Lipid membranes are fundamental elements of cells, serving as barriers that protect the cell interior from the external environment. DNA nanostructures, which can engineer lipid membranes' signal transduction and substance exchange properties, attract interest for their potential applications in the biomedical field. The interaction between DNA nanostructures and lipid membranes is of scientific and technological interest. Here, we investigate the interaction between DNA nanopores (DNPs) and model supported lipid bilayers (SLBs) using real-time quartz crystal microbalance with energy dissipati

Herein we describe one-dimensional electron-spin arrays consisting of two different organic radicals with the designed arrangement based on the DNA sequence. Two mismatch-binding ligands carrying 2,2,6,6-tetramethylpiperidine N-oxide (TEMPO) and nitronyl nitroxide selectively bind to the predetermined sites on double stranded DNA. By using the two mismatch-binding ligands carrying the organic radicals as the glue for DNA, electron-spin assembly could be successfully synchronized with the hybridization. Periodically and tandemly arranged, two kinds of organic radical molecules at designed posit

Structural DNA nanotechnology offers a unique self-assembly toolbox to construct soft materials of arbitrary complexity, through bottom-up approaches including DNA origami, brick, wireframe, and tile-based assemblies. This toolbox can be expanded by incorporating interactions orthogonal to DNA base-pairing such as metal coordination, small molecule hydrogen bonding, π-stacking, fluorophilic interactions, or the hydrophobic effect. These interactions allow for hierarchical and long-range organization in DNA supramolecular assemblies through a DNA-minimal approach: the use of fewer unique DNA se

Reversible functionalization of DNA nanostructures is essential for their integration into dynamic biological constructs and biomedical applications. Current strategies typically rely on strand displacement or stimuli-responsive sequences, while approaches based on dynamic covalent chemistry remain largely unexplored. Yet, dynamic covalent bonds combine covalent stability with reversible exchange under mild conditions, offering adaptable and biologically compatible routes to modulate DNA nanostructures. Here, we report iminoboronate conjugation as a strategy for the reversible functionalizatio

The emerging carbon nanomaterial-based field-effect transistor (FET) biosensing technologies promise bridging the performance gap between experiments and applications in the field of point-of-care testing (POCT) due to the advantages of ultrahigh sensitivity, fast, label-free detection, and potential for integration. The specific recognition and signal transduction of these sensors are achieved by a biomolecular interface layer at the solid-liquid interface, in which the arrangement and controllable assembly of this layer are crucial for developing the sensor potential. Rheumatoid arthritis (RA) remains a challenging autoimmune disease due to its complex and heterogeneous pathophysiology, which complicates therapeutic and diagnostic efforts. Advances in DNA nanotechnology have introduced DNA nanomaterials as promising tools to overcome these barriers. This review focuses on three primary categories of DNA nanomaterials applied in RA: DNA nanostructures, DNA aptamers, and DNA-modified nanoparticles. DNA nanostructures, such as tetrahedral framework nucleic acids (tFNAs) and DNA origami, demonstrate anti-inflammatory properties and facilitate precise, controlle

A novel phthalonitrile derivative (a) containing three functional groups (hexyl, aminated ester, phenoxy) was synthesized and subsequently cyclotetramerized in the presence of the corresponding metal chloride salts to obtain hexadeca-substituted metal {M = Cu(II) and Co(II)} phthalocyanines (b and c). The water-soluble phthalocyanines (d and e) were prepared by treating the newly synthesized complexes (b and c) with methyl iodide. Moreover, gold nanoparticles (1) and silver nanoparticles (2) were prepared, and their surfaces were modified with quaternary phthalocyanines (d and e). The biologic

DNA nanostructures are promising drug delivery platforms but often suffer from limited structural stability and high production complexity. Cisplatin can cross-link DNA and act as a therapeutic agent, offering a simplified approach to stabilize DNA nanostructures while incorporating therapeutic functionality. Here, we explore cisplatin-mediated compaction of a single-stranded DNA scaffold to form stable nanoparticles and evaluate their potential as radiosensitizers. Cisplatin-induced cross-linking produced compact DNA nanoparticles with size and morphology controlled by mixing ratio and incuba

The liquid-liquid phase separation of biomolecules is an important process for intracellular organization. Biomolecular sequence combinatorics leads to a large variety of proteins and nucleic acids which can interact to form a diversity of dense liquid ("condensate") phases. The relationship between sequence design and the diversity of the resultant phases is therefore of interest. Here, we explore this question using the DNA nanostar system which permits the creation of multiphase condensate droplets through sequence engineering of the sticky end bonds that drive particle-particle attraction.

DNA self-assembly is a well-understood nanotechnology to obtain extremely ordered structures from the nanometer to the hundreds of microns scale. In contrast, DNA hydrogels rely on the disordered assembly of DNA building blocks to reach macroscopic volumes. However, in order to hold the promise of DNA bulk materials, the sequence designer needs a systematic understanding of how macroscopic properties emerge from disorder. Here, we show a method to study systematically the mechanical response of a simple DNA nanostar hydrogel. This method mobilises bulk rheology, dynamic light scattering micror

Sequence-specific detection of double-stranded DNA (dsDNA) under physiological conditions remains challenging because the complementary strand is inaccessible to conventional oligonucleotide probes, often requiring thermal denaturation or protein-assisted strand separation. Here, we developed a binary DNAzyme (BiDz) sensor for sequence-specific dsDNA detection at 37 °C. While the all-DNA BiDz failed to recognize dsDNA efficiently, incorporating alternating locked nucleic acid (LNA)/DNA residues into the analyte-binding arms enabled efficient strand invasion and markedly improved signal generat

Nucleic acid nanotechnology has emerged as a versatile tool for the rational design of smart nanoscale architectures to function within cells or at the cell surface. Key features that make these nanostructures particularly well suited for intracellular use include their predictable geometry, modular design, precise spatial addressability, large repertoire of functional components, biocompatibility, capacity to perform molecular logic operations, and stimuli-responsive cargo capture and release. In this review, we highlight recent advances in the use of nucleic acid nanotechnology as synthetic

The lack of organelle-targeted neurotransmitter probes limits understanding of their intracellular roles. Here we created an organelle-targeted neurotransmitter nanoprobe using specific molecule-trapped DNA nanostructures for multiple recognition effects. In particular, we designed phenylboronic acid derivatives for chemical reaction and hydroxymethyl groups for forming hydrogen bonding with norepinephrine (NE), which were confined into tetrahedral DNA nanostructures with the optimized spatial effects, achieving the specific and rapid NE identification. Moreover, cyanine 3 providing built-in c

Reactive oxygen species (ROS) at elevated levels trigger oxidative DNA damage, which is a significant factor in psoriasis exacerbation. However, normal ROS levels are essential for cell signaling, cell growth regulation, differentiation, and immune responses. To address this, we developed ROS control strategies inspired by compensatory effects. DNA nanostructures with the advantage of being more stable than linear nucleic acid molecules in physiological environments were exquisitely fabricated and incorporated into microneedles (MN). These nanostructures regulate ROS levels and facilitate the

Functionalizing DNA nanostructures for stimuli-responsive reconfiguration has been a key area of development in DNA nanotechnology. Here, we demonstrate programmable control over the structural integrity of a tetrahedral DNA nanostructure using an external DNA stimulus that is modulated through an intermediate CRISPR-Cas12a reaction. We designed toehold-containing DNA tetrahedra that can be disassembled by a DNA or RNA invading strand. Activation of the Cas12a enzyme by double-stranded DNA provides precise control over the disassembly of the DNA tetrahedra by digesting the DNA invading strand

DNA nanostructures are programmable, biocompatible platforms for biomedicine; however, their mechanical properties are coupled to sequence and structural dimensions, limiting control over stiffness. Here, we report a paradigm shift from static design to mechanical engineering of DNA nanostructures. Using a nanoemulsion interfacial-confined coordination assembly strategy, hollow DNA nanocapsules (HDCs) were constructed with mechanical properties decoupled from chemical identity, without hard templates. By tuning the nanoemulsion-to-DNA-solution ratio, HDCs with sizes (∼160 nm) and surface charg

Hepatitis B virus (HBV) infection is associated with the progression of liver disease. Occult HBV infection (OBI) is defined as the existence of detectable HBV DNA in HBV surface antigen negative individuals. However, HBV DNA is negative in serum while HBV surface antigen remains positive in incompletely cured chronic HBV infection. Hence, combined detection of HBV surface antigen and HBV DNA is essential for the accurate detection and rehabilitation of HBV infection. Therefore, a multiplex detection strategy based on branched DNA nanostructures was developed. The single-stranded segment of a

A multilayer core-shell architecture capable of selectively capturing distinct nanomaterials into a predetermined layer is anticipated to facilitate multifunctional isolation or cascade processes at the microscale. However, fabricating single-crystal architectures featuring numerous shells with intricate distribution patterns remains challenging. In this study, we employed DNA nanotechnology to construct microscale multilayer core-shell DNA origami crystals. The internal layers exhibit tunable properties, including size, quantity, self-similarity, and morphology. The shell size can be regulate

Tailoring the properties of 2D materials through precise control of both local arrangement and long-range order on surfaces remains a central challenge in materials science. While surface-assisted assembly of symmetric, non-interacting DNA origami nanostructures offers a facile route to 2D superlattices, existing approaches typically yield close-packed arrays with limited control over inter-origami spacing. Here, we present a tunable and reproducible strategy for regulating the lateral spacing of DNA origami during self-assembly into macroscopic 2D lattice patterns. By controlling the growth o

Owing to doxorubicin's high DNA binding affinity, doxorubicin-loaded DNA origami nanostructures (DOX-DONs) are promising nanocarriers against cancer. However, understanding the interactions between doxorubicin (DOX) and DNA origami nanostructures (DONs) is important to ensure the quality of DOX-DONs. This interaction is often taken for granted and the influence of DOX loading conditions is poorly characterized. Exploiting the inherent fluorescence of DOX, steady-state and time-resolved fluorescence anisotropy spectroscopy techniques are used for characterizing nondestructively the binding betw
