Structural basis for membrane binding by coagulation factors V and VIII and their specificity for phosphatidylserine-containing membranes.

Phosphatidylserine on the surface of activated cells serves as a signal for coagulation factor binding and the membrane-dependent assembly of enzyme complexes that drive the accelerated host response to vascular damage. Here, we use single-particle cryo-EM of liposome-bound proteins to establish common mechanisms utilized by coagulation factors V and VIII for membrane binding and phospholipid specificity. The interface between these peripheral proteins and the membrane shows contacts between the discoidin C1 and C2 domains and the interfacial phospholipid headgroup region of the membrane bilay

Enhanced ferroptosis sensitivity promotes the formation of highly myopic cataract via the DDR2-Hippo pathway.

Highly myopic cataract (HMC) is a leading cause of blindness among the working-age individuals, with its pathogenesis poorly understood. This study aimed to elucidate the role of ferroptosis in HMC development as well as the underlying mechanisms. In HMC lens epithelia, levels of Fe2+ and lipid peroxidation were found elevated, with increased vulnerability towards ferroptosis as revealed by transmission electron microscopy. Mechanistically, RNA sequencing of HMC lens epithelial samples identified up-regulated expression of discoidin domain receptor tyrosine kinase 2 (DDR2) as a key factor, whi

EDIL3 induced by GLIS2 suppresses the anti-tumor activity of CD8+ T cells and expedites epithelial-mesenchymal transition in thyroid cancer.

Despite the discoveries of new and promising therapeutics, effective treatments for advanced and metastatic thyroid cancer (THCA) are still lacking. Epithelial-to-mesenchymal transition (EMT) is crucial for developing an invasive phenotype in tumor cells and, therefore, a hallmark of metastatic disease. We here investigate the effect of EGF-like repeat and discoidin I-like domain-containing protein 3 (EDIL3) on EMT in THCA and the mechanism involved. THCA cells with EDIL3 knockdown were generated to analyze the effect on EMT, proliferation, migration, invasion, and angiogenesis. THCA cells wit

Milk Fat Globule Epidermal Growth Factor VIII Fragment Medin in Age-Associated Arterial Adverse Remodeling and Arterial Disease.

Medin, a small 50-amino acid peptide, is an internal cleaved product from the second discoidin domain of milk fat globule epidermal growth factor VIII (MFG-E8) protein. Medin has been reported as the most common amylogenic protein in the upper part of the arterial system, including aortic, temporal, and cerebral arterial walls in the elderly. Medin has a high affinity to elastic fibers and is closely associated with arterial degenerative inflammation, elastic fiber fragmentation, calcification, and amyloidosis. In vitro, treating with the medin peptide promotes the inflammatory phenotypic shif

Circ-LAMP1 promotes T-cell lymphoblastic lymphoma progression via acting as a ceRNA for miR-615-5p to regulate DDR2 expression.

Circular RNAs (circRNAs) act as pivotal functions in tumor progression. Nevertheless, the functions and mechanism of circRNAs in T-cell lymphoblastic lymphoma (T-LBL) remain unclear. In this work, we first screened the differentially expressed circRNAs between T-LBL tissues and normal infantile thymus and circ-LAMP1 was identified the highest expressed circRNA in cancerous tissues. qRT-PCR further verified its upregulation in T-LBL tissues and cell lines. Cell counting kit-8 (CCK-8) experiment proved the cell proliferation-promoting role of circ-LAMP1. This effect is partially dependent on its

Mutations in DDR2 gene cause SMED with short limbs and abnormal calcifications.

The spondylo-meta-epiphyseal dysplasia [SMED] short limb-hand type [SMED-SL] is a rare autosomal-recessive disease, first reported by Borochowitz et al. in 1993.(1) Since then, 14 affected patients have been reported.(2-5) We diagnosed 6 patients from 5 different consanguineous Arab Muslim families from the Jerusalem area with SMED-SL. Additionally, we studied two patients from Algerian and Pakistani ancestry and the parents of the first Jewish patients reported.(1) Using a homozygosity mapping strategy, we located a candidate region on chromosome 1q23 spanning 2.4 Mb. Targeting DDR1 with novel synthesized antagonists delays cellular aging and enhances wound healing in mesenchymal stem cells.

Mesenchymal stem cells (MSCs) are central to regenerative medicine; however, their clinical utility is constrained by reduced proliferative capacity and replicative senescence during ex vivo expansion. Here, we report on the rational design and biological characterization of novel phthalimide-based inhibitors targeting discoidin domain receptor 1 (DDR1). Structure-activity relationship optimization efforts led to the identification of AC-4067 and AC-4061 as potent DDR1 inhibitors, with IC₅₀ values of 30.9 nM and 53.6 nM, respectively. AC-4067 demonstrated 18-fold selectivity for DDR1 over DDR2

DDR2-regulated arginase activity in ovarian cancer-associated fibroblasts promotes collagen production and tumor progression.

Ovarian cancer has poor survival outcomes particularly for advanced stage, metastatic disease. Metastasis is promoted by interactions of stromal cells, such as cancer-associated fibroblasts (CAFs) in the tumor microenvironment (TME), with tumor cells. CAFs play a key role in tumor progression by remodeling the TME and extracellular matrix (ECM) to result in a more permissive environment for tumor progression. It has been shown that fibroblasts, in particular myofibroblasts, utilize metabolism to support ECM remodeling. However, the intricate mechanisms by which CAFs support collagen production

DDR1 Modulates Cytoskeletal Remodeling and Podosome Formation in Renal Fibroblasts.

Discoidin domain receptor 1 (DDR1) has been implicated in fibrotic progression in multiple organs, including the kidney. However, its role in regulating cytoskeletal organization and matrix remodeling in renal fibroblasts remains unclear. Here, we investigated how DDR1 expression is regulated by profibrotic stimulation and extracellular matrix stiffness, and how DDR1 influences cytoskeletal organization and collagen remodeling. Single-cell RNA sequencing of murine kidneys subjected to unilateral ureteral obstruction (UUO) revealed enrichment of Ddr1 expression in transitional fibroblast popula

The Matrix Reloaded: The Hepatic Matrisome as a Therapeutic Opportunity to Fight Liver Fibrosis.

Liver fibrosis is the excessive accumulation of extracellular matrix (ECM) that occurs in most types of chronic liver diseases (CLDs) as a response to sustained liver injury. While the ECM comprises different proteins, collagen being the most abundant, the term matrisome refers to a plethora of ECM-related molecules, including collagen-associated proteins, growth factors, cytokines, enzymes and their endogenous inhibitors. The hepatic matrisome undergoes significant qualitative and quantitative changes during liver fibrosis. Despite intense research over recent years, our understanding of the

WRG-28 attenuates matrix degradation and chondrocyte calcification by inhibiting DDR2 signaling in osteoarthritis.

Osteoarthritis (OA) is a common degenerative joint disease characterized by cartilage degradation and abnormal bone remodeling. Discoidin Domain Receptor 2 (DDR2) has emerged as a critical mediator of OA pathogenesis by disrupting extracellular matrix (ECM) homeostasis. This study investigates the therapeutic potential of DDR2 inhibition via selective allosteric inhibitor WRG-28. To evaluate its in vivo efficacy, an OA rat model was established, followed by intra-articular injections of WRG-28. The therapeutic effects were assessed using μCT, Safranin O/Fast Green staining, immunohistochemistr

DDR2 alleviates retinal vaso-obliteration and pathological neovascularization by modulating microglia M1/M2 phenotypic polarization in a mouse model of proliferative retinopathy.

Retinopathy of prematurity (ROP), a leading cause of blindness in premature infants, is characterized by retinal vaso-obliteration during hyperoxia and pathological neovascularization (NV) in relative hypoxia phase. Current treatments, which focus on the late stages of pathological neovascularization, are associated with numerous side effects. Studies demonstrated that discoidin domain receptor 2 (DDR2), a collagen-binding receptor tyrosine kinase, inhibits the experimental choroidal neovascularization and participates in tumor angiogenesis. However, the role of DDR2 in ROP and underlying mech

EDIL3 regulates pulmonary artery smooth muscle cell proliferation and migration via integrin αVβ3/ERK1/2 axis in pulmonary hypertension.

The abnormal expression of matricellular proteins involved in pulmonary vascular remodelling can accelerate the development of pulmonary hypertension (PH). Epidermal growth factor-like repeats and discoidin I-like domains 3 (EDIL3) is a matrix protein associated with the onset and progression of various diseases, yet its role in PH remains unexamined. This research aims to assess the expression of EDIL3 in PH and to investigate its underlying mechanisms in the condition. Our results indicated that EDIL3 levels are elevated in patients with idiopathic pulmonary arterial hypertension (iPAH), as

Disorganisation of basement membrane zone architecture impairs melanocyte residence in vitiligo.

The basement membrane zone is the interface between the epidermis and dermis, and it is disrupted in several skin conditions. Here, we report the results of a comprehensive investigation into the structural and molecular factors of the basement membrane zone in vitiligo, a dermatological disorder characterised by depigmented patches on the skin. Using electron microscopy and immunofluorescence staining, we confirmed abnormal basement membrane zone morphology and disrupted basement membrane zone architecture in human vitiliginous skin. Furthermore, we identified elevated expression of matrix me
