[Effect of potent analgesics on the ventilatory CO2 response and on the mouth occlusion pressure (author's transl)].

The depressant effects of four potent analgesics on the respiration were studied in 9 healthy young persons by determining the CO2 response (Read's method) and the mouth occlusion pressure (Whitelaw's method) before and after administration of the drugs and with or without interposition of additional inspiratory resistance (Rin). The analgesics tested were: Mr. 1268 = M1; pentazocine = M2; tramadol = M3; Mr. 1353 = M4. M1 and M2 induced a marked reduction in the ventilatory CO2 response (56.2 per cent +/- 21.8 for M1 and 53.8 per cent +/- 17.1 for M2). Interposition of Rin reduced the response

Investigation of Cannabidiol in the Mouse Drug Discrimination Paradigm.

Introduction: Cannabidiol (CBD) has gained considerable public and scientific attention because of its known and potential medicinal properties, as well as its commercial success in a wide range of products. Although CBD lacks cannabimimetic intoxicating side effects in humans and fails to substitute for cannabinoid type-1 receptor (CB1R) agonists in laboratory animal models of drug discrimination paradigm, anecdotal reports describe it as producing a "pleasant" subjective effect in humans. Thus, we speculated that this phytocannabinoid may elicit distinct subjective effects. Accordingly, we i

Gulypyrones A and B and Phomentrioloxins B and C Produced by Diaporthe gulyae, a Potential Mycoherbicide for Saffron Thistle (Carthamus lanatus).

A virulent strain of Diaporthe gulyae, isolated from stem cankers of sunflower and known to be pathogenic to saffron thistle, has been shown to produce both known and previously undescribed metabolites when grown in either static liquid culture or a bioreactor. Together with phomentrioloxin, a phytotoxic geranylcyclohexenetriol recently isolated from a strain of Phomopsis sp., two new phytotoxic trisubstituted α-pyrones, named gulypyrones A and B (1 and 2), and two new 1,O- and 2,O-dehydro derivatives of phomentrioloxin, named phomentrioloxins B and C (3 and 4), were isolated from the liquid c

A detailed characterization of the effects of four cannabinoid agonists on operant lever pressing.

The present experiments were conducted to investigate the effects of four cannabimimetics on detailed temporal parameters of operant responding. In this study, the behavioral output during performance of a fixed ratio 5 schedule of reinforcement was recorded by a computer program that measured the response initiation time (IT; time interval between the offset of one lever press and the onset of the next) and the response duration (the amount of time that elapses from the onset to the offset of one lever press) of each lever press. ITs were further partitioned into fast responses (IT=0.0-1.0 s)

Volatilomics Reveals Potential Biomarkers for Identification of Renal Cell Carcinoma: An In Vitro Approach.

The identification of noninvasive biomarkers able to detect renal cell carcinoma (RCC) at an early stage remains an unmet clinical need. The recognition that altered metabolism is a core hallmark of cancer boosted metabolomic studies focused in the search for cancer biomarkers. The present work aims to evaluate the performance of the volatile metabolites present in the extracellular medium to discriminate RCC cell lines with distinct histological subtypes (clear cell and papillary) and metastatic potential from non-tumorigenic renal cells. Hence, volatile organic compounds (VOCs) and volatile

Antioxidative, Anti-Inflammatory, and Anti-Aging Properties of Mycosporine-Like Amino Acids: Molecular and Cellular Mechanisms in the Protection of Skin-Aging.

Prolonged exposure to ultraviolet (UV) radiation causes photoaging of the skin and induces a number of disorders, including sunburn, fine and coarse wrinkles, and skin cancer risk. Therefore, the application of sunscreen has gained much attention to reduce the harmful effects of UV irradiation on our skin. Recently, there has been a growing demand for the replacement of chemical sunscreens with natural UV-absorbing compounds. Mycosporine-like amino acids (MAAs), promising alternative natural UV-absorbing compounds, are a group of widely distributed, low molecular-weight, water-soluble molecule

1, 8-Cineol Protect Against Influenza-Virus-Induced Pneumonia in Mice.

1,8-Cineol is a major monoterpene principally from eucalyptus essential oils and has been shown to exert anti-inflammatory, antiviral, and inhibitory of nuclear factor (NF)-kB effect. In the present study, we evaluated the effect of 1,8-cineol on mice infected with influenza A virus. We found that 1,8-cineol protects against influenza viral infection in mice. Moreover, 1,8-cineol efficiently decreased the level of IL-4, IL-5, IL-10, and MCP-1 in nasal lavage fluids and the level of IL-1β, IL-6, TNF-α, and IFN-γ in lung tissues of mice infected with influenza virus. The results also showed that

Effect of ring-constrained phenylpropyloxyethylamines on sigma receptors.

A series of ring-constrained phenylpropyloxyethylamines, partial opioid structure analogs and derivatives of a previously studied sigma (σ) receptor ligand, was synthesized and evaluated at σ and opioid receptors for receptor selectivity. The results of this study identified several compounds with nanomolar affinity at both σ receptor subtypes. Compounds 6 and 9 had the highest selectivity for both σ receptor subtypes, compared to μ opioid receptors. In addition, compounds 6 and 9 significantly reduced the convulsive effects of cocaine in mice, which would be consistent with antagonism of σ re

[Acute poisoning with a narcotic (Tramadol) in an infant of five weeks].

Accidental rectal administration of 27 mg/kg of the narcotic analgesic Tramadol to a five-week-old infant resulted in severe cerebral depression, which had to be treated with Naloxone during 48 hours. The severity and the duration of this intoxication are not explained solely by the high dosage of Tramadol. A decreased elimination kinetics and an increased permeability of the blood-cerebrospinal fluid barrier probably also account for the pattern of the present intoxication.

The mucolytic effect of Sputolosin in horses with respiratory disease.

The effect of the mucolytic drug Sputolosin on the clinical signs of respiratory disease among 28 matched pairs of horses was examined. Compared with the untreated group, the treated group showed a significant decrease in the frequency of coughing and a decreased time to resolution of both cough and nasal discharge. The results indicate that the drug is potentially useful in the management of respiratory disease characterised by an abnormal or increased production of mucus.

Sex-specific mechanisms of tolerance for the cannabinoid agonists CP55,940 and delta-9-tetrahydrocannabinol (Δ9-THC).

RATIONALE: Tolerance to cannabinoids could limit their therapeutic potential. Male mice expressing a desensitization-resistant form (S426A/S430A) of the type-1 cannabinoid receptor (CB1R) show delayed tolerance to delta-9-tetrahydrocannabinol (∆9-THC) but not CP55,940. With more women than men using medical cannabis for pain relief, it is essential to understand sex differences in cannabinoid antinociception, hypothermia, and resultant tolerance. OBJECTIVE: Our objective was to determine whether female mice rely on the same molecular mechanisms for tolerance to the antinociceptive and/or hypo

Linalool oxide: generalist plant based lure for mosquito disease vectors.

BACKGROUND: Lack of effective vaccines and therapeutics for important arboviral diseases such as Rift Valley fever (RVF) and dengue, necessitates continuous monitoring of vector populations for infections in them. Plant-based lures as surveillance tools has the potential of targeting mosquitoes of both sexes and females of varied physiological states; yet such lures are lacking for vectors of these diseases. Here, we present evidence of the effectiveness of linalool oxide (LO), a single plant-based lure previously developed for malaria vectors in trapping RVF vectors, Aedes mcintoshi and Aedes

Highly recyclable and efficient electrocatalytic synthesis of adipic acid catalyzed by ultra-alkali-resistant [Ni36] nanocages in MOF.

As a pivotal industrial chemical, adipic acid (AA) is synthesizable through the electrocatalytic oxidation of cyclohexanol under ambient conditions, utilizing electrons as a clean oxidant. This approach circumvents the energy-intensive and environmentally detrimental nature of conventional petrochemical processes, thereby offering a sustainable alternative for AA production. However, existing electrocatalysts are often plagued by instability in alkaline media, and their structure-activity relationships remain poorly understood. Here, we report a novel [Ni36] nanocage-based metal-organic framew

Biomass-to-biomass strategy: sulphonic acid functionalized covalent organic frameworks for metal-free esterification of biomass-derived acids.

A sulphonic acid functionalized Covalent Organic Framework (COF-SO3H) was synthesized via an ultrasound-assisted aqueous-phase Schiff base condensation, providing a green and efficient route to incorporate Brønsted acidic sites directly into the framework. Characterisation techniques showed that the resulting material exhibits a partially crystalline, hierarchically porous structure with a surface area of 185 m2 g-1. DFT calculations performed using B3LYP reveal a well-defined hexagonal pore topology with uniformly distributed -SO3H groups along the framework edges. The catalytic activity of C
