Development of a recombinase-aided amplification assay for detection of orf virus.

Orf, caused by orf virus (ORFV), is an important zoonotic disease that infects goat and sheep, leading to huge economic losses. ORFV can also cause cutaneous lesions in people who come in close contact with the diseased animals. Although accurate diagnostic methods for ORFV infection exist, there is a need for a rapid, specific, and sensitive method for easy clinical application. Here, we successfully established a recombinase-aided amplification (RAA) assay for rapid detection of ORFV. The analytical sensitivity of the assay for ORFV detection is 1 × 101 copies per reaction. Moreover, no cros

Transplacental (vertical) transmission of Orf virus in mice (Mus musculus).

Contagious ecthyma, caused by Orf virus (Parapoxvirus), is enzootic in sheep and goats across Asia, Africa, and other regions of the world. Due to the self-limiting nature of the disease, it has been neglected for decades, causing huge economic losses to sheep and goat farming industries. Young animals are most widely affected by this disease. Several outbreaks of Orf virus disease have been reported in previous years in many provinces of China. By using the specific pathogen-free mice (C57BL/6), molecular detection (targeting the conserved region of the viral B2L gene), histopathological eval

Polymerase chain reaction for laboratory diagnosis of orf virus infections.

BACKGROUND: The orf virus of sheep and goats is one of several zoonotic parapoxviruses. In the ovine/caprine host it causes contagious ecthyma (contagious pustular dermatitis, scabby mouth), but in humans it normally causes solitary or clustered orf lesions, typically on hands, arms or face. In addition to disease in the animals, the virus can be quite a nuisance as an occupational hazard in farmers and butchers. Clinical diagnosis is often possible, but laboratory diagnosis is sometimes necessary. For virus isolation, primary ovine or bovine cells, not routinely present, are needed. Serologic

Mastitis and other diseases of the goat's udder.

Udder problems of modern dairy goats are similar to those seen in dairy cows. Anomalies of the goat's udder and teats are common, and many may be hereditary. Skin diseases of the udder include viral infections, mange, sunburn, wounds, and staphylococcal dermatitis. There are numerous known causes of caprine mastitis. These include streptococci, hemolytic and nonhemolytic staphylococci, corynebacteria, and mycoplasmas. Diagnosis of mastitis in goats is often difficult, as the udder secretion may remain grossly normal and somatic cell counts in nonmastitic goats are higher than the recognized no

Important Diseases of Small Ruminants in Sub-Saharan Africa: A Review with a Focus on Current Strategies for Treatment and Control in Smallholder Systems.

Sheep and goats are an important source of livelihood for smallholder farmers in sub-Saharan Africa (SSA). These livestock are almost entirely managed by resource-poor, smallholder farmers and pastoralists. Despite the large number of sheep and goats in SSA, their productivity is low, mainly due to diseases, poor feed, and inferior breeds. This review aims to summarize the most important diseases in small ruminants in SSA, with a focus on current treatment and control strategies. The following diseases were identified as the most significant constraints for small ruminant farmers: helminthoses

Causation and epidemiological features of proliferative lip and facial lesions in Lao goats.

Past surveys and studies have identified a syndrome of lip and facial dermatitis as common in Lao goats and identified Orf virus (ORFV) as the likely cause. To more definitively determine the cause of this syndrome, a matched case-control study was implemented in 70 smallholder goat farms and a large-scale goat holding from Savannakhet province, Laos. Forty-one scab samples from goats with lip and facial dermatitis (cases) and 41 paired samples from unaffected animals (controls) were collected from 31 goat holdings over 12 months. Samples were tested using quantitative PCR assays targeting pos

The clinical aspects of anthrax.

Cutaneous anthrax is usually easy to diagnose provided the doctor thinks of anthrax. The lesion most likely to be confused with anthrax is accidental vaccinia. Orf lacks the characteristic central eschar. Cutaneous anthrax responds to antibiotic therapy: rare complications are meningitis and hypoxic hypertension. Pulmonary anthrax is highly fatal: its incidence is related to the number and size of anthrax-containing particles which are inhaled. Artificial mists containing lethal doses of anthrax bacilli can be manufactured. Intestinal anthrax may present as gastroenteritis or as ulceration of

A novel duplex TaqMan qPCR assay for rapid differential diagnosis and co-infection screening of Orf virus and Brucella spp. in goats.

Orf and brucellosis are significant infectious diseases that pose serious threats to the goat farming industry. Despite their co-occurrence in goat herds, no TaqMan-based duplex qPCR assay has been reported for their simultaneous detection. In this study, we developed a duplex TaqMan real-time qPCR assay for the simultaneous detection of Orf virus (ORFV) and Brucella spp., facilitating rapid differential diagnosis and co-infection screening. Specific primers and probes targeting conserved regions of the ORFV B2L gene and Brucella spp. Bcsp31 gene were designed, and reaction conditions were opt

The re-emerging of orf virus infection: A call for surveillance, vaccination and effective control measures.

Orf disease is known to be enzootic among small ruminants in Asia, Africa, and some other parts of the world. The disease caused by orf virus is highly contagious among small ruminant species. Unfortunately, it has been neglected for decades because of the general belief that it only causes a self-limiting disease. On the other hand, in the past it has been reported to cause huge cumulative financial losses in livestock farming. Orf disease is characterized by localized proliferative and persistent skin nodule lesions that can be classified into three forms: generalized, labial and mammary or

Lamb pays lip service: two cases of ecthyma contagiosum (orf).

Ecthyma contagiosum (orf) is caused by a parapox virus, which results in ulcerative stomatitis of mainly sheep and goats. The disease may be transmitted to humans through direct contact. Complications are rare in healthy individuals, who rarely report the disease. Two married, recreational sheep farmers, were bitten on their index fingers by an affected lamb. While the husband made an uneventful recovery after oral flucloxacillin, his wife was admitted to hospital with necrosis of her finger, cellulitis and lymphangitis requiring intravenous clindamycin. She subsequently developed a generalize

A novel ORF127/ORF130 gene deletion Orf virus vaccine candidate: construction and evaluation.

BACKGROUND: Orf is a zoonotic infectious disease caused by the Orf virus (ORFV), posing a serious threat to the health of ruminants and humans. Vaccination remains the most effective strategy for disease control. However, the limitations of conventional vaccines necessitate the development of safer and more efficient alternatives. METHODS: Based on the previously attenuated Orf vaccine strain (HB-TS09F65) as the parental virus, two gene-deletion strains, rHB09-ΔORF127 and rHB09-ΔORF130, were successfully constructed by deleting the ORF130 and ORF127 genes via homologous recombination. Construction and Biological Characteristics of a Quadruple Gene-Deleted Strain of Orf Virus as a Vaccine Candidate.

(1) Background: Contagious ecthyma, also known as orf, is an epitheliotropic zoonotic disease caused by the orf virus (ORFV), primarily affecting the skin and mucous membranes of ruminants such as goats and sheep, leading to the formation of papules and pustules. Vaccination is the most effective way to prevent this disease in susceptible animals; however, traditional attenuated vaccines carry the potential risk of reversion to virulence. Therefore, there is an urgent need to develop safe and effective vaccines for the prevention and control of orf. (2) Methods: In this study, building upon th

Molecular insights of immune evasion by Orf Virus: the interaction and functional impact of viral OH1 phosphatase on STAT1.

Orf virus, a member of the Parapoxvirus genus within the Poxvirus family, is the causative agent of Contagious Ecthyma, a zoonotic infection primarily affecting goats and sheep. The immune response against Orf virus is short-lived, enabling the virus to repeatedly infect animals, regardless of their vaccination status. Several virulence factors, including the OH1 tyrosine phosphatase, are responsible for the modulation of the host immune response. Here, we report the direct interaction between the viral OH1 and the human transcription factor STAT1 identified as a physiological OH1 substrate. I

A review of mutual transmission of important infectious diseases between livestock and wildlife in Europe.

Oral vaccination of red foxes against rabies has been practiced in Europe since 1978 and has succeeded in greatly reducing the occurrence of this disease in foxes: this is an example of coordinated activity against a disease that affects both wild and domestic animals as well as humans. Some examples of diseases that affect both domestic and wild animals in Europe are: classical swine fever (hog cholera) in wild boars and domestic swine; myxomatosis and rabbit hemorrhagic disease in domestic and wild rabbits; bovine viral diarrhea (BVD) in cattle and roe deer; contagious ecthyma in domestic sh

[Histopathology of the more common viral skin infections].

We describe the histopathological characteristics of viral skin infections. Herpes simplex virus and varicella-zoster virus produce an intraepidermal vesicle with variable degrees of epithelial necrosis. Typical findings include keratinocytes with ballooned nuclei with a ground-glass appearance and giant multinucleated keratinocytes. In the endothelial cells of the dermal blood vessels, cytomegalovirus produces large eosinophilic nuclear inclusions surrounded by a clear halo. Human herpes virus 8 is etiologically associated with Kaposi sarcoma. In its early stages, this tumor contains blood ve
