[Study of the fine structure of human deciduous dentin with dentinogenesis imperfecta, with special reference to the mantle dentin].

A lower deciduous incioer exhibiting dentinogenesis imperfecta (D.I) obtained from a 6-year-old boy with osteogenesis imperfecta (Shields' Type I) was examined by means of light microscopy (LM), scanning electron microscopy (SEM), and X-ray microanalysis (XMA). With LM, the dentin displayed a sparse and irregular tubular pattern near the dentino-enamel junction (DEJ) and only few or no tubular structures in the area corresponding to the circumpulpal dentin. Between these two areas, cleft-like structures were characteristically noted. Structural irregularities in the dentinal tubules were also

Efficacy of a desensitizing toothpaste containing arginine and calcium carbonate on dentin surface pore structure and dentin morphology.

PURPOSE: To investigate the laboratory changes in dentin tubule occlusion morphology during short term use of desensitizing products as evaluated by electron microscopy and an image analysis. METHODS: Freshly extracted human third molar teeth were collected at random and 40 dentin discs were prepared. These dentin samples were then divided in to four groups (n=10). The test treatment consisted of undiluted Colgate Sensitive Pro-Relief Toothpaste containing 8.0% arginine and calcium carbonate that was applied on the dentin surface under a brushing cycle of 200 strokes, 2 times/day, for 10 days

MEPE/OF45, a new dentin/bone matrix protein and candidate gene for dentin diseases mapping to chromosome 4q21.

Substantial progress has been made regarding the molecular etiology of the dentin diseases dentinogenesis imperfecta types II and III, and dentin dysplasia type II. Genetic linkage studies have identified the critical loci for these diseases on human chromosome 4q21. Located within an overlapping segment of these disease loci is a dentin/bone gene cluster that includes osteopontin, bone sialoprotein, dentin matrix protein 1, dentin sialophosphoprotein, and a new gene MEPE also known as OF45, renamed osteoregulin. In this paper, we report the location of MEPE/OF45 in relationship to the other m

Correlative transmission electron microscopy examination of nondemineralized and demineralized resin-dentin interfaces formed by two dentin adhesive systems.

The resin-dentin interface formed by two dentin adhesives, Optibond (OPTI, Kerr) and Scotchbond Multi-Purpose (SBMP, 3M), was ultramorphologically examined by transmission electron microscopy (TEM). Ultrastructural information from nondemineralized and demineralized sections was correlated. It was hypothesized that the different chemical formulations of the two adhesives would result in a different morphological appearance of the hybrid layer. Ultrastructural TEM examination proved that each of the two dentin adhesive systems was able to establish a micromechanical bond between dentin and resi

Fabrication of a Vertically Stacked Dentin-Pulp Complex Organ-on-a-Chip Device Using a Human Dentin Disc.

Conventional preclinical models used in dental research, including animal models and static cell cultures, present important limitations in reproducing the physiological conditions of the dentin-pulp interface and predicting human biological responses to dental biomaterials. Organ-on-a-chip (OoC) technology provides an alternative approach by enabling the recreation of tissue-specific microenvironments within controlled microfluidic systems. This manuscript describes a step-by-step protocol for the fabrication and assembly of a dentin-pulp complex OoC device incorporating a human dentin disc w

Dentin sialoprotein, dentin phosphoprotein, enamelysin and ameloblastin: tooth-specific molecules that are distinctively expressed during murine dental differentiation.

Dentin sialophosphoprotein [designated DSPP and cleaved into dentin sialoprotein (DSP) and dentin phosphoprotein (DPP)], enamelysin and ameloblastin are each expressed in unique fashions during tooth development. It is possible that these components participate in cell differentiation and the conversion of unmineralized matrix into mineralized structures. In order to delineate the timing and the positioning of these three molecules in a physiological context, we compared their expression profiles by performing in situ hybridization experiments on consecutive sections in developing mouse tissue

Immunohistochemical localization of Type III collagen in the dentin of patients with osteogenesis imperfecta and hereditary opalescent dentin.

The immunohistochemical localization of Type III collagen was studied in the dentin from nine patients with dominantly inherited osteogenesis imperfecta (O.I.) and six patients with hereditary opalescent dentin (H.O.D.). The most consistent localization of Type III collagen was found in patients with O.I. Among these patients an indirect relationship existed between the severity of dentinal involvement and the presence of Type III antibody localization. Although a similar but less intense staining could be observed in some cases of H.O.D., the severity of dentinal alterations in H.O.D. was not

Effects of a potassium nitrate mouthwash on dentinal tubules--a SEM analysis using the dentine disc model.

The concept of tubular occlusion as a method of dentine desensitisation is a logical conclusion from the hydrodynamic hypothesis put forth by Brannström. The aim of this study was therefore to investigate qualitatively by SEM whether a 3% potassium nitrate/0.2% sodium fluoride mouthwash occluded tubule orifices, and by x-ray microanalysis, to characterise the nature of the deposits if any, following application. Following the 'dentine disc model' methodology 1mm thick tooth sections from unerupted molars were obtained. These were treated with the test and control mouthwashes and subjected to s

Phosphoprotein analysis of sequential extracts of human dentin and the determination of the subsequent remineralization potential of these dentin matrices.

Phosphoprotein appears to play an important role in the mineralization of dentin during tooth development and remineralization after demineralization by dental caries. To better understand this role, we describe the extraction and characterization of phosphoprotein from immature, human root apex dentin during and after EDTA demineralization. The extraction procedure included dissociation of the demineralized dentin matrix by guanidine hydrochloride (Gdn.HCl) followed by subsequent digestion with cyanogen bromide (CNBr) and collagenase. Characterization of these extracts included 'Stains-All' s

Magnetic Janus Nanomotor for Dentin Hypersensitivity Treatment.

Reoccluding the exposed dentinal tubules is crucial for treating dentin hypersensitivity (DH). However, the narrow structure and high-density negative charges of dentinal tubules impede the penetration of filling repair materials to effectively achieve deep-induced mineralization for DH treatment. Herein, a morphologically controllable Fe3O4@SiO2 magnetic Janus nanomotors (MJNs) were synthesized via a seeded emulsion-mediated surface growth strategy. The as-achieved MJNs exhibit magnetic response, with a head diameter of about 200 nm, and snake-like morphology with a tunable tail length regula

Pulp/Dentin Regeneration: It Should Be Complicated.

Stem cell-mediated regenerative endodontics has reached the human clinical trial phase; however, many issues still exist that prevent such technology to be a widely used clinical practice. These issues are not straightforward and are complicated. They should be because pulp regeneration is dealing with a small dead-end space. In addition, when regeneration is needed, the space is often heavily infected. The true standard of pulp regeneration should be everything except generation of some fibrous connective tissue and amorphous mineral deposit. As of now, we are still far short of reaching the

Brazilian dentists' perception of dentin hypersensitivity management.

The aim of this study was to investigate how Brazilian dentists perceive and manage dentin hypersensitivity (DH) in their clinical routine. A 13-item questionnaire-based survey was developed and sent electronically to a convenience sample of dentists. The questionnaire assessed the personal and dental practice characteristics of the sample, the occurrence of DH in their daily clinical practice, and management strategies. The data were analyzed descriptively and together with the chi-square test (a = 0.05). A total of 353 responses were obtained from September 2017 to March 2018. Of all the res

Mutations in COL1A1 Gene Change Dentin Nanostructure.

Although many studies have attempted to associate specific gene mutations with dentin phenotypic severity, it remains unknown how the mutations in COL1A1 gene influence the mechanical behavior of dentin collagen and matrix. Here, we reported one osteogenesis imperfecta (OI) pedigree caused by two new inserting mutations in exon 5 of COL1A1 (NM_000088.3:c.440_441insT;c.441_442insA), which resulted in the unstable expression of COL1A1 mRNA and half quantity of procollagen production. We investigated the morphological and mechanical features of proband's dentin using atomic force microscope (AFM)

Unusual forms of collagen in human dentin.

Unusual fibrillar and segmental forms of collagen have been documented in several tissues, including rodent dentin - but not human dentin, which was analyzed here for the presence of such structures. Of six normal human permanent and deciduous teeth examined, cusps of two deciduous molars displayed atypical formations, designated here symmetrical collagen segments (SCSs). They were detected inside of dentinal tubules. It was evident that the 264-nm long, cross-banded SCSs were occasionally arranged vertically and laterally in a staggered manner into thick, irregularly shaped aggregates. Two de

Etchant composition and bond strength to dentin.

This in vitro study examined the effects of three different types of acid etchants on the bond strengths of three dentin bonding systems. The etchant free of both silica and oxalate provided the highest bond strengths for all three systems. Use of the etchant containing oxalate resulted on the lowest bond strengths for all three systems. It is suggested that bond strengths vary as a consequence of deposition onto the dentin surface of various minerals or salts found in the etchants. It is recommended that if treatment of dentin with acids is indicated it should probably be done with those etch

Heritable dentin defects: nosology, pathology, and treatment.

Heritable dentin defects have been divided into 2 main categories: dentinogenesis imperfecta (DI) and dentin dysplasia (DD). Recent studies have shown that they share many features in common. Of the connective tissue diseases, only osteogenesis imperfecta (OI) has been linked to these disorders. So far, no definitive relation between the type of OI and the dental involvement can be established. Familial occurrence of DI with OI cannot be comprehensively explained by mutations in type I collagen genes. No information about the gene defects in DD is available. At the ultrastructural level, the o

[Initial experiences with the dentin-ossicle prosthesis].

The results of 73 implantations performed from 1.7.1986-30.6.1988 are reported in a first follow-up study. 82% reached an air-bone gap of less than 30 dB, 55% of less than 20 dB. In comparison with other materials recently reported the functional results are a little less favourable due to the high percentage of revision surgery in our patients. Only few complications had to be noticed: no single extrusion was observed, but in one case a nearly complete resorption of the DOP was seen. Two DOPs, removed by revision surgery, were examined histologically. Beside resorptive processes, bony changes

Intraoral bacterial growth on tetracycline-impregnated dentin.

Previous studies have shown that tetracyclines react with enamel and dentin in vitro to form slightly soluble compounds with a pronounced and long-lasting antimicrobial capacity. The purpose of the present project was to study the effect of doxycycline-impregnation of dentin on the growth of oral microorganisms in vivo. Slabs of native human dentin were immersed in aqueous solutions of doxycycline HCl, 10 mg/ml, for 10 min, and were subsequently ligated to orthodontic brackets placed bucally on maxillary molars in seven individuals. Untreated native dentin specimens were similarly placed on co

Interradicular dentin dysplasia associated with amelogenesis imperfecta.

Very few reports are available on the occurrence of dysplasia of both enamel and dentin. This report concerns a rare association of amelogenesis imperfecta with a dysplasia of dentin in the interradicular area in sisters of Japanese descent who have no other morphologic anomalies. Retarded tooth eruption was also a clinical feature in both sisters. Histologic examination of several teeth revealed that the anomalous interradicular dentin consisted of a mass of small, onion-like calcified bodies. The absence of any dental abnormalities in both parents, who are related as first cousins, supports
