Use of the laboratory rat as a model in endocrine disruptor screening and testing.

The screening and testing program the US Environmental Protection Agency (EPA) is currently developing to detect endocrine-disrupting chemicals (EDCs) is described. EDCs have been shown to alter the following activities: hypothalamic-pituitary-gonadal (HPG) function; estrogen, androgen, and thyroid hormone synthesis; and androgen and estrogen receptor-mediated effects in mammals and other animals. The value and limitations of mammalian in vivo assays are described that involve the use of the laboratory rat, the EPA Endocrine Disruptor Screening and Testing Advisory Committee species of choice.

Endocrine disruptors and sexually dimorphic behaviors: a question of heads and tails.

Early brain development seems destined to be the territory where questions about the neurotoxicity of endocrine disruptors will be contested, so we ought to clarify how we propose to invade it. Everything else we have learned tells us that the most sensitive indices are likely to be the most subtle, a term we often use to describe the phenomena and domain of neurobehavioral toxicology. It might mean an effect that is small in amplitude, or that requires sensitive measures to detect. One class of effects that might result from environmental exposure to endocrine disruptors reflects both kinds o

Effects of bisphenol-A and other endocrine disruptors compared with abnormalities of schizophrenia: an endocrine-disruption theory of schizophrenia.

In recent years, numerous substances have been identified as so-called "endocrine disruptors" because exposure to them results in disruption of normal endocrine function with possible adverse health outcomes. The pathologic and behavioral abnormalities attributed to exposure to endocrine disruptors like bisphenol-A (BPA) have been studied in animals. Mental conditions ranging from cognitive impairment to autism have been linked to BPA exposure by more than one investigation. Concurrent with these developments in BPA research, schizophrenia research has continued to find evidence of possible en

Human health impact of endocrine disruptors: time to regain our stolen future!

In the latter half of the 20th century, humans have produced various synthetic chemicals, several of which had serious adverse effects on ecosystems and human health. Colborn et al. noticed that some chemicals disrupt the hormonal system of living organisms. They named these chemicals endocrine-disrupting chemicals (EDCs) and published the book Our Stolen Future. In classical toxicology, toxins directly act on cellular components, such as proteins, DNA, and cell membranes; cause damage and exhibit toxicity; and demonstrate a linear dose-response relationship, thus allowing safe thresholds. EDC

Endocrine disruptors: Unravelling the link between chemical exposure and Women's reproductive health.

An Endocrine Disrupting Chemical (EDC) is any compound that disrupts the function of the endocrine system in humans and is ubiquitous in the environment either as a result of natural events or through anthropogenic activities. Bisphenol A, phthalates, parabens, pesticides, triclosan, polychlorinated biphenyls, and heavy metals, which are frequently found in the pharmaceutical, cosmetic, and packaging sectors, are some of the major sources of EDC pollutants. EDCs have been identified to have a deteriorating effect on the female reproductive system, as evidenced by the increasing number of repro

Identification of known and novel nonpolar endocrine disruptors in human amniotic fluid.

BACKGROUND: Prenatal exposure to endocrine-disrupting compounds (EDCs) may contribute to endocrine-related diseases and disorders later in life. Nevertheless, data on in utero exposure to these compounds are still scarce. OBJECTIVES: We investigated a wide range of known and novel nonpolar EDCs in full-term human amniotic fluid (AF), a representative matrix of direct fetal exposure. METHODS: Gas chromatography high-resolution mass spectrometry (GC-HRMS) was used for the targeted and non-targeted analysis of chemicals present in nonpolar AF fractions with dioxin-like, (anti-)androgenic, and (

Prenatal Exposure to Endocrine Disruptors: A Developmental Etiology for Polycystic Ovary Syndrome.

Polycystic ovary syndrome (PCOS) is one of the most common and complex endocrinopathies among reproductive-age women. Polycystic ovary syndrome is characterized by symptomatology of oligomenorrhea and androgen excess, with or without presence of polycystic ovarian morphology. The etiology of PCOS is multifactorial, including genetic and environmental components. It has been previously established that prenatal androgen exposure results in a PCOS phenotype in experimental animal models and epidemiologic human studies. Investigators hypothesize that prenatal exposure to endocrine-disrupting chem

Role of personal care products as endocrine disruptors affecting reproductive age women.

The impact of endocrine-disrupting chemicals (EDCs) on public health is growing due to their wide-ranging consequence and likelihood of morbidity on human health. Humans are confronted with EDCs through their skin and drinks, as well as by inhaling. EDCs are extensively dispersed in the environment. EDCs have been shown to primarily influence puberty, the reproductive system, embryonic growth, the hypothalamus-pituitary-gonadal axis (HPG) neuroendocrine axis, and gender differentiation in the foetus, despite their capacity to influence a variety of hormone systems. Deleterious Biological Effects of Endocrine Disruptors: An Insight into Human Health Risks.

Endocrine-disrupting chemicals (EDCs) are environmental pollutants. Since EDCs are present in various consumer products, contamination of human beings is very common. EDCs have deleterious effects on various systems of the body, especially the endocrine and reproductive systems. EDCs interfere with the synthesis, metabolism, binding, or cellular responses of natural estrogens and alter various pathways. Biological samples such as blood, saliva, milk, placental tissue, and hair are frequently used for biomonitoring and the detection of EDCs. Early detection and intervention may help in preventi

Antiepileptic drugs are endocrine disruptors for the human fetal testis ex vivo.

Valproic acid (VPA) has long been the most widely used antiepileptic drug (AED) for the treatment of epilepsy, bipolar psychiatric disorders, and migraine. However, long-term VPA treatment has several adverse effects on the male reproductive system notably on endocrine functions and/or spermatic parameters. In utero exposure of the fetus to VPA is well known to be associated with a higher risk of several congenital malformations including those of male reproductive organs. Subsequent generations of AEDs, such as carbamazepine (CARB) and lamotrigine (LAM), are considered safer and are currently

[Endocrine disruptors: hormone-active chemicals from the environment: a risk to humans?].

Many substances from the technical and natural environment can cause damage to the endocrine system. Animal tests show that so-called endocrine disruptors (ED), such as pesticides, fungicides, plasticizers (phthalates), bisphenol A (BPA), and organotin compounds can interfere with the endocrine system. In humans, it is difficult to attribute such changes to specific ED. Nevertheless, in vitro studies with human cells and tissues clearly show that ED are able to interfere with endogenous hormones, i. e. affecting the steroid hormone metabolism and intracellular signaling. Several clinical studi

[Exposure to endocrine disruptors and male urogenital tract malformations [cryptorchidism and hypospadias]].

BACKGROUND: It has been proposed that the intrauterine period of child-mother exposure to hormonally active compounds (endocrine disruptors) is of critical importance. OBJECTIVE: We reviewed all Spanish and English-language epidemiological studies published between 1990 and the first trimester of 2006 that studied the possible association between human exposure to chemical compounds and the risk of cryptorchidism and/or hypospadias. METHODS: The MEDLINE (PubMed) database was searched using the following key words: endocrine disruptors, organochlorine pesticides, xenoestrogens, cryptorchidism

Effects of medical therapy, alcohol, smoking, and endocrine disruptors on male infertility.

Infertility affects up to 15% of the sexually active population, and in 50% of cases, a male factor is involved, either as a primary problem or in combination with a problem in the female partner. Because many commonly encountered drugs and medications can have a detrimental effect on male fertility, the medical evaluation should include a discussion regarding the use of recreational and illicit drugs, medications, and other substances that may impair fertility. With the knowledge of which drugs and medications may be detrimental to fertility, it may be possible to modify medication regimens o

Disruption of hormonal balance and the potential development of cancer - the impact of endocrine disruptors.

BACKGROUND: Endocrine-disrupting chemicals (EDCs) constitute a heterogeneous group of substances capable of interfering with hormonal regulation and thereby influencing the risk of initiation and progression of hormone-sensitive cancers. Their mechanisms of action include interactions with hormone receptors, modulation of hormone synthesis and metabolism, epigenetic alterations, and activation of specific signalling pathways. In recent years, the adverse outcome pathway (AOP) framework has gained prominence, as it enables linking molecular initiating events affected by EDCs to clinical outcome

Global research on endocrine disruptors as emerging hazards for human health and the environment.

Endocrine disruptors (EDs) contaminate nearly every ecosystem and are significantly associated with different neurological and neurodevelopmental disorders. To date, there is no comprehensive literature on global publication efforts. Since there are many unknown substances, modes of action, and risks of EDs, it is necessary to provide detailed insight into global publication patterns from temporal, regional, and socioeconomic perspectives. Hence, this review article provides background information for all stakeholders, from scientists to clinicians and policymakers. A disproportionate increase

The Impact of Endocrine Disruptors on the Female Genital Tract Microbiome: A Narrative Review.

BACKGROUND/OBJECTIVES: Endocrine disruptors (EDs) are xenobiotic chemicals that disrupt hormone signaling and homeostasis within the human body. Accumulative evidence proposes that EDs could affect systemic hormone balance and local microbial communities, including the female genital tract (FGT) microbiome. The FGT microbiome, and especially the vaginal microbiota, contributes significantly to reproductive health maintenance, defense against infection, and favorable pregnancy outcomes. Disruption of the delicate microbial environment is associated with conditions like bacterial vaginosis, infe

BPA Endocrine Disruptor Detection at the Cutting Edge: FPIA and ELISA Immunoassays.

BPA is a chemical commonly used in the production of polymer-based materials that can have detrimental effects on the thyroid gland and impact human reproductive health. Various expensive methods, such as liquid and gas chromatography, have been suggested for detecting BPA. The fluorescence polarization immunoassay (FPIA) is an inexpensive and efficient homogeneous mix-and-read method that allows for high-throughput screening. FPIA offers high specificity and sensitivity and can be carried out in a single phase within a timeframe of 20-30 min. In this study, new tracer molecules were designed

The impact of endocrine disruptor chemicals on oocyte/embryo and clinical outcomes in IVF.

The negative impact of endocrine-disrupting pesticides on human fertility is now a key issue in reproductive health. There are much fewer literature data about the impact of pesticide exposure on women than on men and very few studies of women participating in an in vitro fertilization (IVF) programme. In the present review, we found that (1) various pesticides with an endocrine-disrupting action are associated with poor oocyte maturation and competency, embryonic defects and poor IVF outcomes, and (2) some pesticide compounds are linked to specific causes of female infertility, such as premat

Effects of elevated glucocorticoids on reproduction and development: relevance to endocrine disruptor screening.

This article reviews the influence of the hypothalamo-pituitary-adrenocortical (HPA) axis on mammalian male and female reproduction and development of offspring and its potential impact on the identification of endocrine disruptive chemicals by in vivo assays. In the adult male rat and baboon, stress suppresses testosterone secretion via a direct inhibitory effect of elevated glucocorticoids on Leydig cells. In adult female sheep, stress disrupts reproductive function via multi-stage mechanisms involving glucocorticoid-mediated suppression of LH secretion, LH action on the ovary and the action
