BACKGROUND: Birch witches' broom is an abnormal plant structure caused by infection of birch trees with the dimorphic fungus Taphrina betulina Rostr., (1883). Although classified as a disease, witches' broom has traditionally been used in folk medicine to treat skin disorders. This suggests that fungal infection may induce metabolic reprogramming in host tissues, leading to the production and accumulation of biologically active secondary metabolites. We hypothesized that infection-driven metabolic changes in witches' broom tissue could generate compounds capable of modulating keratinocyte viab

Bisdemethoxycurcumin (BDC) is an important ingredient derived from the food spice turmeric. Although BDC exhibits various pharmacological effects, it is characterized by poor water solubility and limited stability under light. Considering that the high specific surface area of porous starch (PS) renders it an ideal carrier for the encapsulation of active compounds, and polyoxyethylene‑hydrogenated castor oil RH40 (RH40) is an effective solubilizer for BDC, the current study optimized the PS/RH40/BDC formulation to combine these advantages. Consequently, a PS/RH40/BDC ratio of 10:3:1 was identi

There is a growing interest for curcuminoids in the general population and the scientific research community. Curcuminoids, derived from turmeric spice, are lipophiles and therefore have a low solubility in water which hence have a low bioavailability in the human plasma. To circumvent this issue, a natural product developed by Biodroga Nutraceuticals combined curcuminoids with omega-3 fatty acids (OM3) esterified in monoglycerides (MAG). The objective was to perform a 24 h pharmacokinetics in humans receiving a single dose of curcuminoid formulated by three different means, and to compare the

Type 2 diabetes mellitus (T2DM) is associated with pancreatic β-cell dysfunction and insulin resistance. Islet amyloid polypeptide (IAPP) aggregation is found to induce islet β-cell death in T2DM patients. Recently, we demonstrated that yakuchinone B derivative 1 exhibited inhibitory activity against IAPP aggregation. Thus, in this study, a series of synthesized yakuchinone B-inspired compounds were tested for their anti-IAPP aggregation activity. Four of these compounds, 4e-h, showed greater activity than the lead compound 1, in the sub-μM range (IC50 = 0.7-0.8 μM). The molecular docking anal

BACKGROUND: Colorectal cancer (CRC) remains a major global health challenge, driven by aberrant activation of the Wnt/β-catenin pathway and persistent inflammatory signaling. Natural bioactive compounds capable of simultaneously targeting these molecular drivers represent an attractive avenue for safer and more effective chemoprevention. This study aimed to evaluate the anti-carcinogenic potential of Gingerenone A (GinA), a diarylheptanoid derived from ginger, by assessing its effects on Wnt/β-catenin signaling, inflammatory mediators, oxidative stress, and apoptosis during colorectal tumorige

Glioblastoma multiforme (GBM) is the most lethal primary brain tumor in adults, with median survival rarely exceeding 15 months under current therapy with surgery, radiotherapy, and temozolomide (TMZ). Resistance mechanisms, tumor heterogeneity, and glioma stem cells (GSCs) remain major obstacles. Curcumin has shown pleiotropic anticancer effects, but its poor bioavailability and limited blood–brain barrier (BBB) penetration restrict clinical utility. Demethoxycurcumin (DMC), a natural curcuminoid analogue, provides greater chemical stability and potent multi-targeted activity. In vitro studie

The increased concentration of glutamate in synaptic vesicles, mediated by the vesicular glutamate transporter (VGLUT), is an initial vital step in glutamate synaptic transmission. Evidence indicates that aberrant overexpression of VGLUT is involved in certain pathophysiologies of the central nervous system. VGLUT is subject to inhibition by various types of agents. The most potent VGLUT-specific inhibitor currently known is Trypan Blue, which is highly charged, hence membrane-impermeable. We have sought a potent, VGLUT-specific agent amenable to easy modification to a membrane-permeable analo

While mustard seeds offer a prime store of biologically active compounds, there is insufficient data regarding the impact of melatonin (MT) priming on remodelling the chemical patterns, metabolic pathways, and biological attributes of mustard sprouts. Accordingly, the current work is directed to investigate the metabolic shifts in mustard sprouts post MT-priming treatments using UPLC-MS/MS and chemometric analyses. Given this, MT-primed sprouted samples witnessed a significant elevation of phenolic acids, specifically hydroxybenzoic, ferulic, syringic, and chlorogenic acids, with approximately

Turmeric (Curcuma longa) contains curcumin, demethoxycurcumin (DMC), and bisdemethoxycurcumin (BDMC). Nevertheless, curcumin is the most researched active ingredient for its numerous pharmacological effects. We investigated the impact of these curcuminoids found in Ryudai gold, an approved cultivar of Curcuma longa, on wound healing, inflammation, and diabetes. Sub-planter injections of carrageenan induced acute paw inflammation in rats. The wound-healing ability of 1% curcuminoids was examined by making a 6 mm round wound on the shaved dorsum of the mice with a biopsy punch. A single intraper

Curcumin (Cur), is a pigment with antiproliferative activity but has some pharmacokinetic limitations, which led researchers to look for more effective structure analogs. This work investigated the effects of Cur and compared them with the two analogs, demethoxycurcumin (DeMC) and dimethoxycurcumin (DiMC), to elucidate their mechanisms of action. The cytotoxic, antiproliferative, and genotoxic effects these compounds were correlated based on gene expression analysis in the human renal adenocarcinoma cells (786-O). Cur decreased CYP2D6 expression and exhibited cytotoxic effects, such as inducin
