomes. METHODS: This

Success in Achieving Relatively High Targeted Convection Volumes in Multi-Ethnic Individuals Receiving Hemodiafiltration in Singapore.

BACKGROUND: Hemodiafiltration has demonstrated improved outcomes in end-stage kidney disease, particularly with higher convection volumes than conventional hemodialysis. However, data on multiethnic Asian populations remain limited. This study evaluated the feasibility of achieving relatively high targeted convection volumes in hemodiafiltration in patients with end-stage kidney disease in Singapore. METHODS: This retrospective cohort analysis included 1404 patients undergoing hemodiafiltration between 2019 and 2023 at Fresenius Kidney Care clinics in Singapore using data obtained from the Eu

Aerosol Physicochemical Mixing State and Cloud Nucleation Potential during Tracking Aerosol Convection Interactions Experiment (TRACER) Campaign.

Aerosol-cloud interactions remain a major source of uncertainty in estimating global radiative forcing due to the complex nature of the aerosol physicochemical properties. This study investigates the physicochemical characteristics of aerosols collected during the DOE's TRacking Aerosol Convection interactions Experiment (TRACER) campaign, conducted from June to September 2022 in the Greater Houston area. Aerosols were sampled at coastal (Galveston) and inland (Hempstead and Jersey Village) sites during sea-breeze initiated convection and outflow events and analyzed using Raman microspectrosco

Convection enhanced delivery of light responsive antigen capturing oxygen generators for chemo-phototherapy triggered adaptive immunity.

In recent years, combination therapy has emerged as the cornerstone of clinical practice in treating glioblastoma multiforme. However, their ability to trigger and leverage the body's adaptive immunity has rarely been studied. Tumour heterogeneity, the presence of the blood-brain barrier, and an immunosuppressive tumor microenvironment play a crucial role in the 90% local tumor recurrence post-treatment. Herein, we report an improved combination therapy approach capable of stimulating an immune response that utilizes Light responsive antigen-capturing oxygen generators (LAGs). The engineered L

Convection-enhanced delivery for diffuse intrinsic pontine glioma: a single-centre, dose-escalation, phase 1 trial.

BACKGROUND: Diffuse intrinsic pontine glioma is one of the deadliest central nervous system tumours of childhood, with a median overall survival of less than 12 months. Convection-enhanced delivery has been proposed as a means to efficiently deliver therapeutic agents directly into the brainstem while minimising systemic exposure and associated toxic effects. We did this study to evaluate the safety of convection-enhanced delivery of a radioimmunotherapy agent targeting the glioma-associated B7-H3 antigen in children with diffuse intrinsic pontine glioma. METHODS: We did a phase 1, single-arm

Visualizing calcium responses to acetylcholine convection along endothelium of arteriolar networks in Cx40BAC-GCaMP2 transgenic mice.

Acetylcholine evokes endothelium-dependent vasodilation subsequent to a rise in intracellular calcium. Despite widespread application in human and animal studies, calcium responses to intravascular ACh have not been visualized in vivo. Microiontophoresis of ACh in tissue adjacent to an arteriole activates abluminal muscarinic receptors on endothelial cells within a "Локально" region of diffusion, but it is unknown whether ACh released in such fashion gains access to the flow stream resulting in further actions downstream. To test this hypothesis and provide new insight into calcium signaling in v

Molecular structure dependent convective control in binary solvents: a femtosecond thermal lens study.

Using femtosecond time-resolved thermal lens spectroscopy, we demonstrate that the molecular architecture of dyes dictates their interaction strength with binary solvents, thereby controlling convective heat transport. We investigated two structurally distinct dye classes, xanthenes (rhodamine 6G, rhodamine 101) and triarylmethanes (Malachite Green, Brilliant Green), in water-methanol mixtures. Strongly interacting xanthene dyes consistently increased steady-state thermal lens strength across all solvent fractions, which we attribute to dye-solvent cluster formation that enhances local viscosi

Direct convective delivery of adeno-associated virus gene therapy for treatment of neurological disorders.

Molecular biological insights have led to a fundamental understanding of the underlying genomic mechanisms of nervous system disease. These findings have resulted in the identification of therapeutic genes that can be packaged in viral capsids for the treatment of a variety of neurological conditions, including neurodegenerative, metabolic, and enzyme deficiency disorders. Recent data have demonstrated that gene-carrying viral vectors (most often adeno-associated viruses) can be effectively distributed by convection-enhanced delivery (CED) in a safe, reliable, targeted, and homogeneous manner

Safety and efficacy of convection-enhanced delivery of gemcitabine or carboplatin in a malignant glioma model in rats.

OBJECT: Convection-enhanced delivery (CED) can be used safely to perfuse regions of the central nervous system (CNS) with therapeutic agents in a manner that bypasses the blood-brain barrier (BBB). These features make CED a potentially ideal method for the distribution of potent chemotherapeutic agents with certain pharmacokinetic properties to tumors of the CNS. To determine the safety and efficacy of the CED of two chemotherapeutic agents (with properties ideal for this method of delivery) into the CNS, the authors perfused naive rats and those harboring 9L gliomas with carboplatin or gemcit

Feasibility of convection-enhanced delivery of unlabeled 225Ac for glioblastoma: Transport, dosimetry, and clonogenic survival modeling.

BACKGROUND: Glioblastoma (GB) is an aggressive primary brain tumor with local recurrence rates exceeding 90% and a five-year survival rate of approximately 5%. Standard treatments such as surgical resection, external beam radiotherapy (EBRT), and chemotherapy often leave residual tumor cells that are resistant to therapy. Local targeted alpha therapy (TAT) with actinium-225 (225Ac) offers high-linear energy transfer (LET) cytotoxicity but is limited by short diffusion range and elevated intratumoral pressure. Convection-enhanced delivery (CED), which applies mild hydraulic pressure to drive bu

Convection-enhanced delivery of cetuximab conjugated iron-oxide nanoparticles for treatment of spontaneous canine intracranial gliomas.

Cetuximab conjugated iron-oxide nanoparticles (cetuximab-IONPs) have shown both in-vitro and in-vivo anti-tumor efficacy against gliomas. The purpose of this pilot study was to evaluate the safety and potential efficacy of cetuximab-IONPs for treatment of spontaneously occurring intracranial gliomas in canines after convection-enhanced delivery (CED). The use of CED allowed for direct infusion of the cetuximab-IONPs both intratumorally and peritumorally avoiding the blood brain barrier (BBB) and limiting systemic effects. A total of eight dogs participated in the study and only two developed m

Finding the striatum in sheep: use of a multi-modal guided approach for convection enhanced delivery.

Our goal is delivery of a long-term treatment for Huntington's disease. We administer intracerebrally in sheep adeno-associated virus (AAV) to establish optimal safety, spread and neuronal uptake of AAV based therapeutics. Sheep have large gyrencephalic brains and offer the opportunity to study a transgenic Huntington's disease model. However, lack of a relevant brain stereotactic atlas and the difficulty of skull fixation make conventional stereotaxy unreliable. We describe a multi-modal image-guidance technique to achieve accurate placement of therapeutics into the sheep striatum.

Biodistribution of adeno-associated virus type-2 in nonhuman primates after convection-enhanced delivery to brain.

A combination treatment of AAV2-hAADC with oral levodopa is a novel therapeutic approach that is being developed for late-stage Parkinson's disease. Biodistribution of AAV2-hAADC was assessed over a wide range of vector dose in 12 monkeys with parkinsonian syndrome, 6 months after intraputamenal infusion. Quantitative PCR (Q-PCR) from all the major neuroanatomical regions of the brain indicated a dose-dependent increase in vector DNA, with 99% being detected in the target site and other basal ganglia tissues. Within these tissues, the distribution varied widely between the putamen (PT) and the

Salirasib (farnesyl thiosalicylic acid) for brain tumor treatment: a convection-enhanced drug delivery study in rats.

Our aim was to assess the ability of convection-enhanced drug delivery (CED), a novel approach of direct delivery of drugs into brain tissue and brain tumors, to treat brain tumors using salirasib (farsnesyl thiosalicylic acid). CED was achieved by continuous infusion of drugs via intracranial catheters, thus enabling convective distribution of high drug concentrations over large volumes while avoiding systemic toxicity. Several phase II/III CED-based trials are currently in progress but have yet to overcome two major pitfalls of this methodology (the difficulty in attaining efficient CED and

Entropy analysis over radiative marangoni convective nanofluid flow on an infinite porous disk.

Entropy refers to the randomness and disorderness of the system, which significantly affects the efficiency and performance of thermal systems. The motive of the research article is to carry out the comprehensive analysis of entropy along with the effect of thermal radiation on the Marangoni convective flow of nanofluid over an infinite porous disk using a passive flow control technique. In this analysis, the unit effect thermal slip condition and Stefan's blowing have also been considered. Mathematical modeling has been done with the help of partial differential equation (PDE) which has been

Impact of Heavy Slow Resistance vs. Convectional Exercise in Lateral Epicondylitis-A Pilot Study.

BACKGROUND: The extensor carpi radial muscle may become imbalanced compared to other muscles in the upper arm and can disrupt the overall muscle balance in the upper limb. This muscular imbalance may then cause inflammation or irritation in the tendon. Regular physical activity is rarely used as the only treatment for lowering limb edema. Heavy slow resistance (HSR) training is effective compared to regular exercises. HSR affects functional abilities and physical refinement differently from conventional workouts. This study evaluated the effects of HSR and conventional exercise (CE) on subject

The Influence of Size on the Intracranial Distribution of Biomedical Nanoparticles Administered by Convection-enhanced Delivery in Minipigs.

Because of the blood-brain barrier (BBB), successful drug delivery to the brain has long been a key objective for the medical community, calling for pioneering technologies to overcome this challenge. Convection-enhanced delivery (CED), a form of direct intraparenchymal microinfusion, shows promise but requires optimal infusate design and real-time distribution monitoring. The size of the infused substances appears to be especially critical, with current knowledge being limited. Herein, we examined the intracranial administration of polyethylene glycol (PEG)-coated nanoparticles (NPs) of vario
