The rat cytochrome c oxidase subunit IV gene family: tissue-specific and hormonal differences in subunit IV and cytochrome c mRNA expression.

We have isolated three members of the rat cytochrome c oxidase subunit IV gene family: one functional gene and two processed pseudogenes. The pseudogenes appear to represent the only other closely related sequences in this family. The functional gene encodes an isoform which is expressed in all tissues examined and has features characteristic of 'housekeeping' genes. These include multiple transcription start sites mapped to within an approximately 50 bp region and a GC-rich promoter lacking typical CCAAT or TATAA sequences. Although the subunit IV gene is expressed at its highest levels in ca

Catalytic activity of cytochrome oxidase and cytochrome c in apolar solvents containing phospholipids and low amounts of water.

Cytochrome c and cytochrome oxidase, in bovine heart submitochondrial particles and in their purified forms, were transferred to a ternary system that contained phospholipids (10 mg/ml toluene), the apolar solvent toluene, and water at concentrations of 13-15 microliters (high water) and 3 microliters (low water) per milliliter of toluene. When the enzymes were transferred back to an all water system, they exhibited full catalytic capacity. In the low water ternary system, cytochrome c could be reduced by ascorbate introduced via inverted micelles. Also in this system, cytochrome oxidase was r

Resolution of the electronic transitions of cytochrome c oxidase: evidence for two conformational states of ferrous cytochrome alpha.

Second-derivative absorption spectra are reported for a variety of oxidation and ligation states of bovine cytochrome c oxidase (ferrocytochrome-c:oxygen oxidoreductase, EC 1.9.3.1). The high resolving power of the second-derivative method allows us to assign the individual electronic transitions of cytochrome alpha and cytochrome alpha 3 in many of these states. In the fully reduced enzyme, one observes a single electronic transition at 444 nm, corresponding to the Soret transition for both ferrous cytochrome alpha and ferrous cytochrome alpha 3. When the cytochrome alpha 3 site is occupied b

The structure of the cytochrome a3-CuB site of mammalian cytochrome c oxidase as probed by MCD and EPR spectroscopy.

The nature of the complexes formed between cytochrome c oxidase and the three inhibitory ligands N3-, CN-, and S2- have been investigated by a combination of MCD and EPR spectroscopy. CN- forms a linear bridge between the Fe III a3 and CuB II, suggesting that the distance between these centers in the oxidized enzyme is between 5 and 5.25 A. This distance is too short to permit N3- to form a linear bridge and the evidence suggests this to be bent. In contrast S2- or SH- is unable to form any bridge and it seems likely that two SH- ions are bound by the bimetallic site, one to Fe III a3 and the

Quantifying Redox Dynamics of c-Type Cytochromes in a Living Cell Suspension of Dissimilatory Metal-reducing Bacteria.

To quickly and accurately quantify the redox dynamics of c-type cytochromes (c-Cyts) in a living cell suspension, diffuse-transmission UV/visible (DT-UV/Vis) and normal UV/Vis spectroscopy were used to record spectra of c-Cyts in living Shewanella oneidensis MR-1 bacteria. DT-UV/Vis showed a higher absorbance of c-Cyts and lower background compared with normal UV/Vis, because interference from cell surface scattering was removed. The extinction coefficients of oxidized c-Cyts (410 nm) and reduced c-Cyts (419 and 552 nm) were observed. Using this method and the obtained c-Cyts extinction coeffi

The redox couple of the cytochrome c cyanide complex: the contribution of heme iron ligation to the structural stability, chemical reactivity, and physiological behavior of horse cytochrome c.

Contrary to most heme proteins, ferrous cytochrome c does not bind ligands such as cyanide and CO. In order to quantify this observation, the redox potential of the ferric/ferrous cytochrome c-cyanide redox couple was determined for the first time by cyclic voltammetry. Its E0' was -240 mV versus SHE, equivalent to -23.2 kJ/mol. The entropy of reaction for the reduction of the cyanide complex was also determined. From a thermodynamic cycle that included this new value for the cyt c cyanide complex E0', the binding constant of cyanide to the reduced protein was estimated to be 4.7 x 10(-3) L M(

A pathogenic 15-base pair deletion in mitochondrial DNA-encoded cytochrome c oxidase subunit III results in the absence of functional cytochrome c oxidase.

A 15-base pair, in-frame, deletion (9480del15) in the mitochondrial DNA (mtDNA)-encoded cytochrome c oxidase subunit III (COX III) gene was identified previously in a patient with recurrent episodes of myoglobinuria and an isolated COX deficiency. Transmitochondrial cell lines harboring 0, 97, and 100% of the 9480del15 deletion were created by fusing human cells lacking mtDNA (rho(0) cells) with platelet and lymphocyte fractions isolated from the patient. The COX III gene mutation resulted in a severe respiratory chain defect in all mutant cell lines. Cells homoplasmic for the mutation had no

[Cytochrome P-450 content and activity of NADPH-cytochrome P-450 c-reductase in experimental subarachnoid hemorrhage].

The condition was studied of the brain monooxigenase system (MOS) in experimental subarachnoidal hemorrhage in rats. The results show that the above hemorrhage leads to a significant depression of activity of brain MOS enzymes (cytochrome P-450, NADPN-cytochrome C-reductase). Depression of activity of the enzymic system is manifest from day 1 and has a tendency toward reduction but at day 14 and 21 the indices are still below the control values. Under the exposure to phenobarbital the reduction of activity of the enzyme system in subarachnoidal hemorrhage gets accelerated, with normalization o

Components of the cytochrome system of Alcaligenes sp. N.C.I.B., 11015, with special reference to particulate bound c-type cytochromes.

1. The cytochrome type of the particulate fraction from cells of Alcaligenes sp. was examined. The particulate fraction of aerobically-grown cells contained b-, c-, a-type and o-type like cytochromes, whereas that from nitrate-grown cells contained b-, c-type and o-type like cytochromes. 2. Two kinds of c-type cytochromes were extracted and purified from the particulate fraction of nitrate-grown cells of Alcaligenes sp. One of them showed absorption maxima at 415.5, 522, and 551 nm is the reduced form, and at 410.5 nm in the oxidized form; the other at 417.5, 523, and 556 nm in the reduced for

Copper-Aβ and cytochrome c: a potential Janus in Alzheimer's disease.

Aβ peptides and cytochrome c (Cyt c) co-exist in the mitochondria. The interplay between Aβ and Cyt c is thus, crucial in the context of Alzheimer's disease pathology, where oxidative stress and mitochondrial dysfunction are central features. However, the details of interaction between Cu bound Aβ and Cyt c remain unexplored at a molecular level. Previously it was demonstrated that reduced Cu-Aβ can react with O2 to result in H2O2. This H2O2 produced can oxidize neurotransmitters and this process is catalysed by the oxidized Cu-Aβ. Here spectroscopic analyses reveal that reduced Cu-Aβ transfer

A computational study of the fold and stability of cytochrome c with implications for disease.

Cytochrome c (Cyt-c), encoded by the CYCS gene, is crucial for electron transport, peroxidase activity, and apoptosis. Mutations in CYCS cause thrombocytopenia 4 (THC4), a disorder with low platelet counts. We have, for instance, recently described six Italian families with five different heterozygous missense CYCS variants. These mutations likely enhance peroxidase and apoptotic activities, yet the mechanisms causing reduced platelet production and increased apoptosis are unclear. This study investigates clinically-related Cyt-c variants using an integrated bioinformatics approach. Our findin

A Cytochrome c-Chlorotoxin Hybrid Protein as a Possible Antiglioma Drug.

Malignant gliomas are the most lethal form of primary brain tumors. Despite advances in cancer therapy, the prognosis of glioma patients has remained poor. Cytochrome c (Cytc), an endogenous heme-based protein, holds tremendous potential to treat gliomas because of its innate capacity to trigger apoptosis. To this end, a hybrid cytochrome c-chlorotoxin (Cytc-CTX) protein was biosynthesized to enable cellular uptake of the cell impenetrable Cytc using CTX transporters. A nucleotide sequence containing 1 : 1 Cytc and CTX was constructed and separated by a hexahistidine-tag and an enterokinase cl

Inhibitors of cytochrome c release with therapeutic potential for Huntington's disease.

Release of mitochondrial cytochrome c resulting in downstream activation of cell death pathways has been suggested to play a role in neurologic diseases featuring cell death. However, the specific biologic importance of cytochrome c release has not been demonstrated in Huntington's disease (HD). To evaluate the role of cytochrome c release, we screened a drug library to identify new inhibitors of cytochrome c release from mitochondria. Drugs effective at the level of purified mitochondria were evaluated in a cellular model of HD. As proof of principle, one drug was chosen for in depth evaluati

Influence of copper on the interaction between cytochrome c and sulfite in vitro.

The quantitative influence of copper on the interaction between cytochrome c and sulfite, which is a derivate of sulfur dioxide in the human body, has been studied by fluorescence spectrometry and ultraviolet absorption spectrometry in vitro. The results indicate that copper may intensely combine with protein component and ferroporphyrin component in cytochrome c at the concentration of 0.1 mM, and the respective association constants (K(A)) are 3.77 x 10(4) L mol(-1) and 9.38 x 10(3) L mol(-1). Sulfite has little interaction with the protein component in cytochrome c (K(A) = 0.094 L mol(-1)),

Bax, reactive oxygen, and cytochrome c release in neuronal apoptosis.

Half of all neurons produced during embryogenesis undergo apoptotic death shortly before birth or soon thereafter. Two cell culture models have been used extensively to investigate the cellular and molecular mechanisms underlying apoptosis during neuronal development: (a) sympathetic neurons deprived of their required neurotrophic factor, nerve growth factor, and (b) cerebellar granule neurons deprived of serum in low-potassium medium. A dramatic increase in mitochondrial-derived reactive oxygen species (ROS) occurs during the apoptotic death of both of these cell types. These ROS lie downstre

Aspirin induces apoptosis through mitochondrial cytochrome c release.

Aspirin and other non-steroidal anti-inflammatory drugs induce apoptosis in many cell types. Although the involvement of caspases has been demonstrated, the mechanism leading to caspase activation remains unknown. We have studied the role of the mitochondrial pathway in aspirin-induced apoptosis. The apoptotic effect of aspirin was analyzed in different cell lines (Jurkat, MOLT-4, Raji and HL-60) showing induction of mitochondrial cytochrome c release and caspases 9, 3 and 8 processing. Furthermore, early aspirin-induced cytochrome c release was not affected by the caspase inhibitor Z-VAD x fm

Cytochrome c oxidase deficiencies in the muscle of patients with inflammatory myopathies.

We studied mitochondrial function in inflammatory myopathies, using cytochrome c oxidase (COX) reaction on muscle biopsy samples from 30 patients (15 with dermatomyositis, 12 with polymyositis, and 3 with inclusion body myositis) and 30 age-matched controls. We also performed immunocytochemistry for COX II and COX IV subunits in 7 of these patients who had COX deficiency. COX-deficient fibers were a constant finding in patients or controls older than 65 years and the percentage of COX-deficient fibers correlated with age in both patients and controls. Focal COX deficiency was found in 24 patie
