Molecular evidence of St. Louis encephalitis virus infection in patients in Buenos Aires, Argentina.

We report two cases of St. Louis encephalitis where the virus was detected in patients' sera directly by molecular techniques allowing subsequent typing. Phylogenetic analysis of both samples showed that NS5 sequences clustered with viruses previously classified as genotype III.

[Expression, purification and identification of St. Louis encephalitis virus-like particles in eukaryotic cells].

OBJECTIVE: To express St. Louis encephalitis virus-like particles in mammalian cells, it will provide a prerequisite for further immunological diagnostic studies. METHODS: 293T-cell were transiently transfected with recombinant PreM-E plasmid. Expression and antigenicity of the purified protein were determined by transmission electron microscope (TEM), Western-Blot, immunofluorescence assay and ELISA. RESULTS: Recombinant subviral particles, about 50 nm in diameter, were observed by TEM in the supernatant of transfected cells. The results of Western-Blot, IFA and ELISA showed the recombinant

Serologic diagnosis of West Nile and St. Louis encephalitis virus infections in domestic chickens.

Adult domestic chickens were infected with West Nile virus (WNV) or St. Louis encephalitis virus (SLEV) and challenged with homologous or heterologous virus at 21 or 56 days postinfection (dpi). Sera were collected at selected time points after infection and assayed by enzyme immunoassay (EIA), plaque reduction neutralization test (PRNT), and a Western blot (WB) alternative to PRNT. EIA results were sensitive and accurate (few false positives) but not specific, requiring a confirmatory test to determine virus infection history. PRNT results generally were specific until challenge, after which

Eared dove (Zenaida auriculata, Columbidae) as host for St. Louis encephalitis virus (Flaviviridae, Flavivirus).

St. Louis encephalitis virus (SLEV) is an emerging Flavivirus in South American countries. Its ecology and biological transmission cycles are scarcely known. Eared doves (Zenaida auriculata) have frequently been found infected by SLEV, and therefore, could be suspected as SLEV hosts. Thirty post-hatch-year eared doves were subcutaneously inoculated with the genotype V SLEV 78V-6507 viral strain and subsequently bled. No deaths or clinical signs of illness were observed in the inoculated doves. The viremia titers ranged from 2 to 5.5 log(10) plaque-forming units (PFU)/mL during 1-7 days postino

Mutation analysis of the fusion domain region of St. Louis encephalitis virus envelope protein.

The immune response to flavivirus infections produces both species-specific and flavivirus cross-reactive antibodies. The presence of cross-reactive antibodies complicates serodiagnosis of flavivirus infections, especially secondary infections caused by a heterologous virus. A successful public health response to the growing global threat posed by flaviviruses necessitates the development of virus-specific diagnostic antigens. The flavivirus envelope (E) glycoprotein is the principle antigen stimulating protective immunity during infection. Using recombinant St. Louis encephalitis virus-like p

St. Louis encephalitis virus: first isolation from a human in São Paulo State, Brazil.

This paper reports the isolation of St. Louis encephalitis virus (SLEV) from a febrile human case suspected to be dengue, in São Pedro, São Paulo State. A MAC-ELISA done on the patient's acute and convalescent sera was inconclusive and hemagglutination inhibition test detected IgG antibody for flaviviruses. An indirect immunofluorescent assay done on the C6/36 cell culture inoculated with the acute serum was positive for flaviviruses but negative when tested with dengue monoclonal antibodies. RNA extracted from the infected cell culture supernatant was amplified by RT-PCR in the presence of NS

Predicting St. Louis encephalitis virus epidemics: lessons from recent, and not so recent, outbreaks.

St. Louis encephalitis virus was first identified as the cause of human disease in North America after a large urban epidemic in St. Louis, Missouri, during the summer of 1933. Since then, numerous outbreaks of St. Louis encephalitis have occurred throughout the continent. In south Florida, a 1990 epidemic lasted from August 1990 through January 1991 and resulted in 226 clinical cases and 11 deaths in 28 counties. This epidemic severely disrupted normal activities throughout the southern half of the state for 5 months and adversely impacted tourism in the affected region. The accurate forecast

Multiple genotypes of St. Louis encephalitis virus (Flaviviridae: Flavivirus) circulate in Harris County, Texas.

St. Louis encephalitis (SLE) is endemic in Harris County, Texas. The disease is a public health concern in Houston, the largest city in Harris County, and in the state. Consequently, intensive surveillance for SLE virus in local mosquito populations is carried out by the Harris County Mosquito Control Division each year. In this study, we examined genetic variation among SLE isolates obtained during routine virus surveillance over a 13-year time period (1986-1999). St. Louis encephalitis virus isolates were tested for genetic variation using reverse transcription-polymerase chain reaction foll

St Louis encephalitis virus establishes a productive, cytopathic and persistent infection of Sf9 cells.

The Sf9 cell line, commonly used for gene expression by recombinant baculovirus, has been productively infected by St Louis encephalitis (SLE) virus, a flavivirus. SLE viral infection produced a c.p.e. in the Sf9 cells characterized by giant cells and the presence of 10-fold fewer cells in the infected cultures after the first week of infection compared with uninoculated control cultures. Infected Sf9 cells expressed SLE viral antigens, and intracellular virus particles were observed by electron microscopy. Titres of cell-associated SLE virus rose slightly over an 8 week period, whereas titres

Phenotypes of St Louis encephalitis virus mutants produced in persistently infected mosquito cell cultures.

Viral mutants that appeared during long-term persistent infections of mosquito cell cultures (Aedes albopictus, A. dorsalis and Culex tarsalis) with St Louis encephalitis virus were characterized. Evidence was obtained for the presence of temperature-sensitive mutants in the A. dorsalis and C. tarsalis persistently infected cultures, and small plaque mutants were predominant in all cultures except one of two cell cultures of C. tarsalis. Virus from persistently infected A. albopictus cell cultures was growth-restricted in Vero and C. tarsalis cells. One of two persistently infected A. dorsalis

Viremic enhancement due to transovarially acquired antibodies to St. Louis encephalitis virus in birds.

Adult house sparrows (Passer domesticus) were captured and experimentally inoculated with St. Louis encephalitis (SLE) virus to produce high concentrations of circulating antiviral antibody. Nestlings, 5-7 and 14-16 days of age, from SLE immune adult females and challenged with SLE virus, exhibited viremic enhancement by producing viremias of greater duration and magnitude than did controls. Nestlings possessing maternal antibody and challenged with SLE virus between 8 and 13 days of age did not produce viremias differing significantly from controls in magnitude, duration, or temporal appearan

Genetic characterization of St. Louis encephalitis virus isolated from human in São Paulo, Brazil.

The molecular characterization of SPH253157, a new strain of St. Louis encephalitis virus (SLEV), isolated in 2004 from the first case of human infection recognized in the state of São Paulo, Brazil, is reported. The patient, presenting a febrile illness without neurological involvement, was hospitalized as a probable case of dengue fever. Genomic RNA was isolated from the supernatant of C6/36 cells infected with acute phase-serum specimen of the patient and the envelope gene was amplified by reverse-transcription-polymerase chain reaction. The complete nucleotide sequence of the envelope gene

Molecular typing of West Nile Virus, Dengue, and St. Louis encephalitis using multiplex sequencing.

We report the development of an assay to simultaneously identify three of the clinically important flaviviruses (West Nile Virus, Dengue, and St. Louis encephalitis). This assay is based on the nucleotide sequence variations within a 266-bp region of the non-structural protein 5. Further, based on the nucleotide variations in the same region of the non-structural protein 5, four of the present Dengue serotypes were identified. To identify some of the subtypes of WNV we have developed a second assay using multiplex sequencing technology. The format of the result of this assay is an electrophero

Immunisation with DNA polynucleotides protects mice against lethal challenge with St. Louis encephalitis virus.

In vivo transfection by intramuscular injection with plasmids expressing the immunogenic proteins of microbial pathogens has considerable potential as a vaccination strategy against many pathogens of both man and animals. Here we report that weanling mice given a single intramuscular injection of 50 micrograms of a plasmid, pSLE1 expressing the St. Louis encephalitis virus (SLE) prM/E protein under the control of the cytomegalovirus immediate early protein promoter produced SLE-specific antibody and were protected against lethal challenge with the virulent virus. EFFECT OF PREGNANCY UPON THE IMMUNITY OF MICE VACCINATED AGAINST ST. LOUIS ENCEPHALITIS VIRUS.

1. Virgin and pregnant Swiss mice are equally susceptible to intracerebral inoculation of St. Louis encephalitis virus. 2. Following subcutaneous vaccination with the St. Louis virus, the great majority of virgin Swiss mice become immune to subsequent intracerebral injection of 10,000 M.L.D. of the virus. 3. The majority of mice vaccinated during pregnancy do not become immune to even as little as 500 intracerebral M.L.D. of the virus. The depression of the ability to acquire immunity against the virus is most marked when the vaccination is carried out late in pregnancy, but it is also demonst
