Characterizing seasonal persistence of low levels of Pseudo-nitzschia australis in the Gulf of Maine with environmental DNA.

The potent neurotoxin domoic acid is produced by some species of diatoms in the genus Pseudo-nitzschia, which can lead to amnesic shellfish poisoning (ASP) events in marine food webs. Of the toxin-producing Pseudo-nitzschia, P. australis has caused the most toxic and widespread events, making it a species of concern. Toxic and non-toxic Pseudo-nitzschia species cannot be distinguished easily by light microscopy, necessitating molecular testing. We designed a real-time quantitative PCR assay for rapid and accurate detection of P. australis in environmental DNA (eDNA) samples, targeting the inte

Analysis of Environmental DNA and Edaphic Factors for the Detection of the Snail Intermediate Host Oncomelania hupensis quadrasi.

BACKGROUND: The perpetuation of schistosomiasis japonica in the Philippines depends to a major extent on the persistence of its intermediate host Oncomelania hupensis quadrasi, an amphibious snail. While the malacological survey remains the method of choice in determining the contamination of the environment as evidenced by snails infected with schistosome larval stages, an emerging technology known as environmental DNA (eDNA) detection provides an alternative method. Previous reports showed that O. hupensis quadrasi eDNA could be detected in water, but no reports have been made on its detecti

Structural and thermodynamic consequences of the replacement of zinc with environmental metals on estrogen receptor α-DNA interactions.

Estrogen receptor α (ERα) acts as a transcription factor by virtue of the ability of its DNA-binding (DB) domain, comprised of a tandem pair of zinc fingers, to recognize the estrogen response element within the promoters of target genes. Herein, using an array of biophysical methods, we probe the structural consequences of the replacement of zinc within the DB domain of ERα with various environmental metals and their effects on the thermodynamics of binding to DNA. Our data reveal that whereas the DB domain reconstituted with divalent ions of zinc, cadmium, mercury, and cobalt binds to DNA wi

Diversity and potential environmental risks of DNA viruses on international ships' ballast water at Shanghai port, China.

Over 10 billion tons of ballast water is transferred annually globally, transporting many microorganisms such as bacteria and viruses into new environments and harming local ecology, economy and human health. Numerous studies on bacteria in ballast water have shown its remarkable bacterial diversity and potential risks in recent years. However, the diversity of viruses present in ballast water and their potential environmental implications remain extremely limited in our understanding. In this investigation, we utilized viral metagenomic sequencing to evaluate the viral diversity and distribut

Programmable DNA-mediated coordination polymer nanospheres as highly specific catechol oxidase mimics for biosensing and environmental monitoring.

We leverage adenine-rich single-stranded DNA as both a structural director and a coordination scaffold to synthesize Cu-coordination nanospheres. These nanospheres act as highly selective catechol oxidase mimics, exhibiting no peroxidase-like activity toward TMB or ABTS, thus overcoming the catalytic promiscuity inherent to conventional oxidase nanozymes.

Typhoon Khanun-driven stormwater runoff enhances the detection of terrestrial mammalian environmental DNA in a forest stream.

Monitoring terrestrial mammalian biodiversity remains challenging due to the limitations of conventional survey methods, including high labour requirements and detection bias. This study investigated how an extreme rainfall event, Typhoon Khanun (August 2023), influences the detection of terrestrial mammalian environmental DNA (eDNA) in a stream ecosystem. Samples were collected during the typhoon-induced runoff and again after hydrological conditions returned to baseline. Mammalian eDNA was successfully detected only during the rainfall event, whereas no mammalian DNA was detected in post-eve

Environmental Detection of Pathogenic Leptospira DNA in Agricultural Ecosystems from a Mediterranean-Climate Region of Central Chile.

Although pathogenic Leptospira DNA has been detected in water and soil from different climatic regions, information from Mediterranean-climate agricultural systems remains limited. This study characterized the environmental detection of pathogenic Leptospira DNA in water and soil samples from irrigated agroecosystems of central Chile, evaluating spatial and seasonal variation and associations with selected physicochemical variables. A total of 605 samples were collected from eight agricultural sites during spring 2019, summer 2020, and winter 2021. Samples were analyzed by real-time PCR target

Whole Blood DNA Damage Precedes a Diagnosis of Canine Multicentric Lymphoma and Correlates With Environmental Chemical Exposures.

Canine multicentric lymphoma is a common cancer in dogs without evidence-based prevention measures. While breed accounts for part of lymphoma risk, environmental exposures might also contribute. Human non-Hodgkin lymphoma (NHL), which resembles canine lymphoma, is associated with exposures to volatile organic compounds (VOCs) and herbicides. Pet dogs are commonly exposed to these genotoxic chemicals, but it is unknown whether such exposures are associated with in vivo DNA damage as a potential contributor to lymphoma in dogs or whether early DNA damage can be detected before lymphoma diagnosis

Elucidation of the life cycle of the trematode Curtuteria arguinae (Digenea: Himasthlidae), using environmental DNA detection methods.

Detection approaches based on environmental DNA (eDNA) are widely used for free-living species but remain underutilized for parasite species. This study applies eDNA detection methods to elucidate the life cycle of the trematode Curtuteria arguinae, which infects the socioeconomically and ecologically important edible cockle (Cerastoderma edule) as its second intermediate host along the northeastern Atlantic coast, including Arcachon Bay, France. The first intermediate and definitive hosts remained unknown. To identify these hosts - presumed to be a gastropod and a shorebird - we developed a q

Methodological assessment for efficient collection of Schistosoma mansoni environmental DNA and improved schistosomiasis surveillance in tropical wetlands.

Schistosomiasis, caused by trematodes of genus Schistosoma, is among the most seriously neglected tropical diseases. Although rapid surveillance of risk areas for Schistosoma transmission is vital to control schistosomiasis, the habitat and infection status of this parasite are difficult to assess. Environmental DNA (eDNA) analysis, involving the detection of extra-organismal DNA in water samples, facilitates cost-efficient and sensitive biomonitoring of aquatic environments and is a promising tool to identify Schistosoma habitat and infection risk areas. However, in tropical wetlands, highly

Impact assessment of ephemeral discharge of contamination downstream of two legacy base metal mines using environmental DNA.

Mining and processing metalliferous ores can degrade the environment well beyond the footprint of the mine, particularly where on-site containment and post-mining remediation has been insufficient to prevent releases of solid and aqueous mine wastes. In this study, we investigated the potential of sediment and water chemistry coupled with environmental (e)DNA metabarcoding to evaluate discrete and cumulative ecological impacts of two legacy base metal (copper (Cu), zinc (Zn), lead (Pb)) mines (Peelwood and Cordillera) which discharge metals via ephemeral tributaries into perennial Peelwood Cre

Profiling of environmental Legionella pneumophila strains by randomly amplified polymorphic DNA method isolated from geographically nearby buildings.

Legionella pneumophila (L. pneumophila) which is also known as etiologic agent Legionnaires Disease lives in natural water and man made water systems. These bacteria belonging to Legionellaceae family are divided 15 serogroups. Phenotypical methods used for the identification of Legionella isolates are not very discriminatory. In this study we investigated genotypic features of eight L. pneumophila serogroup 1 and 18 L. pneumophila serogroup 2-14 strains isolated from different buildings in Istanbul by randomly amplified polymorphic DNA (RAPD) method. Eight L. pneumophila serogroup 1 strains (

Applications of environmental DNA monitoring for seaweed reproductive phenology: A case study with giant kelp (Macrocystis pyrifera).

Monitoring the seasonal reproductive cycles of seaweeds is crucial for effective population and ecosystem management, as well as mariculture seedstock collection. Traditional methods, such as visual monitoring by SCUBA diving or snorkeling, are costly, labor-intensive, and limited in temporal and spatial coverage. This study explores substituting these methods with environmental DNA (eDNA) techniques for giant kelp (Macrocystis pyrifera, order Laminariales). This laboratory study aimed to determine the minimum detectable concentration of zoospores and sporophyte tissue needed for detecting the

Comparative Assessment of Environmental DNA and Bulk-Sample Metabarcoding in Biosecurity Surveillance for Detecting Biting Midges (Ceratopogonidae).

Biting midges, Culicoides spp. (Diptera: Ceratopogonidae), are significant vectors capable of transmitting arboviruses, such as bluetongue virus, to livestock. New Zealand is free of Culicoides, and a national surveillance programme is in place for the early detection of an incursion. Traditionally, insect trap samples from the surveillance programme are analyzed using morphology-based diagnostics under microscopes, which is time-consuming and relies on specialized taxonomic expertise. Here, we assessed the effectiveness of DNA metabarcoding using insect bulk samples and environmental DNA (eDN

Environmental detection of eumycetoma pathogens using multiplex real-time PCR for soil DNA in Sennar State, Sudan.

BACKGROUND: Mycetoma is a chronic disease affecting the skin and subcutaneous tissue endemic in the tropical and subtropical regions. Several bacteria and fungi can cause mycetoma, but fungal mycetoma (eumycetoma) is challenging because the treatment requires a combination of a long-term antifungal agent and surgery. Although the transmission route has not yet been elucidated, infection from the soil is a leading hypothesis. However, there are few soil investigation studies, and the geographical distribution of mycetoma pathogens is not well documented. Here, we used multiplex real-time PCR te

A systematic survey of environmental DNA in Palau's lakes and waterfalls reveals an increase in Leptospira levels after flooding.

OBJECTIVE: Leptospirosis is an important bacterial zoonosis which is widespread in tropical and subtropical islands and influences human and animal health which has secondary economic effects. Although leptospirosis is endemic in Palau, an Oceanian Pacific Island country, few systematic surveys of potential risk factors for Leptospira infection, such as weather and host animals, have been conducted in the natural environment. We used environmental DNA metabarcoding to assess the distribution, species diversity, and abundance of pathogenic Leptospira in this endemic region to investigate the po

Use of passive sampling in environmental DNA metabarcoding technology: Monitoring of fish diversity in the Jiangmen coastal waters.

Environmental DNA (eDNA) metabarcoding technology is proving to be the most up-to-date and promising method for monitoring marine fish diversity. Fish eDNA is usually collected on a filter membrane after the filtration of water. Not only does this require the use of specialized equipment, but the amount of filtered water needed is also difficult to meet. The recently proposed passive eDNA collection method can expand the sampling scale, providing new perspectives for monitoring marine biodiversity. The role of collection methods in eDNA surveys, however, remains unclear. In this study, a low-c
