Nephroprotective effects of honokiol in a high-fat diet-streptozotocin rat model of diabetic nephropathy.

AIMS: Diabetic nephropathy (DN) is the foremost basis of end-stage kidney failure implicating endoplasmic reticulum (ER) stress and dysregulation of Rho kinase/Rock pathway. Magnolia plants are used in traditional medicine systems in Southeast Asia owing to bioactive phytoconstituents. Earlier, honokiol (Hon) exhibited therapeutic potential in experimental models of metabolic, renal, and brain disorders. In the present study, we evaluated potential of Hon against DN and possible molecular mechanisms. MAIN METHODS: In the existing experiments, high-fat diet (HFD) (17 weeks) and streptozotocin

A low-carbohydrate diet in place of SGLT2i therapy in a patient with diabetic cardiomyopathy.

SUMMARY: In patients with diabetes mellitus, the toxic milieu caused by abnormal glucose and free fatty acid handling can lead to heart failure (HF). Referred to as diabetic cardiomyopathy (DMCM), this syndrome often exists in the absence of conventional risk factors for HF such as history of myocardial infarction or hypertension. Low-carbohydrate diets (LCDs) have recently been endorsed as an efficacious therapeutic dietary approach to prevent and reverse cardiometabolic disease including type 2 diabetes mellitus (T2DM). LCDs improve systemic insulin resistance (IR), reverses cardiac remodell

Lansoprazole enhances the antidiabetic effect of dapagliflozin in fortified diet-fed streptozotocin-treated diabetic rats.

Dapagliflozin (DAPA) is used for treating type 2 diabetes, whereas lansoprazole (LPZ) is used as a traditional antiulcer drug. The present study investigated the possible antidiabetic effects of LPZ on fortified diet-fed streptozotocin (FDF/STZ)-induced insulin-resistant diabetic rats. On the basis of the current results, it can be concluded that LPZ could be used as an add-on drug along with the conventional treatment for T2D as it showed beneficial effects in the current experimental model of insulin resistance.

Improved glucose tolerance in gestational diabetic women on a low fat, high unrefined carbohydrate diet.

In a randomized cross-over study 5 gestational diabetic women were tested on a low fat, high unrefined carbohydrate (HC) diet and a low carbohydrate (LC) diet for a period of 4 days each. Glucose tolerance was shown to be significantly improved on the HC diet compared to the LC diet (p less than 0.05). Urinary glucose output was 50% lower on the HC diet (1.3 +/- 1.1 mmol/d) than on the LC diet (2.6 +/- 3.0 mmol/d), although this difference was not statistically significant. Fasting free fatty acid levels were significantly lower on the HC diet (HC 590 +/- 270 mumol/l, LC 690 +/- 270 mumol/l; p

Diet control to achieve euglycemia induces significant loss of heart and liver weight via increased autophagy compared with ad libitum diet in diabetic rats.

Intensive glucose control increases the all-cause mortality in type 2 diabetes mellitus (T2DM); however, the underlying mechanisms remain unclear. We hypothesized that strict diet control to achieve euglycemia in diabetes damages major organs, increasing the mortality risk. To evaluate effects on major organs when euglycemia is obtained by diet control, we generated a model of end-stage T2DM in 13-week-old Sprague-Dawley rats by subtotal pancreatectomy, followed by ad libitum feeding for 5 weeks. We divided these rats into two groups and for the subsequent 6 weeks provided ad libitum feeding t

Anti-Diabetic Effects of Ethanol Extract from Sanghuangporous vaninii in High-Fat/Sucrose Diet and Streptozotocin-Induced Diabetic Mice by Modulating Gut Microbiota.

Type 2 diabetes mellitus (T2DM) may lead to abnormally elevated blood glucose, lipid metabolism disorder, and low-grade inflammation. Besides, the development of T2DM is always accompanied by gut microbiota dysbiosis and metabolic dysfunction. In this study, the T2DM mice model was established by feeding a high-fat/sucrose diet combined with injecting a low dose of streptozotocin. Additionally, the effects of oral administration of ethanol extract from Sanghuangporous vaninii (SVE) on T2DM and its complications (including hypoglycemia, hyperlipidemia, inflammation, and gut microbiota dysbiosis

Dioscorea zingiberensis ameliorates diabetic nephropathy by inhibiting NLRP3 inflammasome and curbing the expression of p66Shc in high-fat diet/streptozotocin-induced diabetic mice.

OBJECTIVES: Diabetic nephropathy (DN) is a severe diabetic complication. Dioscorea zingiberensis (DZ) possesses excellent pharmacological properties with lower toxicity. The purpose of this study was to investigate the efficacy and mechanism of DZ in DN. METHODS: DN was established by the high-fat diet combining intraperitoneal injection of streptozotocin in mice. The DZ (125 and 250 mg/kg/day) were intragastrical administered for 8 consecutive weeks. After treatment, blood, urine and kidney tissue were collected for biological detection, renal morphology, fibrosis and molecular mechanism res

EXPERIMENTAL STUDIES ON DIABETES : SERIES I. PRODUCTION AND CONTROL OF DIABETES IN THE DOG. 2. EFFECTS OF CARBOHYDRATE DIETS.

1. The injurious effects of excessive carbohydrate diet are demonstrable in partially depancreatized dogs, in the same manner as in human patients. With severe diabetes there is rapid progress of emaciation and weakness and early death. 2. With milder diabetes, there is frequently a transitional state following operation, when the fate depends on the diet. If the tolerance is spared for a time, recovery sometimes occurs to such extent that diabetes cannot be produced by any kind or quantity of feeding, but only by removal of a small additional fragment of pancreatic tissue. The proper degree o

Protective effect of orientin on diabetic nephropathy in rat models of high-fat diet and streptozotocin-induced diabetes.

Diabetic nephropathy (DN) represents the primary cause of chronic kidney disease (CKD) worldwide. Orientin is a natural bioactive flavonoid with profound immunomodulatory, anti-inflammatory, and antioxidative effects. This study aimed to investigate the nephroprotective effect of orientin on rat prototypes of high-fat diet (HFD) and streptozotocin (STZ)-induced DN. 75 male rats were divided into 5 groups of 15 rats each. Rats were fed a HFD for 4 weeks, injected with a single dose of STZ 30 mg/kg, and continued on HFD for 15 weeks. Orientin was administered daily at 40 mg/kg for 15 weeks. The

Effects of a high protein diet on the evolution of diabetes in streptozotocin-induced and spontaneously diabetic "BB" Wistar rats.

Recently, we demonstrated a reduction in the diabetogenic action of streptozotocin (STZ) in rats previously adapted to a high protein (HP) diet. These data suggested that amelioration of diabetes resulted from the combination of two effects of the HP diet: initial protection against the diabetogenic action of the drug at the time of exposure and subsequent improvement of the induced diabetic condition. The present study evaluated the effects of a HP diet on the evolution of the metabolic condition in rats with STZ-induced or spontaneous diabetes (BB Wistar rats). Two days after STZ injection,

Traditional diets and cultural beliefs affecting diabetes prevention and control in Africa.

The burden of type 2 diabetes mellitus in Africa is growing rapidly due to socio-economic changes, urbanization, and substitution of traditional and nutritious, high-fiber diets with ultra-processed, high-energy foods. This review explains the protective value of traditional African diets that contain native grains and fermented foods, but are under threat of westernization and cultural transitions. Moreover, cultural beliefs, social practices, dietary limitations, and gender roles strongly influence diabetes prevention and management practices on the continent. Community-based programs, schoo

High-Fat Diet: A Paradoxical Factor in the Setting of Type 1 Diabetes.

Type 1 diabetes (T1D) stems from the autoimmune attack on insulin-producing pancreatic islet β cells. Progression of T1D is fundamentally dictated by genetic predisposition and modified by environmental and dietary factors. Although patients with T1D are typically characterized by weight loss, overweight and obesity have become more prevalent in recent years. This phenomenon is related to the intake of fat beyond the recommended limit, and a high-fat diet (HFD) is speculated to elevate the risk of T1D along with the development of diabetic complications. By contrast, results of studies in anim

Molecular mechanisms of enteric neuropathies in high-fat diet feeding and diabetes.

BACKGROUND: Obesity and diabetes are associated with altered gastrointestinal function and with the development of abdominal pain, nausea, diarrhea, and constipation among other symptoms. The enteric nervous system (ENS) regulates gastrointestinal motility. Enteric neuropathies defined as damage or loss of enteric neurons can lead to motility disorders. PURPOSE: Here, we review the molecular mechanisms that drive enteric neurodegeneration in diabetes and obesity, including signaling pathways leading to neuronal cell death, oxidative stress, and microbiota alteration. We also highlight potenti

Low-carbohydrate diets in type 1 diabetes: balancing benefits and risks.

PURPOSE OF REVIEW: Interest in the use of calorie restriction with low-carbohydrate diets for patients with type 1 diabetes appears to be increasing despite physicians' discomfort about its longer term outcomes. A divergence in opinion regarding the balance of benefits and safety may lead to patient disengagement from conventional medical supervision. This review describes the current evidence regarding the benefits and risks of these diets and suggests a way forward to addressing this potential misalignment between the aims of patients and their physicians. RECENT FINDINGS: Benefits on glyca

The Mediterranean Diet for Adolescents with Type 1 Diabetes: A Prospective Interventional Study.

The Mediterranean diet (MED) is highly recommended. Medical nutrition therapy is the cornerstone of diabetes treatment. The primary outcome was to evaluate the change in micronutrient intake of youth with type 1 diabetes before and after a 6-month MED intervention; we also assessed adherence and glycemic control. Twenty adolescents, median age 18 years (interquartile range: 15.5-21), median diabetes duration 9 years (7-14), using continuous glucose monitoring devices, received personalized diet regimes based on MED. At 6 months post-intervention, the caloric intake remained unchanged; however,
