Endocrine disruption in crustaceans due to pollutants: a review.

The main endocrine-regulated processes of crustaceans have been reviewed in relation to the effects of endocrine-disrupting compounds (EDCs). Molting has been shown to be inhibited by several organic pollutants, such as xenoestrogens and related compounds, as well as by some pesticides. Most of these disrupters are thought to interfere with ecdysone at target tissues, although only for a few has this action been demonstrated in vitro. The heavy metal cadmium appears to inhibit some ecdysone secretion. Juvenoid compounds have also been shown to inhibit molting, likely by interfering with the st

Localization of the mosquito insulin receptor homolog (MIR) in reproducing yellow fever mosquitoes (Aedes aegypti).

The female mosquito takes a blood meal to produce a batch of eggs. Initiation of egg maturation and growth of oocytes is governed by several endocrine factors. Peptide factors from the brain are involved in this process and some are also responsible for the induction of ecdysone secretion. The latter appears to be required to maintain a high rate of vitellogenin synthesis. By analogy with the known functions of insulin-like molecules (e.g. bombyxins) which in insects activate the secretion of ecdysteroids, we have postulated that there is an insulin receptor homolog responsible for activation

Acetylation enables a dual-targeting fungal effector to inhibit insect immunity and development.

Pathogenic microbes deploy secreted effectors to subvert host immunity and physiology across kingdom boundaries, yet the molecular mechanisms that maintain effector competence during this transition remain poorly defined. Here, we identify MrNIS1, an effector from the insect-pathogenic fungus Metarhizium robertsii whose stability and secretion are governed by K24 acetylation through physical association with the acetyltransferase MrKAT1 and the deacetylase MrSIR2. Once inside the insect, MrNIS1 acts as a dual-targeting effector to disrupt distinct physiological pathways. It binds the serine pr

Conserved cellular signals and mechanisms accompany the reproductive shutdown of honey bee (Apis mellifera) queens for dispersal.

Temporal reproductive plasticity is a central life-history trait for most iteroparous animals, yet its underlying molecular mechanisms remain poorly understood. We utilize the reversible reproductive arrest experienced by honey bee queens during colony swarming to investigate how these reproductive specialists dynamically adapt their physiology. We demonstrate that pre-swarming nutritional restriction triggers a resource reallocation, whereby ovarian mass declines as flight muscle function is enhanced, providing empirical support for a flight-fecundity trade-off. This transition is accompanied

Bollworm responses to release of genetically modified Helicoverpa armigera nucleopolyhedroviruses in cotton.

Helicoverpa armigera single nucleocapsid nucleopolyhedrovirus (HaSNPV) has been developed as a commercial biopesticide to control the cotton bollworm, H. armigera, in China. The major limitation to a broader application of this virus has been the relative long time to incapacitate the target insect. Two HaSNPV recombinants with improved insecticidal properties were released in bollworm-infested cotton. One recombinant (HaCXW1) lacked the ecdysteroid UDP-glucosyltransferase (egt) gene and in another recombinant (HaCXW2), an insect-selective scorpion toxin (AaIT) gene replaced the egt gene. In a

Barren female Strongyloides stercoralis from occult chronic infections are rejuvenated by transfer to parasite-naive recipient hosts and give rise to an autoinfective burst.

It is widely assumed that barren Strongyloides stercoralis occurring in chronically infected carriers can become fecund when immunity wanes. Evidence for this involves corticosteroid treatment of hosts harboring occult infections that subsequently return to patency. However, nematodes have ecdysteroid receptors, and it has been suggested that corticosteroids act directly on the parasite, inducing autoinfective development, rather than indirectly by suppressing host immunity. To test these competing concepts, barren females were recovered from donor dogs when the dogs' fecal examinations turned

Comparative transcriptology reveals effects of circadian rhythm in the nervous system on precocious puberty of the female Chinese mitten crab.

In juvenile Chinese mitten crabs, ovarian maturation occurring in the first year is known as precocity, and can cause huge economic losses to crab breeding. To discover the molecular mechanisms underlying the regulation of the nervous system of female precocious crabs, eyestalk, brain, and thoracic ganglion transcriptome data were obtained in normal and precocious crabs via high-throughput sequencing technology. A total of 81, 4276, and 22,684 differentially-expressed genes were obtained from the eyestalk, brain, and thoracic ganglion groups, respectively. Functional analysis showed that these

Transcriptome profiling to decipher molecular mechanisms of diapause in Chilo partellus (Swinhoe) (Lepidoptera: Crambidae).

Diapause is a critical adaptive strategy in insects to survive unfavourable environmental conditions. The present study investigated molecular mechanisms underlying diapause in Chilo partellus, using high-throughput RNA sequencing and de novo transcriptome assembly. Six transcriptome libraries from diapause and non-diapause larvae yielded over 50 million reads per sample, with >98% mapping efficiency. The Evidential Gene-refined assembly produced 67,116 high-quality transcripts, and BUSCO analysis confirmed its completeness. DEG analysis identified 1204 DEGs, including 660 upregulated and 544

The amphipod ecdysone receptor as a complementary tool for environmental risk assessment: from functional analysis to proof of concept study.

Endocrine-disrupting chemicals (EDCs) pose significant risks to aquatic wildlife by disrupting hormonal signaling pathways, notably those mediated by nuclear receptors (NRs) - key regulators of endocrine processes. For decades, in vitro bioassays based on the interaction between NRs and EDCs have been used to investigate EDC activity in chemical monitoring and risk assessment frameworks. Yet, current guidelines are mostly restricted to vertebrate species, failing to capture the diversity of chemical-induced NR-driven effects in aquatic animals - particularly arthropods, whose endocrine systems

Two insulin-like peptide family members from the mosquito Aedes aegypti exhibit differential biological and receptor binding activities.

Insects encode multiple ILPs but only one homolog of the vertebrate IR that activates the insulin-signaling pathway. However, it remains unclear whether all insect ILPs are high affinity ligands for the IR or have similar biological functions. The yellow fever mosquito, Aedes aegypti, encodes eight ILPs with prior studies strongly implicating ILPs from the brain in regulating metabolism and the maturation of eggs following blood feeding. Here we addressed whether two ILP family members expressed in the brain, ILP4 and ILP3, have overlapping functional and receptor binding activities. Our resul

Recent advances in epidemiological field techniques in filariasis.

There have been very few advances in epidemiological field techniques for filariasis in recent years. Advances in other areas of study of filariasis have been published and a summary of some of those which have some application to field technology will be given. Studies on vectors involving host influences on the development of filariae and the use of DNA probes for vector speciation were reported as well as the development of techniques that yielded species-specific identification of larvae in mosquito vectors. Experiments on animal transmission potentials also have been described. Mathematic

A novel strategy for the protective effect of ginsenoside Rg1 against ovarian reserve decline by the PINK1 pathway.

CONTEXT: The decline in ovarian reserve is a major concern in female reproductive health, often associated with oxidative stress and mitochondrial dysfunction. Although ginsenoside Rg1 is known to modulate mitophagy, its effectiveness in mitigating ovarian reserve decline remains unclear. OBJECTIVE: To investigate the role of ginsenoside Rg1 in promoting mitophagy to preserve ovarian reserve. MATERIALS AND METHODS: Ovarian reserve function, reproductive capacity, oxidative stress levels, and mitochondrial function were compared between ginsenoside Rg1-treated and untreated naturally aged fem

Epidermal carbonic anhydrase activity and exoskeletal metal content during the molting cycle of the blue crab, Callinectes sapidus.

During the crustacean molting cycle, the exoskeleton is first mineralized in postmolt and intermolt and then presumably demineralized in premolt in order for epidermal retraction to occur. The mineralization process calls for divalent metal ions, such as Ca(2+) and Mg(2+) , and bicarbonate ions whereas protons are necessary for dissolution of carbonate salts. Carbonic anhydrase (CA) has been suggested to be involved in exoskeletal mineralization by providing bicarbonate ions through catalyzing the reaction of carbon dioxide hydration. However, results of earlier studies on the role of epiderma

Analysis of molecular markers for metamorphic competency and their response to starvation or feeding in the mosquito, Aedes aegypti (Diptera: Culicidae).

The nutritional condition of fourth instar larvae of the yellow fever mosquito, Aedes aegypti, governs female longevity and egg production, both are key determinants of pathogen transmission. As well, nutrition provisions larval growth and development and attains its greatest pace in the last larval instar in preparation for metamorphosis to an adult. These developmental processes are regulated by a complex endocrine interplay of juvenile hormone, neuropeptides, and ecdysteroids that is nutrition sensitive. We previously determined that feeding for only 24h post-ecdysis was sufficient for four

On the control of ecdysone biosynthesis by the central nervous system of blowfly larvae.

Ecdysone was found to be the major secreted steroid of ring glands dissected from blowfly larvae and incubated in vitro. Other secretory products such as 3-dehydroecdysone and 20-deoxy-makisterone A could not be detected when the glands were labelled with tritiated cholesterol. Ecdysone synthesis and secretion were found to be tightly coupled. The highest rate of secretion was observed a few hours before pupariation. In vitro, the rate of ecdysone secretion by ring glands was affected significantly by coincubation with the central nervous system (CNS). Modulating effects from the CNS to the gl

RNAi-Mediated Knockdown of the Sodium Channel Auxiliary Subunit TipE Increases Pyrethroid-Associated Mortality and Reveals Metabolic Transcriptional Responses in Megalurothrips usitatus.

Background: The bean flower thrips, Megalurothrips usitatus, is a destructive agricultural pest with increasing resistance to pyrethroid insecticides. Most mechanistic studies emphasize mutations in the pore-forming voltage-gated sodium channels α-subunit, whereas the contribution of insect-specific sodium-channel auxiliary subunits remains unclear. Methods: We used dsRNA feeding, pyrethroid bioassays, RNA sequencing, and RT-qPCR validation to evaluate the role of temperature-induced paralytic E (TipE) in M. usitatus. Results: The optimized RNAi condition was 400 μg/mL of dsTipE for 48 h, whic

Ovarian transcriptome analyses indicate that weak juvenile hormone signaling underlies the molecular basis of oogenesis deficiencies in mosquitoes.

BACKGROUND: Juvenile hormone (JH) is synthesized by the corpora allata (CA) and controls development and reproduction in insects. We recently used CRISPR/Cas9 to establish a line lacking the enzyme that catalyzes the final step of JH biosynthesis in mosquitoes, a P450 epoxidase. The CA of the epox-/- mutants do not synthesize epoxidized JH III but methyl farneosate (MF), a weak agonist of the JH receptor. Female epox-/- mosquitoes have reduced JH signaling and show a substantial loss of reproductive fitness. To understand the molecular basis of this loss of fitness, we constructed ovarian mRNA

Ovarian microrna transcriptome of Macrobrachium rosenbergii after eyestalk ablation: unveiling molecular switches in reproductive regulation.

BACKGROUND: Macrobrachium rosenbergii is an economically important freshwater prawn species in China. The X-organ–sinus gland complex in crustacean eyestalks regulates reproduction through neurohormones, with ablation significantly promoting ovarian maturation. However, the miRNA mediated molecular mechanisms underlying this eyestalk ovary regulatory axis remain poorly characterized in M. rosenbergii. RESULTS: RNA sequencing of 9 ovarian samples produced 207 million paired-end reads (62.25 Gb total), averaging > 6 Gb per sample with Q30 scores exceeding 94.42%. The reads showed 84.32% alignmen

Metabolomic analysis of combined exposure to microplastics and methylmercury in the brackish water flea Diaphanosoma celebensis.

Owing to their widespread distribution and high bioaccumulation, microplastics (MPs) and mercury (Hg) are considered major threats to the ocean. MP interacts with Hg because of its high adsorption properties. However, their toxicological interactions with marine organisms, especially combined effects at the molecular level, are poorly understood. This study investigated the single and combined effects of MP and Hg on the metabolic profile of the brackish water flea Diaphanosoma celebensis. A total of 238 metabolites were significantly affected by MP, Hg, or MP + Hg. Metabolite perturbation pat

Asymptomatic Strongyloides stercoralis hyperinfection in an alcoholic patient with intense anemia.

Strongyloides stercoralis infection is endemic in many tropical and subtropical areas. The parasite has the unusual ability to multiply inside the host due to the transformation of rhabditiform larvae into infective filariforms. Several studies have shown that chronic alcoholism is an important factor that predisposes to strongyloidiasis. The increased susceptibility to S. stercoralis infections seen in alcoholic individuals could be explained by their increased exposure to the parasite, malnutrition, breakdown of local immune responses, and/or alterations in intestinal barriers. Moreover, eth
