Ginsenoside Rb1 restores dental pulp by promoting odontogenic differentiation of dental pulp stem cells via the TGF-β/Smad signaling pathway.

BACKGROUND: Dental surgery is a common treatment for pulpitis, which can lead to decreased pulp vitality. A novel approach is needed to induce reparative dentin formation without pulp removal. Ginsenoside Rb1 (G-Rb1) is known for its regenerative properties in various diseases. This study aimed to examine the odontogenic differentiation-promoting effects of G-Rb1 in human dental pulp stem cells (hDPSCs) and a rat pulpotomy model, and explore the underlying molecular mechanisms. METHODS: hDPSCs were extracted from supernumerary teeth and cultured with 10 μg/mL G-Rb1. Odontoblastic differentiat

[Research progress in autophagy in the development of pulp inflammation and dental pulp injury reparation and regeneration].

Pulpitis is an infectious disease characterized by persistent inflammation of dental pulp and severe pain of patients, root canal treatment increases the risk of tooth fracture, discoloration and reinfection. Therefore, pulp injury repair and pulp regeneration become the new targets of pulpitis treatment. Autophagy is considered as an important defense and protective mechanism, thus plays an important role in preventing the host from excessive inflammatory reaction. There are few reports on the regulative mechanisms and therapeutic strategies of autophagy on pulp inflammation progression, ther

A Decellularized Matrix Hydrogel Derived from Human Dental Pulp Promotes Dental Pulp Stem Cell Proliferation, Migration, and Induced Multidirectional Differentiation In Vitro.

INTRODUCTION: Dental pulp is a major composition in the pulp-dentin complex, which serves as protective system against dental trauma/infection. Functional dental pulp regeneration is highly desirable after pulpitis or pulp necrosis. However, endodontic regeneration has remained challenging for decades because of the deconstructive microenvironment and the lack of functional cells within the root canal system. The present study developed a decellularized matrix hydrogel derived from human dental pulp (hDDPM-G), which might serve as a growth-permissive microenvironment for dental pulp regenerati

Potential dental pulp revascularization and odonto-/osteogenic capacity of a novel transplant combined with dental pulp stem cells and platelet-rich fibrin.

Our aim is to investigate the cytobiological effects of autologous platelet-rich fibrin (PRF) on dental pulp stem cells (DPSCs) and to explore the ectopic and orthotopic possibilities of dental pulp revascularization and pulp-dentin complex regeneration along the root canal cavities of the tooth by using a novel tissue-engineered transplant composed of cell-sheet fragments of DPSCs and PRF granules. Canine DPSCs were isolated and characterized by assaying their colony-forming ability and by determining their cell surface markers and osteogenic/adipogenic differentiation potential. The biologic

Inhibition of lipoxygenase of rat dental pulp and human platelets by phenolic dental medicaments.

The effects of phenolic dental medicaments on lipoxygenase activities of rat dental pulp and human platelets were studied. The major product derived from [14C] arachidonic acid by the homogenate of rat dental pulp was 12-HETE (15-HETE). Eugenol and p-chlorophenol dose-dependently inhibited HETEs formation. The IC50 values of eugenol and p-chlorophenol were 0.62 and 0.34 mM respectively. The concentrations of these compounds that inhibit lipoxygenase were similar to those required to inhibit cyclooxygenase. These compounds also inhibited 12-lipoxygenase of human platelets with a similar range o

Dental Pulp Vascular Response to Early Stages of Caries.

OBJECTIVES: During caries progression, dental pulp is increasingly pathologically affected. Since the accurate assessment of pulp is of vital importance in clinical decision-making, this study aimed to evaluate pulpal condition in the early stages of caries via laser Doppler (LD) flowmetry and histologic analysis and determine their agreement. METHODS: Fourteen patients with severe dental crowding were included. Prior to extractions and orthodontic treatment, dental pulp condition of 52 premolars was evaluated via LD flowmetry. Teeth were assessed for the presence of caries and lesions were g

Intercellular mitochondrial transfer alleviates pyroptosis in dental pulp damage.

Mitochondrial transfer is emerging as a promising therapeutic strategy for tissue repair, but whether it protects against pulpitis remains unclear. Here, we show that hyperactivated nucleotide-binding domain and leucine-rich repeat protein3 (NLRP3) inflammasomes with pyroptotic cell death was present in pulpitis tissues, especially in the odontoblast layer, and mitochondrial oxidative stress (OS) was involved in driving this NLRP3 inflammasome-induced pathology. Using bone marrow mesenchymal stem cells (BMSCs) as mitochondrial donor cells, we demonstrated that BMSCs could donate their mitochon

Down-regulated genes in mouse dental papillae and pulp.

Important factors involved in odontogenesis in mouse dental papillae disappear between the pre- and post-natal stages of development. Therefore, we hypothesized that certain genes involved in odontogenesis in dental papillae were subject to pre-/post-natal down-regulation. Our goal was to identify, by microarray analysis, which genes were down-regulated. Dental papillae were isolated from embryonic 16-day-, 18-day- (E16, E18), and post-natal 3-day-old (P3) murine first mandibular molar germs and analyzed by microarray. The number of down-regulated genes was 2269 between E16 and E18, and 3130 b

Terrein reduces pulpal inflammation in human dental pulp cells.

Terrein is a bioactive fungal metabolite whose anti-inflammatory properties are virtually unknown. The purpose of this study was to determine the effects of terrein on lipopolysaccharide (LPS)-induced expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) in human dental pulp cells and to determine the mechanism of the observed effects. The LPS-induced expression of ICAM-1 and VCAM-1 was inhibited by terrein in both a time- and dose-dependent manner. LPS-stimulated translocation of nuclear factor kappa B (NF-kappaB) into the nucleus, which was b
