Potassium and magnesium aspartate (K,Mg aspartate) is used in treating and preventing cardiac disruptions caused by electrolytic disturbances, primarily low potassium (K) and magnesium (Mg) levels (e.g. in the treatment with cardiac glycosides and diuretic drugs). Widely used, K,Mg aspartate is synthesized from aspartic acid representing a racemic mix of L- and D-stereoisomers. Differences in metabolism and utilisation of D- and L-amino acids probably have an effect on the pharmacological properties of K,Mg L- and D-aspartates, and what is more, pharmacological doses of magnesium and potassium

The complete amino acid sequences of two postsynaptic neurotoxins (toxin-3 and toxin-5) isolated from Malayan cobra (Naja naja sputatrix) venom were determined by direct automated Edman degradation of peptides obtained from digests with various proteases. Toxin-3 and toxin-5 are both short-chain neurotoxins and their amino acid sequences are highly homologous to Naja naja atra and Naja naja philippinensis neurotoxin, respectively. Toxin-3 is unique in possessing aspartic acid (D) as the fifth residue, while all other homologous short-chain neurotoxins have asparagine (N) at the corresponding p

The effects of 8 selective neuroeffector agonists and antagonists were measured in mice in order to identify specific functional changes associated with behavioral sensitization to amphetamine-induced stereotypy. The changes observed included a decreased convulsive threshold to N-methyl-DL-aspartic acid, an increased convulsive threshold to bicuculline, and an increased head-twitch response to 5-hydroxytryptophan. Of these effects, only the persistence of the two convulsive threshold changes correlated with the persistence of the behavioral sensitization. The induction of behavioral sensitizat

In forensic dental medicine, one of the most important endeavors is the estimation of age. We have studied the method of age assessment using the D/L ratio of aspartic acid from the dentin. The age of deciduous teeth has been estimated by measuring the racemization reaction of this amino acid. The results of the present study indicated that the measured D/L ratio for the dentin of deciduous teeth was highly correlated with the real age (r = 0.824 - 0.981 (sigma = +/- 0.5 to +/- 1.3). These results indicate that age can be accurately estimated from the dentin of deciduous teeth, as it can when

Patients with Duchenne muscular dystrophy (DMD) may experience neurobehavioral and cognitive concerns, including psychiatric symptoms, due to the absence of full-length dystrophin (Dp427), frequently accompanied by deficiencies in shorter isoforms. The lack of dystrophin affects neurophysiological processes from the uterine phase, impacting neural circuitry in brain regions such as the prefrontal cortex, hippocampus, and cerebellum. This leads to reduced inhibitory GABAergic transmission and altered hippocampal glutamatergic signaling. The resulting imbalance between inhibitory and excitatory

We investigated the pharmacological actions of a slow-releasing H2S donor, GYY 4137; a substrate for the biosynthesis of H2S, L-cysteine and its precursor, N-acetylcysteine on potassium (K+; 50 mM)-evoked [3H]D-aspartate release from bovine isolated retinae using the Superfusion Method. GYY 4137 (10 nM-10 µM), L-cysteine (100 nM-10 µM) and N-acetylcysteine (10 µM-1 mM) elicited a concentration-dependent decrease in K+-evoked [3H]D-aspartate release from isolated bovine retinae without affecting basal tritium efflux. At equimolar concentration of 10 µM, the rank order of activity was as follows

The K+-stimulated release of [3H]-D-aspartate and [14C]-GABA from synaptosomal (P2) fractions prepared from rat cerebellum was studied. Muscimol enhanced the release of [3H]-D-aspartate by 60-75% and the release of [14C]-GABA by 20-35%. Muscimol also enhanced the release of [3H]-D-aspartate from P2 fractions prepared from swine and mouse cerebellum. Pentobarbital, an anesthetic barbiturate, had no effect on basal or K+-stimulated release of [3H]-D-aspartate or [14C]-GABA but potentiated the enhancement of [3H]-D-aspartate release by muscimol. The EC50 was approx. 50 microM. The S(-)-isomer of

It has been reported that both activation of N-methyl-D-aspartate receptors and acid-sensing ion channels during cerebral ischemic insult contributed to brain injury. But which of these two molecular targets plays a more pivotal role in hypoxia-induced brain injury during ischemia is not known. In this study, the neuroprotective effects of an acid-sensing cation channel blocker and an N-methyl-D-aspartate receptor blocker were evaluated in a rat model of cardiac arrest-induced cerebral hypoxia. We found that intracisternal injection of amiloride, an acid-sensing ion channel blocker, dose-depen

1 The effect of various concentrations of thiopentone, pentobarbitone, methohexitone, hydroxydione, alphaxalone/alphadolone, ketamine, alpha-chloralose, and urethane on the transport of radiolabelled gamma-aminobutyric acid (GABA) and D-aspartate was investigated. 2 Uptake of the amino acids was weakly inhibited, if at all, by the anaesthetics and it is unlikely that such effects contribute significantly to their physiological function. 3 The spontaneous efflux of GABA and D-aspartate was not detectably altered by any of the drugs tested. 4 Thiopentone, pentobarbitone, methohexitone and hydrox

The effects of a convulsant barbiturate, 5(2-cyclohexylidine-ethyl)-5-ethyl barbituric acid (CHEB), and phenobarbitone (PhB) on the release of exogenous D-aspartate and GABA from slices of rat cerebral cortex were investigated. While PhB inhibited potassium-evoked release of D-aspartate more so than that of GABA, CHEB potently inhibited potassium-evoked GABA release and stimulated evoked D-aspartate release, in a concentration-dependent manner. These actions are consistent with the observed in vivo convulsant and anticonvulsant properties of these barbiturates. CHEB, but not PhB also elevated

The involvement of the excitatory amino acids glutamate and aspartate in cerebral ischemia and excitotoxicity is well-documented. Nevertheless, the role of non-excitatory amino acids in brain damage following a stroke or brain trauma remains largely understudied. The release of amino acids by necrotic cells in the ischemic core may contribute to the expansion of the penumbra. Our findings indicated that the reversible loss of field excitatory postsynaptic potentials caused by transient hypoxia became irreversible when exposed to a mixture of just four non-excitatory amino acids (L-alanine, gly

Aleutian mink disease parvovirus (ADV) is the etiological agent of Aleutian disease of mink. Several ADV isolates have been identified which vary in the severity of the disease they elicit. The isolate ADV-Utah replicates to high levels in mink, causing severe Aleutian disease that results in death within 6 to 8 weeks, but does not replicate in Crandell feline kidney (CrFK) cells. In contrast, ADV-G replicates in CrFK cells but does not replicate in mink. The ability of the virus to replicate in vivo is determined by virally encoded determinants contained within a defined region of the VP2 gen

BACKGROUND: Hypertension is a significant global health concern and serves as a critical risk factor for cardiovascular diseases, stroke, and renal failure. Traditional Chinese Medicine (TCM) has been utilized for an extended period to address hypertension, with the syndrome known as Yin deficiency and Yang hyperactivity syndrome (YDYHS) frequently observed in individuals with elevated blood pressure. This syndrome is characterized by symptoms including dizziness, tinnitus, irritability, and insomnia, which are thought to result from an imbalance between Yin, representing the cooling and nouri

The influence of intracaudate administration of N-methyl-D-aspartic acid (NMDA) and of the competitive NMDA receptor antagonist, DL-2-amino-5-phosphonovaleric acid (AP-5) was studied on thyrotropin-releasing hormone (TRH)-induced scratching in rabbits. NMDA (28 nmol) significantly increased the latency of TRH-induced scratching but did not modify the duration of this behaviour. Conversely, AP-5 (0.5 mumol) significantly potentiated scratching duration. Since TRH-induced scratching has been reported to be a dopamine-dependent behaviour, these results suggest that NMDA receptor ligands modulate

Aging has strong genetic components and the list of genes that may regulate the aging process is collected in the GenAge database. There may be characteristic patterns in the amino acid sequences of aging-related proteins that distinguish them from other proteins and this information will lead to a better understanding of the aging process. To test this hypothesis, human protein sequences are extracted from the UniProt database and the relative frequency of every amino acid residue in aging-related proteins and the remaining proteins is calculated. The main observation is that the mean relativ

Snake venom L-aminoacid oxidase, while inactive on 4C and 5C dicarboxylic alpha-amino acids, may oxidize their 6C and 7C homologues alpha-aminoadipic and alpha-aminopimelic acid. It has been demonstrated that the enzyme also oxidizes S-carboxyethyl-L-cysteine and S-carboxymethyl-L-homocysteine, two analogues of alpha-aminopimelic acid with the gamma or delta methylene groups substituted by a sulfur atom. As oxidation products the corresponding ketoacids were obtained. Thus substrate specificity of L-aminoacid oxidase for dicarboxylic alpha-aminoacids seems highly dependent on the carbon chain

Mercuric chloride (MC) and methylmercury (MeHg) were found to increase amino acid release from astrocytes. This suggests interaction with sulfhydryl (-SH) groups which are controlled by glutathione [GSH] levels. In the present study, we evaluated the effects of alterations in intracellular glutathione concentrations [GSH]i on the outcome of MC and MeHg treatment. [GSH]i were increased in a time-dependent fashion by incubating the astrocytes with 1 mM L-2-oxothiazolidine-4-carboxylic acid (OTC), a cysteine precursor. OTC attenuated the release of [2,3-3H]D-aspartic acid from astrocytes exposed

Cultured neocortical neurons, which predominantly consist of GABAergic neurons exhibit a pronounced stimulus-coupled GABA release. Since the cultures may contain a small population of glutamatergic neurons and the GABAergic neurons have a high content of glutamate it was of interest to examine if glutamate in addition to gamma-aminobutyric acid (GABA) could be released from these cultures. The neurons were preloaded with [(3)H]D-aspartate and subsequently its release was followed during depolarization induced by a high potassium concentration or the alpha-amino-3-hydroxy-5-methyl-4-isoxazolepr
