The effects of various lethal mutations were studied when they were present in the hemizygous state as sectors in genetic mosaics (gynandromorphs) of Drosophila melanogaster. Sixty percent of the lethals studied can survive to adulthood in combination with normal tissue in mosaics, whereas the remainder cause the death of such mosaic animals. Some of the gynandromorph-viable lethals appeared to be locally active, in that they allowed the survival of only those mosaics in which the male tissue was present in the abdomen. The regions affected by these lethals were identified more precisely by an…

The transformation of the eye primordium into a monolayer eye epithelium with identical ommatidial units requires corporate and coordinated efforts of the retinal determination genes. During the process, the same set of retinal determination genes is utilized reiteratively to fulfill distinct functions concerning eye specification, eye patterning, cell proliferation, and cell survival. To achieve this sophisticated goal, instructions sent from the key developmental pathways and the dorsal-ventral patterning genes are incorporated, and connections are built among the retinal determination genes

Visual systems transform photoreceptor inputs into rich perceptual representations through hierarchical networks that extract features along parallel pathways. Although this architecture is conserved across species, how visual information is routed throughout an entire brain remains elusive in any animal. Using the male Drosophila connectome, we trace signals from photoreceptors through the optic lobes-layered, retinotopic regions containing two-thirds of the brain's neurons-and onward into the central brain. Network-based analyses reveal a multilayered architecture of pathway classes with dis…

The basement membrane (BM) is a specialized extracellular matrix that provides crucial structural support to tissues and organs. Mutations in BM genes can affect this structural support and are often associated with human diseases, including fibrosis, diabetes, and cancer, and are considered a hallmark of aging. However, how the BM maintains this support in organs with constant mechanical stress is largely unknown. In the Drosophila ovary, the BM provides the mechanical cues that are required for egg development. We discovered that the glycoprotein Ndg is important for maintaining organ integr…

The continuity of a species depends on germ cells. Germ cells are different from all the other cell types of the body (somatic cells) as they are solely destined to develop into gametes (sperm or egg) to create the next generation. In this review, we will touch on 4 areas of embryonic germ cell development in Drosophila melanogaster: the assembly and function of germplasm, which houses the determinants for germ cell specification and fate and the mitochondria of the next generation; the process of pole cell formation, which will give rise to primordial germ cells (PGCs); the specification of p…

The Drosophila melanogaster intestine has emerged as a powerful model system for studying the fundamental mechanisms underlying cancer biology, including tumorigenesis, metastasis, and paraneoplastic syndromes such as cachexia. The key signaling pathways implicated in human colorectal cancers play similar roles in the Drosophila intestinal system. Dysregulation of these pathways in the Drosophila intestine leads to overproliferation of intestinal stem cells, loss of cell differentiation, and potential transdifferentiation, mirroring tumorigenic processes in humans. Recent research utilizing Dr…

Late-stage cancer patients often exhibit cachexia, a wasting syndrome characterized by the loss of muscle and adipose tissues. However, the extent to which these wasting effects directly contribute to mortality remains unclear. Drosophila, a well-conserved model organism, has been instrumental in demonstrating systemic energy wasting and mortality in the context of tumors, similar to mice and humans. Research in Drosophila has elucidated the underlying mechanisms by which tumor-associated secreted proteins cause muscle atrophy and lipid loss, ultimately leading to energy wasting. Nevertheless,…

Gamete formation is essential for sexual reproduction in metazoans. Meiosis in males gives rise to spermatids that must differentiate and individualize into mature sperm. In Drosophila melanogaster, individualization of interconnected spermatids requires the formation of individualization complexes that synchronously move along the sperm bundles. Here, we show that Mob4, a member of the Mps-one binder family, is essential for male fertility but has no detectable role in female fertility. We show that Mob4 is required for proper axonemal structure and its loss leads to male sterility associated…

BACKGROUND: Dermatophytosis is one of the most common fungal infections worldwide. The distribution of dermatophytes varies across continents, but the genera Trichophyton and Microsporum have emerged as the main isolated agents in humans and animals.
OBJECTIVES: To validate Drosophila melanogaster flies as a fast and feasible model to study dermatophytic infections.
METHODS: Wild-type (WT) and Toll-deficient D. melanogaster flies were infected by Trichophyton rubrum, T. mentagrophytes, Microsporum canis and Nannizzia gypsea by pricking with a needle previously dipped in inoculum concentratio…

Telomeres in Drosophila melanogaster, which have inspired a large part of Sergio Pimpinelli work, are similar to those of other eukaryotes in terms of their function. Yet, their length maintenance relies on the transposition of the specialized retrotransposons Het-A, TART, and TAHRE, rather than on the activity of the enzyme telomerase as it occurs in most other eukaryotic organisms. The length of the telomeres in Drosophila thus depends on the number of copies of these transposable elements. Our previous work has led to the isolation of a dominant mutation, Tel1, that caused a several-fold el…

Fluid clearance from the respiratory system during developmental transitions is critically important for achieving optimal gas exchange in animals. During insect development from embryo to adult, airway clearance occurs episodically each time the molt is completed by performance of the ecdysis sequence, coordinated by a peptide-signaling cascade initiated by ecdysis-triggering hormone (ETH). We find that the neuropeptide Kinin (also known as Drosokinin or Leukokinin) is required for normal respiratory fluid clearance or "tracheal air-filling" in Drosophila larvae. Disruption of Kinin signaling…

Human genetic studies support that loss-of-function mutations in the SH3 domain and ankyrin repeat containing family proteins (SHANK1-3), the large synaptic scaffolding proteins enriched at the postsynaptic density of excitatory synapses, are causative for autism spectrum disorder and other neuropsychiatric disorders in humans. To better understand the in vivo functions of Shank and facilitate dissection of neuropathology associated with SHANK mutations in human, we generated multiple mutations in the Shank gene, the only member of the SHANK family in Drosophila melanogaster Both male and fema…

Tauopathies are a group of neurodegenerative disorders that include hereditary frontotemporal dementias (FTDs) such as FTD with parkinsonism linked to chromosome 17 (FTDP-17), as well as sporadic variants of FTDs like progressive supranuclear palsy (PSP), corticobasal degeneration (CBD), and Pick's disease. These diverse diseases all have in common the presence of abnormally phosphorylated tau aggregates. In this review, we will summarize key features of transgenic Drosophila models of tauopathies and a number of insights into disease mechanisms as well as therapeutic implications gained from …

Proteins, nucleic acids, and lipids are three major components of the cell. Despite a few basic metabolic pathways, we know very little about lipids, compared with the explosion of knowledge about proteins and nucleic acids. How many different forms of lipids are there? What are the in vivo functions of individual lipid? How does lipid metabolism contribute to normal development and human health? Many of these questions remain unanswered. For over a century, the fruit fly Drosophila melanogaster has been used as a model organism to study basic biological questions. In recent years, increasing …
