Unroughing the cat's tongue mushrooms: Four new species of Pseudohydnum from Brazil based on morphological and molecular phylogenetic evidence.

Pseudohydnum, commonly known as cat's tongue mushrooms, is a monophyletic assemblage within Auriculariales, which encompasses species with gelatinous basidiomata, spathulate, flabellate, or shell-shaped pileus, hydnoid hymenophore, globose to ellipsoidal basidiospores, and longitudinally cruciate-septate basidia. According to the available literature, 16 species have been described in Pseudohydnum, mostly represented in temperate-boreal forests of the Northern Hemisphere. However, the limited morphological, molecular, and ecological information, especially from the Southern Hemisphere ecosyste

The lichen symbiont Wadeana represents an independent origin of hysterothecial fungi in the Eurotiomycetes.

The genus Wadeana was erected almost fifty years ago to accommodate an unusual pair of lichen fungal symbionts with hysterothecial ascomata and polysporous asci associated with the algal genus Trentepohlia. The evolutionary position of Wadeana has long been unclear, and the genus has variably been treated as a dothideomycete, as incertae sedis or, recently, within the subclass Acarosporomycetidae of the Lecanoromycetes. Here we present multilocus evidence from metagenome-assembled genomes from the generitype, Wadeana dendrographa, that place it with strong support as sister to all genera curre

Enhancing ascitic fungal infection diagnosis through next-generation sequencing: a pilot study in surgical ICU patients.

OBJECTIVES: Ascites, often associated with critical pathologies such as liver cirrhosis or bowel perforation, can be complicated by fungal infection, increasing mortality especially in intensive care settings and demanding rapid diagnosis and adequate treatment. Traditional microbiological diagnostic methods have limited sensitivity in accurately identifying fungal pathogens in ascitic fluid. Alternative diagnostic methods may offer important insights to enable guiding of antifungal therapy and refining empirical treatment strategies. The objective of this study was to evaluate the potential o

[Identification of the agents of coccidioidomycosis using polymerase chain reaction].

Two pairs of primers for diagnosis of coccidioidomycosis using the method of PCR were constructed. One pair was used for identification of the two species of Coccidioides (C. immitis and C. posadasil) on the basis of MBP-1 gene. The other pair was chosen on the basis of SOWgp82 gene, which encodes an immunodominant, spherule outer wall glycoprotein for detecting only C. posadasii. The used primers allowed the agents of coccidioidomycosis to be detected using PCR with high sensitivity and specificity. The effective method of isolation of fungus DNA from soil contaminated with arthroconidia of C

Contamination of liposomal bupivacaine during multi-dose usage in a clinical setting evaluated by culture and PCR.

INTRODUCTION: Liposomal bupivacaine, a long-acting local anesthetic, is sold in single-dose vials at a cost of approximately $200/20 mL vial. As many veterinary patients are not dosed an entire vial, the vials have been used for multiple doses at our institution to provide cost savings. Multiple punctures of a vial can lead to increased opportunity for contamination of the contents. This study aims to describe our institutional procedure for multi-dose use of single-dose liposomal bupivacaine vials and to evaluate clinically utilized liposomal bupivacaine for bacterial and fungal contamination

DNA Aptamers Targeting BcSOD1: A Novel Strategy for Controlling Botrytis cinerea in Sustainable Agriculture.

Botrytis cinerea, the necrotrophic fungus responsible for grey mould disease, is a major threat to global crop production. Control strategies mainly rely on chemical fungicides, but resistance development limits their long-term effectiveness. This study introduces, for the first time in crop protection, the use of DNA aptamers as a novel and sustainable strategy. Aptamers are short, single-stranded DNA molecules that bind specific targets with high affinity, acting like 'chemical antibodies'. Using SELEX technology, two aptamers, SOD9.14F and SOD9.26F, were designed to target BcSOD1, a superox

Microbial exposure and diversity in Norwegian shrimp processing plants.

Seafood processing workers have a high prevalence of respiratory symptoms and occupational asthma, primarily attributed to allergenic protein exposure. However, exposure to airborne microorganisms from raw materials can also contribute to allergic sensitization and other respiratory ailments. This study aimed to assess microbial exposure in shrimp processing plants and identify susceptible work tasks. Full-shift personal air samples were collected from two Norwegian shrimp processing plants across five distinct work processes: thawing, truck driving, cooking-peeling (technician), packing, and

The development of a multiplex recombinase polymerase amplification reaction to detect the most common causative agents of eumycetoma.

PURPOSE: Mycetoma is a neglected tropical disease that affects the subcutaneous tissue. The disease can be caused by over 90 different pathogens, including both bacteria (actinomycetoma) and fungi (eumycetoma). While diagnostic tools for eumycetoma causative agents are available, these are generally not well suited for use in endemic regions. This study aims to develop an isothermal based multiplex recombinase polymerase amplification reaction (RPA), that can be integrated in the diagnostic workflow of endemic regions. METHODS: The RPA was designed targeting the Internal Transcribed Spacer (I

Identification of hypocrealean reptile pathogenic isolates with MALDI-TOF MS.

Biotyper analysis of Nannizziopsis guarroi, a fatal fungal pathogen in lizards, was described recently. Hypocrealean fungal infections in captive reptiles appear with an increasing frequency during the last decade. Therefore, the aim of this study was to proof Matrix-assisted laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF MS) as diagnostic tool for the identification of reptile pathogenic hypocrealean fungi. Ten fungal isolates obtained from nine reptiles with fungal glossitis, disseminated visceral mycosis, pneumomycosis, and fungal keratitis were analyzed. Phylogeny

Detection of fungal and bacterial organisms from clinical uterine samples of horses using next-generation sequencing in comparison with culture.

Fungal and bacterial culture is currently the primary method for pathogen detection and identification. Next-generation sequencing is a powerful method for detecting and identifying the presence of microbial DNA in samples. We evaluated the correlation between fungal and bacterial culture with next-generation sequencing in equine uterine samples. Fungal cultures (n=63) were evaluated based on their culture results. In culture positive (n=16) samples, next-generation sequencing identified the same organism in 10 samples (62.5%), 5 samples did not identify fungal agents, and 1 sample identified

Onychomycosis insensitive to systemic terbinafine and azole treatments reveals non-dermatophyte moulds as infectious agents.

BACKGROUND: Dermatophytes are the main cause of onychomycoses, but various non-dermatophyte filamentous fungi are often isolated from abnormal nails. OBJECTIVE: Our aim was the in situ identification of the fungal infectious agent in 8 cases of onychomycoses which could not be cured after systemic terbinafine and itraconazole treatment. METHODS: Fungal DNA was extracted from nail samples, and infectious fungi were identified by restriction fragment length polymorphism (RFLP) of amplified fungal ribosomal DNA using a previously described PCR/RFLP assay. RESULTS: PCR/RFLP identification of fu

First molecular record of Panaeolus subbalteatus (Psathyrellaceae, Agaricales) from Pakistan.

Understanding the distribution of agaric fungi through molecular phylogeny is critical for biodiversity assessment. This comprehensive investigation documented the presence of Panaeolus subbalteatus, a previously unreported agaric fungus, addressing a significant gap in the country's knowledge of fungal diversity. This psilocybin-containing mushroom species, known for its neurotoxic properties, was collected from humus-rich soil in the Gujranwala district of Punjab province. The study employed integrated morpho-anatomical and molecular approaches, including detailed microscopic examination of

[Use of rifampicin for directed modification of the nucleotide composition of the cells of Bacillus megaterium and Candida utilis].

The object of this work was to study the effect of rifampicin on the physiological state of Bacillus megaterium and Candida utilis in chemostat and batch cultures. When B. megaterium is cultivated in a medium containing 1 microgram of rifampicin per litre, its metabolism changes, the pool of free nucleotides rises, and the content of RNA drops in response to the specific action of the inhibitor. The modified state is observed throughout many generations. The action of the inhibitor is also non-specific: protein content falls and polyhydroxybutyric acid is accumulated. The morphology of B. mega

ERIC- and REP-PCR amplify non-repetitive fragments from the genome of Drechslera avenae and Stemphylium solani.

In the present study, the homology relationships among fragments amplified by ERIC (Enterobacterial Repetitive Intergenic Consensus) and REP (Repetitive Extragenic Palindromic) primers in Drechslera avenae and Stemphylium solani causing leaf blotch and black spot of oats and leaf blight of cotton, respectively, were investigated by Southern hybridization followed by DNA sequencing. ERIC- and REP-PCR were performed for 24 isolates of D. avenae and 33 isolates of S. solani. The banding patterns obtained were transferred to nylon membranes and a total of 16 fragments, four ERIC and four REP bands

The value of CMV and fungal PCR for monitoring for acute leukaemia and autologous stem cell transplant patients.

Both CMV and fungal infections have been suggested to be causes of fever of unknown origin (FUO) in neutropenic patients. The aim of this prospective, blinded study was to use nucleic acid techniques for monitoring of 20 acute leukemia and 15 autologous stem cell transplant (SCT) patients. Blood samples were taken weekly and examined for fungal and CMV DNA by PCR and CMV mRNA by NASBA. 387 samples were analysed. Fungal DNA was detected in 9 samples. Four samples were positive for Aspergillus and 6 for Candida DNA (1 sample positive for both). Candida PCR was positive in 2 patients with FUO, 1

Rapid identification of fungi by using the ITS2 genetic region and an automated fluorescent capillary electrophoresis system.

Invasive fungal disease often plays an important role in the morbidity and mortality of immunocompromised patients. The poor sensitivity of current fungal blood culture and histological practices has led to the development of highly sensitive and specific molecular techniques, such as the PCR. Sequence variability of the internal transcribed spacer 2 (ITS2) region of fungi is potentially useful in rapid and accurate diagnosis of clinical fungal isolates. PCR with fungus-specific primers targeted toward conserved sequences of the 5.8S and 28S ribosomal DNA (rDNA) results in amplification of the

Anticancer drugs targeting topoisomerase II for antifungal treatment.

Fungal topoisomerase II (TopoII) has been identified as essential for viability. Thus, our research aimed to investigate the potential of fungal TopoII as a novel target for antifungal chemotherapy. We conducted studies on eleventh antitumor compounds targeting human topoisomerase II, either approved by the U.S. Food and Drug Administration (FDA) or currently under clinical trials to evaluate their potential for use in other therapeutic applications. While most of the compounds we analyzed are potent inhibitors of yeast TopoII, only a few exhibited antifungal activity. Idarubicin emerged as th

Detection of Histoplasma DNA from Tissue Blocks by a Specific and a Broad-Range Real-Time PCR: Tools to Elucidate the Epidemiology of Histoplasmosis.

Lack of sensitive diagnostic tests impairs the understanding of the epidemiology of histoplasmosis, a disease whose burden is estimated to be largely underrated. Broad-range PCRs have been applied to identify fungal agents from pathology blocks, but sensitivity is variable. In this study, we compared the results of a specific Histoplasma qPCR (H. qPCR) with the results of a broad-range qPCR (28S qPCR) on formalin-fixed, paraffin-embedded (FFPE) tissue specimens from patients with proven fungal infections (n = 67), histologically suggestive of histoplasmosis (n = 36) and other mycoses (n = 31).
