Electron microscopic demonstration of virus particles in hand, foot and mouth disease.

Early skin and oral mucous membrane lesions of hand, foot and mouth disease (HFMD) were biopsied and investigated by light and electron microscopy. Histology showed spongiosis, intraepidermal splits indicating beginning vesicle formation, exocytosis of lymphoid cells and macrophages, and individual necrotic keratinocytes. Electron microscopy revealed naked viruses with a mean diameter of 24 nm which were arranged in sheets in the cytoplasm. Many keratinocyte nuclei contained tonofibrils. Cellular injury ranged from slight cytoplasmic edema to keratinocyte necrosis. Some nuclei showed dissoluti

Ultrastructure of the mammalian pinealocyte under natural and experimental conditions: quantitative aspects.

This review briefly summarizes data accumulated on the quantitative aspects of the ultrastructure of the mammalian pinealocyte. Quantitative changes have been demonstrated under natural and experimental conditions in pinealocyte cell organelles in various species. Special attention is paid to two cytoplasmic components most frequently studied by means of quantitative electron microscopy, namely, dense-core vesicles and "synaptic" ribbons.

Recruiting ESCRT to single-chain heterotrimer peptide MHCI releases antigen-presenting vesicles that stimulate T cells selectively.

Immune cells naturally secrete extracellular antigen-presenting vesicles (APVs) displaying peptide:MHC complexes to facilitate the initiation, expansion, maintenance, or silencing of immune responses. Previous work has sought to manufacture and purify these vesicles for cell-free immunotherapies. In this study, APV assembly and release is achieved in nonimmune cells by transfecting a single-chain heterotrimer (SCT) peptide major histocompatibility complex I (pMHCI) construct containing an ESCRT- and ALIX-binding region (EABR) sequence appended to the cytoplasmic tail; this EABR sequence recrui

Presence, Tissue Localization, and Gene Expression of the Adiponectin Receptor 1 in Testis and Accessory Glands of Male Rams during the Non-Breeding Season.

Adiponectin (ADIPOQ) is a member adipocytokines, and its actions are supported by two receptors, ADIPOQ receptor 1 and -2, respectively (ADIPOR1 and -R2). Our study was performed to evaluate the ADIPOR1 presence and location and its gene expression in reproductive tissues of the male ram, during its non-breading season. The different portions of the male ram reproductive system (testis, epididymis, seminal vesicle, ampoule vas deferens, bulb-urethral gland) were collected in a slaughterhouse. Immunohistochemistry showed ADIPOR1 positive signals in the cytoplasm of all the glandular epithelial

The thyroid gland of the woodchuck, Marmota monax: a morphological study of seasonal variations in the follicular cells.

The morphology of the thyroid gland of the woodchuck, Marmota monax, was studied during the four seasons of the year. In the spring the thyroid is extremely heterogenous in appearance. Some follicular cells appear quite active. They contain a well defined Golgi apparatus, abundant large colloid droplets and pseudopodia but few, if any, apical vesicles. Other less active cells have poorly defined rough surfaced endoplasmic reticulum and lack a well developed Golgi apparatus. They do not contain apical vesicles or colloid droplets. Summer thyroids have uniformly small follicles which are lined b

Retinol-binding protein 4 downregulation during osteogenesis and its localization to non-endocytic vesicles in human cranial suture mesenchymal cells suggest a novel tissue function.

Craniosynostosis is a developmental disorder of the skull arising from premature bony fusion of cranial sutures, the sites of skull bone growth. In a recent gene microarray study, we demonstrated that retinol-binding protein 4 (RBP4) was the most highly downregulated gene in suture tissue during the pathological process of premature bony fusion. To gain insight into the function of RBP4 in cranial sutures, we analysed primary cells cultured from human cranial suture mesenchyme. These cells express RBP4 but not CRBP1, cellular retinol-binding protein 1, the typical cytoplasmic retinol storage p

[The ultrastructural study of choroid plexus papillomas (author's transl)].

Four cases of choroid plexus papilloma (CPP) obtained at the time of surgical excision were examined by light and electron microscopy and compared with normal choroid plexus (CP) of mouse and chick. Ultrastructurally CPPs were the same as CP except in the following two points: In CCPs 1) pinocytotic vesicles of the vascular endothelium and 2) cytoplasmic filaments were more increased in number than in CP. In one case, which showed malignant changes in some parts, we found prominently increased spot desmosomes and cytoplasmic filaments as compared with other three cases, microcysts at the apica

A freeze-fracture study of muscle fibres infected with Trichinella spiralis.

The muscle fibres of mice containing the infective-stage larvae of the nematode Trichinella spiralis have been studied by means of the freeze-fracturing technique. The larva lies in what appears to be a fluid-filled cavity within the cytoplasm of an altered muscle fibre. There is no membrane separating the cytoplasm of the nurse cell from the cavity surrounding the larva which is therefore truly intracellular, unlike many parasites that reside within a membrane-lined parasitophorous vacuole within the host cell. This altered muscle fibre, known as a nurse cell, lacks myofilaments but does cont

Identification, proteomic profiling, and origin of ram epididymal fluid exosome-like vesicles.

Small membranous vesicles, between 25- and 75-nm diameter, were collected by high-speed centrifugation from the ram cauda epididymal fluid and were found to be normal constituents of this fluid and of the seminal plasma. The SDS-PAGE protein pattern of these vesicles was specific and very different from that of the caudal fluid, seminal plasma, sperm extract, and cytoplasmic droplets. After two-dimensional electrophoresis separation and mass spectrometry analysis, several proteins were identified and grouped into i) membrane-linked enzymes, such as dipeptidyl peptidase IV (DPP-IV), neprilysin

Calcium handling by the cat carotid body--a pyroantimonate study.

Subcellular regulation mechanisms of calcium concentrations related to oxygen sensing in the carotid body are unclear. In the present study, we investigated the ultrastructural distribution patterns of calcium in carotid body cells and its changes evoked by hypoxia. Carotid bodies were dissected from anesthetized cats exposed in vivo to normoxic or acute hypoxic conditions. We used the oxalate-pyroantimonate technique that yields an electron-opaque calcium precipitate. X-ray microanalysis and appropriate controls confirmed the presence of calcium in the precipitate. Calcium precipitates were f

Cystic fibrosis of the pancreas. A study in cell culture.

103 individuals from 16 families with cystic fibrosis and 87 individuals without family history of cystic fibrosis have been studied using the methods of cell culture. Skin fibroblast cultures derived from 19 affected children, and fibroblast cultures from 11 different organs obtained at autopsy from two affected children, showed cellular metachromasia. The morphological appearance and the intracellular mucopolysaccharide content enabled these cultures to be divided into two distinct classes. Class I had discrete cytoplasmic metachromatic vesicles and a mucopolysaccharide content similar to th

Time-resolved quantitative proteomics implicates the core snRNP protein SmB together with SMN in neural trafficking.

The biogenesis of splicing snRNPs (small nuclear ribonucleoproteins) is a complex process, beginning and ending in the nucleus of the cell but including key stages that take place in the cytoplasm. In particular, the SMN (survival motor neuron) protein complex is required for addition of the core Sm proteins to the snRNP. Insufficiency of SMN results in the inherited neurodegenerative condition, spinal muscular atrophy (SMA). Details of the physical organization of the cytoplasmic stages of snRNP biogenesis are unknown. Here, we use time-resolved quantitative proteomics to identify proteins th

Organic secretion by striated ducts.

In addition to their role in electrolyte homeostasis, striated ducts in the parotid and submandibular glands of many mammalian species engage in secretion of organic products. This secretion usually is in the form of serous-like granules that lack substructure. Usually, the granules are in the 2.0-2.5 mm range, but granules smaller than 0.1 mm and larger than 12 mm have been observed. In mustelids, striated duct cells contain rhomboidal crystalloids in their apical cytoplasm; in dogs and at least two bat species, the apical plasmalemma is festooned with perpendicularly-oriented rods. Rather th

Inseparable/IER3IP1 are essential for cytokinesis in Drosophila neuroblasts and human cells.

To unveil the molecular players that maintain neural stem cell (NSC) homeostasis, we conducted a genetic screen in Drosophila and isolated an uncharacterized gene that we named Inseparable (Insep). Insep is the Drosophila homologue of human IER3IP1, a gene associated with Microcephaly, Epilepsy, and Neonatal Diabetes Syndrome-1 (MEDS-1). We show that Insep loss leads to early larval lethality with small brains and these phenotypes can be rescued by expressing IER3IP1 indicating that their biological function is conserved through evolution. The Insep deficient neuroblasts fail to complete cytok

Distribution of sodium, magnesium, phosphorus, sulphur, chlorine, potassium, calcium and iodine in mammalian thyroid gland.

Anhydrous specimen preparation techniques and X-ray microanalysis were used to study the elemental composition of the rat thyroid follicular cells and colloid in the follicular lumen. Analysis were performed in morphologically defined follicular cells and adjacent colloid. Use of semithin cryosections allowed us to selectively analyze cell nucleus and dense vesicles in the cytoplasm of follicular cells. The analytical results indicate that the basolateral and apical cell membrane of follicular cells act differently in the transport of iodine and other ions, thus creating ionic gradient in the

Kinase partner zinc-finger protein modulates pollen tube growth by influencing tip vesicle trafficking.

The polar growth of pollen tubes requires the precise coordination of membrane trafficking processes, such as exocytosis and endocytosis, and the orderly deposition of cell wall materials at the growing tip. We previously identified 2 plasma membrane (PM)-localized receptor kinases, LePRK1 and LePRK2, which form a dynamic complex with the small GTPase-activating protein kinase partner protein and actin-binding proteins to regulate pollen tube growth. Here, we show that kinase partner zinc-finger protein (KPZP) in tomato (Solanum lycopersicum), whose N-terminus interacts with LePRK1 and LePRK2,

Angiotensin II Receptor Antagonism Reduces Transforming Growth Factor Beta and Smad Signaling in Thoracic Aortic Aneurysm.

BACKGROUND: The expression of transforming growth factor beta (TGF-β) and Smad3 regulates extracellular matrix homeostasis and inflammation in aortic aneurysms. The expression of Smad3 depends on signaling by angiotensin II (AngII) receptor pathways through TGF-β receptor-dependent and -independent pathways. METHODS: To determine the expression of AngII type 1 (AT1R) and type 2 receptors (AT2R), TGF-β, and Smad3 in thoracic aortic aneurysms, we performed immunohistochemistry testing on tissue and cultured cells derived from subjects with Marfan syndrome (MFS) and bicuspid aortic valve (BAV) m

Secretion by striated ducts of mammalian major salivary glands: review from an ultrastructural, functional, and evolutionary perspective.

In addition to their role in electrolyte homeostasis, striated ducts (SDs) in the major salivary glands of many mammalian species engage in secretion of organic products. This phenomenon usually is manifested as the presence of small serous-like secretory granules in the apical cytoplasm of SD cells. The composition of these granules is largely unknown, except in the case of the cat and rat submandibular gland, where the granules have unequivocally been shown to contain kallikrein. In some species, the apical cytoplasm of SD cells contains variable numbers of vesicles, both spherical and elong
