Ultrastructural correlates of luminescence in Porichthys photophores. II. Effects of metabolic inhibitors.

Prolonged, bright luminescent glows in Porichthys photophores are elicited by administration of 2,4-dinitrophenol (DNP) and potassium cyanide (KCN). Ultrastructural alterations of varicose nerve endings precede photocyte changes during such luminescent activity. Common alterations of nerve profiles include mitochondrial disruptions, flattening and depletion of synaptic vesicles, formation of large vacuolar cisternae, and invaginations in the contour of axolemma. Protracted luminescent activity in response to DNP results in depletion of photocyte vesicle material while vesicle and ER membranes

The Computational Functions of the Inferior Olive: Clues from Evolution.

Precerebellar structures, mainly studied in weakly electric fish, act to remove the effects of the fish's own movements on the electrical signals they receive, but do not directly control movement, nor do they have a clear error signal. The cerebellum does directly control movement - arguably the most prominent difference in function between the two structures. The most pronounced anatomical difference between precerebellar structures and the cerebellum itself is that the latter possesses an inferior olive. This conjunction suggests that the inferior olive carries out a function that is necess

Opioids potentiate electrical transmission at mixed synapses on the Mauthner cell.

Opioid receptors were shown to modulate a variety of cellular processes in the vertebrate central nervous system, including synaptic transmission. While the effects of opioid receptors on chemically mediated transmission have been extensively investigated, little is known of their actions on gap junction-mediated electrical synapses. Here we report that pharmacological activation of mu-opioid receptors led to a long-term enhancement of electrical (and glutamatergic) transmission at identifiable mixed synapses on the goldfish Mauthner cells. The effect also required activation of both dopamine

Effects of adenosine on the contractility of normoxic rainbow trout heart.

Temperature strongly affects oxygen solubility in water, oxygen convection in the blood and locomotor activity of the fish. Since oxygen supply and demand are temperature dependent, it was hypothesized that the purinergic control of the heart, one of the most important mediators in oxygen-limited conditions, might also show temperature dependence. Therefore, the present study examines the effects of adenosine (Ado), a purinergic agonist, on the contractile and electrical activity of the thermally acclimated trout ( Oncorhynchus mykiss Walbaum) heart. The fish were acclimated to either 4 degree

Marine Nematodes in Seafood: A Review of Food Safety Concerns, Advanced Detection Techniques, and Mitigation Strategies.

Marine nematodes are important food-borne parasites infecting many species of marine fish and cephalopods. This review provides a comprehensive discussion of the food safety concerns associated with marine nematodes in seafood, along with emerging methods for their detection. The potential prevention strategies for these seafood-borne parasites were also explored. According to the results, anisakiasis is the most serious nematode disease humans acquire from marine fish, chiefly due to Anisakis simplex larvae and, to a lesser degree, Pseudoterranova decipiens. Several analytical methods are emp

Differential Regulation of Gonadotropins as Revealed by Transcriptomes of Distinct LH and FSH Cells of Fish Pituitary.

From mammals to fish, reproduction is driven by luteinizing hormone (LH) and follicle-stimulating hormone (FSH) temporally secreted from the pituitary gland. Teleost fish are an excellent model for addressing the unique regulation and function of each gonadotropin cell since, unlike mammals, they synthesize and secrete LH and FSH from distinct cells. Only very distant vertebrate classes (such as fish and birds) demonstrate the mono-hormonal strategy, suggesting a potential convergent evolution. Cell-specific transcriptome analysis of double-labeled transgenic tilapia expressing GFP and RFP in

Electrical resonance of isolated hair cells does not account for acoustic tuning in the free-standing region of the alligator lizard's cochlea.

The cochlea of the alligator lizard is divided into two morphologically and physiologically distinct regions. In the "tectorial region," hair bundles of hair cells are draped by a tectorial membrane, whereas in the "free-standing region," hair bundles are said to be free-standing because there are no overlying tectorial structures. The acoustic tuning of the free-standing region depends at least in part on mechanical resonances of the hair bundles. In the turtle cochlea, in contrast, acoustic tuning depends in large part upon the electrical properties of the hair cells. We have investigated th

Rabbitfish ("aras"): an unusual source of ciguatera poisoning.

BACKGROUND: Ciguatera poisoning is the commonest fish-borne seafood intoxication. It is endemic to warm water tropical areas and is caused by consumption of bottom-dwelling shore reef fish, mostly during spring and summer. The causative agent, ciguatoxin, is a heat-stable ester complex that becomes concentrated in fish feeding on toxic dinoflagellates. The common clinical manifestations are a combination of gastrointestinal and neurologic symptoms. Severe poisoning may be associated with seizures and respiratory paralysis. OBJECTIVE: To describe a series of patients who sustained ciguatera po

Plasticity in an electrosensory system. II. Postsynaptic events associated with a dynamic sensory filter.

1. This report summarizes studies of the changes in postsynaptic potentials that occur as pyramidal cells within the primary electrosensory processing nucleus learn to reject repetitive patterns of afferent input. The rejection mechanism employs "negative image inputs" that oppose or cancel electroreceptor afferent inputs or patterns of pyramidal hyperpolarization or depolarization caused by intracellular current injection. Feedback pathways carrying descending electrosensory as well as other types of information provide the negative image inputs. This study focuses on the role of a directly d

Interactive effects of mercury exposure and hypoxia on ECG patterns in two Neotropical freshwater fish species: Matrinxã, Brycon amazonicus and traíra, Hoplias malabaricus.

Hypoxia and mercury contamination often co-occur in tropical freshwater ecosystems, but the interactive effects of these two stressors on fish populations are poorly known. The effects of mercury (Hg) on recorded changes in the detailed form of the electrocardiogram (ECG) during exposure to progressive hypoxia were investigated in two Neotropical freshwater fish species, matrinxã, Brycon amazonicus and traíra, Hoplias malabaricus. Matrinxã were exposed to a sublethal concentration of 0.1 mg L-1 of HgCl2 in water for 96 h. Traíra were exposed to dietary doses of Hg by being fed over a period of

Ampullary sense organs, peripheral, central and behavioral electroreception in chimeras (Hydrolagus, Holocephali, Chondrichthyes).

Ampullary sense organs are distributed in groups over the head of Hydrolagus colliei with their pores in clusters and innervated by the buccal, hyomandibular and superficial ophthalmic branches of the anterior lateral line nerve. The ampullae contain ciliated sense cells in an alveolate-shaped epithelium, which communicates to the surface through a jelly-filled tube. The sense cells synapse at their bases with the afferent nerve fibers that terminate in the dorsal nucleus of the anterior lateral line lobe of the medulla. The anatomy and ultrastructure support the homology with the ampullae of

Polyphenol Conductive Nanozyme-Enhanced Redox Hydrogel Coupled with Vagus Nerve Electrical Stimulation for Diabetic Wound Repair.

Diabetic wound repair is complicated by excessive oxidative reactions, bacterial infection, and persistent inflammation. Nanozyme-incorporated hydrogels have shown potential to facilitate diabetic wound repair. This study developed a polyphenol-conductive nanozyme-enhanced redox hydrogel composed of dopamine-grafted fish gelatin, methacrylated silk fibroin, and dopamine-mediated poly(3,4-ethylenedioxythiophene) (PDA-Fe-PEDOT) conductive nanozymes. The PDA-Fe-PEDOT conductive nanozyme confers electrical conductivity to the hydrogel, enabling it to couple endogenous electrical signals at the wou

Three types of gonadotrophin-releasing hormone neurones and steroid-sensitive sexually dimorphic kisspeptin neurones in teleosts.

In general, the gonadotrophin-releasing hormone (GnRH) neuronal systems of vertebrates consist of one group of hypothalamic neuroendocrine and one or two group(s) of extrahypothalamic neuromodulatory GnRH neurones. By taking advantage of the brains of dwarf gourami and GnRH-green fluorescent protein transgenic medaka, the spontaneous electrical activities of all three different types of GnRH neurones have now been characterised. The hypophysiotrophic preoptic area-gnrh1 neurones show irregular and episodic spontaneous electrical activities, whereas extrahypothalamic midbrain gnrh2 and terminal

Organochlorine chemicals in seafood: occurrence and health concerns.

The cheap availability of chlorine gas, together with the development of industrial chlorinating procedures in the 20th century, led to the production of a wide range of organochlorine compounds many with a variety of commercial applications, including usage as insecticides and defoliants and polychlorinated biphenyls (PCBs) used as coolants in electricity supply transformers. However, it was soon found that many of these technologically valuable chemicals suffered from a major disadvantage in that they resisted biodegradation and that the continued use of these compounds would lead to their p

Mathematical analysis of the stimulus for the lateral line organ.

Behavioral studies have shown that a blind fish is capable of detecting and recognizing stationary objects in its surroundings. It is proposed that the displacement of water caused by the fish as it moves is the basis for this detection capability. Alterations in the displacement of water around the fish, caused by the obstacle, act as stimuli for the lateral line organ. The question of how these stimuli acting on the skin of the fish, image the environment and what information is thus made available to the fish is the concern of this paper. The stimuli for the lateral line organ are derived m

A mechanism for differential control of axonal and dendritic spiking underlying learning in a cerebellum-like circuit.

In addition to the action potentials used for axonal signaling, many neurons generate dendritic "spikes" associated with synaptic plasticity. However, in order to control both plasticity and signaling, synaptic inputs must be able to differentially modulate the firing of these two spike types. Here, we investigate this issue in the electrosensory lobe (ELL) of weakly electric mormyrid fish, where separate control over axonal and dendritic spikes is essential for the transmission of learned predictive signals from inhibitory interneurons to the output stage of the circuit. Through a combination

Twenty Years of Research in Seabass and Seabream Welfare during Slaughter.

The behavioural responses of fish to a stressful situation must be considered an adverse reaction caused by the perception of pain. Consequently, the handling prior to stunning and the immediacy of loss consciousness following stunning are the aspects to take into account during the slaughtering process. The most common commercial stunning method in seabream and seabass is based on hypothermia, but other methods such as electrical stunning, carbon dioxide narcosis or anaesthetic with clove oil, are discussed in relation to the time to reach the unconsciousness stage and some welfare indicators

The effects of pulsing magnetic fields on pineal melatonin synthesis in a teleost fish (brook trout, Salvelinus fontinalis).

Based on findings in various mammalian species, where exposure to electromagnetic fields decreased the nocturnal synthesis of the pineal secretory product melatonin, we investigated the effects of magnetic field (MF) exposure in a teleost fish, the brook trout (Salvelinus fontinalis). Fields were generated by Helmholtz coils (maximum flux density 40 microT, frequency 1 Hz, 200 ms on, 800 ms off). Melatonin concentrations were estimated by a specific radioimmunoassay. MF exposure significantly increased night-time pineal (P < 0.001) and serum (P < 0.01) melatonin levels, as compared with the co

Connections of the olfactory bulb in the gymnotiform fish, Apteronotus leptorhynchus.

This work examines the connectivity of the olfactory bulb in the gynmotiform fish Apteronotus leptorhynchus. Wheat germ agglutinin conjugated horseradish peroxidase was iontophoresed in different areas and depths of the bulb in order to define its efferent and afferent connections. The olfactory bulb projects bilaterally via the medial (medial and centromedial fascicles) and lateral olfactory (lateral and centrolateral fascicles) tracts. The nervus terminalis courses through the ventromedial aspect of the bulb to terminate in parts of the medial subpallium and hypothalamus. Its telencephalic c

Anesthesia Overdose Versus Rapid Cooling for Euthanasia of Adult Zebrafish.

The rapid increase in zebrafish use needs to be accompanied by research into the refinement of procedures. The European (EU) Directive lists three possible euthanasia methods for fish: anesthetic overdose, electrical stunning, and concussion. However, for small fish such as zebrafish, concussion and electrical stunning are difficult to perform, leaving anesthetic overdose as the most used method. Our aim was to test the efficacy and side effects of anesthesia overdose using different anesthetics and the rapid cooling method to euthanize adult zebrafish. Adult mixed-sex AB zebrafish were random
