Quadruple the Considerations: Four Genetic Conditions Unveiled in the Setting of Developmental Delays Including a Complex DMD Rearrangement, Telomere Biology Disorder, and Neurodevelopmental Disorders.

We present a patient with dystrophinopathy and additional complex genetic variants to highlight potential challenges in directing the care and discussion of treatment options. A 12-month-old male child presented to neurology with global delays, including gross motor and speech delay, macrocephaly with frontal bossing, short stature, and hypertelorism. He underwent extensive genetic workup, which identified 4 genetic diagnoses including an intragenic DMD inversion flanked by single exon deletions, a telomere biology disorder, and 2 neurodevelopmental conditions. Our case highlights the need for

Swimming Upstream to Understand Congenital Anomalies of the Kidney and Urinary Tract: Zebrafish Models for Developmental Biology, Disease Mechanisms, and Functional Interpretation of Genetic Variation.

Congenital anomalies of the kidney and urinary tract (CAKUT) are the leading cause of pediatric chronic kidney disease (CKD) and comprise a heterogeneous group of developmental disorders with a substantial genetic contribution. Advances in next-generation sequencing have facilitated the identification of numerous candidate genes and rare variants associated with CAKUT. However, establishing causality and defining the biological functions of implicated genes remain major challenges. Functional validation is therefore essential to bridge the gap between gene discovery and mechanistic understandi

Whole-Exome sequencing and systems biology approaches revealed pathogenicity of compound heterozygote variants of NAGLU gene manifesting developmental regression, brain atrophy, intellectual disability, and ADHD.

BACKGROUND: Sanfilippo syndrome type B results from NAGLU mutations which cause progressive cognitive impairments and central nervous system degeneration. A 10-year-old boy presented with developmental regression, brain atrophy, intellectual disability, attention-deficit/hyperactivity disorder, and restlessness. His parents were non-consanguineous and asymptomatic. METHODS: Whole-exome sequencing (WES) was performed, and variants were confirmed by Sanger sequencing. Downstream analyses integrated protein-protein interaction (PPI), gene-microRNA interaction (GMI), and drug-disease association

Sir Joseph Barcroft, Cambridge placental and fetal research (1933-1966) and inter-generational Science.

The nature of Cambridge (UK) placental and fetal research in the middle third of the twentieth century is reviewed on the basis of published literature and personal recollection. Joseph Barcroft is a central figure who came to fetal research late in an extremely productive career which is briefly sketched. Contemporaneous Cambridge academics in the field included J.D. Boyd (the authors father), J. Hammond, F.H.A. Marshall, R.A. McCance, J. Needham, A.S. Parkes and Elsie Widdowson. The then current Cambridge academic geography is explained and features of its scientific life such as funding, in

Morphological and histological study of larval development of the Senegal sole Solea senegalensis: an integrative study.

This study provides a comprehensive description of the main morphological and histological events that take place during larval and post-larval development of Senegal sole Solea senegalensis in order to establish a reference for its normal developmental organogenesis. Five stages have been described. Before gill development at the onset of metamorphosis (eye migration process, stage 4c), the skin was the main site of gas and ion exchange, whereas during stages 3 and 4, the skin begins differentiating into the definitive juvenile structure. The timing of development of the endocrine system depe

Obtaining Xenopus laevis Embryos.

The embryos of the African clawed frog, Xenopus laevis, are a powerful substrate for the study of complex fundamental biological and disease mechanisms in neurobiology, physiology, molecular biology, cell biology, and developmental biology. A simple and straightforward technique for generating a large number of developmentally synchronized embryos is in vitro fertilization (IVF). IVF permits simultaneous fertilization of thousands of eggs but requires the death of the parental male, which may not be feasible if the male comes from a stock of precious animals. An alternative to euthanizing a pr

Gene-environment interactions in development and disease.

Developmental geneticists continue to make substantial jumps in our understanding of the genetic pathways that regulate development. This understanding stems predominantly from analyses of genetically tractable model organisms developing in laboratory environments. This environment is vastly different from that in which human development occurs. As such, most causes of developmental defects in humans are thought to involve multifactorial gene-gene and gene-environment interactions. In this review, we discuss how gene-environment interactions with environmental teratogens may predispose embryos

Morphometrics, 3D Imaging, and Craniofacial Development.

Recent studies have shown how volumetric imaging and morphometrics can add significantly to our understanding of morphogenesis, the developmental basis for variation, and the etiology of structural birth defects. On the other hand, the complex questions and diverse imaging data in developmental biology present morphometrics with more complex challenges than applications in virtually any other field. Meeting these challenges is necessary in order to understand the mechanistic basis for variation in complex morphologies. This chapter reviews the methods and theory that enable the application of

[Evolution of teeth].

True teeth developed approximately 230 million years ago. Mammals have an accurate dental occlusion, whereby the renewal of teeth has reduced to two sets of teeth. Since the shape of teeth does not change after eruption, they form a good subject for developmental biology research. Research on fossil material and current ecology of mammals has opened up new perspectives into the research of tooth shape, food, ecology and climate.

Nematode model systems in evolution and development.

The free-living nematode Caenorhabditis elegans is one of the most important model organisms in all areas of modern biology. Using the knowledge about C. elegans as a baseline, nematodes are now intensively studied in evolution and development. Evolutionary developmental biology or for short, 'evo-devo' has been developed as a new research discipline during the last two decades to investigate how changes in developmental processes and mechanisms result in the modification of morphological structures and phenotypic novelty. In this article, we review the concepts that make nematode evo-devo a s

Schistosomiasis as a disease of stem cells.

Schistosomiasis is a devastating parasitic disease caused by flatworms of the genus Schistosoma. The complex life cycles and developmental plasticity of these parasites have captured the attention of parsitologists for decades, yet little is known on the molecular level about the developmental underpinnings that have allowed these worms to thrive as obligate parasites. Here, we describe basic schistosome biology and highlight how understanding the functions of stem cells in these worms will transform our understanding of these parasites. Indeed, we propose that schistosomiasis is fundamentally

Zebrafish: a model for understanding diabetic complications.

Diabetes mellitus causes several vascular complications in patients, such as macrovascular problems including myocardial infarction, peripheral artery diseases and stroke and microvascular problems including nephropathy and retinopathy. Likewise, diabetes mellitus is associated with other complications such as neuropathy and delayed wound healing. The zebrafish has been used for decades as a model organism for studies in developmental biology. In fact several common and important developmental mechanisms have been identified in zebrafish which are similar in mammals. The zebrafish has short ge

[Embryonic development of the liver: basic biology and pediatric pathology].

At birth a number of children suffer from hepatic enzyme deficiencies or from biliary malformations. These diseases can be better understood, and in some instances better treated, with the help of knowledge gained from developmental biology studies. Here we review the key steps of liver development and illustrate how developmental biology can contribute to improve diagnosis and treatment of hepatic diseases, and at the same, time benefits from the input from clinical sciences.

The developmental-genetics of canalization.

Canalization, or robustness to genetic or environmental perturbations, is fundamental to complex organisms. While there is strong evidence for canalization as an evolved property that varies among genotypes, the developmental and genetic mechanisms that produce this phenomenon are very poorly understood. For evolutionary biology, understanding how canalization arises is important because, by modulating the phenotypic variation that arises in response to genetic differences, canalization is a determinant of evolvability. For genetics of disease in humans and for economically important traits in

The people behind the papers - Veronica Biga and Cerys Manning.

Notch target genes HES1 and HES5 mutually repress one another, while also exhibiting oscillatory expression across tissues, to regulate cell fate specification, during mammalian neural tube development. In their work, Veronica Biga, Cerys Manning and colleagues show that there is a mechanistic link between oscillatory expression of the cross-repressive factors HES1 and HES5, and fate resolution in the developing spinal cord. To learn more about their work, we talked to the first author Veronica Biga, as well as the corresponding author, Cerys Manning, Presidential Fellow in the School of Medic

Developmental toxicology: methods and protocols. Volume overview: introduction.

The origins and history of the study of teratology and developmental toxicology span centuries of human experience. Anatomical malformations observed at birth have been described across many generations but the root causes of these events have been enigmatic and difficult to understand. Many factors may contribute to the lack of mechanistic understanding, but the miniscule size, inaccessibility, and the consequences of ethical concerns contribute heavily to the unique restrictions on developmental toxicology research. Classic teratogens, such as vitamin A, Rubella virus, thalidomide, and methy
