Expression profiling of the gibberellin 2-oxidase gene CsGA2ox2 (Cucumis sativus L.) reveals stage- and tissue-specific transcript accumulation during fruit development.

BACKGROUND: Gibberellins (GAs) regulate fruit set and expansion, and their endogenous levels are controlled in part through catabolic inactivation. GA2ox enzymes convert bioactive GAs into inactive forms, but the expression of GA2ox genes during cucumber fruit development has not been characterised in a tissue-resolved manner. METHODS: Cucumber (Cucumis sativus L.) '9930' fruit were sampled at 0, 7, 14, 21 and 28 days after pollination. The inner fleshy tissue (pulp) and the outer fruit wall (pericarp) were separated, and CsGA2ox2 transcript abundance was quantified by quantitative real-time

Univariate and multivariate QTL analyses provide insights into genetic architecture of variation of key volatile aroma compounds in cucumber (Cucumis sativus L.).

Univariate and multivariate (PC-based) QTL analyses in multiple segregating populations and environments identify 284 QTL and 52 PC-QTL for 24 volatile aroma compounds in cucumber, including two environmentally stable QTL clusters, each harboring 26 and 25 QTL for major C6/C9 alcohols and aldehydes, respectively. Consumers enjoy cucumber for its refreshing sensation, which is primarily associated with volatile organic compounds (VOCs) produced by the lipoxygenase biosynthesis pathway. However, our knowledge of cucumber flavor genetics remains limited. Here, we report QTL mapping of VOCs in mul

Evaluation and selection of superior snap melon (Cucumis melo var. momordica Duth. & Full.) genotypes using morphological, yield, and selected fruit quality traits.

The snap melon (Cucumis melo var. momordica Duth. & Full.), recognized for its characteristic fruit splitting at maturity, is a nutritious and resilient cucurbit with considerable potential for crop improvement. However, systematic breeding efforts remain limited due to low genetic variability present in available germplasm and the relationships among yield and selected fruit quality traits. This study evaluated 30 snap melon genotypes to quantify genetic variability, heritability, trait correlations, and genetic divergence for the identification of superior breeding materials. Substantial phe

Genome-Wide Identification and Characterization of the TBL Gene Family and Temporal Expression Dynamics During Powdery Mildew Infection in Cucumber (Cucumis sativus).

Cell-wall polysaccharide O-acetylation contributes to cell-wall assembly, organ development, and plant-pathogen interactions, but the cucumber TBL gene family remains poorly characterized. Here, 37 CsTBL genes were identified genome-wide and analyzed using phylogenetic, syntenic, conserved-motif, gene-structure, promoter, protein-structure, Gene Ontology, and transcriptome approaches, followed by RT-qPCR analysis after powdery mildew inoculation. All CsTBL proteins contained the conserved GDS and DxxH motifs, whereas accessory motifs and predicted structural features varied among clades. Intra

β-Aminobutyric Acid and Powdery Mildew Infection Enhanced the Activation of Defense-Related Genes and Salicylic Acid in Cucumber (Cucumis sativus L.).

Powdery mildew disease, caused by Sphaerotheca fusca, is a major disease affecting cucumbers cultivated in greenhouses. This study was conducted to find defense genes induced by β-aminobutyric acid (BABA) and powdery mildew in cucumber. Disease severities of 25% and 5% were exhibited by the 2000 and 5000 mg/L BABA-treated cucumber, respectively. BABA did not affect the spore germination of the powdery mildew pathogen, showing that BABA is not an antifungal agent against the pathogen. In quantitative real-time PCR analysis, BABA-treated cucumber upregulated the transcriptional levels of the def

Salicylic acid alleviates cadmium-induced inhibition of growth and photosynthesis through upregulating antioxidant defense system in two melon cultivars (Cucumis melo L.).

Cadmium (Cd) is a widespread toxic heavy metal that usually causes deleterious effects on plant growth and development. Salicylic acid (SA), a naturally existing phenolic compound, is involved in specific responses to various environmental stresses. To explore the role of SA in the tolerance of melon (Cucumis melo L.) to Cd stress, the influence of SA application on the growth and physiological processes was compared in the two melon cultivars Hamilv (Cd-tolerant) and Xiulv (Cd-sensitive) under Cd stress. Under 400-μM Cd treatment, Hamilv showed a higher biomass accumulation, more chlorophyll

Glutathione is required for nitric oxide-induced chilling tolerance by synergistically regulating antioxidant system, polyamine synthesis, and mitochondrial function in cucumber (Cucumis sativus L.).

In this paper, we discussed the physiological mechanism of enhanced chilling tolerance with combined treatment of nitric oxide (NO) and reduced glutathione (GSH) in cucumber seedlings. With prolonged low temperature (10 °C/6 °C), oxidative stress improved, which was manifested as an increase the hydrogen peroxide (H2O2) and malondialdehyde (MDA), causing cell membrane damage, particularly after 48 h of chilling stress. Exogenous sodium nitroprusside (SNP, NO donor) enhanced the activity of nitric oxide synthase NOS-like, the contents of GSH and polyamines (PAs), and the cellular redox state, t

Effects of seedling-stage LED supplementary lighting on the eating quality and textural properties of oriental melon (Cucumis melo L. var. makuwa Makino) fruits at maturity.

INTRODUCTION: Low-light stress during winter compromises the sensory quality of off-season oriental melons (Cucumis melo L. var. makuwa Makino) in cultivation. Although LED supplemental lighting is commonly used to alleviate low-light stress in horticulture, its long-term programming effects on fruit quality remain largely unknown. METHODS: This study investigated whether early light signals from different LED spectra (red:blue ratios of 1:1 and 5:1) and intensities (full‑spectrum white at 18, 48, and 60 W) could persistently shape the texture and eating quality of mature fruits in two cultiv
