First isolation of Japanese encephalitis from Culex quinquefasciatus in Thailand.

Isolation of Japanese encephalitis (JE) virus using C6/36 cell and immunofluorescence virus antigen detection techniques was attempted from female mosquitoes collected with CDC gravid traps in Samut Songkhram Province in the central region and in Phuket Province in southern Thailand, in 2003. One thousand and eighty female mosquitoes including 6 species of the Culicidae family (Culex quinquefasciatus, Cx. gelidus, Cx. tritaeniorhynchus, Cx. whitmorei, Cx. vishnui complex, Cx. s.g. culiciomyia) (pooled by specific specimen), were processed for virus isolation. Two pools of Cx. quinquefasciatus

Venereal transmission of Japanese encephalitis virus in Culex bitaeniorhynchus mosquitoes.

Experiments were conducted in the laboratory on C. bitaeniorhynchus to determine the possible role of males in maintaining or sustaining the Japanese encephalitis virus activity in nature. It was found that infected males are capable of passing on the virus to female mosquitoes while mating. The occurrence of phenomenon of venereal transmission of this virus may have epidemiological importance.

Growth of Japanese encephalitis virus in Culex tritaeniorhynchus cell cultures.

Ovarian cells from the mosquito Culex tritaeniorhynchus established in continuous tissue cultures have been found to support the growth of Japanese encephalitis virus with titers reaching as high as 10(7.36) MICLD50 per 0.03 ml. Virus-cell cultures were serially subcultured 60 times over 300 days and the virulence of the virus was determined after each passage. A gradual loss in virus titer was observed, with titers always higher in the medium than in the cells. Cytopathogenic effects were not observed in the infected cell cultures, either in primary or subsequent passages. However, the cell g

Identification of Neotropical Culex Mosquitoes by MALDI-TOF MS Profiling.

The mosquito (Diptera: Culicidae) fauna of French Guiana encompasses 242 species, of which nearly half of them belong to the genus Culex. Whereas several species of Culex are important vectors of arboviruses, only a limited number of studies focus on them due to the difficulties to morphologically identify field-caught females. Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) has been reported as a promising method for the identification of mosquitoes. Culex females collected in French Guiana were morphologically identified and dissected. Abdomens wer

West Nile Virus lineage-2 in Culex specimens from Iran.

OBJECTIVE: Screening of mosquitoes for viruses is an important forecasting tool for emerging and re-emerging arboviruses. Iran has been known to harbour medically important arboviruses such as West Nile virus (WNV) and dengue virus (DENV) based on seroepidemiological data. However, there are no data about the potential mosquito vectors for arboviruses in Iran. This study was performed to provide mosquito and arbovirus data from Iran. MATERIALS AND METHODS: A total of 32 317 mosquitos were collected at 16 sites in five provinces of Iran in 2015 and 2016. RT-PCR for detection of flaviviruses wa

Wing variation in Culex nigripalpus (Diptera: Culicidae) in urban parks.

BACKGROUND: Culex nigripalpus has a wide geographical distribution and is found in North and South America. Females are considered primary vectors for several arboviruses, including Saint Louis encephalitis virus, Venezuelan equine encephalitis virus and Eastern equine encephalitis virus, as well as a potential vector of West Nile virus. In view of the epidemiological importance of this mosquito and its high abundance, this study sought to investigate wing variation in Cx. nigripalpus populations from urban parks in the city of São Paulo, Brazil. METHODS: Female mosquitoes were collected in s

Culex pipiens quinquefasciatus: a potential vector to transmit Zika virus.

Zika virus (ZIKV) has become a threat to global health since the outbreak in Brazil in 2015. Although ZIKV is generally considered an Aedes-transmitted pathogen, new evidence has shown that parts of the virus closely resemble Culex-transmitted viruses. Therefore, it is important to evaluate the competence of Culex species for ZIKV to understand their potential as vectors. In this study, female Culex pipiens quinquefasciatus were orally exposed to ZIKV. Mosquito midguts, salivary glands and ovaries were tested for ZIKV to measure infection and dissemination at 2, 4, 6, 8, 12, 16 and 18 days pos

Vector competence of Culex neavei (Diptera: Culicidae) for Usutu virus.

Usutu virus (USUV), a flavivirus belonging to the Japanese encephalitis serocomplex, was isolated for the first time from a Culex neavei mosquito in 1959 in South Africa. Despite multiple isolations of USUV from Cx. neavei in Africa, its vector competence remains unproven. Therefore, we infected Cx. neavei orally with the USUV reference strain and used reverse transcription-polymerase chain reaction and an indirect immunofluorescence assay to detect virus in bodies, legs, wings, and saliva of mosquitoes. We demonstrated the susceptibility of Cx. neavei mosquitoes for the USUV reference strain,

Isolation of Japanese encephalitis virus from Culex annulirostris in Australia.

Mosquitoes were collected using CO2 and octenol-baited light traps during an outbreak of Japanese encephalitis (JE) on Badu Island in the Torres Strait, Australia in April 1995. A total of 13,300 mosquitoes comprising 12 species were processed for virus isolation. Eight isolates of JE virus were obtained from Culex annulirostris, with a carriage rate of 2.97:1,000; this mosquito also yielded one Sindbis virus isolate. A reverse transcriptase-polymerase chain reaction was used to sequence the JE viruses, which were compared with published sequences. The eight isolates were 90% homologous with k

Experimental Wuchereria bancrofti infection of Culex quinquefasciatus and Aedes aegypti.

Laboratory-derived females of Culex quinquefasciatus and Aedes aegypti were experimentally infected with Wuchereria bancrofti by allowing the insects to feed on infected volunteers of known microfilariae density between 24.00 and 2.00 hours. Of the 240 mosquitoes used for the experiments only 67 (55.8%) of C. quinquefasciatus and 83 (69.2%) of the Ae.aegypti actually fed on the volunteers' blood. After 16 days post-infection period, 50 (74.62%) of the engorged C. quinquefasciatus harboured infective L3 larval stages of W. bancrofti, while 16 (23.9%) and 1 (1.49%) harboured L2 and L1 pre-infect

Influence of the distribution of host species on adult abundance of Japanese encephalitis vectors Culex vishnui subgroup and Culex gelidus in a rice-cultivating village in northern Vietnam.

A field study was conducted in a village in northern Vietnam to investigate how host distribution influences Japanese encephalitis (JE) vector abundance. Indoor and outdoor collections were conducted from 50 compounds. We collected three JE vector species--Culex tritaeniorhynchus and Culex vishnui that comprised the Culex vishnui group, and Culex gelidus. Spatial autocorrelation was not observed in the mosquito assemblies at any scale larger than the house compounds. Multivariate analyses revealed that the Cx. gelidus density correlated positively with both the host proximity to the breeding s

Effect of Climate Warming on Culex Mosquito Population Dynamics in Newfoundland.

Mosquitoes are important vectors of infectious diseases affecting humans and animals. In North America, Culex mosquitoes transmit West Nile virus and other arboviruses, while their persistence in temperate regions depends strongly on seasonal processes such as diapause. Temperature, rainfall, and photoperiod jointly regulate mosquito development, survival, reproduction, and seasonal activity, yet many climate-driven models do not explicitly represent diapause or interacting environmental drivers. We develop a mechanistic, stage-structured model for temperate Culex populations that includes egg

Study on the temporal and spatial distribution of Culex mosquitoes in Hanoi, Vietnam.

Arboviruses transmitted by mosquitoes, including Japanese encephalitis virus (JEV), present a substantial global health threat. JEV is transmitted by mosquitoes in the genus Culex, which are common in both urban and rural areas in Vietnam. In 2020, we conducted a 1-year survey of Culex mosquito abundance in urban, suburban, and peri-urban areas of Hanoi using CDC-light traps. Mosquitoes were identified to species and sorted into pools based on species, sex, and trap location. The mosquito pools were also investigated by RT-qPCR for detection of JEV. In total, 4829 mosquitoes were collected ove

A deeper insight into the sialome of male and female Culex quinquefasciatus mosquitoes.

INTRODUCTION: During evolution, blood-feeding arthropods developed a complex salivary mixture that can interfere with host haemostatic and immune response, favoring blood acquisition and pathogen transmission. Therefore, a survey of the salivary gland contents can lead to the identification of molecules with potent pharmacological activity in addition to increase our understanding of the molecular mechanisms underlying the hematophagic behaviour of arthropods. The southern house mosquito, Culex quinquefasciatus, is a vector of several pathogenic agents, including viruses and filarial parasites
