Covalent Plant Natural Product that Potentiates Antitumor Immunity.

Despite the unprecedented therapeutic potential of immune checkpoint antibody therapies, their efficacy is limited partly by the dysfunction of T cells within the cancer microenvironment. Combination therapies with small molecules have also been explored, but their clinical implementation has been met with significant challenges. To search for antitumor immunity activators, the present study developed a cell-based system that emulates cancer-attenuated T cells. The cell-based screening of 232 natural products containing electrophilic reactive functional groups led to the identification of arve

Targeted inhibition of IGF2BP1 effectively suppresses HBV replication via an m6A-dependent manner.

Elucidation of the regulation mechanism of hepatitis B virus (HBV) replication will provide potential targets for the development of novel anti-HBV therapeutics. It has been reported that the N6-methyladenosine (m6A) modification of HBV RNA plays a crucial role in the HBV life cycle. However, the mechanisms underlying the regulation of this modification remain incompletely understood. In this study, combining loss- and gain-of-function genetic analyses, we defined the role of IGF2BP1, an m6A reader, in facilitating HBV replication. Mechanistic studies revealed that IGF2BP1 stabilizes HBV RNAs

The downregulation of PEPCK mediated cuproptosis is involved in triclosan induced reproductive damage in mouse testes via triggering ROS overproduction.

Triclosan (TCS) is a widely used broad-spectrum antimicrobial agent that has been demonstrated to induce damage to the male reproductive system. However, the specific mechanisms of testicular injury induced by TCS are not fully understood. In the present study, mice were treated with 0, 20 mg/kg and 40 mg/kg per day TCS from PND 56 to PND 84. Decreased sperm counts, increased sperm deformity rates, downregulation of Phosphoenolpyruvate carboxykinase (PEPCK) and cuproptosis activation were observed. In vitro, TM4 cells were exposed to 0, 5 nM and 10 nM TCS for 24 h, which induced oxidative stre

Helicobacter pylori-Induced Persistent IGF2BP1 Activation Promotes Ferroptosis Resistance in Gastric Tumorigenesis.

Helicobacter pylori (H. pylori) infection is a major risk factor for gastric cancer, yet how a transient bacterial insult induces durable oncogenic reprogramming in gastric epithelial cells remains incompletely understood. Here, we identify insulin-like growth factor 2 mRNA-binding protein 1 (IGF2BP1) as a key mediator of sustained ferroptosis resistance during gastric tumorigenesis. We show that H. pylori infection induces IGF2BP1 expression in gastric epithelial cells, murine models, and human gastric tissues, and that IGF2BP1 upregulation can persist following bacterial eradication, consist

STAT3 signalling pathway is implicated in keloid pathogenesis by preliminary transcriptome and open chromatin analyses.

Keloids are wounding-induced fibroproliferative human tumor-like skin scars of complex genetic makeup and poorly defined pathogenesis. To reveal dynamic epigenetic and transcriptome changes of keloid fibroblasts, we performed RNA-seq and ATAC-seq analysis on an early passage keloid fibroblast cell strain and its paired normal control fibroblasts. This keloid strain produced keloid-like scars in a plasma clot-based skin equivalent humanized keloid animal model. RNA-seq analysis reveals gene ontology terms including hepatic fibrosis, Wnt-β-catenin, TGF-β, regulation of epithelial-mesenchymal tra

Three-dimensional cell cultures as an in vitro tool for ovarian cancer modeling and natural product discovery.

Ovarian cancer (OC) remains the most common cancer among women, with high incidence and mortality rates, prompting extensive investigation across in vivo, in vitro, and clinical settings. In this context, in vitro cell culture models have been widely used to study drug response, tumor progression, and the development of novel therapeutic strategies. Among these, three-dimensional (3D) culture systems have emerged as advanced platforms capable of more accurately recapitulating tumor complexity. Concurrently, the exploration of complementary therapeutic approaches, especially natural products, h

The Role of Natural Products in Liver Cancer: Focus on Angiogenesis, Inflammation, Oxidative Stress, and Apoptosis.

Hepatocellular carcinoma (HCC) is the primary liver tumor that arises from various risk factors, including hepatitis, excessive alcohol consumption, and obesity. These risk factors induce pathophysiological alterations in the liver, such as inflammation, fibrosis, cirrhosis, fatty liver, abnormal regeneration, and angiogenesis. Although procedures like ablation and transplantation hold promise for addressing this malignancy, the recurrence and metastasis rates limit the long-term efficacy of these surgical interventions. Thus, natural compounds exhibit great potential in enhancing clinical int

Photobiomodulation reduces spinal cord edema by decreasing the expression of AQP4 in the astrocytes of male spinal cord injury rats via the JAK2/STAT3 signaling pathway.

BACKGROUND: Spinal cord swelling commonly occurs following SCI. Previous studies suggest that PBM may reduce inflammation and scar formation after SCI. However, whether PBM can alleviate post-spinal cord injury edema and its underlying mechanisms have not yet been reported. This study aims to investigate the effects of PBM on spinal cord swelling in rats following SCI and explore the underlying mechanisms. METHODS: A rat model of SCI was established, and the rats received continuous PBM therapy for two weeks. Tissue hydration, motor function, AQP4 expression, and pathological changes in the s
