Deciphering the determinants of recombinant protein expression across the human secretome.

Protein secretion is an essential process of mammalian cells. In biomanufacturing, this process can be optimized to enhance production yields and biotherapeutic quality. While cell line engineering and bioprocess optimization have yielded high protein titers for some recombinant proteins, many remain difficult to express. Here, we investigated factors influencing protein expression in Chinese hamster ovary (CHO) cells, expressing 2,135 Human Secretome Project proteins. While the abundance of mRNA from recombinant proteins explained less than 1% of observed variation in secretion titers, analys

Elucidation of Proteoforms of Chinese Hamster Ovary (CHO) Phospholipase B-Like 2 (PLBL2) Captured From a Monoclonal Antibody.

Removing host cell proteins (HCPs) during biotherapeutic manufacturing is essential to ensure patient safety and supply continuity. Yet, this goal remains exceptionally challenging for some HCP species. For example, phospholipase B-like-2 (PLBL2) from Chinese hamster ovary (CHO) cells plagues process engineers by evading typical purification strategies. Separation challenges for CHO PLBL2 and other HCPs are due in large part to a dearth of detailed biochemical characterization for HCPs, such as their proteoforms or variants. Herein, we present as a case study the elucidation of proteoforms of

Unraveling productivity-enhancing genes in Chinese hamster ovary cells via CRISPR activation screening using recombinase-mediated cassette exchange system.

Chinese hamster ovary (CHO) cells, which are widely used for therapeutic protein production, have been genetically manipulated to enhance productivity. Nearly half of the genes in CHO cells are silenced, which are promising targets for CHO cell engineering. To identify novel gene targets among the silenced genes that can enhance productivity, we established a genome-wide clustered regularly interspaced short palindromic repeats activation (CRISPRa) screening platform for bispecific antibody (bsAb)-producing CHO (CHO-bsAb) cells with 110,979 guide RNAs (gRNAs) targeting 13,812 silenced genes us

Sequence and Configuration of a Novel Bispecific Antibody Format Impacts Its Production Using Chinese Hamster Ovary (CHO) Cells.

There are a number of new format antibody-inspired molecules with multiple antigen binding capabilities in development and clinical evaluation. Here, we describe the impact of the sequence and configuration of a unique bispecific antibody format (termed BYbe) using a panel of four BYbe's and the three IgG1s from which they were derived on their production in a Chinese hamster ovary (CHO) cell expression system. Following transfection and selection, one bispecific antibody format yielded fewer mini-pools in comparison to the other bispecific cell pools. When the top 12 expressing stable mini-po

Utilizing targeted integration CHO pools to potentially accelerate the GMP manufacturing of monoclonal and bispecific antibodies.

Monoclonal antibodies (mAbs) are effective therapeutic agents against many acute infectious diseases including COVID-19, Ebola, RSV, Clostridium difficile, and Anthrax. mAbs can therefore help combat a future pandemic. Unfortunately, mAb development typically takes years, limiting its potential to save lives during a pandemic. Therefore "pandemic mAb" timelines need to be shortened. One acceleration tool is "deferred cloning" and leverages new Chinese hamster ovary (CHO) technology based on targeted gene integration (TI). CHO pools, instead of CHO clones, can be used for Phase I/II clinical ma

Plasmodium vivax Gametocytes Adherence to Bone Marrow Endothelial Cells.

In a Plasmodium vivax infection, it was shown a proportionally increased on gametocyte distribution within the bone marrow aspirant, suggesting a role of this organ as a reservoir for this parasite stage. Here, we evaluated the ex vivo cytoadhesive capacity of P. vivax gametocytes to bone marrow endothelial cells (HBMEC) and investigated the involvement of some receptors in the cytoadhesion process by using transfected CHO cells (CHO-ICAM1, CHO-CD36 and CHO-VCAM), wild type (CHO-K1) or deficient in heparan and chondroitin sulfate (CHO-745). Ex-vivo cytoadhesion assays were performed using a to

Activity of the mouse Notch ligand DLL1 is sensitive to C-terminal tagging in vivo.

OBJECTIVE: The mammalian Notch ligand DLL1 has essential functions during development. To visualise DLL1 in tissues, for sorting and enrichment of DLL1-expressing cells, and to efficiently purify DLL1 protein complexes we tagged DLL1 in mice with AcGFPHA or Strep/FLAG. RESULTS: We generated constructs to express DLL1 that carried C-terminal in-frame an AcGFPHA tag flanked by loxP sites followed by a Strep/FLAG (SF) tag out of frame. Cre-mediated recombination replaced AcGFP-HA by SF. The AcGFPHAstopSF cassette was added to DLL1 for tests in cultured cells and introduced into endogenous DLL1 i

Genetic rearrangement during site specific integration event facilitates cell line development of a bispecific molecule.

Site specific integration (SSI) expression systems offer robust means of generating highly productive and stable cell lines for traditional monoclonal antibodies. As complex modalities such as antibody-like molecules comprised of greater than two peptides become more prevalent, greater emphasis needs to be placed on the ability to produce appreciable quantities of the correct product of interest (POI). The ability to screen several transcript stoichiometries could play a large role in ensuring high amounts of the correct POI. Here we illustrate implementation of an SSI expression system with a

Direct observation of nystatin binding to the plasma membrane of living cells.

Nystatin is an antifungal polyene macrolide which is widely applied to treat yeast infections. Nystatin has also been used as a laboratory tool to inhibit endocytic processes in mammalian cells. The interaction of nystatin with model membranes has been studied thoroughly by various spectroscopic methods, making use of its weak fluorescence in the ultraviolet (UV). Studying its interaction with cells would require direct imaging, which, so far, required attachment of a fluorophore to nystatin. Using UV-sensitive microscopy, we show here how to visualize the interaction of nystatin with the plas

CHO genome mining for synthetic promoter design.

Synthetic promoters are an attractive alternative for use in mammalian hosts such as CHO cells as they can be designed de novo with user-defined functionalities. In this study, we describe and validate a method for bioprocess-directed design of synthetic promoters utilizing CHO genomic sequence information. We designed promoters with two objective features, (i) constitutive high-level recombinant gene transcription, and (ii) upregulated transcription under mild hypothermia or late-stage culture. CHO genes varying in transcriptional activity were selected based on a comparative analysis of RNA-

Evidence of Clostridium perfringens epsilon toxin associated with multiple sclerosis.

BACKGROUND: It was recently reported that, using Western blotting, some multiple sclerosis (MS) patients in the United States had antibodies against epsilon toxin (Etx) from Clostridium perfringens, suggesting that the toxin may play a role in the disease. OBJECTIVE: We investigated for serum antibodies against Etx in UK patients with clinically definite multiple sclerosis (CDMS) or presenting with clinically isolated syndrome (CIS) or optic neuritis (ON) and in age- and gender-matched controls. METHODS: We tested sera from CDMS, CIS or ON patients or controls by Western blotting. We also te

Effective isotope labeling of proteins in a mammalian expression system.

Isotope labeling of biologically interesting proteins is a prerequisite for structural and dynamics studies by NMR spectroscopy. Many of these proteins require mammalian cofactors, chaperons, or posttranslational modifications such as myristoylation, glypiation, disulfide bond formation, or N- or O-linked glycosylation; and mammalian cells have the necessary machinery to produce them in their functional forms. Here, we describe recent advances in mammalian expression, including an efficient adenoviral vector-based system, for the production of isotopically labeled proteins. This system enables

Assessment of antimutagenic effects of stobadine dihydrochloride on MNNG-induced mutations in Chinese hamster cells V79.

Mutagenicity of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and antimutagenic effect of antioxidant stobadine (STB) were investigated by so called HGPRT/V79 system. Cells were treated by STB before, during and after MNNG-treatment. Our results showed that the highest antimutagenic effect of STB was observed if the drug was given as a pretreatment before exposure of cells to MNNG. This effect was not concentration-dependent within the framework of 1.5-9 mmol. All other combinations of MNNG- and STB-treatment led to the weaker but statistically significant decrease of 6-TGr mutations. Inhibition

Metabolic and morphological changes of the heart in Chinese hamsters (CHAD strain) with spontaneous long-term diabetes.

We studied the effect of spontaneous long-term (9-10 months) diabetes on the heart of Chinese hamsters (CHAD strain) to elucidate the relationship between diabetes mellitus and cardiomyopathy. The diabetic hamsters, aged approximately 11 months, showed body weight loss, hyperglycemia (mean fasting plasma glucose 402 mg/dl), hypoinsulinemia, hyperlipidemia and ketonemia. The diabetic hamsters showed reduced activities of cytoplasmic glycolytic key enzymes; hexokinase, pyruvate kinase and phosphofructokinase, increases in cardiac glycogen and glucose-6-phosphate contents and a 40% decrease in ca

A temperature sensitive mutant of a Chinese hamster cell line exhibiting high chromosomal breakage.

A temperature sensitive mutant exhibiting a very high level of chromosomal aberrations has been isolated from the Chinese hamster cell line E36. The chromosome aberrations, which include chromosome and chromatid breaks, multiradial configurations, dicentrics, and pulverizations, start to appear 1 h after a shift in temperature from 34 degrees C to 40.5 degrees C. The rate of sister chromatid exchange is not increased in this mutant. Analysis of somatic cell hybrids indicates that the mutation in this temperature sensitive mutant is dominant.

Cytological effects of kepone on Chinese hamster cells.

Cytological and cytotoxic effects of kepone on Chinese hamster cells (M3-1) were investigated. Cells treated with 2 micrograms, 4 micrograms, and 6 micrograms/ml of kepone did not show any morphological abnormalities. However, cytological observations showed that chromosome breaks, chromatid breaks, dicentric chromosomes, and chromosome interchanges were produced by these treatments. Cytotoxic studies revealed dose response and time response reactions to kepone. Cell toxicity was greater at the 30 micrograms/ml concentration, producing 100 percent cell death within 24 hours.

Strategies for removing two "problematic" host cell proteins, using an "end-to-end" approach.

Host cell proteins (HCPs) are critical process-related impurities of recombinant protein biopharmaceuticals that have the potential to impact product safety and efficacy. In this study, two residual HCPs, heat shock protein 90 beta and perilipin-4-like, produced from a CHO cell line, were identified during the development of a late-stage platform Immunoglobulin G subclass 1 (IgG1) monoclonal antibody process. A risk assessment was performed that included identification of HCPs, homology with their human protein counterparts, and prior non-clinical and clinical experience. The outcome deemed th

Assessment of in vitro toxicity of chloronitramide anion (CNAM).

In 2024, the chloronitramide anion (CNAM) was identified as a degradation product of inorganic chloramines, more than 40 years since its existence was suggested. However, to our knowledge, no studies have evaluated the toxicity of CNAM. In this manuscript, we evaluate CNAM in vitro toxicity using the Chinese hamster ovary (CHO) cells chronic cytotoxicity assay. CNAM exhibited a LC50 value of 9.99 × 10-5 M (99.9 µM; 9.45 mg/L), where LC50 denotes the concentration causing 50% cell death. These results indicate that CNAM is more toxic than regulated disinfection by-products (DBPs), including tri

Systematic identification of genomic hotspots for high-yield protein production in CHO cells.

The efficient and stable production of therapeutic proteins in Chinese hamster ovary (CHO) cells hinges on robust cell line development (CLD). Traditional methods relying on random transgene integration often result in clonal variability, requiring extensive and resource-intensive screening. To address this limitation, we established a systematic, multiomics-driven framework that integrates 202 RNA-sequencing datasets and whole-genome sequencing data to identify genomic "hotspot" loci for precise and high-yield transgene integration. From an initial pool of 20 candidate loci, 5 top-performing

A Novel Strategy to Rapidly Profile and Quantify High-Risk Host Cell Proteins Using Integrated DDA-PRM-dMRM Mode.

Host cell proteins (HCPs) are critical process-related impurities in biotherapeutics that threaten drug stability and safety. The substantial dynamic range (>5 orders of magnitude) between therapeutic proteins and residual HCPs often leads to difficulty in detecting high-risk species. Herein, we developed a novel strategy integrating data-dependent acquisition (DDA), parallel reaction monitoring (PRM), and multiple reaction monitoring (MRM) techniques to comprehensively profile and accurately quantify high-risk HCPs. We constructed a Chinese hamster ovary (CHO) cell spectral library by DDA tha
