Transcriptional profiling identification of inflammatory signaling pathways in ulcerative colitis.

Ulcerative colitis (UC) is a chronic type of inflammatory bowel disease (IBD). This study identified core genes and pathways involved in UC by performing transcriptional profiling of colon biopsies from UC patients and healthy controls using data from the Gene Expression Omnibus (GEO) database. A total of 202 samples, including 129 UC patients and 73 healthy controls, were analyzed, measuring the expression of 40,991 genes using a 44K formatted microarray. Differential gene expression (DGE) analysis and gene set enrichment analysis (GSEA) identified several biomarkers potentially involved in U

Drosophila melanogaster mitigates gastro-oral infections by stimulating pathogen expulsion.

BACKGROUND: Food-borne pathogens, particularly belonging to the Enterobacteriaceae family, are a major cause of gastrointestinal infections in humans. While Drosophila melanogaster has been widely explored to study antimicrobial responses, most studies rely on septic injury or direct injection routes that bypass the gut. A coordinated set of antimicrobial defenses acts to counteract the invading bacteria, with some variations depending on the entry route into the host. Herein, we tracked the dynamics of Enterobacteriaceae pathogens in Drosophila using a natural infection route. RESULTS: Most

De novo DUOX2 expression in neutrophil subsets shapes the pathogenesis of intestinal disease.

Infiltrating neutrophils are key effector cells in inflammatory bowel disease (IBD) while providing antimicrobial defense and tissue restitution in the intestine. The complexity of neutrophil functions in local environments underscores our limited understanding of how their adaptation in tissues influences disease progression. Here, we demonstrate that neutrophils recruited in murine colitis and infection models, idiopathic IBD, and chronic granulomatous disease-associated IBD undergo extensive transcriptional reprogramming, resulting in the emergence of neutrophil populations that feature uni

Dual oxidase2 is expressed all along the digestive tract.

The dual oxidase (Duox)2 flavoprotein is strongly expressed in the thyroid gland, where it plays a critical role in the synthesis of thyroid hormones by providing thyroperoxidase with H2O2. DUOX2 mRNA was recently detected by RT-PCR and in-situ hybridization experiments in other tissues, such as rat colon and rat and human epithelial cells from the salivary excretory ducts and rectal glands. We examined Duox2 expression at the protein level throughout the porcine digestive tract and in human colon. Western blot analysis identified Duox2 as the same two molecular species (M(r) 165 and 175 kDa)

Eicosatetraynoic Acid and Butyrate Regulate Human Intestinal Organoid Mitochondrial and Extracellular Matrix Pathways Implicated in Crohn's Disease Strictures.

BACKGROUND: Perturbagen analysis of Crohn's disease (CD) ileal gene expression data identified small molecules including eicosatetraynoic acid (ETYA), which may exert an antifibrotic effect. We developed a patient-specific human intestinal organoid (HIO) model system to test small molecule regulation of mitochondrial and wound-healing functions implicated in stricturing behavior. METHODS: HIOs were made from CD induced pluripotent stem cells with and without a loss-of-function haplotype in the DUOX2 gene implicated in ileal homeostasis and characterized under basal conditions and following ex

Genetic disorders coupled to ROS deficiency.

Maintaining the redox balance between generation and elimination of reactive oxygen species (ROS) is critical for health. Disturbances such as continuously elevated ROS levels will result in oxidative stress and development of disease, but likewise, insufficient ROS production will be detrimental to health. Reduced or even complete loss of ROS generation originates mainly from inactivating variants in genes encoding for NADPH oxidase complexes. In particular, deficiency in phagocyte Nox2 oxidase function due to genetic variants (CYBB, CYBA, NCF1, NCF2, NCF4) has been recognized as a direct cau

Human Nasal and Bronchial Epithelium in Cystic Fibrosis: Differences in Expression of Airway pH Regulatory Proteins.

BACKGROUND: Decreased apical bicarbonate transport into the airway surface liquid (ASL) has been associated with decreased ASL pH, which can have adverse respiratory effects. However, the human CF epithelium can normalize ASL pH. Thus, we hypothesized that pH regulatory proteins other than the CFTR could be altered in the CF epithelium. METHODS: Primary human nasal and bronchial epithelial cells from healthy controls and CF subjects were grown at air-liquid interface until fully ciliated. Western blot was used to measure proteins known to affect human epithelial pH: carbonic anhydrases (CA) 1

Contactin 6, A Novel Causative Gene for Congenital Hypothyroidism, Mediates Thyroid Hormone Biosynthesis Through Notch Signaling.

Background: Congenital hypothyroidism (CH) is the most common neonatal metabolic disorder. In patients with CH in China, thyroid dyshormonogenesis is more common than thyroid dysgenesis; however, the genetic causes of CH due to thyroid dyshormonogenesis remain largely unknown. Therefore, we aimed at identifying novel candidate causative genes for CH. Methods: To identify novel CH candidate genes, a total of 599 patients with CH were enrolled and next-generation sequencing was performed. The functions of the identified variants were confirmed using HEK293T and FTC-133 cell lines in vitro and in

Roscovitine Worsens Mycobacterium abscessus Infection by Reducing DUOX2-mediated Neutrophil Response.

Persistent neutrophilic inflammation associated with chronic pulmonary infection causes progressive lung injury and, eventually, death in individuals with cystic fibrosis (CF), a genetic disease caused by biallelic mutations in the CF transmembrane conductance regulator (CFTR) gene. Therefore, we examined whether roscovitine, a cyclin-dependent kinase inhibitor that (in other conditions) reduces inflammation while promoting host defense, might provide a beneficial effect in the context of CF. Herein, using CFTR-depleted zebrafish larvae as an innovative vertebrate model of CF immunopathophysio

Phagocyte NADPH oxidase, oxidative stress and lipids: Anti- or pro ageing?

The role of NADPH oxidase in ageing is debated because of the dual roles of free radicals, toxic though necessary. In this paper we summarize some results about two aspects linked to the regulation of the activity of phagocyte NADPH oxidase (Nox2), encountered frequently in elderly people: inflammation and hypercholesterolemia. In the presence of a high amount of reactive oxygen species (ROS) created by itself or by any other source, the enzyme activity is mostly lowered. Oxidation of the membrane and/or of one of the cytosolic partners could be responsible for this loss of activity. However u

Virtual and in vitro bioassay screening of phytochemical inhibitors from flavonoids and isoflavones against xanthine oxidase and cyclooxygenase-2 for gout treatment.

Synthetic drugs such as allopurinol and benzbromarone are commonly used to treat the complex pathogenesis of gout, a metabolic disease that results from an inflammation of the joints caused by precipitation of uric acid. We seek to discover novel phytochemicals that could treat gout, by targeting the xanthine oxidase and cyclooxygenase-2 enzymes. In this study, we report the screening of nine compounds of flavonoids from the ZINC and PubChem databases (containing 2092 flavonoids) using the IGEMDOCK software tool against the xanthine oxidase and cyclooxygenase-2 3D protein structures. GPRC6A-Duox1 Axis Regulates the Hair Cycle Through H2O2 Generation.

BACKGROUND: Emerging evidence suggests that G-protein-coupled receptor family C group 6 member A (GPRC6A) and Dual oxidase 1 (Duox1) mediate a non-classical testosterone signaling pathway. However, the molecular mechanism in which testosterone mediates the GPRC6A-Duox1 cascade in the hair cycle was unclear. Therefore, this study aimed to elucidate the molecular role of the testosterone-GPRC6A-Duox1 signaling axis in regulating hair cycle progression and testosterone-mediated hair loss. METHODS: GPRC6A-deficient and Duox1-deficient keratinocytes were prepared and stimulated with testosterone t

Cu-V-Ag tri-metal mesoporous nanozyme for high-efficiency UV-vis/RGB monitoring of CrO42- and amoxicillin residue.

To monitor toxic CrO42- and amoxicillin residue efficiently, tri-metal Cu-V-Ag mesoporous nanospheres (Cu-V-Ag MNS) were identified in this work. Based on the synergistic effect among Ag0, V2O5, CuOx and bovine serum albumin (BSA), Cu-V-Ag MNS expressed outstanding oxidase-like activity to catalyze 3,3',5,5'-tetramethylbenzidine (TMB) oxidization with Vmax/Km of 0.0326 mM/2.86×10-8 M·s-1. Trace CrO42- or amoxicillin could exclusively enhance or deteriorate its oxidase-like activity with clear hyperchromic or hypochromic effect. Under optimized conditions, Cu-V-Ag MNS was applied for dual-mode

NADPH oxidase and endoplasmic reticulum stress is associated with neuronal degeneration in orbitofrontal cortex of individuals with alcohol use disorder.

Many disorders of the central nervous system (CNS), including alcohol use disorder (AUD), are associated with induction of proinflammatory neuroimmune signalling and neurodegeneration. In previous studies, we found increased expression of Toll-like receptors (TLRs), activated NF-κB p65 (RELA), and other proinflammatory signalling molecules. Proinflammatory NADPH oxidases generate reactive oxygen species, which are linked to neurodegeneration. We tested the hypothesis that AUD increased RELA activation increases NADPH oxidase-oxidative stress and endoplasmic reticulum (ER) stress cell death cas

Sexual dimorphism of thyroid reactive oxygen species production due to higher NADPH oxidase 4 expression in female thyroid glands.

BACKGROUND: Dual oxidases (DUOX1 and DUOX2) are NADPH oxidases (NOX) involved in hydrogen peroxide production necessary for thyroid hormonogenesis, but recently, the NOX4 has also been described in the thyroid gland. The prevalence of thyroid disease is higher in women, and the basis for this difference might involve a higher oxidative stress level in the female thyroid gland. Hence, we aimed at evaluating whether the function and the expression of enzymes involved in the thyroid redox balance differ between females and males. METHODS: DUOX1, DUOX2, NOX4, glutathione peroxidase (GPx), and cat

Inhibition of the thyroid hormonogenic H2O2 production by Duox/DuoxA in zebrafish reveals VAS2870 as a new goitrogenic compound.

Thyroid hormone (TH) synthesis requires extracellular hydrogen peroxide generated by the NADPH oxidases, DUOX1 and DUOX2, with maturation factors, DUOXA1 and DUOXA2. In zebrafish, only one duox and one duoxa gene are present. Using a thyroid-specific reporter line, we investigated the role of Duox and Duoxa for TH biosynthesis in zebrafish larvae. Analysis of several zebrafish duox and duoxa mutant models consistently recovered hypothyroid phenotypes with hyperplastic goiter caused by impaired TH synthesis. Mutant larvae developed enlarged thyroids and showed increased expression of the EGFP r

NADPH oxidase promotes neutrophil extracellular trap formation in pulmonary aspergillosis.

NADPH oxidase is a crucial enzyme in antimicrobial host defense and in regulating inflammation. Chronic granulomatous disease (CGD) is an inherited disorder of NADPH oxidase in which phagocytes are defective in generation of reactive oxidant intermediates. Aspergillus species are ubiquitous, filamentous fungi, which can cause invasive aspergillosis, a major cause of morbidity and mortality in CGD, reflecting the critical role for NADPH oxidase in antifungal host defense. Activation of NADPH oxidase in neutrophils can be coupled to the release of proteins and chromatin that comingle in neutroph

Implantable hydrogel-based scaffold integrating cascaded chemotherapy and self-amplified immunotherapy to suppress postsurgical tumor recurrence.

Tumor recurrence following surgical resection remains a formidable clinical challenge, as conventional adjuvant therapies are often constrained by chemoresistance and chemotherapy-associated complications. To circumvent these limitations, we engineered a modular, three-dimensional (3D)-printed hydrogel-based scaffold for synergistic chemotherapy and immunotherapy. The scaffold, composed of soybean isolate protein, polyvinyl alcohol, sodium alginate, and magnesium acetate, was loaded with doxorubicin (DOX) and tirapazamine (TPZ) to establish a sequential therapeutic cascade.
