Dopamine not effective for treatment of hypotension in chlorpromazine overdose: first case report.

Dopamine is a popular and effective vasopressor agent. However, in the event of overdose with a potent alpha 1-receptor blocker, dopamine may not be effective in treatment of hypotension. A vasopressor that stimulates only alpha-receptors, such as norepinephrine or phenylephrine, may be more appropriate. In our case, dopamine was clearly ineffective, whereas norepinephrine worked well. Our case also demonstrates the importance of carefully monitoring the effectiveness of any vasopressor therapy.

Investigations on the interactions of scorpion neurotoxins with the predicted structure of D1 dopamine receptor by protein-protein docking method. A bioinformatics approach.

Dopamine receptors play a critical role in the cell signalling process responsible for information transfer in neurons functioning in the nervous system. Development of improved therapeutics for disorders like Parkinson's disease and schizophrenia would be significantly enhanced with the availability of the 3D structure for the dopamine receptors. Scorpion neurotoxins are unique source of structural templates from which new therapeutic agents might be developed. We report here the 3D structure of the human D1 dopamine receptor, predicted from primary sequence using computational techniques. Th

Norepinephrine Versus Dopamine as Initial Vasoactive Therapy in Neonatal Septic Shock: A Randomized Controlled Trial in a Tertiary Care Neonatal Intensive Care Unit in Eastern India.

Background Septic shock remains a major cause of neonatal morbidity and mortality, particularly in preterm and low birth weight neonates. Although dopamine was traditionally used as the first-line vasoactive agent in neonatal shock, norepinephrine has emerged as a potential alternative agent of late because of its potent vasoconstrictive effects and favourable hemodynamic profile. However, evidence comparing the efficacy of these agents in septic shock in neonates remains limited. Objective To compare the efficacy of norepinephrine and dopamine as the initial vasopressor in neonatal septic sho

Dopamine D2 and D4 receptor ligands: relation to antipsychotic action.

Since the discovery that the antipsychotic action of phenothiazines was mediated by dopamine D2 receptors, the dopamine system has been scrutinized for schizophrenia related abnormalities. The focus has been to create neuroleptics with improved antipsychotic profiles and reduced side effects. With the identification of multiple dopamine receptor subtypes, the hypotheses regarding the role of dopamine in schizophrenia and antipsychotic action of neuroleptics have been refined. Even after the molecular identification of newer dopamine D2-like receptor subtypes (D3 and D4), the dopamine D2 recept

[Current role of antidopaminergic drugs in pediatrics].

The treatment of gastrointestinal disorders in pediatrics is based on the use of prokinetic agents; amongst these prokinetic compounds, cholinomimetic drugs and dopamine antagonists (metoclopramide, domperidone) are principally available. Metoclopramide is an antidopaminergic benzamide with mainly antiemetic effects, due to the interaction with dopamine receptors in the central trigger zone. Another effect is to enhance the transit of material through the gastrointestinal tract. Disadvantages in the use of metoclopramide are neurological effects (asthenia, sleepiness), extrapyramidal dyskineti

Effects of temperature on the response of chemoreceptor fibers to chemical agents.

The effects of stimulating (NaCN, ACh, eserine and haloperidol) and depressant (zero [Ca2+]o, mecamylamine and dopamine) agents of carotid body chemoreceptors were tested at different temperatures. Temperature was varied between 29 and 39 degrees C and Arrhenius plots of the sensory discharge frequency against the reciprocal of absolute temperature were constructed before and during application of the chemicals. NaCN, ACh and zero [Ca2+]o did not change the apparent activation energy (mu) whereas eserine, haloperidol and probably mecamylamine depressed mu. Dopamine probably increased mu. Thus,

Effects of short-term reserpine treatment on generalized and focal dystonic-dyskinetic syndromes.

We report that low dose reserpine treatment (0.25 mg/day for 7 days) improves the dystonic-dyskinetic disability score in patients affected by idiopathic dystonias, iatrogenic dystonic-dyskinetic syndromes and Sydenham's chorea. The results support the idea that the particular antidopaminergic activity exerted by reserpine or other dopamine-depleting agents may produce good therapeutical efficacy, although reserpine can precipitate Parkinsonism and induce dopamine-receptor supersensitivity.

Multi-Step Amperometry for Detecting Dopamine Using Boron-Doped Diamond Microelectrodes in the Striatum of the Mouse Brain.

The real-time measurement of dopamine concentration in the brain is crucial in elucidating dopamine-related physiological functions and pathologies and guiding therapeutic development. Boron-doped diamond (BDD) electrodes have emerged as promising tools for use in biological measurement because of their exceptional durabilities, sensitivities, and biocompatibilities, but electrochemically distinguishing dopamine from norepinephrine, which exhibits a similar chemical structure, remains challenging. Other substances that interfere with dopamine detection include tyrosine, L-3,4-dihydroxyphenylal

Complicating Symmetric Peripheral Gangrene after Dopamine Therapy to Patients with Septic Shock.

Dopamine is commonly used as a first-line agent in the treatment of patients with septic shock. The use of dopamine rarely causes symmetric peripheral dry gangrene. If the symmetric peripheral dry gangrene occurs in the patient after dopamine injection, it easily leads to disagreement between doctors and patients. A 60-year-old woman who had sudden septic shock was sent to intensive care unit (ICU). She was received dopamine injection according to the routine during treatment. Over the next 3 months, her limbs developed to dry gangrene and required amputation. The result shows that the occurre

Dopamine agonists and antipsychotics.

There can potentially be a number of clinical interactions that could adversely affect patient outcomes in a patient with a prolactinoma and psychiatric disease that might require antipsychotic and dopamine agonist treatment. Dopamine agonists stimulate the dopamine D2 receptor, resulting in a decrease in prolactin (PRL) levels and in prolactinoma size but action on dopamine receptors in the meso-limbic system may rarely cause psychosis and more commonly cause impulse control disorders. The psychiatric benefits of antipsychotic agents involve blocking the D2 and other dopamine receptors but th

Dopaminergic mechanisms in the teleost retina. I. Dopamine-sensitive adenylate cyclase in homogenates of carp retina; effects of agonists, antagonists, and ergots.

A specific dopamine-sensitive adenylate cyclase has been identified in homogenates of the teleost (carp) retina. Maximal stimulation by 100 microM-dopamine resulted in a 5--10-fold increase in adenylate cyclase activity with half-maximal stimulation occurring at a concentration of 1 microM. l-Noradrenaline and l-adrenaline were some 10 times less potent than dopamine whilst the alpha- and beta-adrenoreceptor agonists, l-phenylephrine and dl-isoprenaline were inactive. Apomorphine elicited a partial stimulation of adenylate cyclase activity whilst various ergot alkaloids produced mixed agonist/

Locomotor stereotypy produced by dexbenzetimide and scopolamine is reduced by SKF 83566, not sulpiride.

Like amphetamine, scopolamine produces locomotor stereotypy (repetitive routes of locomotion) in an open field. To determine whether locomotor stereotypy is a common behavioral effect of anticholingeric agents, several doses of the anticholinergic dexbenzetimide were tested for the ability to produce locomotor stereotypy; like scopolamine, dexbenzetimide produced locomotor stereotypy. To investigate a possible role of dopamine in anticholinergic-induced locomotor stereotypy, we tested the ability of the dopamine D1 antagonist SKF 83566 and the D2 antagonist sulpiride to block the locomotor ste

Delirium is a palliative care emergency where patients experience changes in perception, awareness, and behavior. Common features include changes in the sleep-wake cycle, emotional lability, delusional thinking, and language and thought disorders. Delirium results from neurotransmitter imbalances involving several neurotransmitters such as dopamine, glutamate, norepinephrine, acetylcholine, gamma-aminobutyric acid, and serotonin. Untreated delirium causes significant morbidity and mortality. Nonpharmacologic and pharmacologic approaches treat delirium. Current pharmacologic management of delir

Nanoplasmonic Aptasensing Enables Real-Time Optical Monitoring of Neurotransmitters in Living Brainstem Organoids.

Disruption of dopamine signaling is central to Parkinson's disease (PD) pathology and a primary target of symptomatic therapy. However, direct measurement of dopamine dynamics in human neural tissue with high sensitivity, temporal resolution, and compatibility with three-dimensional models remains technically challenging. Here, an organoid-on-a-chip platform that integrates a cavity-coupled nanoplasmonic aptasensor with human brainstem organoids (hBSOs) and microfluidic control is employed for label-free monitoring of the extracellular dopamine response. The plasmonic aptasensor achieves a lim

Susceptibility to a parkinsonian toxin varies during primate development.

Symptoms of Parkinson's disease typically emerge later in life when loss of nigrostriatal dopamine neuron function exceeds the threshold of compensatory mechanisms in the basal ganglia. Although nigrostriatal dopamine neurons are lost during aging, in Parkinson's disease other detrimental factors must play a role to produce greater than normal loss of these neurons. Early development has been hypothesized to be a potentially vulnerable period when environmental or genetic abnormalities may compromise central dopamine neurons. This study uses a specific parkinsonian neurotoxin, 1-methyl-4-pheny

A high-quality homology model for the human dopamine transporter validated for drug design purposes.

The human dopamine transporter (hDAT) plays many vital functions within the central nervous system and is thus targeted by many pharmaceutical agents. Dopamine-related therapies are in current development for individuals with dopamine-related disorders including depression, Parkinson's disease, and psychostimulant addictions such as cocaine abuse. Yet, most efforts to develop new dopamine therapies are within costly structure-activity relationship studies. Through structure-based drug design techniques, the binding site of hDAT can be utilized to develop novel selective and potent dopamine the

Inhibition of dopamine uptake into PC-12 cells by analogues of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP).

The inhibition of uptake of radioactive dopamine into PC-12 cells by analogues of MPTP with systematic variations of structure was studied in order to assess the steric constraints of the transmembrane dopamine transporter. Of 15 analogues tested, 11 were found to inhibit dopamine uptake, all being competitive inhibitors. Major changes to the phenyl ring of MPTP, e.g. replacement of the phenyl by a naphthyl group, had a relatively modest effect on the inhibition of uptake of dopamine. Minor modifications to the N-methyl moiety reduced the ability to inhibit uptake, although the unmethylated an

Decrease in tobacco consumption after treatment with topiramate and aripiprazole: a case report.

INTRODUCTION: A large part of research into drug addiction focuses on mesolimbic dopamine circuitry; however, both alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid and/or kainate and dopamine D2 receptors can play a role in maintaining the established addiction. CASE PRESENTATION: We report the case of a 34-year-old man who compulsively smoked 80 to 100 cigarettes each day. After receiving treatment with topiramate and aripiprazole, his tobacco consumption was dramatically reduced. CONCLUSION: Alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid and/or kainate blocking agents and

Imaging dopamine transporters in Parkinson's disease.

The dopamine transporter (DAT) is responsible for clearance of dopamine from the synaptic cleft after its release. Imaging DAT availability provides a measure of dopamine terminal function and a method for detecting striatal dopamine deficiency states present in idiopathic Parkinson's disease and atypical neurodegenerative Parkinsonian disorders such as multiple system atrophy and progressive supranuclear palsy. DAT imaging with PET or single photon emission computed tomography can be used to support a diagnosis of dopamine-deficient parkinsonism in cases where this is suspected and rationaliz

Hyperprolactinaemia and DNA synthesis in the pituitary gland of the rat.

Hyperprolactinaemia produced in rats by the transplanted prolactin-secreting tumours MtTW15 and 7315a significantly (P less than 0.01) inhibited by 70% the incorporation of [3H]thymidine into the pituitary DNA of the host animals. The weight and the DNA content of the glands were significantly (P less than 0.01) reduced by 30%. The administration of haloperidol, a dopamine receptor blocking agent, to the tumour-bearing rats increased the suppressed DNA replication in the anterior pituitary glands by approximately 560% in the MtTW15-bearing rat and by 100% in the 7315a-bearing animals.
